Berberine이 구속 스트레스 부가후 Mouse의 혈중 Catecholamine 함량에 미치는 영향 (Effects of Berberine on Serum Levels of Catecholamines after Immobilization Stress in Mice)

  • Berberine, a protoberberine isoquinoline alkaloid, showed inhibitory effects on dopamine content in PC12 cells $(53.8\%\;inhibition\;at\;20\;{\mu}M)$. Tyrosine hydroxylase, the rate-limiting enzyme in the catecholamine biosynthetic pathway, was inhibited at $20\;{\mu}M)$ of berberine by $21.8\%$ relative to control. Thus, we hypothesized that the inhibition of tyrosine hydroxylase by berberine might be partially contributed to the decrease in dopamine content in PC12 cells. Furthermore, we investigated the effects of berberine on catecholamine content of serum after immobilization and cold stress in mice. Adult male mice were either subjected to 30 min of restraint or to 2 hr of cold chamber at $4-6^{\circ}C$. Serum norepinephrine, 16.8 pmol/ml, in control mice was increased to 28.8 pmol/ml by immobilization and the stress-induced rise in serum norepinephrine was partially blocked by the treatment of berberine (10 mg/kg, i.p.) once a day for 6 days. Berberine (10 mg/kg/day for 3 days, i.p.) also inhibited the increase in serum norepinephrine by cold stress in mice. These results suggest that berberine may be developed as the promising antistress agent.

Panax ginseng exerts antidepressant-like effects by suppressing neuroinflammatory response and upregulating nuclear factor erythroid 2 related factor 2 signaling in the amygdala

  • Background: Depression is one of the most commonly diagnosed neuropsychiatric diseases, but the underlying mechanism and medicine are not well-known. Although Panax ginseng has been reported to exert protective effects in various neurological studies, little information is available regarding its antidepressant effects. Methods: Here, we examined the antidepressant effect and underlying mechanism of P. ginseng extract (PGE) in a chronic restraint stress (CRS)-induced depression model in mice. Results: Oral administration of PGE for 14 d decreased immobility (depression-like behaviors) time in forced swim and tail suspended tests after CRS induction, which corresponded with attenuation of the levels of serum adrenocorticotropic hormone and corticosterone, as well as attenuated c-Fos expression in the amygdala. PGE enhanced messenger RNA expression level of brain-derived neurotrophic factor but ameliorated microglial activation and neuroinflammation (the level of messenger RNA and protein expression of cyclooxygenase-2 and inducible nitric oxide synthase) in the amygdala of mice after CRS induction. Interestingly, 14-d treatment with celecoxib, a selective cyclooxygenase-2 inhibitor, and $N_{\omega}$-nitro-L-arginine methyl ester hydrochloride, a selective inducible nitric oxide synthase inhibitor, attenuated depression-like behaviors after CRS induction. Additionally, PGE inhibited the upregulation of the nuclear factor erythroid 2 related factor 2 and heme oxygenase-1 pathways. Conclusion: Taken together, our findings suggest that PGE exerts antidepressant-like effect of CRS-induced depression by antineuroinflammatory and antioxidant (nuclear factor erythroid 2 related factor 2/heme oxygenase-1 activation) activities by inhibiting the hypothalamo-pituitary-adrenal axis mechanism. Further studies are needed to evaluate the potential of components of P. ginseng as an alternative treatment of depression, including clinical trial evaluation.

Chronic Administration of Monosodium Glutamate under Chronic Variable Stress Impaired Hypothalamic-Pituitary-Adrenal Axis Function in Rats

  • The hypothalamic-pituitary-adrenal (HPA) axis is the primary endocrine system to respond to stress. The HPA axis may be affected by increased level of corticotrophin-releasing factors under chronic stress and by chronic administration of monosodium glutamate (MSG). The purpose of this study was to investigate whether chronic MSG administration aggravates chronic variable stress (CVS)-induced behavioral and hormonal changes. Twenty-four adult male Sprague-Dawley rats, weighing 200~220 g, were divided into 4 groups as follows: water administration (CON), MSG (3 g/kg) administration (MSG), CVS, and CVS with MSG (3 g/kg) administration (CVS+MSG). In addition, for the purpose of comparing the effect on plasma corticosterone levels between chronic stress and daily care or acute stress, 2 groups were added at the end of the experiment; the 2 new groups were as follows: naive mice (n=7) and mice exposed to restraint stress for 2 h just before decapitation (A-Str, n=7). In an open field test performed after the experiment, the CVS+MSG group significant decrease in activity. The increase in relative adrenal weights in the CVS and CVS+MSG group was significantly greater than those in the CON and/or MSG groups. In spite of the increase in the relative adrenal weight, there was a significant decrease in the plasma corticosterone levels in the CVS+MSG group as compared to all other groups, except the naive group. These results suggest that impaired HPA axis function as well as the decrease in the behavioral activity in adult rats can be induced by chronic MSG administration under CVS rather than CVS alone.

