Determination of metabolites of prostanozol in human urine by LC/ESI/MS and GC/TOF-MS (LC/ESI/MS와 GC/TOF-MS를 이용한 인체뇨시료에서의 Prostanozol 대사체 검출)

  • This research examined prostanozol and its metabolites in urine of women who took the medicine (prostanozol). Prostanozol and its metabolites were successfully separated and detected by using LC/ESI/MS and GC/TOF-MS. Mass spectrum of LC/ESI/MS estimated molecular weight of Prostanozol and its metabolites and that of GC/TOF-MS verified them. For M1, carbon number 17 of Prostanozol substituted to a keto group and it is called 17-keto-Prostanozol. M2 turned out to be hydroxy-17-keto-Prostanozol. It came from substitution of one hydroxyl group of pyrazole nucleus and A-ring of M1. Substitution of one hydroxyl group of B-ring or C-ring became M3, hydroxy-17-keto-Prostanozol. M4 was found to be a hydroxy-17-keto-Prostsnozol transposed from one hydroxyl group to a D-ring. M5 has a hydroxyl group of carbon number 17. One hydroxyl group is substituted from B-ring or C-ring and it is assumed to be hydroxy-17-hydroxy-Prostanozol. M6 was turned out to be dihydroxy-17-keto-Prostanozol transposed from one hydroxyl group to pyrazole nucleus or A-ring and to B-ring or C-ring. Like M6, M7 has a keto group at carbon number 17 and was identified as dihydroxy-17-keto-Prostanozol. M7 has one hydroxyl group at pyrazole nucleus or A-ring and also at D-ring. At last M8 was found to be dihydroxy-17-hydroxy-Prostanozol. Pyrazole nucleus or A-ring has got one hydroxyl group and other rings were substituted to another hydroxyl group. From above, M5, M7 and M8 were verified as new metabolites that were not discovered yet. Prostanozol and all of the 8 metabolites formed glucuronic conjugates as a result of conjugation reaction test in human body. Some of 8 metabolites were excreted without forming conjugates. Particularly M6 and M7 were excreted as sulfate conjugates.

Evaluation of Odors and Odorous Compounds from Liquid Animal Manure Treated with Different Methods and Their Application to Soils (액상 가축분뇨의 처리 및 토양환원에 따른 악취 및 악취물질의 평가)

  • To comply with stricter regulations provoked by increasing odor nuisance, it is imperative to practice effective odor control for sustainable livestock production. This study was conducted to assess odor and odorous compounds emitted from liquid animal manure with different treatment methods such as Fresh Manure(without treatment, FM), Anaerobic Digestion(AD) and Thermophilic Aerobic Digestion(TAD) and their application to soil. Air samples were collected at the headspace of liquid manure, upland and paddy soil, and analyzed for odor intensity and offensiveness using an olfactometry; odor concentration index using odor analyser; nitrogen-containing compound such as ammonia(NH3) using fluorescence method; and sulfur containing compounds such as hydrogen sulfide(H2S), methyl mercaptan(MeSH), dimethyl sulfide(DMS) and dimethyl disulfide(DMDS) using gas chromatography-pulsed flame photometric detector, respectively. Odor intensity, offensiveness and concentration index from TAD liquid manure was statistically lower than those from FM and AD(p<0.01). Mean concentrations of H2S, MeSH, DMS, DMDS and NH3 were 65.93ppb, 18.55ppb, 5.26ppb, 0.33ppb and 10.57ppm for liquid manure with AD; and 5.15ppb, 0.97ppb, 0.80ppb, 0.56ppb and 1.34ppm for liquid manure with TAD, respectively. More than 60% of malodorous compounds related to nitrogen and sulfur were removed by heterotrophic microorganisms during TAD treatment. When liquid manure was applied onto upland and paddy soil, NH3 removal efficiencies ranged from 51 to 94% and 22 to 91% for AD and TAD liquid manure, respectively. The above results show that liquid manure with TAD is superior to AD and FM with respect to the odor reduction and odor problem caused by land applied liquid manure is directly related to the degree of odor generated by the manure treatment method.

