High Level of Trans-Resveratrol, a Natural Anti-Cancer Agent, Found in Korean Noul Red Wine

  • Resveratrol (trans-3,5,4'-trihydroxystilbene), a phenolic substance present in both grapes and wines, has been reported to have certain pharmacological effects. Using an ethylacetate-phase extraction followed by a high performance liquid chromatographic analysis, the concentration of trans-resveratrol was measured in 9 red wines commercially available in Korea, including the Korean red wine (Noul). Noul red wine with a 1998 vintage had a trans-resveratrol concentration of 3.3 ㎎/l, which was rather higher than the concentrations found in other wines (0.19-2.45 ㎎/l) with the exception of the French wine, J. P. Chenet (3.39 ㎎/l). In addition, the grapes of Vitis labrusca cv. Sheridan cultivated in the Kyungsan area showed a trans-resveratrol content of 6.4 ㎎/㎏.

Candicidal Action of Resveratrol Isolated from Grapes on Human Pathogenic Yeast C. albicans

  • Resveratrol (3,5,4'-trihydroxystilbene) is a naturally occurring, multi-biofunctional chemical existing in grapes and various other plants as a polyphenol type, and it is one of the best known natural anticancer and antiatherosclerosis reagents. In this study, we investigated the antifungal action by resveratrol in Candida albicans, which is a human infectious fungi as an agent of candidiasis. Resveratrol displayed potent fungicidal activity in an energy-dependent manner, without any hemolytic effects against human erythrocytes. It was found that the serum-induced mycelial forms, which playa crucial role in the pathogenesis of C. albicans during host tissue invasion, were disrupted by resveratrol. To understand the correlation between lethal effects and resveratrol action, we examined the physiological changes of C. albicans. A significant accumulation of intracellular trehalose was induced by stress responses to resveratrol action, and a remarkable arrest of cell-cycle processes at the S-phase in C. albicans occured. Therefore, the fungicidal effects of resveratrol demonstrate that this compound is a potential candidate as an antifungal agent in treating infectious diseases by candidal infections.

Effects of Resveratrol and trans-3,5,4'-Trimethoxystilbene on Glutamate-Induced Cytotoxicity, Heme Oxygenase-1, and Sirtuin 1 in HT22 Neuronal Cells

  • Resveratrol (trans-3,5,4'-trihydroxystilbene) has received considerable attention recently for the potential neuroprotective effects in neurodegenerative disorders where heme oxygenase-1 (HO-1) and sirtuin 1 (SIRT1) represent promising therapeutic targets. Resveratrol has been known to increase HO-1 expression and SIRT1 activity. In this study, the effects of resveratrol and trans-3,5,4'-trimethoxystilbene (TMS), a resveratrol derivative, on cytotoxicity caused by glutamate-induced oxidative stress, HO-1 expression, and SIRT1 activation have been investigated by using murine hippocampal HT22 cells, which have been widely used as an in vitro model for investigating glutamate-induced neurotoxicity. Resveratrol protected HT22 neuronal cells from glutamate-induced cytotoxicity and increased HO-1 expression as well as SIRT1 activity in a concentration-dependent manner. Cytoprotection afforded by resveratrol was partially reversed by the specific inhibition of HO-1 expression by HO-1 small interfering RNA and the nonspecific blockage of HO-1 activity by tin protoporphyrin IX, but not by SIRT1 inhibitors. Surprisingly, TMS, a resveratrol derivative with methoxyl groups in lieu of the hydroxyl groups, and trans-stilbene, a non-hydroxylated analog, failed to protect HT22 cells from glutamate-induced cytotoxicity and to increase HO-1 expression and SIRT1 activity. Taken together, our findings suggest that the cytoprotective effect of resveratrol was at least in part associated with HO-1 expression but not with SIRT1 activation and, importantly, that the presence of hydroxyl groups on the benzene rings of resveratrol appears to be necessary for cytoprotection against glutamate-induced oxidative stress, HO-1 expression, and SIRT1 activation in HT22 neuronal cells.

Resveratrol Anglog 3,5,2',4'-Tetramethoxy-trans-stilbene, Potentiates the Inhibotion of Cell Growth and Induces Apoptosis in Human Cancer Cells

  • Resveratrol, a trihydroxystilbene found in grapes and several plants, has been shown to be active in inhibiting multistage carcinogenic process. Using resveratrol as the prototype, we synthesized several analogs and evaluated their growth inhibitory effect using cultured human cancer cells. In the present report we show that one of the resveratrol analogs, 3, 5,2',4'-tetramethoxy-trans-stilbene, potentiated the inhibition of cancer cell growth. Prompted by the strong growth Inhibitory activity of the compound ($IC_{50}$; $0.8{\mu}$ g/ml) compared to resveratrol ($IC_{50}$; $18{\mu}$ug/ml) in cultured human colon cancer cells (Col2), we performed an action mechanism study using the compound. The compound induced the accumulation of cellular DNA contents in the sub-CO phase DNA contents of the cell cycle by in a time-dependent manner. The morphological changes were also consistent with an apoptotic process. This result indicated that the compound induced apoptosis of cancer cells, and may be a candidate for use in the development of potential cancer chemotherapeutic or cancer chemopreventive agents.

