  • In sino-atrial node cells which act as the normal pacemaker of the heart, K conductance in resting state is minimal due to the absence of inward rectifier K channels K conductance only increases when the membrane is depolarized by the activation of the delayed rectifier K current I$\_$k/. Signaling Pathways Controlling Microglia Chemotaxis

  • Microglia are the primary resident immune cells of the central nervous system (CNS). They are the first line of defense of the brain's innate immune response against infection, injury, and diseases. Microglia respond to extracellular signals and engulf unwanted neuronal debris by phagocytosis, thereby maintaining normal cellular homeostasis in the CNS. Pathological stimuli such as neuronal injury induce transformation and activation of resting microglia with ramified morphology into a motile amoeboid form and activated microglia chemotax toward lesion site. This review outlines the current research on microglial activation and chemotaxis.

Alkane 유도체의 미생물학적 산화 제1보 (Oxidation of Alkane Derivatives by Corynebacterium sp.)

  • Twelve Microorganisms capable of utilizing diaminododecane were isolated from the soil by enrichment culture technique. Seven strains of these were identified as Corynebacterium. The isolated strains were tested for the ability to utilize as carbon source, 10 different kind of alkane derivatives containing CN, $NH_2$, Cl, and SH groups. Laurylcyanide, dicyanooetane, chlorodecane, and dichlorodecane were not utilized by any of the isolated strains; putrescine dihydrochloride, cadaverine dihydrochloride, diaminododecane, and n-dodecane were utilized by all of the isolated strains; and all of the isolated strains except DAD 2-3 could utilize dodecylmercaptan. The alkane derivatives that did not serve as ,growth substrates were tested further in oxidation tests using resting cell preparation. Alkane derivatives that are being oxidized by all of the isolated strains are laurylcyanide and dichlorodecane. Dicyanooctane was also oxidized by all of the isolated strains except DAD 30L, chlorodecane was the only oxidized by the three isolated strains. The most remarkable substrate that is being oxidized is dichlorodecane containing CN groups diterminally. Evidence obtained with thin layer chromatography of ,ethyl acetate extracts of culture broth of isolated strains grown in some alkane derivatives shows that these alkane derivatives are degraded.

  • The present study examined that in Vivo/Vitro test is investigated in normotensive sham-operated rats (NSR) and aldosterone-analogue deoxycorticosterone acetate (DOCA)-salt hypertensive rats (ADHR) and that the antihypertensive effect was induced by silver spike point (SSP) electrical stimulation at meridian points(CV-3, -4, Ki-12, SP-6, LR-3, BL-25, -28, -32, -52), specifically, such as aldosterone in 24 hour urine analysis from healthy volunteer. The gross examination and morphometric-histological changes, such as hypertrophy, production of necrotic tissues, and the changes of cell arrangement on the kidney, and adrenal gland were markedly observed in aldosterone-analogue DOCA-salt hypertensive rats compared with those from normotensive sham-operated rats. The systolic blood pressure, weight of kidney and adrenal gland were significantly increased in ADHR than that in NSR. The required time of PSS-induced resting tone was significantly increased in ADHR than that in NSR. However, the voltage-dependent K+ (Kv) currents were significantly decreased in ADHR than that in NSR. The urine analysis showed that the concentration of aldosterone was significantly decreased in resting state from the elderly people compared with those from the adolescent healthy volunteer. The current of 1 Hz continue type of SSP electrical stimulation significantly decreased in the concentration of aldosterone of 24 hour urine from the elderly people. These results suggest that the development of aldosterone analogue-induced hypertension is associated with changed the weight of kidney and adrenal gland, blood pressure, resting tone and Kv currents, which directly affects blood pressure. Therefore, the hypertension is a risk factor on cerebrovascular disease. Moreover, these results suggest that the diminished responsiveness to SSP electrical stimulation, especially current of 1Hz continue type, in elderly people may be, in part, related by the increased of antihypertensive effects.

