Effect of Stilbene Derivatives from Rheum undulatum on Carrageenan-Induced Acute Edema in Rats (Carrageenan 유발 족부종에 미치는 종대황 스틸벤 유도체의 효과)

  • In order to examine the anti-ohyul (anti-inflammatory) activity of rhubarb, we investigated the effects of stilbene derivatives obtained from rhizomes of Rheum undulatum on the carrageenan-induced hind paw edema in rats. Administration of stilbene aglycon (rhapontigenin) at doses of 50 and 100 mg/kg exhibited the anti- edema effect in dose dependent manner and stilbene glycosides $(rhaponticin,\;piceatannol-3'-O-{\beta}-D-glucopyranoside)$ at a dose of 100 mg/kg also showed inhibitory effect on the carrageenan-induced edema. These inhibitory effects may ascribed, at least in part, to the anti-ohyul activity of rhubarb.

Translational Study on a Chapter of Taeeum-Disease[太陰病篇] in "The Golden Mirror of Medicine.The Notes of Treatise on Cold-Induced Diseases(醫宗金鑑.傷寒論注)" ("의종금감(醫宗金鑑).정정중경전서상한론주(訂正仲景全書傷寒論註)" "변태음병맥증병치전편(辨太陰病脈證幷治全篇)"에 대한 번역연구)

  • "The Golden Mirror of Medicine(醫宗金鑑)" was compiled by the medical officers of the Cheong(淸) government headed by Ogyeom(吳謙: 1736-1795) in 1742, and was adopted as a textbook by the Institute of Imperial Physicians(太醫院) in 1749. This book provides a good summary of academic contents and clinical experiences from before the Cheong(淸) dynasty, and serves as a convenient and practical guide book. "The Notes of Treatise on Cold-Induced Diseases(傷寒論注)" is one part of "The Golden Mirror of Medicine(醫宗金鑑)", and this is placed at the beginning of the book, indicating its importance. The chapter on taeeum-disease[太陰病篇], which is the third part of "The Notes of the Treatise on Cold-Induced Diseases(傷寒論注)", has not yet been translated into Korean. Therefore, in this study, the characteristics of Ogyeom's(吳謙) notes are inspected through a comparative study of the chapter of taeeum-disease[太陰病篇] based on translation and the notes of famous scholars. The texts first provide an outline of taeeum-disease[太陰病], which is followed by diarrhea, vomiting and therapeutic methods of syndrome involving both the exterior and interior[表裏兼證], as well as abdominal distension and pain. The prognoses are then explained in succession. The eight texts that have been shown in the chapter of taeeum-disease[太陰病篇] of original text were relocated and the seven texts that existed in the chapters of taeyang(太陽), yangmyeong(陽明) and gwol-eum(厥陰) were moved to this chapter. Furthermore, Ogyeom(吳謙) moved the cold-dysphagia[寒格] text from a chapter of gwol-eum-disease[厥陰病] to a chapter of taeeum-disease[太陰病] and explained vomiting due to pathogenic cold. The origins of taeeum-disease[太陰病] are purported to occur through the yang-channel[陽經] to the eum-channel[陰經], and taeeum-disease[太陰病] was reported to include both interior-deficiency-cold-syndrome[裏虛寒證] and interior-excess-heat-syndrome[裏實熱證]. In the case of diarrhea-more-severe-symptoms[自利益甚], he thought it induced by faultpurgation[誤下], and in indication for decoction of cinnamon with peony[桂枝加芍藥湯] and decoction of cinnamon with rhubarb[桂枝加大黃湯], he thought it included the exterior syndrome of taeyang-disease[太陽表證], and rhubarb was used in purgation of taeeum-excess[太陰實].

  • As a series of the studies on cultivated Korean rhubarb rhizomes (Rheum undulatum), five stilbene derivatives were screened for inhibitory activity against $5{\alpha}-reductase$. Of these, desoxyrhapontigenin, rhapontigenin, and piceatannol exhibited inhibition, whereas, two other stilbene glycosides, rhaponticin and $piceatannol-3'-O-{\beta}-D-glucopyranoside$ did not show the inhibition.

Rhapontigenin Production by Bioconversion and Inhibition of Melanin Synthesis (생물전환에 의한 Rhapontigenin의 생산 및 멜라닌 합성저해)

  • Rhapontin is the glycosylated stilbene compound, and comprising major component of rhubarb root extract. Rhapontin has been used as a raw material of skin-whitening cosmetics in Korea. Rhapontigenin, the aglycone of rhapontin, has been suggested to be more active than its glycosylated form. Therefore, the rhubarb root extract was treated with commercial enzyme, Pectinex to remove glycosylated moiety of rhapontin and rhapontigenin was prepared. The resulting material was analysed and identified as rhapontigenin by proton NMR and MALDI-Mass. Rhapontigenin exhibited tyrosinase inhibitory activity with an $IC_{50}$ of $126.72{\mu}g/mL$. The tyrosinase inhibitory activity of rhapontigenin was six times higher than that of rhapontin. In melanin biosynthesis inhibition assay using Streptomyces bikiniensis, rhapontigenin showed wider inhibition zone than that of rhapontin. From these results, we expect that rhapontigenin has stronger skin whitening effect than rhapontin and has advantages in cosmetic industry.

