Aquaporin 4 expression is downregulated in large bovine ovarian follicles

  • Aquaporin channels (AQPs) are known to play an important role in the development of ovarian follicles through their function in water transport pathways. Compared to other AQPs, research on the role of AQP4 in female reproductive physiology, particularly in cattle, remains limited. In our previous study, gene chip microarray data showed a downregulation of AQP4 in bovine cystic follicles. This study was performed to validate the AQP4 expression level at the protein level in bovine follicles using immunohistochemistry, Western blotting, and immunoprecipitation assays. Immunostaining data showed that AQP4 was expressed in granulosa and theca cells of bovine ovarian follicles. The ovarian follicles were classified according to size as small (< 10 mm) or large (> 25 mm) in diameter. Consistent with earlier microarray data, semi-quantitative PCR data showed a decrease in AQP4 mRNA expression in large follicles. Western blot analysis showed a downregulation of the AQP4 protein in large follicles. In addition, AQP4 was immunoprecipitated and blotted with anti-AQP4 antibody in small and large follicles. Accordingly, AQP4 exhibited a low expression in large follicles. These results show that AQP4 is downregulated in bovine ovarian large follicles, suggesting that the downregulation of AQP4 expression may interfere with follicular water transport, leading to bovine follicular cysts.

Estimation of Heading Date of Paddy Rice from Slanted View Images Using Deep Learning Classification Model

  • Estimation of heading date of paddy rice is laborious and time consuming. Therefore, automatic estimation of heading date of paddy rice is highly essential. In this experiment, deep learning classification models were used to classify two difference categories of rice (vegetative and reproductive stage) based on the panicle initiation of paddy field. Specifically, the dataset includes 444 slanted view images belonging to two categories and was then expanded to include 1,497 images via IMGAUG data augmentation technique. We adopt two transfer learning strategies: (First, used transferring model weights already trained on ImageNet to six classification network models: VGGNet, ResNet, DenseNet, InceptionV3, Xception and MobileNet, Second, fine-tuned some layers of the network according to our dataset). After training the CNN model, we used several evaluation metrics commonly used for classification tasks, including Accuracy, Precision, Recall, and F1-score. In addition, GradCAM was used to generate visual explanations for each image patch. Experimental results showed that the InceptionV3 is the best performing model in terms of the accuracy, average recall, precision, and F1-score. The fine-tuned InceptionV3 model achieved an overall classification accuracy of 0.95 with a high F1-score of 0.95. Our CNN model also represented the change of rice heading date under different date of transplanting. This study demonstrated that image based deep learning model can reliably be used as an automatic monitoring system to detect the heading date of rice crops using CCTV camera.

Proteomic Analysis of Differentially Expressed Proteins in Bovine Endometrium with Endometritis

  • Endometritis is one of the primary reasons for reproductive failure. In order to investigate endometritis-associated marker proteins, proteomic analysis was performed on bovine endometrium with endometritis. In bovine endometritis, desmin, $\alpha$-actin-2, heat-shock protein (HSP) 27, peroxiredoxin-6, luteinizing hormone receptor isoform 1, collectin-43 precursor, deoxyribonuclease-I (DNase-I), and MHC class I heavy chain (MHC-Ih) were up-regulated. In contrast, transferrin, interleukin-2 precursor, hemoglobin $\beta$ subunit, and potassium channel tetramerisation domaincontaining 11 (KCTD11) were down-regulated in comparison to normal endometrium. The proteomic results were validated by semiquantitative-PCR and immunoblot analysis. The mRNA levels of desmin, transferrin, $\alpha$-actin-2, HSP27, KCTD11, and MHC-Ih were up-regulated by over 1.5-fold, and showed a pattern similar to their proteomic profiles. Desmin and $\alpha$-actin-2 protein showed positive correlations between proteomic analysis and immunoblot analysis. These results suggest that desmin and $\alpha$-actin-2 may play important roles in endometritis-related function, and could be useful markers for the diagnosis of bovine endometritis.

Influence of aqueous extract of Annona muricata leaves in Tris-egg yolk extender on storage of spermatozoa from West African Dwarf goat (Capra hircus)

  • Background: Because oxidative stress can induce decreased quality of caprine semen during the storage, there has been limitation for the use of stored semen in the assisted reproductive technologies. The present study, therefore, assesses the potential of Annona muricata (A. muricata) to reduce semen storage associated-damages. Methods: Semen was collected by electro-ejaculation from ten bucks, and extended with Tris-egg yolk (TEY) supplemented with A. muricata leaf aqueous extract (SAE) at 20 (SAE20), 40 (SAE40), and 80 (SAE80) ㎍/mL. Sperm variables including motility, motion characteristics, viability, membrane functionality, and DNA integrity were assessed at different storage periods (6, 24, 48, and 72 hr). In addition, oxidative stress indicators in the extender supplemted with SAE were also assessed for each group. Results: By adding SAE at 80 ㎍/mL in TEY, the storage of goat buck semen was improved, resulting in reduced loss of sperm motility, viability, DNA fragmentation, and membrane integrity during chilled storage at 4℃ for up to 72 hr. In addition, enrichment of TEY extender with SAE significantly (p < 0.05) reduced malondialdehyde, an indicator of oxidative stress, compared to the negative control. Conclusions: Supplementation of SAE in TEY extender can reduce buck spermatozoa liquid storage associated damages due to oxidative stress.

