가미귀용탕(加味歸茸湯)이 새끼 생쥐의 면역반응(免疫反應) 및 성장(成長)에 미치는 영향(影響) (Effect of Kamikwiryongtang on Immune Response and Growth in a Young Mouse)

  • Objective: The purpose of this study was to investigate the effects of Kamikwiryongtang (KKT) on the immune response and growth in a young mouse (3 weeks mice). Methods The viability of thymocytes and splenocytes in vivo and in vitro system, the population of helper T (Th) cells and cytotoxic T (Tc) cells in thymocytes and increased the population of T-lymphocytes and the population of Th cells in splenocytes, the production of ${\gamma}$ -interferon, interleukin-2 and interleukin-4 in splenocytes was investigated. KKT (500mg/kg) was administerd p.o. once a day for 7 days. Results: KKT increased the viability of thymocytes and splenocytes in vivo, but did not affect the viability of thymocytes and enhanced the viability of splenocytes in vitro system. In addition, KKT did not affect the population of helper T (Th) cells and cytotoxic T (Tc) cells in thymocytes and increased the population of T -lymphocytes and the population of Th cells in splenocytes. Also, KKT increased the production of ${\gamma}$-interferon, interleukin-2 and interleukin-4 in splenocytes. Furthermore, KKT increased the production of nitric oxide in vivo, but did not affect the production of nitric oxide in vitro system. KKT enhanced the phagocytic activity of peritoneal macrophages in vivo, but decreased the phagocytic activity in vitro system: KKT increased the body weight of a young mouse. Conclusions: KKT stimulates the specific immune response via increase of, the viability of thymocytes and splenocytes and the non-specific immune response via increase of phagocytic activity of peritoneal macrophages and stimulates the growth of a young mouse.

Relative Performance and Immune Response in White Leghorn Layers Fed Liquid DL-methionine Hydroxy Analogue and DL-methionine

  • The relative performance and immune response was evaluated in White Leghorn layers fed liquid DL-methionine hydroxyl analogue-free acid (MHA-FA) relative to dry DL-methionine (DLM) in maize-soybean-sunflower based diets. Three graded levels of methionine (Met) from DLM or MHA-FA were added to the basal diet containing 0.27% Met on an equimolar basis to achieve 0.30, 0.36 and 0.42% Met in the diet. Each diet was fed ad libitum to 25 replicates of one bird (individual feeding) each, from 24 to 40 weeks of age. A regime of 16 h light was provided and all the layers were kept under uniform management throughout the experimental period. None of the parameters studied were influenced by the interaction between source and level of Met in diets. Similarly, the majority of parameters, except for daily feed consumption and immune response (influenced by level) and egg specific gravity and shell thickness (influenced by source), were not affected by either source or level of Met in the diets. Feed consumption was significantly lower in the birds fed a diet containing 0.42% Met compared to those fed lower levels of Met. The cutaneous basophilic hypersensitivity response to PHA-P and antibody titre (32 and 40 wk) to inoculation of sheep red blood cells increased significantly by increasing the concentration of Met in the diet from 0.30 to 0.36%. Thus, the Met requirement for immune competence was higher than for optimum production. The source of Met significantly influenced the egg specific gravity and shell thickness. The specific gravity and shell thickness of eggs increased significantly when MHA-FA was used as the source of Met in the diet compared to DLM. From the study it is concluded that Met requirement for immune competence (360 mg/b/d) is higher than for optimum production (300 mg/b/d). MHA-FA was comparable with DLM as a source of Met for production performance and immunity, when the bioavailability of MHA-FA was considered as 88% of DLM. Further, MHA-FA improved egg shell quality compared to DLM.

  • The aim of this research was to the experimental data using statistical and spectral analyzing method to get the motion reponses of a stern trawler in operation states such as drifting, sailing and trawling according to the wave height. In drifting, the significant and the maximum valuer of roll in beam sea increased according to the wave height, but those of pitch decreased. The response and the period of peak of roll in beam sea were increased, but those of pitch decreased. In navigation, the significant and maximum values of roll increased remarkably according to the wave height, but those of pitch changed a little. The response of roll was highest in quartering sea, beam sea and then following sea, but those of pitch was highest in bow sea, head sea and then beam sea in the order of all wave heights. The period of peak of roll due to the wave height and the wave direction changed from 3.8 to 9.9 seconds, and those of pitch changed from 3.3 to 10.4 seconds. In trawling, the significant and maximum values of roll increased a little according to the wave height, but those of pitch increased significantly. The response of roll was highest in beam sea, bow sea and then quartering sea, but those of pitch was highest in head sea, following sea, and then beam sea in the order. The period of peak of roll due to the wave height and the direction changed from 6.6 to 10.9 seconds, and those of pitch changed from 6.7 to 11.2 seconds.

