Bitter Melon Seed Extract does not Alter Photoperiodic Effects on Reproduction of Male Golden Hamsters

  • Bitter melon (Momordica charantia, MC) has been used in traditional Korean medicine in treating diabetes. In addition, some reports were emerged, showing the antifertility activities of MC in mammals. We investigated the effects of ethanolic MC extract on the reproductive activity of golden hamsters whose spermatogenetic capacity is controlled by their photoperiods. The animals were divided into 4 groups: long photoperiod (LP) control, short photoperiod (SP) control, and LP animals treated with MC. The animals were orally ingested with low (0.03 g/kg) or high (0.15 g/kg) concentrations of the ethanolic extracts for 8 weeks on the daily basis. The control animals received the vehicle. The animals were then mated with age-matched females, experienced pregnancy. As results, the LP control animals showed active large testes but SP control animals displayed remarkably reduced testes. The animals treated with both concentrations of MC extracts demonstrated large testes, indicating fertile activity as animals in LP. LP control animals had litters as expected, but SP controls had no litters at all. MC extract showed the same results as LP animals in generating offsprings. These results suggest that the MC extract does not change the photoperiodic influence on reproductive activity of male golden hamsters.

Effect of Splenectomy on the Blood Picture according to Gestation Periods in Rats (비장적출이 임신 rat의 혈액상에 미치는 영향)

  • This experiment was examined the effect of splenectomy on the hematology in pregnant wistar rat. Only animals that had been shown regular 4-day estrous cycles for more than two cycles were used. The day after mating with the same male animal ws designated Day 0 of pregnancy. Spleen was removed from Day 0(early), 6(middle) and 13(late) of pregnant rat, respectively. Blood sample was collected at Day 1, 7, 14 and 21 of the pregnancy. 1. RBC was increased significantly(P<0.05) to the progress of pregnancy in control rat. The late splenectomized rats were decreased significantly(P<0.05) at Day 21 of pregnancy than control rats. 2. Hb was increased significantly (P<0.05) at 21th day of pregnancy in late splenectomized groups than others group. 3. In the late splenectomized rats, Ht was decreased significant (P<0.05) due to the progress of pregnancy and decreased significantly (P<0.05) at Day 21 of pregnancy in all splenectomized groups. 4. WBC was increased significantly (P<0.05) at Day 1 of pregnancy in splenectomized groups compared with control. 5. In differential leukocyte rate, the Basophils and Monocytes was not significantly changed. Neutrophils was increased significantly(P<0.05) at Day 14 and 21 than Day 1 and 7 of pregnancy in control. Lymphocytes was decreased significantly(P<0.05) in control due to progress of pregnancy. Neutrophils was increased and Lymphocytes was decreased significantly(P<0.05) in splenectomized groups compared with control.

Targeted Suppression of Connexin 43 in Ovine Preimplantation Embryos by RNA Interference Using Long Double-stranded RNA

  • RNA interference (RNAi) is an acknowledged useful and effective tool to study gene function in various cells. Here, we suppressed the Connexin 43 (Cx 43) gene expression during in vitro development of ovine pre-implantation embryos using the RNAi method. The 353 bp Cx 43 double-stranded RNA was microinjected into in vitro fertilized ovine zygotes, and the levels of target mRNA and protein were investigated. Control groups included uninjected zygotes or those injected with RNase-free water. The dsRNA injection resulted in the specific reduction of Cx 43 transcripts as analyzed by quantitative real-time RT-PCR and decreased protein levels as shown by Western blot analysis at the blastocyst stage. Microinjection of Cx 43 dsRNA led to 20.3%, 21.7% and 34.5% blastocyst rates and 19.2%, 37.5% and 41.3% hatched blastocyst rates in Cx 43 dsRNA-injected, water-injected and uninjected groups, respectively. Then the RNAi could not significantly affect cell number and cell death rates of blastocysts. Therefore, suppression of Cx 43 dsRNA and proteins did not apparently affect the development potential of ovine pre-implantation embryos but may play a role in embryo quality. RNAi technology is a promising approach to study gene function in early ovine embryogenesis.

