Distribution, Characterization, and Diversity of the Endophytic Fungal Communities on Korean Seacoasts Showing Contrasting Geographic Conditions

  • This study analyzed the distribution of endophytic fungi in 3 coastal environments with different climatic, geographical, and geological characteristics: the volcanic islands of Dokdo, the East Sea, and the West Sea of Korea. The isolated fungal endophytes were characterized and analyzed with respect to the characteristics of their host environments. For this purpose, we selected common native coastal halophyte communities from three regions. Molecular identification of the fungal endophytes showed clear differences among the sampling sites and halophyte host species. Isolates were also characterized by growth at specific salinities or pH gradients, with reference to previous geographical, geological, and climate studies. Unlike the East Sea or West Sea isolates, some Dokdo Islands isolates showed endurable traits with growth in high salinity, and many showed growth under extremely alkaline conditions. A smaller proportion of West Sea coast isolates tolerate compared to the East Sea or Dokdo Islands isolates. These results suggest that these unique fungal biota developed through a close interaction between the host halophyte and their environment, even within the same halophyte species. Therefore, this study proposes the application of specific fungal resources for restoring sand dunes and salt-damaged agricultural lands and industrialization of halophytic plants.

DNA barcoding of Euphorbiaceae in Korea

  • The Euphorbiaceae family features some of the most economically important plants that are sources of foods, oils, waxes, and medicines. The accurate identification of Euphorbiaceae species is critical in sustainable utilization of plant resources. We examined 234 sequences of nrDNA ITS, cpDNA rbcL and matK loci from 20 species in Euphorbiaceae in Korea and three outgroup taxa to develop efficient DNA barcodes. The three barcode loci were successfully amplified and sequenced for all Euphorbiaceae species. nrDNA ITS locus showed the highest mean interspecific K2P distance (0.3034), followed by cpDNA matK (0.0830), and rbcL (0.0352) locus. The degree of species resolution for individual barcode loci ranged from 75% (rbcL and matK) to 80% (ITS). The degree of species resolution was not enhanced with the different combinations of three barcode loci. The combined data set of the three loci(ITS+rbcL+matK) provided 80% of species resolution. These results confirm that ITS locus, as a single barcode, is the best option for barcoding of the Euphorbiaceae in Korea.

  • Justicia kampotiana Benoist (쥐꼬리망초과)를 베트남 미기록종으로 보고한다. 본 종은 유사종인 J. ingrata Benoist와 달리 외화관에 털이있으며, 화통은 판연 보다 약간 길고 화병보다는 아주 짧다. 종 동정을 위한 기재, 그림도해 및 컬러사진과 함께 본 종과 유사종의 비교를 위한 검색형질들을 제시하였다.

  • 본 연구를 통해 국내미기록속인 까마귀나비속의 푸른줄까마귀왕나비를 최초로 보고한다. 푸른줄까마귀왕나비의 암컷 형태와 생식기, 채집지 등의 정보를 제공하였다. 또한 신속한 종 동정을 위한 DNA 바코드 염기서열을 정보를 제공하였다.

Isolation and Identification of Intestinal Bacteria of C57BL/6 Mice to Assess Biological Activities of Plant Resources

  • Much of bacteria inhabit intestine and affect health. To elucidate the composition of intestinal bacteria and biological activities of plant materials on the bacteria, bacterial strains are need to be isolated and identified. In previous study, we isolated 41 fecal bacteria of BALB/c mice and the strains were identified as 11 species including Lactobacillus murinus and not classified bacterium. To expand the bacterial resources, we tried to isolate more bacteria from C57BL/6 mice. Fresh feces was suspended and serially diluted in distilled water. The aliquots were inoculated on GAM agar plate and incubated anaerobically at $37^{\circ}C$ for 48 h. Each of colony formed was picked up and incubated again on GAM agar plate for stock and sampling. The bacteria gained were analyzed and identified by 16S rRNA gene. The bacterial strain were listed up. Major strain was Lactobacillus murinus which was observed as an abundant strain of BALB/c mice. The resources could be used for experiments of biological activities of plant materials and microbial composition of intestinal contents of experimental animals.

사슴 미토콘드리아 DNA의 염기서열 및 PCR-RFLP분석에 의한 녹용의 종 감별 (Identification of Deer Antler Species Using Sequence Analysis and PCR-RFLP of Mitochondrial DNA)