Analysis of the Effect of Heat Island on the Administrative District Unit in Seoul Using LANDSAT Image (LANDSAT영상을 이용한 서울시 행정구역 단위의 열섬효과 분석)

  • The increase in the rate of industrialization due to urbanization has caused the Urban Heat Island phenomenon where the temperature of the city is higher than the surrounding area, and its intensity is increasing with climate change. Among the cities where heat island phenomenon occurs, Seoul city has different degree of urbanization, green area ratio, energy consumption, and population density in each administrative district, and as a result, the strength of heat island is also different. So It is necessary to analyze the difference of Urban Heat Island Intensity by administrative district and the cause. In this study, the UHI intensity of the administrative gu and the administrative dong were extracted from the Seoul metropolitan area and the differences among the administrative districts were examined. and linear regression analysis were conducted with The variables included in the three categories(weather condition, anthropogenic heat generation, and land use characteristics) to investigate the cause of the difference in heat UHI intensity in each administrative district. As a result of analysis, UHI Intensity was found to be different according to the characteristics of administrative gu, administrative dong, and surrounding environment. The difference in administrative dong was larger than gu unit, and the UHI Intensity of gu and the UHI Intensity distribution of dongs belonging to the gu were also different. Linear regression analysis showed that there was a difference in heat island development intensity according to the average wind speed, development degree, Soil Adjusted Vegetation Index (SAVI), Normalized Difference Built-up Index (NDBI) value. Among them, the SAVI and NDBI showed a difference in value up to the dong unit and The creation of a wind route environment for the mitigation of the heat island phenomenon is necessary for the administrative dong unit level. Therefore, it is considered that projects for mitigating heat island phenomenon such as land cover improvement plan, wind route improvement plan, and green wall surface plan for development area need to consider administrative dongs belonging to the gu rather than just considering the difference of administrative gu units. The results of this study are expected to provide the directions for urban thermal environment design and policy development in the future by deriving the necessity of analysis unit and the factors to be considered for the administrative city unit to mitigate the urban heat island phenomenon.

Changes of Protein Profiles in Cheonggukjang during the Fermentation Period (전통 청국장의 발효 기간 동안 변화하는 수용성 단백질 개요)

  • The fermented soybean product, cheonggukjang, is favored by many people, partly due to its bio-functional ingredients. Since the fermentation process of cheonggukjang is mediated by enzymes, including proteases, produced by microbes, analysis of the proteome profile changes in cheonggukjang during fermentation would provide us with valuable information for fermentation optimization, as well as a better understanding of the formation mechanisms of the bio-functional substances. The soluble proteins from cheonggukjang were prepared by a phenol/chloroform extraction method, in order to remove interfering molecules for high resolution 2-D gel analysis. Proteomic analysis of the cheonggukjang different fermentation periods suggested that most of the soluble soy proteins were degraded into smaller forms within 20hr, and many microbial proteins, such as mucilage proteins, dominated the soluble protein fraction. The proteomic profile of cheonggukjang was very different from natto, in terms of the 2-D gel protein profile. Among the separated protein spots on the 2-D gels, 50 proteins from each gel were analyzed by MALDI-TOF MS and PMF for protein identification. Due to database limitations with regard to soy proteins and microbial proteins, identification of the changed proteins during fermentation was restricted to 9 proteins for cheonggukjang and 15 for natto. From de novo sequencing of the proteins by a tandem MS/MS, as well as by database searches using BLASTP, a limited number of proteins were identified with low reliability. However, the 2-D gel analysis of proteins, including protein preparation methods, remains a valuable tool to analyze complex mixtures of proteins entirely. Also, for intensive mass spectrometric analysis, it is also advisable to focus on a few of the interestingly changed proteins in cheonggukjang.

Isolation and Structure Identification of Photosensitizer from Perilla frutescens Leaves Which Induces Apoptosis in U937 (들깻잎(Perilla frutescens)으로부터 U937 세포에 apoptosis를 유도하는 광과민성 물질의 분리 및 구조동정)

  • In this study, we tried to separate the photosensitizer that induces apoptosis of leukemia cells (U937) from perilla leaves. Perilla leaves (Perilla frutescens Britt var. japonica Hara) are a popular vegetable in Korea, being rich in vitamins (A and E), GABA, and minerals. Dried perilla leaves were extracted with methanol to separate the photosensitizer by various chromatographic techniques. The structure of the isolated compound (PL9443) was identified by 1D-NMR, 2D-NMR, and FAB-mass spectroscopy. Absorbance of the UV-Vis spectrum was highest at 410 nm and was confirmed by the 330, 410, and 668 nm. PL9443 compound was determined to be pheophorbide, an ethyl ester having a molecular weight of 620. It was identified as a derivative compound of pheophorbide structure when magnesium comes away from a porphyrin ring. Observation of morphological changes in U937 cells following cell death induced by treated PL9443 compound revealed representative phenomena of apoptosis only in light irradiation conditions (apoptotic body, vesicle formation). Results from examining the cytotoxicity of PL9443 substance against U937 cells showed that inhibition rates of the cell growth were 99.9% with the concentration of 0.32 nM PL9443. Also, the caspase-3/7 activity was 99% against U937 cells with the concentration of 0.08 nM of PL9443 substance. The result of the electrophoresis was that a DNA ladder was formed by the PL9443. The PL9443 compound is a promising lead compound as a photosensitizer for photodynamic therapy of cancer.