진구가 Collagen유발(誘發) 관절염(關節炎)의 면역반응(免疫反應)에 미치는 영향(影響) (Effects of Gentiana marcrophylla on Immune Response in the Collagen induced Arthritis)

  • To know the effects of Gentiana macrophylla(GM) on the immune response, this study was undertaken. GM is one of the well-known oriental medicines for a lomg time used for the treatment of such diseases as arthralgia, headache, hepatitis, SLE, hemiparesis, and so on. To evaluate the effects of GM on immune response in the collagen induced arthritis, phagocytic activity of macrophages, proliferation of T-lymphocytes, secretion of nitric oxide in urine, and production of ROIs, $TNF-{\alpha}$ and nitric oxide in synoviocytes were measured. The results obtained in this study were as follows: 1. During the progress of CIA, the administration of GM enhanced phagocytic activity of macrophages in vivo and vitro. 2. During the progress of CIA, the administration of GM inhibited production of ROIs in synoviocytes. 3. During the progress of CIA, the administration of GM inhibited production of TNF- a in synoviocytes. 4. During the progress of CIA, the administration of GM inhibited proliferation of T lymphocytes. 5. During the progress of CIA, the administration of GM inhibited production of nitric oxide in synoviocytes. 6. During the progress of CIA, the administration of GM inhibited secretion of nitric oxide in urine. According to the above results, during the progress of CIA, it might be considered that GM has a curative effect on rheumatoid arthritis by controling immune response.

A Three-step Method of Immunotoxicity Assessment

  • The immunosuppressive effects of thirty nine chemicals chosen by their potential toxicity were evaluated using a three-step testing method. The immunotoxicity test method developed in this study consisted of three simple assays of lymphoproliferation, mixed leukocyte response, and interleukin (IL)-2 production. The first step was mitogen-induced proliferation assay. Ten chemicals showed the inhibitory effects on the mitogen (lipopolysaccharide or concanavalin A)-induced proliferation in dose-dependent manners. The second step was mixed lymphocyte response. This step crosschecked the growth-suppressive effects detected at the first step. All of 10 chemicals, which showed suppression of lymphoproliferation, also exhibited the suppressive effects on the mixed lymphocyte response in the similar range of chemical concentration. The third step was planned to determine whether or not this growth suppression was mediated through an early activation of T-cell, which could be represented with IL-2 production. Six out of 10 chemicals decreased the interleukin-2 production in the similar concentration range used in the step 1 and 2. These results suggest that those 6 chemicals might have their targets on the signal transduction path-way toward the IL-2 production. In the meantime the other 4 chemicals might have their targets after the IL-2 production signal. Taken all together, the three-step test would be simple, fast, and efficient to deter-mine whether or not the chemical has immunosuppressive effects.

Influence of Temperature Shifts on Antibody Synthesis in the Oliver Flounder (Paralichthys olivaceus) Immunised with Formalin Killed Edwardsiella tarda Antigen

  • The effects of various temperature shifts on the kinetics of the humoral antibody response in oliver flounder, Paralichthys olivaceus, immunised with formalin-killed Edwardsiella tarda, were determined by measuring the antibody production in vivo and in vitro. When fish acclimated to a high temperature and immunised at that temperature were transferred to a lower temperature (22℃ to 12℃) at a various times after immunisation, the fish showed a weaker immune response than that achieved when the fish were kept at a high environmental temperature. However, in the converse experiment (12℃ to 22℃), the magnitude of the humoral immune response was recovered independent of the time of the transfer after immunisation at low temperature, even though the peak levels of each transferred group did not reach the level found in the positive control group that was maintained and immunised at a high environmental temperature. Hence, these studies provide some evidence that the potential for antibody production in B cells of oliver flounder immunized at high temperature is not impaired by subsequent exposure to low temperature.