  • In summary, progesterone is probably the only naturallly occurring progestational agent of any significance. Small amounts may be synthesized by the cells of the follicle in the provulatory swelling phase; however, the major production is by the corpus luteum cells of the ovary during the luteal phase of the cycle. It is constantly produced in small amounts by the adrenal gland and by the testis in the male. In the adrenal and the testis, it probably serves as the precursor for corticoids and androgens. It is transported in the blood by a specific binding protein and metabolized and conjugated in the liver into sodium pregnanediol glucuronide which also circulates in the blood. Approximately 20% is excreted in the urine as sodium pregnanediol glucronide; pregnanolone represents a minor metabolic product. The pregnanediol which is excreted in the bile is enzymically hydrolyzed by the gut so that the pregnanediol recovered in the feces is in the free form.

  • Impact of microsporidian infection and season on reproductive potential of Bombyx mori L. was investigated in the laboratory. Microsporidian infection significantly (P<0.01) reduced fecundity and hatching and increased sterility and mortality of eggs. Among the microsporidia, Nosema sp. 2 infected silkworm produced eggs with least fecundity and hatching percent as well as highest dead and sterile eggs followed by Nosema sp. 1 and N. bombycis. Microsporidia, in general, significantly reduced fecundity and hatching percent of eggs and increased number of dead and sterile eggs in all the three seasons except N. bombycis in July - August (S3) and Nosema sp. 2 in January February (S1). Since, seed production is the anchor sheet of mulberry sericulture, coefficient of egg lying is considered as an important aspect and the industry quite often facing shortage of disease free layings. The present study indicates that B. mori is more susceptible to microsporidia during S2 followed by S3 and S1 and Nosema sp2. is most virulent followed by Nosema spl. and N. bombycis.

Assessment of Reproductive Health Risk of Polychlorinated Biphenyls by Monitoring the Expression of Claudius and Transepithelial Electrical Resistance in Mouse Sertoli Cells

  • Tight junctions (TJ) between adjacent Sertoli cells in testis are important for the formation of the blood testis barrier (BTB). In an effort to verify the reproductive health risk of endocrine-active chemicals (EACs), changes in the transepithelial electrical resistance (TER) and the expression of TJ genes were examined by co-planar polychlorinated biphenyl (PCB) treatment in cultured mouse Sertoli cells. Although the increase in TER of Sertoli cells was accelerated by 10 nM co-planar PCB, it was downregulated by 100 nM co-planar PCB. The expression of claudin-1 was downregulated by co-planar PCB in a concentration-dependent manner. On the contrary, the expression of claudin-1 was increased in the Sertoli cells by 10 nM co-planar PCB treatment. These results suggest that the structure and function of TJ may be targeted by co-planar PCB in Sertoli cells. Assessment of the structure and function of TJ in Sertoli cells might be useful for screening the reproductive health risk of EACs.

Impact of glycosylation on the unimpaired functions of the sperm

  • One of the key factors of early development is the specification of competence between the oocyte and the sperm, which occurs during gametogenesis. However, the starting point, growth, and maturation for acquiring competence during spermatogenesis and oogenesis in mammals are very different. Spermatogenesis includes spermiogenesis, but such a metamorphosis is not observed during oogenesis. Glycosylation, a ubiquitous modification, is a preliminary requisite for distribution of the structural and functional components of spermatids for metamorphosis. In addition, glycosylation using epididymal or female genital secretory glycans is an important process for the sperm maturation, the acquisition of the potential for fertilization, and the acceleration of early embryo development. However, nonemzymatic unexpected covalent bonding of a carbohydrate and malglycosylation can result in falling fertility rates as shown in the diabetic male. So far, glycosylation during spermatogenesis and the dynamics of the plasma membrane in the process of capacitation and fertilization have been evaluated, and a powerful role of glycosylation in spermatogenesis and early development is also suggested by structural bioinformatics, functional genomics, and functional proteomics. Further understanding of glycosylation is needed to provide a better understanding of fertilization and embryo development and for the development of new diagnostic and therapeutic tools for infertility.

Effects of an Educational Program about Reproductive Health Promotion on Sexual Knowledge and Attitudes of University Students from Korea (생식건강증진 프로그램이 대학생의 성지식과 성태도에 미치는 효과)

  • Purpose: The purpose of this study was to examine the effects of an educational program about reproductive health promotion on sexual knowledge and attitudes of university students. Methods: The sample included 79 students of all grades from 3 universities in Korea. Data were collected from 8th to 11th of January, 2008 by using a structured questionnaire administered before and after the educational program. The data were analyzed using SPSS/WIN 12.0 program. Results: The students' scores on sexual knowledge increased, and attitudes toward sexuality changed at the completion of the educational program. The program was especially effective in 'physiology', 'contraception and abortion' and 'venereal disease and AIDS' among sexual knowledge, and in 'intercourse' and 'sexual morality' among sexual attitudes. Conclusion: The results of this study show that an educational program about reproductive health increases sexual knowledge and changes the sexual attitudes of university students.

Comparative Effects on Secretion of LH, FSH, Prolactin, and Testosterone by Chronic and Direct Hypothalamic Administration of Nonylphenol to Adult Male Rats

  • Nonylphenol (NP) is a widespread environmental pollutant that has been shown to exert both toxic and estrogenic effects on mammalian cells. As the effects of NP on the reproductive system of adult male vertebrates are virtually unknown, we investigated not only the changes of reproductive hormone secretion in serum after chronic exposure to NP but also, in order to identify the site of its action, the reproductive hormone secretion in serum 48 hours after microinfusion of NP within hypothalamic preoptic area (POA). In the chronic exposure, the luteinizing hormone (LH), follicle stimulating hormone (FSH), and testosterone in serum were decreased but prolactin (PRL) concentrations were increased. The LH, FSH, and testosterone in serum were decreased through the direct infusion of NP into POA, while there was no difference in mean serum prolactin between NP and control groups. These observations suggest that NP as endocrine disruptor has modulatory effects on hypothalamo-pituitary-gonadal axis and that the site of action of NP could be hypothalamic POA.