The Effect of Exercise Training on Aβ-42, BDNF, GLUT-1 and HSP-70 Proteins in a NSE/ APPsw-transgenic Model for Alzheimer's Disease. (지구성 운동이 NSE/APPsw 알츠하이머 질환 생쥐의 인지능력, Aβ-42, BDNF, GLUT-1과 HSP-70 단백질 발현에 미치는 영향)

  • Mutations in the APP gene lead to enhanced cleavage by ${\beta}-$ and ${\gamma}-secretase$, and increased $A{\beta}$ formation, which are closely associated with Alzheimer's disease (AD)-like neuropathological changes. Recent studies have shown that exercise training can ameliorate pathogenic phenotypes ($A{\beta}-42$, BDNF, GLUT-1 and HSP70) in experimental models of Alzheimer's disease. Here, we have used NSE/APPsw transgenic mice to investigate directly whether exercise training ameliorates pathogenic phenotypes within Alzheimer's brains. Sixteen weeks of exercise training resulted in a reduction of $A{\beta}-42$ peptides and also facilitated improvement of cognitive function. Furthermore, GLUT -1 and BDNF proteins produced by exercise training may protect brain neurons by inducing the concomitant expression of genes that encode proteins (HSP-70) which suppress stress induced neuron cell damages from APPsw transgenic mice. Thus, the improved cognitive function by exercise training may be mechanistically linked to a reduction of $A{\beta}-42$ peptides, possibly via activation of BDNF, GLUT-1, and HSP-70 proteins. On the basis of the evidences presented in this study, exercise training may represent a practical therapeutic management strategy for human subjects suffering from Alzheimer's disease.

Anti-wrinkle Effect of Cambodian Phellinus linteus Extracts (캄보디안 상황버섯 추출물의 주름개선 효과 연구)

  • The skin of human is constantly being exposed to environmental irritants such as ultraviolet, smoke and chemicals. These irritants cause free radicals and reactive oxygen species which leave serious damages on the cells of skin. The water and ethanol extracts of Cambodian Phellinus linteus were investigated for the activities of anti-lipid peroxidation and anti-wrinkle effects to apply as a functional ingredient for cosmetic products. As the result of evaluation of liquid oxidation rate by add $Fe^{2+}$ and $Cu^{2+}$ to Cambodian Phellinus linteus extracts, Cambodian Phellinus linteus ethanol extracts were higher than Cambodian Phellinus linteus water extracts in the chealting ability of $Fe^{2+}$ and $Cu^{2+}$. The Cambodian Phellinus linteus ethanol extracts exhibited that anti-lipid peroxidation higher than butylated hydroxytoluene (BHT) at the concentration of 0.1 mg/ml, 0.5 mg/ml and 1 mg/ml. Cambodian Phellinus linteus water and ethanol extracts showed a higher inhibitory effect on $Fe^{2+}$-induced lipid peroxidation compared to $Cu^{2+}$-induced lipid peroxidation. In the case of anti-wrinkle effect, the elastase inhibition activity of Cambodian Phellinus linteus ethanol extracts was 50.7%, and it is higher than urosolic acid at the concentration of 0.01 mg/ml. Also, in collagenase inhibition activity, Cambodian Phellinus linteus water extract showed low effect, but Cambodian Phellinus linteus ethanol extract was about 50% at a 0.1 mg/ml. concentration. These results proved that Cambodian Phellinus linteus had anti-lipid peroxidation and anti-wrinkle effect. Therefore, Cambodian Phellinus linteus could be useful as an anti-wrinkle cosmetic ingredient.

A Study of Recognition of Housewives on Wedding Food in Seoul Area (서울지역 주부들의 혼례음식에 대한 인지도 조사)