  • LVDT에 의해 전체변형율이 측정되었고, 국부적 변형율은 변형율게이지에 의해 측정되었다. 또한 공시체에서의 응력 분포를 알기 위하여 유한요소법에 의한 축대칭 탄소성 해석을 하였다. 단면구속영향을 고려하면 H/D=1인 경우에 특히 구속영향을 크게 받는다. 또한 공시체 직경에 대한 공시체 길이가 길어질수록 영향을 적게 받는다는 것을 알 수 있다. 공시체와 재하판과의 접촉면에서 응력 및 변위 분포는 접촉하고 있는 두 재료의 탄성계수비와 접촉면의 마찰저항에 크게 좌우된다. 즉 암석공시체에 대한 재하판의 영향은 경암에서보다 연암에서 더 현저하다 또한 단면구속이 변형율 분포에 미치는 영향 및 치수효과는 강성이 큰 암석에서 현저하게 나타난다.

Design and calibration of a semi-active control logic to mitigate structural vibrations in wind turbines

  • The design of a semi-active (SA) control system addressed to mitigate wind induced structural demand to high wind turbine towers is discussed herein. Actually, the remarkable growth in height of wind turbines in the last decades, for a higher production of electricity, makes this issue pressing than ever. The main objective is limiting bending moment demand by relaxing the base restraint, without increasing the top displacement, so reducing the incidence of harmful "p-delta" effects. A variable restraint at the base, able to modify in real time its mechanical properties according to the instantaneous response of the tower, is proposed. It is made of a smooth hinge with additional elastic stiffness and variable damping respectively given by springs and SA magnetorheological (MR) dampers installed in parallel. The idea has been physically realized at the Denmark Technical University where a 1/20 scale model of a real, one hundred meters tall wind turbine has been assumed as case study for shaking table tests. A special control algorithm has been purposely designed to drive MR dampers. Starting from the results of preliminary laboratory tests, a finite element model of such structure has been calibrated so as to develop several numerical simulations addressed to calibrate the controller, i.e., to achieve as much as possible different, even conflicting, structural goals. The results are definitely encouraging, since the best configuration of the controller leaded to about 80% of reduction of base stress, as well as to about 30% of reduction of top displacement in respect to the fixed base case.

흰쥐의 스트레스성 위궤양에 대한 Alove vera의 항궤양작용 (The Antiulcer Effects of Alove vera on the Stomach ulcer Induced by Stress in rats)

  • This was performed to investigate the antiulcer effects of Aloe vera on the stomach ulcer induced by restraint and water-immersion stress in rats. For this experiment, 60 male Sprague-Dawley strain were used. The experimental groups were divided into five: a control(CA) and 4 aloe treatment groups. Each dose of aloe was 50 mg/kg BW(AA), 100mg/kg BW(AB), 200 mg/kg BW(AC), and 400 mg/kg BW(AD). The rats were allocated to each group by 12 and observed for 4 weeks. The results are as following. 1. The stomach surface pH in each group showed no significant difference, byt the values of aloe treatment groups were higher than the value of the control group. 2. The gastric wall mucus was significantly increased in all aloe treatment groups(p<0.05) compared with the control group. Especially in AC and AD group the differences were higher(p<0.01). 3. At shear rate rate 11.25, 45.0, 90.0, 225.0 sec-1, whole blood viscosity and plasma viscosity were measured. Most of the values of aloe treatment groups were significantly low compared with those of the control group(P<0.05). 4. The ulcer index of aloe treatment groups were significantly low campared with control group(p<0.05). Especially in AC and AD group the differences were more significant(p<0.01). 5. Less severe ulcers were observed in AA and AB group than in the control group. Tissues of AC and AD group had only slight ulcers and necrosis of tissue was not observed in these groups. Especially in AD group, there was more mucus than other groups and it seemed that alove vera stimulated the epithelial regeneration. From the results of this study, it can be concluded that the oral administration of Alove vera results in protection of stomach ulcer by stimulating the secretion of gastric mucus and the circulation of blood.