New Response Surface Approach to Optimize Medium Composition for Production of Bacteriocin by Lactobacillus acidophilus ATCC 4356

  • The objective of this study was to optimize medium composition of initial pH, tryptone, glucose, yeast extract, and mineral mixture for production of bacteriocin by Lactobacillus acidophilus ATCC 4356, using response surface methodology. A response surface approach including new statistical and plotting methods was employed for design and analysis of the experiment. An interiorly augmented central composite design was used as an experimental design. A normal-distribution log-link generalized linear model based on a subset fourth-order polynomial ($R^2$=0.94, Mean Error Deviance=0.0065) was used as an analysis model. This model was statistically superior to the full second-order polynomial-based generalized linear model ($R^2$=0.80, Mean Error Deviance=0.0140). Nonlinear programming determined the optimum composition of the medium as initial pH 6.35, typtone $1.21\%$, glucose $0.9\%$, yeast extract $0.65\%$, and mineral mixture $1.17\%$. A validation experiment confirmed that the optimized medium was comparable to the MRS medium in bacteriocin production, having the advantage of economy and practicality.

Optimization of Culture Media for Enhanced Chitinase Production from a Novel Strain of Stenotrophomonas maltophilia Using Response Surface Methodology

  • Chitinase is one of the most important mycolytic enzymes with industrial significance. This enzyme is produced by a number of organisms including bacteria. In this study, we describe the optimization of media components with increased production of chitinase for the selected bacteria, Stenotrophomonas maltophilia, isolated from soil. Different components of the defined media responsible for influencing chitinase secretion by the bacterial isolate were screened using Plackett-Burman experimental design and were further optimized by Box-Behnken factorial design of response surface methodology in liquid culture. Maximum chitinase production was predicted in medium containing 4.94 g/l chitin, 5.56 g/l maltose, 0.62 g/l yeast extract, 1.33 g/l $KH_2PO_4$, and 0.65 g/l $MgSO_4{\cdot}7H_2O$ using response surface plots and the point prediction tool of the DESIGN EXPERT 7.1.6 (Stat-Ease, USA) software.

Bovine Serum Albumin 투여가 산란계의 품종별 면역반응에 미치는 영향 (The Level of Immune Response on Bovine Serum Albumin(BSA) Injection to Different Breeds of Laying Hen)

  • This experiment was carried out to investigate the effects of the laying hens on the immune response against bovine serum albumin(BSA) in egg yolk. Total 45 laying hens were divided into three groups according to breeds (White Leghorn, ISA Brown, Native hen). They were fed the experimental diet for 12 weeks. Immune response were examind in egg yolk from three groups of hens injected with BSA. The results obtained from this work were summaried as follows : 1. The weight of egg yolk and the percentage of hen-day production in the ISA Brown hens are greater than those in the Native hens and the White Leghons. 2. IgY concentrations in eggs from hens immunized with BSA were not different among the breeds laying hens. 3. The anti-BSA antibody activities determined by enzyme linked immunosorbent assay (ELISA) in the egg yolk were similar between the White-Leghorn and ISA Brown hens, but Native hens tended to decrease in 20∼50 days respectively. Therefore, the weight of egg yolk and the percentage of hen-day production in the ISA Brown hens are greater than those in the Native hens and the White Leghons will be as important factors for an efficient production of IgY.

Application of Response Surface Methodology and Plackett Burman Design assisted with Support Vector Machine for the Optimization of Nitrilase Production by Bacillus subtilis AGAB-2

  • Nitrilases are a hydrolase group of enzymes that catalyzes nitrile compounds and produce industrially important organic acids. The current objective is to optimize nitrilase production using statistical methods assisted with artificial intelligence (AI) tool from novel nitrile degrading isolate. A nitrile hydrolyzing bacteria Bacillus subtilis AGAB-2 (GenBank Ascension number- MW857547) was isolated from industrial effluent waste through an enrichment culture technique. The culture conditions were optimized by creating an orthogonal design with 7 variables to investigate the effect of the significant factors on nitrilase activity. On the basis of obtained data, an AI-driven support vector machine was used for the fitted regression, which yielded new sets of predicted responses with zero mean error and reduced root mean square error. The results of the above global optimization were regarded as the theoretical optimal function conditions. Nitrilase activity of 9832 ± 15.3 U/ml was obtained under optimized conditions, which is a 5.3-fold increase in compared to unoptimized (1822 ± 18.42 U/ml). The statistical optimization method involving Plackett Burman Design and Response surface methodology in combination with an AI tool created a better response prediction model with a significant improvement in enzyme production.