  • This study was carried out to survey the degree of the recognition and necessity of wedding food to include $Ph{\grave{a}}ebak$ and Ibaji foods prepared by bride's parents and sent to the bridgroom's. The participants of the survey were 293 housewives of 20 years old or more residing in Seoul area. The questionnaires used in this research consisted of 10 questions. The findings could be summarized as follows: 1. As to the degree of the recognition of $Ph{\grave{a}}ebak$ food, 55.2 percent of the participants perceive it as 'the food sent to the bridegroomvs house to be used when the newly-weds greet groom's relatives, whereas 35.8 percent regarded the practice as 'a traditional custom from the ancient times,' which is, in fact, correct and exact. As to its importance, 10.2 percent regarded $Ph{\grave{a}}ebak$ food very important, while 48.5 percent, 'important,' and 11.9 percent, 'not important.' 2. As for the items for $Ph{\grave{a}}ebak$ dishes, 77.5 percent answered their preferences for 'jujube prop-nut,' followed by 'ricecake,' 'liquor' and 'Korean cookies,' in descending order. 3. As for the meaning of Ibaji dishes, 48.2% understand them as 'a set of dishes prepared by the bride's parents to be sent to the bridegroom's'; and as for their willingness or necessity of sending the Ibaji dishes, 15.5 percent indicated that they might prepare them, whereas 78.7 percent said that they would prepare them if the situation required. On the other hand 5.8 percent did not feel the necessity. The overall result is that most females surveyed recognized the two types of food important and were inclined to prepare them when the occasion demanded.

Modification of WASP5 for Ungauged Watershed Management and Its Application (미계측 유역관리를 위한 WASP5 모형의 개선 및 적용성 검토)

  • This study was carried out to develop a water quality simulation model for the evaluation of an ungauged watershed. For this purpose, the WASP5 model was selected and modified. The model consists of three sub-models, LOAD-M, DYN-M, and EUT-M. LOAD-M, an empirical model, estimates runoff loadings using point and non-point source data of villages. The Geum River Estuary watershed was selected to calibrate and verify the Modified-WASP5. The LOAD-M model was established using field data of water quality and quantity at the gauging stations of the watershed and was applied to the ungauged watersheds, taking the watershed properties into consideration. The result of water quality simulation using Modified-WASP5 shows that the observed average BOD data from Gongju and Ganggyeong were 2.6 mg/L and 2.8 mg/L, and the simulated data were 2.5 mg/L and 2.4 mg/L, respectively. Generally, simulation results were in good agreement with the observed data. This study focused on formulating an integrated model for evaluating ungauged watersheds. Even though simulation results varied slightly due to limited availability of data, the model developed in this study would be a useful tool for the assessment and management of ungauged watersheds.

Studies on Biological Nitrogen Fixation -III. Influences of organic matter sources, kinds and amount of fertilizer nitrogen on the changes of biological N2-fixation and kjeldahl nitrogen under dark and light condition in submerged paddy soil (생물학적(生物學的) 질소고정(窒素固定)에 관(關)한 연구(硏究) -III. 담수토양(湛水土壤)에서 유기물종류(有機物種類), 질소비종(窒素肥種)과 시비량(施肥量)을 달리했을때 광합성(光合成) 및 타양성질소고정력(他養性窒素固定力)에 미치는 영향(影響))

  • A green house experiment was conducted to find out the differences in the amount of biologically fixed nitrogen and kjeldahl nitrogen on the different soil texture, kinds and amounts of fertilizer nitrogen under light (photosynthetic $N_2$-fixation) and dark (heterotrophic $N_2$-fixation) condition in submerged paddy soil. The reults obtained were summarized as follows; 1. The amount of biologically fixed nitrogen per mg carbon from different organic matter was obtained as 0.13 mg in glucose, 0.09 mg in rice straw, and 0.07 mg in refused mushroom compost and barley straw under 60 days of incubation. 2. Nitrogen fixing activities were decreased with increase of fertilizer nitrogen and those tendency was pronounced more in sandy soil with application of urea than that of ammonium sulfate. 3. The application of ammonium sulfate in sandy soil under light condition was increased the photosynthetic $N_2$-fixation and the applied urea was remarkably reduced the heterotrophic $N_2$-fixation in sandy soil. The proportion of biologically fixed total nitrogen after experiment in sandy soil was obtained as 25% for dark(heterotrophic $N_2$-fixation) and 75% for light (photosynthetic $N_2$-fixation) condition. On the other hand, very similar biological $N_2$-fixing tendency was obtained between kinds of nitrogen fertilizer and two light condition in clayey soil. 4. The kjeldahl nitrogen was remarkably decreased after experiment under dark condition with application of urea than that of light condition with ammonium sulfate, and no remarkable decreasing tendency was obtained in clayey soil between two kinds of fertilizer nitrogen. 5. The high significant positive correlationship was obtained between calculated biological nitrogen fixation by acetylene reducing activity and kjeldahl nitrogen after experiment under light (y=0.8488X-5.9632, $r=0.9928^{**}$, n=21) and dark (y=0.8795X-7.1056, $r=0.9782^{**}$, n=21) condition. In this experiment condition, conversion factors of 6:1 was obtained from biological nitrogen fixation to soil nitrogen.