Characterization of Pectate Lyase Produced by Erwinia rhapontici During Growth in Host Plant Tissue (Erwinia rhapontici가 기주식물 조직에서 생산한 Pectate Lyase의 특성)

  • Erwinia rhapontici causes soft-rot disease in a number of plants such as rhubarb, onion, hyacinth and garlic. Pectate lyase (Pel) depolymerizes pectin and other polygalacturonates, which is though to play a role in bacterial invasion of plants. Pel activity was not detected in E. rhapontici cultured in a minimal salts medium containing glycerol, polygalacturonate, or citrus pectin as a carbon source. However, when sterilized potato tuber and Chinese cabbage slices were added to minimal salts polygalacturonate (0.5%) medium, E. rhapontici produced pectate lyase enzyme. Also Pel activity was consistently detected from macerated potato tubers, Chinese cabbage leaves, lettuce leaves and celery petioles tissue. Pel in the extract of macerated Chinese cabbage caused by E. rhapontici strain 1, resulted in electrolyte loss, tissue maceration and cell death of potato tuber tissue. These results indicate that E. rhapontici produces pectate lyase only in the presence of non-diffusible plant components, and that this enzyme probably contributes to its pathogenicity.

Suppressive Effect of Organic Farming Materials on the Development of Tomato Gray Mold (토마토 잿빛곰팡이병에 대한 유기농업자재의 억제효과)

  • Botrytis cinerea infects stems, leaves and fruits of greenhouse tomato and can cause serious economic losses. This study was conducted to develop organic farming control method against tomato gray mold. Twenty two organic farming materials including mineral and plant extracts were screened for the suppressive activity against Botrytis cinerea, in vitro and in vivo. Among the organic farming materials, sulfur, copper, Chinese twinleaf extract and rhubarb extract decreased by 51.7-90% of the spore germination of Botrytis cinerea. Also, gray mold incidence was reduced more than 90% on tomato stems by treating sulfur, seaweed extracts, rhubarb root extracts and Chinese twinleaf extract. After the selected four organic farming materials were applied on tomato cultivated in greenhouse, their control effects against the tomato gray mold were tested. When the water soluble sulfur was foliar-sprayed on the tomato leaves infected by artificial inoculation with spore suspension of Botrytis cinerea, it showed 87.9% of control value. Also, control activity of the water soluble sulfur was paralleled with chemical fungicide, diethofencarb+carbendazim. The above mentioned results indicate the sulfur formulation can be used as chemical fungicide alternatives for controlling tomato gray mold in the greenhouse.

Fresh-cut사과 전처리에 따른 물성학적 품질 특성

  • 편의와 fresh-like quality를 선호하는 소비 경향에 맞추어 가공한 fresh-cut사과의 선도 연장을 위한 저해제를 탐색하였다. 실험에 사용된 사과는 박피하여 속심을 제거한 사과를 8조각으로 절단하여 저해제에 3분간 침지한 후 물기를 제거하고 각각 LDPE 포장지에 포장하여 실온에서 저장하면서 colorimeter와 rheometer를 이용하여 색도와 물성을 측정하였다. 저해제로는 1% ascorbic acid, citric acid, NaCl, MgCl$_2$, CaCl$_2$, KCl, CaSO$_4$, MgSO$_4$, $Na_2$SO$_4$, oxalic acid, calcium lactate, calcium citrate를 사용하였으며 천연저해제로는 건칡, 맥문동, 목향, Rhubarb, 당귀, 오약, 시체, 지각, 천문동, 청피, 계피, 구기자, 당귀, 진피, 감초, 녹차, 후추 등을 열수추출하여 사용하였다. 물리적인 변화는 시간이 경과함에 따라 감소하는 추세를 보였으며 일부 건조로 인해 texture의 증가를 나타내기도 했다. 처리에 따른 물리적인 변화로는 CaCl$_2$, NaSO$_4$, Cl, 시금치를 처리하였을 때 그 변화가 가장 작았으나 시금치의 경우 시금치로 인한 사과의 색변화로 관능적으로 좋지 않았다. 한약재는 목향, 당귀, 지각, 계피를 처리하였을 때 물리적인 변화가 가장 작았으나 시금치와 마찬가지로 처리 초기에 한약재 자체의 향과 색으로 인하여 관능적으로 좋지 못한 결과를 나타냈다. 갈변 정도를 나타내는 ΔE의 변화는 KCl과 CaCl$_2$, CaSO$_4$를 처리한 구에서 변화가 가장 작았으며 한약재를 처리하였을 경우 당귀, 천문동을 처리한 구에서 효과를 보였다. oxalic acid는 갈변저해에는 효과적이었으나 시간이 경과함에 따라 수분의 용출이 증가하였으며 물성의 변화도 컸다. 또한 ZnCl$_2$를 처리한 구에서는 이취 발생과 수분 용출이 컸으며 색의 변화도 컸다.

The Present Condition of Production and Consumption of Medicinal Plants in Japan (日本における 藥用植物の生産, 消費の 現況)

  • The medicinal plants used traditional medicines are classified into two groups. The first group is formulary medicine which have been recorded on authorized books, for example, kampo drug in Japan. The second group is the fork medicines. These medicinal plants production is 165 billion yen(about 1.5 billion dollar) and which rate to to total pharmaceutical production is about 3% in 1990. The number of important medicinal plants treated in the market is 124 herbs from the total 364 herbs for kampo durgs(210 preparations kampo durgs) and other propritetary drugs (agents for common cold, laxatives, gastrointestional and so on). The production of the Ginseng and Bupleurum Root are increased about ten times in last twelve years(1976 or 1988). The 80% of these medicinal plants are imported from China, Korea and Southern Asian countries. Then the Pharmaceutical Affairs Bureau of Ministry of Health and Welfare published "The Guideline of Medicinal Plant for Cultivation" for a quality control and a stabilization supply of 80 herbs. The first part(1992) of the guideline reported 5 herbs; Coptis Rhizome, Rehmannia Root, Bupleur, Root, Japanese Angelica Root and Rhubarb, The next part will be published following 5herbs; Cnidium Rhizome, Safflower, Coix seed and Fruiet, Platicodon Root and Zedoary.