Research advances in reproduction for dairy goats

  • Considerable progress in reproduction of dairy goats has been made, with advances in reproductive technology accelerating dairy goat production since the 1980s. Reproduction in goats is described as seasonal. The onset and length of the breeding season is dependent on various factors such as breed, climate, physiological stage, male effect, breeding system, and photoperiod. The reproductive physiology of goats was investigated extensively, including hypothalamic and pituitary control of the ovary related to estrus behavior and cyclicity etc. Photoperiodic treatments coupled with the male effect allow hormone-free synchronization of ovulation, but the kidding rate is still less than for hormonal treatments. Different protocols have been developed to meet the needs and expectations of producers; dairy industries are subject to growing demands for year round production. Hormonal treatments for synchronization of estrus and ovulation in combination with artificial insemination (AI) or natural mating facilitate out-of-season breeding and the grouping of the kidding period. The AI with fresh or frozen semen has been increasingly adopted in the intensive production system, this is perhaps the most powerful tool that reproductive physiologists and geneticists have provided the dairy goat industry with for improving reproductive efficiency, genetic progress and genetic materials transportation. One of the most exciting developments in the reproduction of dairy animals is embryo transfer (ET), the so-called second generation reproductive biotechnology following AI. Multiple ovulation and ET (MOET) program in dairy goats combining with estrus synchronization (ES) and AI significantly increase annual genetic improvement by decreasing the generation interval. Based on the advances in reproduction technologies that have been utilized through experiments and investigation, this review will focus on the application of these technologies and how they can be used to promote the dairy goat research and industry development in the future.

Effect of vitamin C on pregnancy rate and 8-OHdG levels during heat stress in post-partum dairy cattle

  • In this study the effect of vitamin C administration on pregnancy rates during summer heat stress in dairy cows was examined. A total of 80 Holstein-Friesian cows were divided into control and treatment groups (n = 40 each). Control group animals were given 10 mL isotonic normal saline, and treatment group, Vitamin C (4 mg/kg) on artificial insemination day (day 0) and 4th, 8th and 12th day post insemination. Pregnancy diagnosis was performed on 30th day post insemination by ultrasonography. Blood samples were randomly taken from 11 animals from each group. Serum P4, GSH, MDA and plasma 8-OHdG levels were determined by using ELISA method. Results showed that 8-OHdG levels were lower in treatment group on day 4, 8 and 12 (p < 0.05) compared with the control group. Similarly, pregnancy rate was higher in treatment group (32.5%) than control (22.5%), respectively. However, MDA, P4 and GSH levels were similar in both groups at 4th, 8th and 12th day. A gradual increase in P4, and MDA levels, and a strong positive correlation between 0, 4th (r = 0.54), 4, 8th (r = 0.59) and 8, 12th (r = 0.51) day was found. Similarly, GSH levels also showed positive correlation at days 0, 4th (r = 0.47) and 4, 8th (r = 0.56). However, a strong negative correlation (r = -0.56) between MDA day 0, and GSH day 8 was found. In conclusion, vitamin C application during insemination period in postpartum cows increases pregnancy rate, and reduces oxidative stress metabolite 8-OHdG levels.

  • This experiment was undertaken to investigate the effect of splenectomy on the hematology and maintenance of pregnant rats. Those animals that had shown regular 4 or 5 day estrous cycles for more than two cycles were used. The day after mating ws designated Day 0 of pregnancy. The total serum protein was increased significantly (P<0.05) to the progress of pregnancy in control rat. All rats that splenectomized groups were decreased significantly(P<0.05) at Day 21 of pregnancy than control rats. 2. Albumin was not significantly changed. 3. Globulin was decreased significantly(P<0.05) between contol and middle treatment at Day 7, 14 of pregnancy. 4. Glucose was increased for the due to the pregress of pregnancy. It was no significance differences among the each groups. 5. Rate of abortion was increased in groups of early and middle of splenectomy compared with control. 6. Period of pregnancy was delayed in middle and late splenectomized gruops companed with control. 7. Litter size was lowed significantly (P<0.05) in early and middle splenectomized groups compared with control.

