Acorenone, a diterpene isolated from Acorus gramineus, showed strong resistance inhibitory activity against multi-drug resistant microorganisms such as Staphylococcus aureus SA2, which has resistance to 10 usual antibiotics including chloramphenicol (Cm). At the level of $5\;{\mu}g/ml$ when combined with $50\;{\mu}g/ml$ of Cm. Bacterial resistance to Cm is due to the presence in resistant bacteria of an enzyme, chloramphenicol acetyltransferase (CAT), which catalyses the acetyl-CoA dependent acetylation of the antibiotic at C-3 hydroxyl group. To elucidate the mechanism of resistant inhibitory effect, the acorenone which had the strongest resistant inhibitory activity, was investigated on the CAT assay. As the result, the combination of Cm and acorenone showed the strongest inhibitory activity on CAT as noncompetitive and dose dependent manner.
This study was conducted to establish the efficient screening method for resistant cabbage to Fusarium wilt caused by Fusarium oxysporum f. sp. conglutinans. The resistance degrees of nine commercial cabbage cultivars to the disease were evaluated. Among them, five cultivars (YR-honam, Ogane, Greenhot, Redmat, and Ccoccoma) showing different resistance to the fungus were selected. Then development of Fusarium wilt of the cultivars according to several conditions including root wounding, dipping period of roots in spore suspension, inoculum concentration, and incubation temperature to develop the disease was investigated. Highly resistant cultivars such as 'YR-honam' and 'Ogane' hardly showed change of resistance to the disease by root wounding, dipping period, and inoculum concentration, while disease severity of Fusarium wilt on the cultivars was changed with incubation temperatures ($20^{\circ}C$, $25^{\circ}C$ and $30^{\circ}C$). When the cabbage cultivars were incubated at $25^{\circ}C$, they represented the most difference of resistance and susceptibility to Fusarium wilt. From above results, we suggest that an efficient screening method for resistant cabbage to F. oxysporum f. sp. conglutinans is to dip the non-cut roots of 14-day-old seedlings in spore suspension of $1{\times}10^7$ conidia/ml for 0.5 hr and to transplant the seedlings to plastic pots with a fertilized soil, and then to cultivate the plants in a growth chamber at $25^{\circ}C$ for 3 weeks to develop Fusarium wilt.

This study was carried out to establish the simple mass-screening methods for resistant tomato to Fusarium wilt caused by Fusarium oxysporum f. sp. lycopersici (FOL). Root dip inoculation method has been used in many studies on the resistance of tomato to disease. On the other hand, in mass-screening for resistant tomato to Fusarium wilt, the inoculation method is time-consuming and laborious procedure. Disease development of two FOL isolates on two cultivars of tomato according to inoculation method including root dip, tip and scalpel methods were investigated. In compatible interaction, tomato seedlings of each cultivar inoculated by tip method showed the lower and more variable disease severities than by root dip method. Whereas the seedlings by scalpel method represented clear resistant and susceptible responses to Fusarium wilt as root dip method. The resistance degree of each cultivar inoculated with FOL isolates by scalpel method was hardly affected by the tested incubation temperature and inoculum concentration. On the basis of the results, we suggest scalpel inoculation method as an efficient mass-screening method for resistant of tomato cultivars to Fusarium wilt. Roots of tomato seedlings at two-leaf stage grown in plastic cell tray were injured with scalpel and then spore suspension (more than $1{\times}10^7\;conidia{\cdot}mL^{-1}$) of FOL was poured directly on the roots. The infected plants were cultivated in a growth room at $25-30^{\circ}C$ for 4 weeks with 12-hours light a day.