Short Heterodimer Partner as a Regulator in OxLDL-induced Signaling Pathway

  • Oxidized low-density lipoprotein (oxLDL) has been shown to modulate transactivations by the peroxisome proliferator activated receptor (PPAR)$\gamma$ and nuclear factor-kappa B (NF$\kappa$B). In this study, the oxLDL signaling pathways involved with the NF$\kappa$B transactivation were investigated by utilizing a reporter construct driven by three upstream NF$\kappa$B binding sites, and various pharmacological inhibitors. OxLDL and its constituent lysophophatidylcholine (lysoPC) induced a rapid and transient increase of intracellular calcium and stimulated the NF-KB transactivation in resting RAW264.7 macrophage cells in an oxidation-dependent manner. The NF$\kappa$B activation by oxLDL or lysoPC was inhibited by protein kinase C inhibitors or an intracellular calcium chelator. Tyrosine kinase or PI3 kinase inhibitors did not block the NF$\kappa$B transactivation. Furthermore, the oxLDL-induced NF$\kappa$B activity was abolished by the PPAR$\gamma$ ligands. When the endocytosis of oxLDL was blocked by cytochalasin B, the NF$\kappa$B transactivation by oxLDL was synergistically increased, while PPAR transactivation was blocked. These results suggest that oxLDL activates NF-$\kappa$B in resting macrophages via protein kinase C- and/or calcium-dependent pathways, which does not involve the endocytic processing of oxLDL. The endocytosis-dependent PPAR$\gamma$ activation by oxLDL may function as an inactivation route of the oxLDL induced NF$\kappa$B signal. Short heterodimer partner (SHP), specifically expressed in liver and a limited number of other tissues, is an unusual orphan nuclear receptor that lacks the conventional DNA-binding domain. In this work, we found that SHP expression is abundant in murine macrophage cell line RAW 264.7 but suppressed by oxLDL and its constituent I3-HODE, a ligand for peroxisome proliferator-activated receptor y. Furthermore, SHP acted as a transcription coactivator of nuclear factor-$\kappa$B (NF$\kappa$B) and was essential for the previously described NF$\kappa$B transactivation by lysoPC, one of the oxLDL constituents. Accordingly, NF$\kappa$B, transcriptionally active in the beginning, became progressively inert in oxLDL-treated RAW 264.7 cells, as oxLDL decreased the SHP expression. Thus, SHP appears to be an important modulatory component to regulate the transcriptional activities of NF$\kappa$B in oxLDL-treated, resting macrophage cells.

동해 북동부해역 제 4기 후기 퇴적물의 규조 산출과 고해양학적 변화 (Occurrence of Diatom in the Late Quaternary Sediments of the Northeastern East Sea (Sea of Japan) and its Paleoceanographic Changes)

  • 동해 북동쪽에 위치한 홋카이도섬 서부해역에서 채취된 중력시추코아 GH98-1223의 45개 퇴적물 시료들로부터 휴면포자를 제외한 총 31속 50종 1아종의 규조류가 동정되었다. 최대 산출종은 Thalassionema nitzschioides로 전시료에서 29${\sim}$59%의 높은 산출빈도를 나타냈으며, 그 외에 비교적 개체 보존이 양호한 Denticulopsis seminae와 Pseudoeunotia doliolus를 비롯한 대부분의 종들은 평균 산출빈도가 5% 이내로 매우 낮게 나타났다 전 시료에서 한류종의 산출빈도가 난류종보다 상대적으로 우세하였으며, 수직분포는 각 종의 생태적 특성에 따라 차이가 있으나 대부분의 층준에서 난류종과 한류종의 산출빈도는 서로 상반되는 경향을 보였다. 대표적인 한류종인 D. seminae와 난류종으로 쿠로시오해류(Kuroshio Current)의 지류인 대마난류(Tsushima Warm Current)의 지시종인 P doliolus의 전체적인 변화양상은 뚜렷하게 상반되며, 이것은 규조온도지수(T$_d$)의 수직적인 변화와도 서로 일치한다. 규조온도지수는 150 cm깊이를 기준으로 코아의 상부에서는 평균 이상의 높은 수치를 나타내며 하부에서는 평균 이하의 낮은 수치를 나타내는데, 이러한 경향은 동해의 북부지역에 위치한 본 연구지역에 대마난류의 영향이 홀로세의 후빙기를 거치면서 점차적으로 증가된 것을 지시하고 있다. 또한, 하층부에서 규조온도지수는 지그재그형의 변화를 나타내는데 이는 규조류가 서식하는 해수의 환경이 불안정하였다는 것을 의미한다. 생산성과 용승의 정도를 지시하는 Chaetoceros resting spores는 5.3${\sim}$40% 정도의 산출빈도를 보이는데 , 최대값은 80 cm 보다 상층부에서 나타났다. 퇴적물의 생물 기원물질의 상대적인 양을 예측하는데 이용되는 Chaetoceros resting spores/Chaetoceroe vegetative cells도 80 cm 보다 상층에서는 높게 나타나 규조온도지수 분포와도 일치하는 경향을 보인다. 이상의 규조군집 분석 결과에 의하면, 홀로세의 후빙기동안 본 연구 지역인 동해 북동부에는 대마 난류의 유입이후 현재와 유사한 환경이 우세하게 발달했으나, 난류종 P. doliolus의 변화는 동해내에서 대마난류의 세기가 반복되었음을 지시하고 있다.

