Rapid molecular authentication of three medicinal plant species, Cynanchum wilfordii, Cynanchum auriculatum, and Polygonum multiflorum (Fallopia multiflorum), by the development of RAPD-derived SCAR markers and multiplex-PCR

  • Definitive identification of original plant species is important for standardizing herbal medicine. The herbal medicines Cynanchi Wilfordii Radix (Baekshuoh in Korean and Beishuwu in Chinese) and Polygoni Multiflori Radix (Hashuoh in Korean and Heshuwu in Chinese) are often misidentified in the Korean herbal market due to morphological similarities and similar names. Therefore, we developed a reliable molecular marker for the identification of Cynanchi Wilfordii Radix and Polygoni Multiflori Radix. We used random amplified polymorphic DNA (RAPD) analysis of three plant species, Polygoni multiflorum, Cynanchum wilfordii, and Cynanchum auriculatum, to obtain several species-specific RAPD amplicons. From nucleotide sequences of these RAPD amplicons, we developed six sequence characterized amplification region (SCAR) markers for distinguishing Polygoni Multiflori Radix and Cynanchi Wilfordii Radix. Furthermore, we established SCAR markers for the simultaneous discrimination of the three species within a single reaction by using multiplex-PCR. These SCAR markers can be used for efficient and rapid authentication of these closely related species, and will be useful for preventing the distribution of adulterants.

Mode identifiability of a multi-span cable-stayed bridge utilizing stabilization diagram and singular values

  • This study investigates the mode identifiability of a multi-span cable-stayed bridge in terms of a benchmark study using stabilization diagrams of a system model identified using stochastic subspace identification (SSI). Cumulative contribution ratios (CCRs) estimated from singular values of system models under different wind conditions were also considered. Observations revealed that wind speed might influence the mode identifiability of a specific mode of a cable-stayed bridge. Moreover the cumulative contribution ratio showed that the time histories monitored during strong winds, such as those of a typhoon, can be modeled with less system order than under weak winds. The blind data Acc 1 and Acc 2 were categorized as data obtained under a typhoon. Blind data Acc 3 and Acc 4 were categorized as data obtained under wind conditions of critical wind speeds around 7.5 m/s. Finally, blind data Acc 5 and Acc 6 were categorized as data measured under weak wind conditions.

  • Post-transcriptional nucleotide sequence modification of transcripts by RNA editing is an important molecular mechanism in the regulation of protein function and is associated with a variety of human disease phenotypes. Identification of RNA editing sites is the basic step for studying RNA editing. Databases and bioinformatics resources are used to annotate and evaluate as well as identify RNA editing sites. No method is free of limitations. Correctly establishing an analytic pipeline and strategic application of both experimental and bioinformatics methods constitute the first step in investigating RNA editing. This review summarizes modern bioinformatics approaches and related resources for RNA editing research.

Identification of Species-Specific Components between Hanwoo and Holstein Meat (한우 및 홀스타인육의 품종간 특이성분의 검색에 관한 연구)

  • Sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) of muscles extracted with distilled water, saline solution, SDS or Trition X-100 showed simular protein patterns between Hanwoo and Holstein meat, indicating that SDS-PAGE technique may not be useful for the identification between Hanwoo and Holstein meat. Lectine blot analysis of muscle extracted with distilled water demonstrated that Hanwoo and Holstein meat had similar affinities for concanavalin A (Con A), ricinus communis agglutinin (RCA-120), ulex europaeus agglutinin (UEA-1) or peanut agglutinin (PNA) lectins. However, approximately 32.1 kDa component of Hanwoo meat showed high affinity for dolichos biflorus agglutinin (DBA) lectin. On the contrary, high molecular weight components of Holstein meat had the specific affinity for wheat germ agglutinin (WGA) lectin. Hanwoo meat-specific components were observed by lectin staining of heat-denatured meat at 100$^{\circ}C$ for 30 sec. Also, the component of heat-denatured meat at 100$^{\circ}C$ for 30 sec, which was slightly smaller than Hanwoo meat-specific component, was concentrated specifically in Holstein meat.

  • The drought is one of the extreme natural disasters observed in any climate zone and it is due to the deficiency in moisture. The flash drought is identified recently as a subdivision of drought and it is an extreme event distinguished by sudden onset and rapid intensification of drought conditions with severe impacts. The main cause for the flash drought is coupled situation due to precipitation deficit and high evapotranspiration. Hence, heat waves plays major role in identification of flash drought. Therefore, this study focused on identifying changes in distribution of heat waves for Korean Peninsula. The daily maximum and minimum temperature data were used in this study. The heat wave, heat wave intensity and heat wave intensity index were derived. The results of the study would be an input for the future studies on identification of flash drought in Korean Peninsula.

  • Korean species of Atypophthalmus Brunetti, 1911 and Thrypticomyia Skuse, 1890 crane flies (Diptera: Limoniidae) were taxonomically revised. New findings of Achyrolimonia Alexander, 1965, Helius Lepeletier and Serville, 1828 (Latreille et al., 1828) and Limonia Meigen, 1803 were discussed. Genera Atypophthalmus and Thrypticomyia, species Helius(Helius) flavus(Walker, 1856) were newly recorded for the Korean Peninsula, and Achyrolimonia neonebulosa (Alexander, 1924) and Limonia juvenca Alexander, 1935 were new records for North Korea. Male terminalia of L. episema Alexander, 1924 were described with detailed photographs. Identification keys, re-descriptions and illustrations of both sexes from all species in Korea were presented.

  • the relationships between rainfall and runoff are analyzed statistically and modelled using discrete linear transfer function, which can be shown with the relations between input and output in hydrologic system. The procedures of identification, estimation and diagnostic checking of model are proposed, and the suitabilith of assume model is determined by the statistics used in time series analysis.