Effects of the Unilateral Ovariectomy on Estrus Cycle, Weights of Ovary, Placenta and Fetus, and Number of Litter in Female Rats (편측난소척출이 흰쥐의 발정주기, 난소.태반.태아 중량 및 산자수에 미치는 영향)

  • This experiment was carried out to investigate the recovery period of estrus cycle, the pseudopregnancy period, weights of ovary, placenta and fetus, and number of litter of female rats after unilateral ovariectomy(ULO). The results obtained are as follows : 1. The estrus cycle was inclined to become normal from the second time. 2. The pseudopregnancy period of rats in unilaterally ovariectomized group and control group, and no significant difference in pseudopregnancy period was shown between the two groups. 3. The ovarian weights of each ULO group were significantly(P<0.01) heavier than that of the control group throughout the observation periods of pregnancy. 4. The placental weights of ULO group were not different from that of the control group in 10 and 15 days of pregnancy. But they became significantly (P<0.05) heavier in 20 days. 5. The weights of fetus in ULO at 10 days of pregnancy was not different from that ofthe control group, but the weights in ULO group at 15 and 20 days of pregnancy were significantly(P<0.01) heavier. 6. The number of litter in ULO was smaller than that of the control groups.

Growth and Reproductive Performance of Small Ruminants under Integrated Livestock-Oil Palm Production System

  • The effects of supplementation with mixed fodder or concentrate were investigated on the performance of a) growth of male lambs and kids grazing extensively under oil palm trees and b) reproduction of ewes and goats stall-fed with native herbage. Supplemented kids and lambs had higher average daily weight gain than controls but the effect was only significant for those which received concentrate (p < 0.05). Final body weight was only significantly different from controls for lambs supplemented with concentrate (p < 0.05). The ages at puberty, first mating, first conception and first kidding of supplemented goats were about 110 days earlier than those for controls (p < 0.05 or better). Supplemented goats had first mating and conceived at lower body weights (p < 0.01) than those in control groups. Different feeding regimes had no effects (p > 0.05) on the reproductive performance of ewes apart from highest body weight of first lambing in animals supplemented with concentrate (p < 0.05). Native herbage available under oil palm trees of 5 years old was sufficient for growth and reproduction of sheep and goats. Concentrate was better than mixed fodder as supplement for enhancing the reproductive performance of goats but both appeared to have limited effects on the performance of reproduction in ewes and growth in male kids.

A Study on The Color Reproduction Properties of Domestic Newspaper Prints (국내 신문 인쇄의 색재현 특성에 대한 평가)

  • Web offset newspaper prints is the most widely used printing process in domestic printing industry, but there are no industry-wide specifications for press control to assure consistent quality across newspaper printing plants. As newspaper printing becomes more of a commodity and less of an art, it is necessary to develop the print quality specification standard suitable for Korea web offset newspaper printing field. There are a few commonly employed printing standards such as, SNAP(Specification for Non-Heatset Publication) of the US, IFRA(INCA-FIEJ Research Association), J-color (Japan printing machinery association). And there exists KS for domestic standards, however, since it has been directly imported from ISO standards, there are discrepancies in printing materials and media. This paper aims to examine the color reproduction quality of domestic web offset newspaper prints by comparing with foreign standards. Printing experiments on this study have been performed at one of the domestic newspaper printing companies. The color reproduction properties of the web offset newspaper prints are measured by means of densitometry and colorimetry. These measures are evaluated comapratively with the international standards by means of analyzing, the major factors that determine the color reproduction quality of web offset newspapar prints.