  • 1. 정상 성인 남자를 대상으로 한 25%의 알콜 100 ml 섭취시 DWP715는 전 대상인에서 $C_{max}$ 및 AUC 등의 감소와 다소 지연된 반감기($T_{1/2}$)를 나타내었으며 2시간에서 최대의 알콜농도 감소효과를 보여 주었다. 2. 3시간 구속스트레스를 통한 피로모델에서 DWP715는 피로부여시 증가된 LDH 수치, GOT수치 및 비장의 무게 등을 정상상태로 유지시켰다. 3. 과산화지질 생성을 위해 $CC1_4$로 유도된 항산화효과 측정 모델에서 DWP715는 대조군에 비해 유의적인 MDA감소를 보여주었다. 이상의 결과로 볼 때 DWP715는 알콜섭취 후 유도되는 간기능 저하에 따른 피로 상태와 아세트알데히드에 의해 형성되는 라디칼에 의한 간조직 손상을 보호해 주며 알콜섭취시 혈중으로의 알콜 흡수를 저하시키는 효능을 갖고 있는 것으로 판단된다. 따라서 DWP715는 혈중 알콜농도 감소와 함께 알콜섭취 후 발생되는 비특이적인 생체반응들에 의한 체내 불균형 상태를 효과적으로 회복시켜 줄 수 있을 것으로 생각된다.

지유가 흰쥐의 급성위염 및 소화성 궤양에 미치는 영향 (The Effects of Ethylacetate Fraction of Sanguisorba officinalis L. on Experimentally-induced Acute Gastritis and Peptic Ulcers in Rats)

  • 본 실험에서 흰쥐에 실험적으로 급성위염, 위궤양 및 십이지장궤양을 유발하고 지유 에틸아세테이트분획을 투여하여 항위염 및 항궤양에 미치는 영향을 검토하였던 바 다음과 같은 결과를 얻었다. 지유 에탄올 추출물을 n-헥산, 클로르포룸, 에틸아세테이트 및 n-부타놀과 물로 계통분획하여 농축하고 in vitro에서 항산화력을 측정한 결과 에틸아세테이트분획이 가장 우수하였다. 지유 에틸아세테이트분획 (SOL & SOH)의 HCI-EtOH급성위염에 대한 억제율은 각각 $27.7\%$, $31.7\%$로 대조약물로 사용된 cimetidine(CMT, $47.2\%$)의 억제효과에는 미치지 못했으나 CON에 비해 우수하였다. Indomethacin유발 위궤양에서, SOL & SOH의 위궤양억제율은 각각 $48.28\%$, $51.08\%$로 대조약물로 사용된 CMT($58.26\%$)의 억제효과에는 미치지 못했으나 CON에 비하며 유의 성있는 억제효과를 나타냈다. Shay 위궤양에서, 지유 에틸아세테이트분획을 십이지장에서 위내로 투여하였을 때 궤양 억제율은 SOL이 $42.86\%$로 CON에 비하여 억제되었으나 유의성 있는 효과는 아니었으며, 용량을 증량하여 투여한 SOH는 $47.71\%$로 대조약물인 omeprazol(OMP)의 저해효과($62.00\%$)에 미치지는 못했으나 CON에 비해 유의적민 효과를 나타내었다. 위액량 및 총위산분비량에서, 지유 에틸아세데이트분획 투여로 CON에 비하여 $32.11\%$$47.69\%$, $24.28\%$$34.69\%$의 감소로 유의성 있는 효과를 나타냈으며 OMP($55.69\%$, $41.62\%$)에 근접하게 감소되었다. pepsin 활성은 지유 에틸아세테이트분획 투여로 CON에 비해 각각 $19.55\%$, $\%21.45$가 저하되었으나 유의성있는 변화는 아니었다. 수침 $\cdot$ 구속 스트레스에 의해 유발된 궤양의 경우, SOL는 궤양억제율이 $9.23\%$ 로 CON와 유사하게 출혈을 동반한 깊은 궤양이 관찰되었으나 용량을 증량하여 투여한 SOH는 $48.45\%$로 CON에 비해 궤양의 깊이도 얕아졌고 출혈을 동반한 궤양도 감소되어 유의적인 억제율을 나타내었다. Cysteamine 유발 십이지장궤양에서, CMT는 유발된 십이지장 궤양을 CON에 비해 $54\%$ 정도 억제하여 우수한 항궤양효과를 나타냈고 시료투여군인 SOL 및 SOH는 CMT보다 우수하지는 않았으나 약 $35\∼42\%$ 정도의 궤양억제율을 나타냈다. 이상의 실험결과에서 지유의 항위염 및 항궤양효과는 항산화물질에 의한 결과로 판단되어지며, 앞으로 위염 및 위궤양 개선을 위한 건강 기능성 식품으로 개발이 기대된다