The soilborne fungus Fusarium oxysporum f. sp. lilii (Fol) is a serious threat to all lily cultivars, especially infecting bulbs and flowers. It has become increasingly important to develop varieties resistant against the bulb rot disease. Genetic diversity of cultivars and reliable screening methods are required for this purpose. Here, an efficient in vitro screening system for evaluating resistance to Fol in 38 in vitro-grown lily plants was investigated. Various factors including culture conditions of Fol, inoculum density, appropriate plant materials, inoculation method and duration, and incubation period of plant materials after inoculation were combined to optimize the screening method. As a result, we suggest optimal conditions for an in vitro screening system for the selection of Fol-resistant lily cultivars as follows. Fol was grown on potato dextrose agar (PDA) medium for 6 days at $25^{\circ}C$ in darkness and used as working inoculation. Spore suspensions were prepared (inoculum density: $1.0{\times}10^4$$spores{\cdot}mL^{-1}$), and then leaf segments $1.5{\times}2.0cm^2$ were inoculated by dipping for 22 hours at $25^{\circ}C$ in dark. Later, leaves were cultured on 0.6% agar plates at $25^{\circ}C$ and 50% humidity with a photoperiod of 16 hours light/8 hours dark (fluence rate of $40{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$) to examine the progress of bulb rot. After 7 days, disease levels were classified into indices 1 (no symptom) to 6 (serious bulb rot). Soil inoculation of Fol carried out with resistant or susceptible lily cultivars that had been selected through in vitro screening confirmed the reproducibility of results. Therefore, the in vitro screening method established in this study is efficient and reliable for selection of lily cultivars resistant against bulb rot disease.
Susceptibilies of tow-spotted spider mite (Tetranychus urticae Koch) collected at 6 locations were assessed to 15 acaricides. The response to acaricides were almost similar in female adults and eggs. There were considerable difference in susceptibility depending on the acaricide treated and the region from which the population was collected. The population showing resistance ratio of more than 20 with respect to certain acaricide was regarded as a resistant population to the acaricide. The resistant populations in terms of female adult were as follows: Suwon population to azocyclotin, cyhexatin, and fenbutatin-oxide; Taejon population to dicofol and fenbutatin-oxide; Chongju population to dicofoll Chinju populatin to cyhexatin, dicofol, and fenbutatin-oxide. The resistant populations in terms of egg were as follows; Suwon population to bifenthrin, clofentezine, hexythiazox, and tetradifon; Kunwi, Chongju, and Kwangju populations to bifenthrin; Taejon population to amitraz and bifenthrin; Taejon populatin to amitraz and bifenthrin; Chinju population to amitraz, bifenthrin, clofentezine, dicofol, and tetradifon. However, the female adults and eggs of all field populations were susceptible to abamectin, chlorfenson, and fenpyroximate. This tendency was also reported previously in the susceptible strain from laboratory.

Root-dipping inoculation method has been used to investigate resistance of radish plants to Fusarium wilt. However, the method requires a lot of labor and time because of complicate procedure. This study was conducted to establish a simple and effective mass-screening method for resistant radish to Fusarium wilt. Radish seedlings of susceptible and resistant cultivars were used to investigate wounding method by scalpel, inoculum concentration, and pathogen-inoculated growth stage of seedlings. We established an efficient mass-screening method based on our results as following: Roots of 14-day-old seedlings of radish are cut with a scalpel at a $90^{\circ}$ angle to a 2 cm-depth at a 1 cm-distance from main stem and then inoculated by pouring with a 10 ml-aliquot of a fungal spore suspension ($1.0{\times}10^7conidia/ml$) on soil. The inoculated plants are cultivated in a growth room at $25^{\circ}C$ for about 4 weeks with 12-hour light a day. The proposed screening method enables to effectively select resistant from mass radish plants cultivars to Fusarium wilt.

