Chiral Separation of ${\beta}-Blockers$ after Derivatization with a New Chiral Derivatization Agent, GATC

  • A new chiral derivatization agent with sugar moiety, 2,3,4,6-tetra-O-acetyl-${\beta}$-D-galactopyranosyl isothiocyanate (GATC) was synthesized. Several ${\beta}-blockers$ were investigated for the possible separation of the enantiomers by reversed-phase HPLC after derivatization with this new chiral derivatization agent (GATC). GATC was reacted readily with ${\beta}-blockers$ at room temperature and the reaction mixture could directly be injected into the HPLC system. The corresponding diastereomers were well resolved on an ODS column with acetonitrile-ammonium acetate buffer as a mobile phase and monitored at UV 254 nm. The optimization of the derivatization procedure (concentration of GATC, reaction temperature and time) and HPLC conditions (pH and ionic strength of mobile phase) were investigated and compared with GITC.

A Study on the Orthogonality Properties of the Eigenmodes of Phase Conjugate Optical Resonator (위상 공액 공진기의 고유 모드 직교성에 관한 연구)

  • In this paper, the orthogonal properties of the eigenmodes of optical resonators which have phase conjugate mirrors at both ends are derived. The modes which propagate in resonators are descdribed by the Huygens integral. Then, the eigeneuqation which is needed in order to prove the orthogonality of the eigenmodes of the resonator is obtained from this representation. When the kernel being a part of the eigenequation is Hermitian as in conventional resonators and in optical resonator with only one phase conjugate mirror, one can show that the eigenmodes have essentially biorthogonal relations, which are satisfied the set of modes propagating in one direcdtion around the resonator and the adjoint set of modes propagating in the reversed direction.

Inhibitory Effects of Transforming Growth Factor and Drynariae Rhizoma on Leukocytosis Associated with the Chronic Phase of Arthritis in Mice

  • Drynariae Rhizoma (DR), an herbal medicine known to clean blood and improve its circulation, frequently appears as the main ingredient in the prescriptions for bone injuries. Currently, it is unclear how contributes pharmacologically to the reformation of bone. Therefore, we have done this study. Systematic administrations of TGFβ1 and water extract of DR diminished the polyarthritis development in rats. TGFβ administration (0.1-2 ㎍/animal) and DR (10-100 ㎍/animal), initiated 1 day before an arthritogenic dose of streptococcal cell wall fragments, virtually eliminated the joint swelling and distortion observed during the acute phase and the chronic phase of the disease. The TGFβ and DR synergistically suppressed the arthritis when the administration was begun after the acute phase of arthritis. Also, the synergistic activity between TGFβ and DR was confirmed in their suppression of arthritis in rats. Consistent with the inhibition of inflammatory cell recruitment into the synovium, TGFβ1 and DR reversed the leukocytosis associated with the chronic phase of the arthritis.

  • Numerical analysis is performed for the unsteady axisymmetric two dimensional phase change problem of freezing of water around a vertical tube. Heat conduction in the tube wall and solid phase, natural convection in liquid phase and volume expansion caused by density difference between solid and liquid phases are included in the numerical analysis. Existing correlation is used for estimating density-temperature relation of water, and the effect of volume expansion is reflected as fluid velocity at the interface and the free surface. As pure water has maximum density at 4.deg. C, it is found that there exists an initial temperature at which the flow direction reverses near the interface and by this effect the slope of interface becomes reversed depending on the initial temperature of water. By considering natural convection and solid-liquid density difference in the calculation, their effects on phase change process are studied and the effects of various parameters are also studied quantitatively.

  • Styrene-acrylamide co-polymer was immobilized on porous partially sub-$2{\mu}m$ silica monolith particles and inner surface of fused silica capillary ($50{\mu}m$ ID and 28 cm length) to result in ${\mu}LC$ and CEC stationary phases, respectively, for separation of anomeric D-glucose derivatives. Reversed addition-fragmentation transfer (RAFT) polymerization was incorporated to induce surface polymerization. Acrylamide was employed to incorporate amide-functionality in the stationary phase. The resultant ${\mu}LC$ and CEC stationary phases were able to separate isomers of D-glucose derivatives with high selectivity and efficiency. The mobile phase of 75/25 (v/v) acetonitrile (ACN)/water with 0.1% TFA, was used for HPLC with a packed column (1 mm ID, 300 mm length). The effects of pH and ACN composition on anomeric separation of D-glucose in CEC have been examined. A mobile phase of 85/15 (v/v) ACN/30 mM sodium acetate pH 6.7 was found the optimized mobile phase for CEC. The CEC stationary phase also gave good separation of other saccharides such as maltotriose and Dextran 1500 (MW~1500) with good separation efficiency (number of theoretical plates ~300,000/m).

  • A new method for the simultaneous determination of heavy metal ions in cigarette material by microwave digestion and reversed-phase high-performance liquid chromatography (RP-HPLC) has been developed. The cigarette material was digested by microwave digestion. Lead, cadmium, mercury, nickel and tin ions in the digested samples were pre-column derivatized with tetra-(2-chlorophenyl)-porphyrin ($T_2$-CPP) to form color chelates, which were then enriched by solid phase extraction with a $C_{18}$ cartridge. The chelates were separated on a Waters Xterra$^{TM}RP_{18}$ column by gradient elution with methanol (containing 0.05 mol/L pyrrolidine-aceticacid buffer salt, pH = 10.0) and acetone (containin0.05 mol/L pyrrolidine-acetic acid buffer salt, pH = 10.0)as mobile phase at a flow rate of 0.5mL/min and analyzed with a photodiode array detector from 350-600 nm. The detection limits of lead, cadmium, mercury, nickel and tin were 4,3,3,8 and 5 ng/L, respectively, in the original samples. This method was afforded good results.

Determination of Chloramphenicol in Milk by High Performance Liquid Chromatography (HPLC를 이용한 우유중의 클로람페니콜의 정량 분석)

  • Seven different sorbents were evaluated for their adsorptivity and desorptivity of antibiotic, chloramphenicol. Among the sorbents studied, Carbopak B was found to be the most efficient in enriching the chloramphenicol from dilute aqueous solution. Interfering components in the milk matrix could be washed off by water and petroleum ether from Carbopak B column, while the chloramphenicol was retained on the surface of Carbopak B. The method of simple and efficient purification and enrichment of chloramphenicol using Carbopak B, followed by quantitative analysis employing $C_{18}$ reversed phase high performance liquid chromatography has been applied to the determination of chloramphenicol in milk.