Effects of tianeptine on symptoms of fibromyalgia via BDNF signaling in a fibromyalgia animal model

  • Previous reports have suggested that physical and psychological stresses may trigger fibromyalgia (FM). Stress is an important risk factor in the development of depression and memory impairments. Antidepressants have been used to prevent stress-induced abnormal pain sensation. Among various antidepressants, tianeptine has been reported to be able to prevent neurodegeneration due to chronic stress and reverse decreases in hippocampal volume. To assess the possible effect of tianeptine on FM symptoms, we constructed a FM animal model induced by restraint stress with intermittent cold stress. All mice underwent nociceptive assays using electronic von Frey anesthesiometer and Hargreaves equipment. To assess the relationship between tianeptine and expression levels of brain-derived neurotrophic factor (BDNF), cAMP response element-binding protein (CREB), and phosphorylated cAMP response element-binding protein (p-CREB), western blotting and immunohistochemistry analyses were performed. In behavioral analysis, nociception tests showed that pain threshold was significantly decreased in the FM group compared to that in the control group. Western blot and immunohistochemical analyses of medial prefrontal cortex (mPFC) and hippocampus showed downregulation of BDNF and p-CREB proteins in the FM group compared to the control group. However, tianeptine recovered these changes in behavioral tests and protein level. Therefore, this FM animal model might be useful for investigating mechanisms linking BDNF-CREB pathway and pain. Our results suggest that tianeptine might potentially have therapeutic efficacy for FM.

외부구속 콘크리트 부재의 초기균열 예측을 위해 온도해석 결과를 이용한 자가변형 및 구속응력 측정 실험의 제안 (A Proposal of Autogenous Deformation and Self-induced Restrained Stress Test Using Thermal Analysis Results to Predict Early-Age Cracks of Externally Restrained Concrete Members)

  • 변형구속부재에서 발생하는 콘크리트 부재의 초기균열은 환경에 민감한 변수가 많아서 예측하기 힘들다. 본 연구에서는 벽체-기초부재에서 수화열에 의한 초기균열 발생 조건을 모사한 시편을 제작하고 자가변형과 자체유발응력을 측정하여 재료시험으로 예측하기 힘든 균열발생 가능성을 판단하는 방법을 제시하였다. 이를 위하여 먼저 전체 구조계의 환경조건을 고려한 부재의 수화열 해석으로 초기재령 콘크리트의 온도분포를 예측하고, 해석 결과와 같이 수화열이 전자제어되는 시편을 제작하였다. 초기재령 콘크리트의 자유변형을 측정하고 이를 보상하는 인장력을 가하여 균열을 유발하는 구속응력의 크기를 구하였다. 본 연구에서 제안한 실험적 방법은 측정값의 분산이 큰 초기재령 콘크리트의 재료시험에 의존하지 않고, 초기재령 콘크리트의 수화열에 의한 온도변화, 자기수축과 릴렉세이션에 의한 자유변형량 및 구속응력을 측정할 수 있으므로 초기 균열의 발생 여부를 보다 정확하게 예측할 수 있다. 본 연구의 방법은 대형 구조물의 시공에서 본격적인 콘크리트 타설 이전에 예비실험으로 적용하면 균열예측의 정도를 높일 수 있으므로 특히 유용하다.