사람의 골수와 제대정맥에서 유래된 중간엽 줄기세포에서 TREK1 통로의 기능적 발현 (Functional expression of TREK1 channel in human bone marrow and human umbilical cord vein-derived mesenchymal stem cells)

  • 사람의 골수 또는 제대정맥에서 유래된 중간엽 줄기 세포 (hBM-MSC 또는 hUC-MSC)는 임상적 치료 적용에 매우 유용한 세포유형으로 알려져 왔다. 우리는 이러한 세포에서 two-pore 도메인 포타슘 (K2P)채널을 조사하였다. K2P 채널은 다양한 세포유형들에서 안정막 전위를 형성하는데 중요한 역할을 한다. 그들 중 TREK1은 수소, 저산소증, 다불포화 지방산, 항우울제 및 신경전달물질들의 표적이다. 우리는 RT-PCR 분석과 팻취고정기법을 이용하여 hBM-MSCs와 hUC-MSC가 기능적인 TREK1 채널을 발현하는지 조사했다. hBM-MSCs와 hUC-MSCs에서 100 pS 단일 채널 전도도를 가진 포타슘채널이 발견되었고, 그 채널은 세포막 신전 (-5 mmHg ~ -15 mmHg), 아라키도닉산 ($10{\mu}M$), 세포내 산성화 (pH 6.0)에 의해 활성화 되었다. 이러한 전기생리학적 성질은 TREK1과 유사하였다. 우리의 결과는 안정막 전위에 기여하는 TREK1 채널이 hBM-MSC와 hUC-MSC에 기능적으로 존재하고 있음을 제시한다.

부탄분해미생물에 의한 가솔린첨가제 MTBE(Methyl tert-Butyl Ether) 분해 (Biodegradation of Gasoline Oxygenate MTBE(Methyl tert-Butyl Ether) by Butane-Utilizing Bacteria)

  • 본 논문에서는 순수균주인 ENV425와 오염된 토양으로부터 분리한 혼합균주를 이용하여 가솔린 산화제인 MTBE에 대한 분해 가능성을 조사했다. MTBE는 n-butane에서 성장한 ENV425와 혼합균주에 의해 공대사적으로 분해가 이루어졌다. 또한 아세틸렌의 첨가에 의해 n-butane과 MTBE의 분해가 완전히 방해되어짐에 따라 두 기질 모두 부탄 분해 효소에 의해 분해되어짐을 알 수 있었다. n-butane에서 성장한 ENV42S와 혼합균주는 MTBE를 분해하고, MTBE의 분해산물로 TBA가 생성되었다. TBA의 생성은 분해된 MTBE에 대하여 ENV425와 혼합균주 각각 54.7%, 58.6%가 관찰되었다. 그러나, Resting cell 실험에서는 ENV425와 혼합균주에 의한 산화 생성물로 TBA와 TBF가 생성되었다. ENV425와 혼합균주에 의한 최대 MTBE 분해속도는 각각 52.3 그리고 62.3 (nmol MTBE degraded/hr/mg TSS), 최대 $T_y$ (Transformation yield)는 각각 44.7, 34.0 (nmol MTBE degraded/$\mu$mol n-butane utilized)으로 나타났고, 최대 $T_c$ (Transformation capacity)는 각각 199, 226 (nmol MTBE degraded/mg TSS used)으로 나타났다.

수소생산을 위한 Rhodopseudomonas sphaeroides의 최적 배양조건 (Optimum Culture Conditions for Hydrogen Production of Rhodopseudomonas sphaeroides)

  • We examined optimum culture conditions of Rhodopseudomonas sphaeroides B5 for effective utilization of substrate and sunlight for hydrogen production. The optimum concentration range of DL-lactate as electron donor for hydrogen production by resting cells was from 5 to 50mM, and optimun CN ratio (lactate/glutamat) for maintenence of hydrogen production activity by growing cultures was from 5 to 6. Hydrogen production by the cultures of low cell density (0.36mg/ml dry cells) was saturated with 10 Klux light intensity. Under constant illumination of 50Klux which was set up as the average medium value of annual variation of sunlight intensity, hydrogen production with various cell densities in the culture resulted in highest production rate (132${\mu}$l/hr/mg dry cells) up to 0.64mg/ml dry cells. However, the amount of total hydrogen production was saturated with cell density of 2.1mg/ml dry cells. In addition to these, the optimum inner thickness pervious to light of the culture vessel for hydrogen production which was measured under sunlight was 5 cm.

Developmental Changes of Gustatory Neurons in Nucleus of Solitary Tract in Rats

  • To learn the developmental changes in intrinsic electrophysiological properties of the second order taste neurons, whole cell recordings from the developing nucleus of the solitary tract neurons were done in brainstem slices of postnatal rats. Rats aged from postnatal 0 to 21 days (P0-P21) were used, being divided into 3 age groups: postnatal first week (P0-P7 days), second week (P8-P14 days), and third week (P15-P21 days). Slices containing gustatory NTS were cut horizontally in the thickness of $300\;{\mu}m.$ Whole cell recordings were obtained from neurons in response to a series of hyperpolarizing and depolarizing current pulses. The intrinsic electrophysiological properties of the rostral NTS (rNTS) neurons were compared among the age groups. Depolarizing current pulses evoked a train of action potentials in all neurons of all age groups. The resting membrane potential and input resistance of the neurons did not show any significant differences during the postnatal 3 weeks. The time constant, however, decreased during the development. Duration of action potential measured at half maximum amplitude was longer in younger age groups. Both the maximum rate of rise and the maximum rate of fall in the action potential increased during the first 3 weeks postnatal. Electrophysiologically more than half neurons were type III. In summary, it is suggested that developmental changes in electrophysiological properties in rNTS occur during the first three weeks in rats.