Mycobacterium tuberculosis is a pathogen responsible for 2-3 million deaths every year worldwide. The emergence of drug-resistant and multidrug-resistant tuberculosis has increased the need to identify new antituberculosis targets. Acetohydroxy acid synthase, (AHAS, EC 2.2.1.6), an enzyme involved in branched-chain amino acid synthesis, has recently been identified as a potential anti-tuberculosis target. To assist in the search for new inhibitors and “receptor-based” design of effective inhibitors of tubercular AHAS (TbAHAS), we constructed four different structural models of TbAHAS and used one of the models as a target for virtual screening of potential inhibitors. The quality of each model was assessed stereochemically by PROCHECK and found to be reliable. Up to 89% of the amino acid residues in the structural models were located in the most favored regions of the Ramachandran plot, which indicates that the conformation of each residue in the models is good. In the models, residues at the herbicide-binding site were highly conserved across 39 AHAS sequences. The binding mode of TbAHAS with a sulfonylurea herbicide was characterized by 32 hydrophobic interactions, the majority of which were contributed by residue Trp516. The model based on the highest resolution X-ray structure of yeast AHAS was used as the target for virtual screening of a chemical database containing 8300 molecules with a heterocyclic ring. We developed a short list of molecules that were predicted to bind with high scores to TbAHAS in a conformation similar to that of sulfonylurea derivatives. Five sulfonylurea herbicides that were calculated to efficiently bind TbAHAS were experimentally verified and found to inhibit enzyme activity at micromolar concentrations. The data suggest that this time-saving and costeffective computational approach can be used to discover new TbAHAS inhibitors. The list of chemicals studied in this work is supplied to facilitate independent experimental verification of the computational approach.
Thirty two rice cultivars which have been cultivated or used as breeding materials in Korea were tested for screening rice cultivars resistant to leaf blast at adult-plant stages in the blast nursery hill plots. When compared on the basis of disease severities in individual leaves at different growth stages of rice plants under natural field infection, the 16 indica-japonica hybrids tested were highly resistant but the 16 japonica cultivars tested showed various degrees of resistance to leaf blast. With aging of rice plants, the quantitative levels of resistance to leaf blast increased in all the cultivars, although the levels of resistance to leaf blast varied according to rice genotypes. The leaf position of rice plants in which changed from susceptible to resistant reactions varied also with rice genotypes. The susceptible reactions of the rice cultivars to Pyricularia oryzae were distinctly changed to a resistant reaction on upper leaves of rice plants. The rice cultivars, in which the quantitative level of resistance to leaf blast was higher, were resistant on the lower leaves of rice plants. The cultivars Akibare, Palkeum, Jinheung, Olchal, Dobong and Ginga which drastically decreased blast infection at late growth stage were evaluated as adult-plant-resistant to leaf blast in the field. The adult-plant-resistant cultivars became resistant to leaf blast, both qualitatively and quantitatively, as rice plants matured.

Conyza canadensis is the weed species which most frequently develops resistance to glyphosate in many agricultural crop fields. The continuous use of glyphosate has resulted in the spontaneous occurrences of resistant biotypes. This research was conducted to investigate the response of suspected C. canadensis biotypes to glyphosate. Seeds of C. canadensis were collected from 18 sites in tangerine orchards in Jeju province of Korea. In the preliminary screening, 6 resistant and 12 susceptible biotypes were found at the recommended glyphosate rate ($3.28kga.i.ha^{-1}$). The susceptible biotypes were completely killed at the field application rate whereas the resistant biotypes were initially injured but recovered 14 days after glyphosate application. This is the first case of glyphosate resistance found in Korea despite the national ban on genetically modified glyphosate tolerant crops cultivation. Extended monitoring should be conducted to understand how widely spread the glyphosate resistant C. canadensis is and to estimate the severity of this weed problem in the tangerine orchards of Korea.
Drug resistance is one of the most significant impediments to successful chemotherapy of cancer. Multidrug-resistance (MDR) is characterized by decreased cellular sensitivity to anticancer agents due to the overexpression of P-glycoprotein. By employing a resistant subline of HCT15 to adriamycin (CL02), we undertook the screening for agents which were effective to multidrug-resistant cancer cells. As a result, a myxobacterial strain JW150 was selected for study since an activity against CL02 cells was discovered in the strain. Cytotoxicity-guided fractionation of the culture broth led to the isolation of cystothiazole A and melithiazole F. The producing organism was identified as Myxococcus stipitatus by taxonomic comparison with type strains of Myxococcus sp. as well as its morphological and physiological characteristics. Cystothiazole A and melithiazole F demonstrated potent cytotoxicity against certain human cancer cells with $IC_{50}$ values ranging from 0.03~ $0.72{\mu}{\textrm{g}}$/ml. Both compounds were interestingly as active against drug-resistant sublines CL02 and CP70 as against the corresponding parental cells.
