  • Diagnosis and treatment of major reproductive failures were carried out for 836 dairy cows of 12 farms in Kimje area from May 1999 to May 2000. Reproductive failures were classified into four categories: uterine diseases, ovarian diseases, combination of uterine and ovarian diseases, and anestrus with corpus luteum (CL), based on the diagnosis by both rectal palpation and ultrasonography (SA 600, Medison, 5.OMHz transducer). 1. Of 102 cows examined, 15 cows were diagnosed as pregnant. Cows with real reproductive failures were therefore 87 cows and the rate of reproductive failures far all the cows of 12 farms was 10.4% (87/836). 2. Of 57 cows with reproductive failures, the cows with uterine diseases were 33 heads (37.9%), with ovarian diseases: 30 heads (34.5%), with combination form: 16 heads (18.4%), and with anestrus with CL: 8 heads (9.2%). 3. Of 87 cows with reproductive failures, 7 cows were slaughtered and 80 cows were treated by hormone or douche. The cows expressed estrus were 74 cows (92.5%, 74/80 heads) and 56 cows were pregnant (70.0%, 56/80 heads) following treatment. 4. The reproductive failures tended to be increased as the period proceeded from 30 days (2.3%, 2/87 heads) to over 120 days (56.3%, 49/87 heads) after parturition. 5. Associated other diseases to reproductive failures were foot diseases(5 cows), joint diseases (5 heads), urovagina (6 heads), and rectovaginal fistula (2 cows) and the rate of occurrence of associated diseases was 20.7% (18/87 heads). 6. No reproductive record was used in 7 farms of 12 farms and 5 farms had no play ground or not used for the cows. These results indicated that diagnosis fur reproductive failures by rectal palpation along with ultrasonography was more accurate than the conventional rectal palpation and brought better result of treatment for major types of reproductive failures. It was also indicated that non-uses of reproductive record and play ground were also important factors in occurrence of reproductive failures.

  • The effect of artificial oocyte activation (AOA) with a calcium ionophore on intracytoplasmic morphologically selected sperm injection (IMSI) was examined in patients with histories of repeated failed implantation attempts. Four singleton pregnancies and one twin pregnancy were obtained after embryos transfer (5/14, 35.7%). Therefore, AOA combined with IMSI can be considered an option for cycles with a fertilization defect and recurrent implantation failures.

  • Diagnosis on reproductive failures of dairy cows by ultrasonography was performed for 151 dairy cows. To diagnose types of reproductive failures, ultrasonography (SA 600, Medison, 5.0 MHz rectal linear transducer) was carried out in combination with rectal examination. Of 151 dairy cows, pregnant cows were 13 and the cows in normal estrual cycle were 40 cows, thereby the cows with reproductive failures were 98 cows. 1. Of 98 cows with reproductive failures, the cows with ovarian diseases were 34 cows (34.7%) and the cows with uterine diseases were 41 cows (41.8%). 2. The diameter of follicle in proestrus was 1.94 cm and it was longer than that of follicle in diestrus (p<0.05). 3. The mean size of corpus luteum of pregnant cows was bigger than that of corpus luteum in normal diestrus (p<0.05). 4. The length of cystic corpus luteum was 3.26 and the width of that was 1.91 cm. The length of corpus luteum tissue was 1.95 and the width of that was 1.91 cm excluding the size of cavity in corpus luteum. 5. The mean length of follicular cyst was 3.31 and the mean width of that was 2.3 cm. 6. The mean length and width of luteal cyst was 3.45 and 2.25 cm, respectively. The mean length and width of corpus luteum tissue was 1.15 and 0.67 cm, respectively, excluding the size of cyst in the luteal cyst. 7. The width of uterine horn associated with endometritis was significantly reduced as the period after parturition was elapsed (p<0.05). The mean width of uterine horn within 40 days after parturition was 4.55 cm. These results indicated that ultrasonography is of great use for accurate diagnosis both on ovarian diseases and uterine diseases and that it is very effective to diagnose endometritis in dairy cows.