Resveratrol Impaired the Morphological Transition of Candida albicans Under Various Hyphae-Inducing Conditions

  • The ability of the human fungal pathogen Candida albicans to undergo the morphological transition from a single yeast form to pseudohyphal and hyphal forms in response to various conditions is known to be important for its virulence. Many studies have shown the pharmacological effects of resveratrol, a phytoalexin polyphenolic compound. In this study, we investigated the antifungal activity of resveratrol against C. albicans. Both yeast-form and mycelial growth of C. albicans were inhibited by resveratrol. In addition, normal filamentation of C. albicans was affected and yeast-to-hypha transition under serum-, pH-, and nutrient-induced hyphal growth conditions was impaired by resveratrol.

Comparative Depigmentation Effects of Resveratrol and Its Two Methyl Analogues in α-Melanocyte Stimulating Hormone-Triggered B16/F10 Murine Melanoma Cells

  • Previous research showed that resveratrol (trans-3,4',5-trihydroxystilbene) and pinostilbene (trans-3-methoxy-4',5-dihydroxystilbene) were able to inhibit tyrosinase directly; however, anti-melanogenic effects of pterostilbene (trans-3,5-dimethoxy-4'-hydroxystilbene) and resveratrol trimethyl ether (RTE) have not been compared. To investigate the hypopigmentation effects of pterostilbene and RTE, melanin contents and intracellular tyrosinase activity were determined by western blot analysis. Firstly, pterostilbene showed the inhibitory effects on ${\alpha}$-melanocyte stimulating hormone (MSH)-induced melanin synthesis stronger than RTE, resveratrol, and arbutin. Pterostilbene inhibited melanin biosynthesis in a dose-dependent manner in ${\alpha}$-MSH-stimulated B16/F10 murine melanoma cells. Specifically, melanin content and intracellular tyrosinase activity were inhibited by 63% and 58%, respectively, in response to treatment with $10{\mu}m$ of pterostilbene. The results of western blot analysis indicated that pterostilbene induced downregulation of tyrosinase protein expression and suppression of ${\alpha}$-MSH-stimulated melan-A protein expression stronger than RTE or resveratrol. Based on these results, our study suggests that pterostilbene can induce hypopigmentation effects more effectively than resveratrol and RTE, and it functions via downregulation of protein expression associated with hyperpigmentation in ${\alpha}$-MSH-triggered B16/F10 murine melanoma cells.

Resveratrol functioned as a superagonist at high concentrations (i.e., produced a greater maximal transcriptional response than estradiol) Among the resveratrol derivatives, 10 compounds showed significant estrogenic activity.

Fungicidal Effect of Resveratrol on Human Infectious Fungi

  • Resveratrol, a phenolic antioxidant found in grapes, has been known to mediate various biological activities on the human body. In the present study, we tested the antifungal a ctivity of resveratrol against human pathogenic fungi before carrying out further studies to elucidate the antifungal mechanism(s) of resveratrol. Resveratrol displayed potent antifungal activity against human pathogenic fungi at concentration levels of 10-20 ${\mu}g$/mL. Furthermore, time-kill curve exhibited fungicidal effect of resveratrol on C. albicans, but the compound had no hemolytic activity against human erythrocytes. The destruction of C. albicans cells by resveratrol was confirmed by scanning electron microscopy. These results suggest that resveratrol could be employed as a therapeutic agent to treat fungal infections of humans.

땅콩 품종 및 생육기별 trans-Resveratrol 함량 (tans-Resveratrol Content of Varieties and Growth Period in Peanut)

  • 본 연구에서는 땅콩의 resveratrol에 대한 체계적인 연구를 위하여 고속액체크로마토그래피HPLC)를 이용한 땅콩의 trans-resveratrol 분석체계 확립과 품종 및 생육시기별 trans-resveratrol 함량 변화를 구명하여 금후 고부가가치 고함유 기능성 품종개발의 기초 자료로 활용하고자 수행한 결과를 요약하면 다음과 같다. 1. HPLC를 이용한 trans-resveratrol 분석조건은 컬럼은 Xterra $\textrm{C}_{18}$, 이동상 용매조성은 (A) glacial acetic acid DCW(52.6 : 900)과 (B) acetonitril solution A(80:20), 용매흐름속도는 1.1 ml/min, 검출기는 UV-Vis detector 308 nm 이었다. 2. 땅콩에서 trans-resveratrol 추출시 추출용매의 경우 80%EtOH과 80%MeOH이 유의차가 없었고, 추출온도의 경우에도 온도간 유의성이 인정되지 않았으며, ICH guideline에 따른 안전성과 경제성을 고려한 trans-resveratrol의 최적추출 조건은 80% EtOH, $25^{\circ}C$, 45분이었다. 3. 땅콩 품종별 trans-resveratrol 함량 분포범위는 0.018-1.125 $\mu\textrm{g}/\textrm{g}$으로 품종별 유의차가 있었으나, 초형과 립 크기에 따른 trans-resveratrol 함량은 일정한 경향을 보이지 않았다. 4. 종실과 종피를 포함한 종실의 trans-resverarol함량은 종실(0.152-0.305 $\mu\textrm{g}/\textrm{g}$) 보다 종피를 포함한 종실(0.188-0.345 $\mu\textrm{g}/\textrm{g}$)에서 더 높았다. 5. 생육시기별 trans-resveratrol 함량은 신남광땅콩(0.227$\mu\textrm{g}/\textrm{g}$ 대광땅콩(0.418 $\mu\textrm{g}/\textrm{g}$)은 파종 후 110일에 팔광땅콩(0.486 $\mu\textrm{g}/\textrm{g}$)과 밀양16호(0.441 $\mu\textrm{g}/\textrm{g}$)는 파종 후 130일에 가장 높았다.