  • 우리나라는 전 세계 녹용의 약 80% 이상을 소비하고 있는 양록 대국이나 최근 국내 녹용시장에서의 녹용 둔갑판매 및 불법유통 현상이 문제점으로 대두되고 있다. 따라서 본 연구는 녹용의 종 감별 기술을 개발하고자 현재 국내에서 유통되고 있는 러시아산 원용, 북미산 대록, 국산화용, 중국산 깔깔이 및 알래스카산 순록 등 5종의 대표적인 녹용들을 대상으로 종간 염기서열 변이성이 매우 높은 유전자로 알려져 있는 mt DNA내 cytochrome b 및 D-loop 유전자 영역의 염기서열 분석 및 종간 변이성 비교분석을 수행하였다. 각 녹용시료에서 mt DNA를 분리하고 cytochrome b와 D-loop유전자의 특정 영역을 포함하는 primer를 설계 합성하고 PCR로 증폭한 후 DNA 증폭산물의 염기서열을 분석하여 종간 유전정보의 동일성 여부를 비교한 결과 녹용 종간에 명확한 차이를 보이는 염기서열 부위가 검출되었고 이러한 종간 염기배열 차이에 근거하여 녹용의 종 감별이 가능하였다. 또한, mt DNA cytochrome b유전자에서 종간 특이적 염기서열을 인지하는 두 종류의 제한효소(NlaIV 및 TaqI)을 이용한 PCR-RFLP 기법으로 녹용으로 인정되지 않는 순록의 종 특이적 RFLP 분자표지를 검출하였고 이를 이용하여 녹용과 순록간의 종 판별이 가능하였다. 한편, D-loop 유전자의 특정 영역 염기서열 분석기법을 이용하여 시중에서 러시아산 원용으로 유통되고 있는 녹용 절편 32개를 무작위표본 추출하여 녹용의 종 감별을 조사한 결과 러시아산 원용으로 인정되는 것은 62.5%에 불과하였고 나머지는 중국산 마록(25.0%)과 엘크 및 순록의 아종으로 추정되는 시료도 일부 검출되었다. 따라서 본 연구를 통해 사슴 녹용 mt DNA 유전자의 염기서열 유전정보 변이 차이를 이용한 염기서열 분석법과 특정 제한효소(NlaIV 및 TaqI)를 이용한 PCR-RFLP 기법은 녹용의 과학적인 종 감별과 이를 바탕으로 녹용 원산지의 추정도 가능할 것으로 기대된다.

  • Considering all the factors involved in beef production individual identification using DNA marker testing is the most appropriate solution to give all the breeders' information to the consumers. After taking into account the genealogical information from the Hanwoo, only animals that did not share some parent or grandparent were analysed 33 from the 305 initially sampled. Ten major microsatellite markers were selected from allele amplified and their frequencies, H(Heterozygosity) and PIC(Polymorphism information content) with Hardy-Weinberg equilibrium. Next, in order to evaluate the power of the markers selected on the individual animal identification, the match probability(MP) and the relatedness coefficient(R) were computed.

덩굴류 공예유물의 종 동정 (Species Identification of Vinery Craftwork Relics)

  • Wood-anatomical studies on vines, one of the main materials for fork craftworks used in the past, were insufficient compared to those for trees. Therefore, it is important to identify the species of vines in order to preserve the vinery craftwork relics. In this study, the species of 31 samples (30 traditional baskets and one sculpture relic) obtained from Korea were identified. Twenty-two baskets among 30 baskets were identified as Cocculus trilobus DC, a perennial vine which has flexible xylem and thick fibers in phloem. The other baskets were identified as $Lespedeza$ spp., a shrub with tough xylem. The sculpture part was $Actinidia$ $arguta$ Planch., a perennial vine with chambered piths.

A checklist of Trichoptera (Insecta) of the Korean Peninsula

  • A revised checklist of Korean Trichoptera is provided for the species recorded from the Korean Peninsula, including both North and South Korea. The checklist includes bibliographic research as well as results after reexamination of some specimens. For each species, we provide the taxonomic literature that examined Korean Trichoptera materials or mentioned significant taxonomic treatments regarding to Korean species. We also provide the records of unnamed species based on larval identification for further study. Based on taxonomic considerations, 20 species among the previously known nominal species in Korea are deleted or synonymized, and three species omitted from the previous lists, Hydropsyche athene Malicky and Chantaramongkol, 2000, H. simulata Mosely, 1942 and Helicopsyche coreana Mey, 1991 are newly added to the checklist. Hydropsyche formosana Ulmer, 1911 is recorded from the Korean Peninsula for the first time by the identification of Hydropsyche KD. In addition, we recognized 14 species of larvae separated with only tentative alphabetic designations. As a result, this new Korean Trichoptera checklist includes 218 currently recognized species in 66 genera and 25 families from the Korean Peninsula.

감시정찰 센서네트워크에서 제대규모 식별 기법 (Echelons Scale Identification Scheme of Surveillance and Reconnaissance Sensor Network)

  • Surveillance and reconnaissance sensor network system is an application system based on ubiquitous sensor network technology. This technique is to avoid accidental close combat, to minimize the consumption of limited military resources and personnel, and to provide battlefield situational awareness information for the unit's future combat missions. In this paper, we have proposed a echelons scale identification scheme based on information obtained from surveillance and reconnaissance sensor network system.