  • Outbreaks of porcine reproductive and respiratory syndrome virus (PRRSV) in vaccinated sow herds from occurrence to stabilization were monitored and analyzed in terms of serology and reproductive performance. Three different conventional pig farms experienced severe reproductive failures with the introduction of a type 1 PRRSV. These farms had adopted mass vaccination of sows using a type 2 PRRSV modified live vaccine (MLV). Therefore, to control the type 1 PRRSV, an alternative vaccination program utilizing both type 1 and type 2 MLV was undertaken. Following whole herd vaccinations with both types of MLV, successful stabilization of PRRS outbreaks was identified based on serological data (no viremia and downward trends in ELISA antibody titers in both sows and suckling piglets) and recovery of reproductive performance. Additionally, through comparison of the reproductive parameters between outbreak and non-outbreak periods, it was identified that PRRSV significantly affected the farrowing rate and the number of suckling piglets per litter at all three pig farms. Comparison of reproductive parameters between periods when the different vaccination strategies were applied revealed that the number of piglets born in total and born dead per litter were significantly increased after the introduction of the type 1 PRRS MLV.

  • The prevalence of some swine reproductive and zoonotic diseases in Colombia is unknown, making their management difficult. This study assessed the prevalence of porcine circovirus type 3 (PCV3), Leptospira interrogans, pseudorabies virus, and porcine gamma-herpesvirus by polymerase chain reaction in sows in the productive stage, from farms with a history of reproductive failures, at the department of Tolima. The prevalence of PCV3 was 2.6% and 12.6% for L. interrogans, with some samples co-infected with PCV2. Owing to the coinfections with PCV2, it is necessary to establish whether the interactions between these pathogens were related to the presence of reproductive problems.

  • Steroid hormone profiles during luteal phases after in vitro fertilization(IVF) and embryo transfer(ET) have been evaluated in 83 cycles stimulated by pure follicle-stimulating hormone/human menopausal gonadotropin/human chorionic gonadotropin, in which 13 patients became pregnant. Serum estradiol($E_2$) and progesterone($P_4$) levels were determined on days 2, 5, 7 and 9 after laparoscopic follicle aspiration. The follicular $E_2$ peak was slightly higher in pregnancies than in failures. Positive correlations were observed between the follicular $E_2$ peaks and the $P_4$ levels on days 5 and 7 of the luteal phase in pregnancies, but no correlation was found in failures. The $E_2$ and $P_4$ levels on days 5 and 7 of the luteal phase were significantly higher in pregnancies than in failures, but not different on days 2 and 9. Values of the $P_4/E_2$ ratio were similar between the two groups. The luteal phase durations were 12 to 19 days and no correlation was observed between the lengths of luteal phase and the luteal $E_2$ or $P_4$ concentrations. These data suggest that high $P_4$ levels in the mid-luteal phase, which have positive correlations with the follicular $E_2$ peaks, might have a favorable influence on the pregnancy success in human IVF.

  • Objective: To evaluate whether T helper 1 (Th1) immune response is predominant in women with reproductive failures (recurrent spontaneous abortion and recurrent implantation failure) and the activation of T cell is related to Th1 propensity. Methods: Women with a history of recurrent implantation failure or recurrent spontaneous abortion comprise the study group (n=37). Controls are normal fertile women without a history of infertility or pregnancy losses (n=11). Th1/Th2 ratios of interferon (INF)-$\gamma$/interleukin (IL)-10 and tumor necrosis factor (TNF)-$\alpha$/IL-10 expression on $CD3^+/4^+$ cells, CD154, and CD69 expression on T cells are measured by flow cytometric analysis. Results: The ratios of TNF-$\alpha$ to IL-10 expressing on $CD3^+/4^+$ cells (Th1/Th2 cell ratios) are significantly higher in study group ($42.1{\pm}2.3$) as compared with that of controls ($28.7{\pm}2.7$) (p=0.002). The overall trend of CD154 and CD69 expression on T cells are elevated in study group than those of controls. The proportion (%) of $CD3^+/4^+/154^+$ cells ($1.7{\pm}0.5$ vs. $0.3{\pm}0.2$, p=0.038) and the % of $CD3^+/8^+/154^+$ cells ($0.6{\pm}0.2$ vs. $0.1{\pm}0.0$, p=0.024) are significantly higher in study group. The % of $CD3^+/69^+$ cells ($5.6{\pm}1.9$ vs. $1.3{\pm}5.4$, p=0.046) and % of $CD3^+/8^+/69^+$ cells ($4.8{\pm}1.3$ vs. $1.8{\pm}0.2$, p=0.035) among $CD3^+/8^+$ cells are significantly increased in study group. Conclusion: Women with reproductive failures have Th1 propensity with increased T cell activation. These finding means that activated T cell has a harmful effect on early pregnancy and implantation by induction of Th1 immunity.
