카드뮴유발 흰쥐의 간손상에 대한 산사(山査)추출물의 보호효과 (Hepatoprotective Activity of Crataegii Fructus Water Extract against Cadmium-induced Toxicity in Rats)

  • Crataegii Fructus is commonly used as a improving digestion, removing retention of food, promoting blood circulation and resolving blood stasis agent in East Asia. Cadmium (Cd) is widely distributed in the environment due to its use in industry. An exposure to Cd causes dysuria, polyuria, chest pain, hepatic and renal tubular diseases. The liver is the most important target organ when considering Cd-induced toxicity because Cd primarily accumulates in the liver. This study investigated the protective effect of Crataegii Fructus water extract against cadmium ($CdCl_2$, Cd)-induced liver toxicity in H4IIE cells, a rat hepatocyte-derived cell line and in rats. Cell viability was significantly reduced in Cd-treated H4IIE cells in a time and concentration-dependent manner. However, Crataegii Fructus water extract (CFE) protected the cells from Cd-induced cytotoxicity via inhibition of PARP cleavage. To induce acute toxicity in rats, Cd (4 mg/kg body weight) was dissolved in normal saline and intravenously injected into rats. The rats then received either a vehicle or silymarin (as a positive control) or CFE (50, 100 mg/kg/day) for 3 days, and were subsequently exposed to a single injection of Cd. Alanine aminotransferase (ALT), aspartate aminotransferase (AST) and lactate dehydrogenase (LDH) were significantly increased by Cd treatment. In contrast, pretreatment with CFE reduced ALT, AST and LDH. In histopathological analysis, CFE reduced the hepatic degenerative regions and the number of degenerative hepatocytes. These are considered as direct evidences that Crataegii Fructus has favorable inhibitory effects on the Cd-intoxicated liver damages. The efficacy of Crataegii Fructus shows slight lower than that of silymarin in the present study.

루꼴라(Eruca sativa) 추출물의 멜라닌 생성 저해 효과 (Anti-melanogenic effect of Eruca sativa extract)

  • 본 연구에서 루꼴라(E. sativa)에 대한 다양한 생리활성을 조사하여 기능성소재 응용가능성을 검토하였다. 루꼴라 추출물은 멜라노마 세포에 대하여 낮은 세포독성을 나타냈다. 세포독성이 거의 없는 농도에서 루꼴라 추출물 처리 시 항산화 활성($ID_{50}$, 17.60 mg/L)이 매우 우수하게 나타났으며 루꼴라 추출물 처리시 높은 tyrosinase 활성저해($ID_{50}$, 132.54 mg/L) 및 멜라닌 함량 저하($ID_{50}$, 158.90 mg/L)를 보여주었다. 또한, 루꼴라 추출물 농도 의존적으로 tyrosinase 발현이 억제되었으며, 이는 루꼴라 추출물이 직접적인 tyrosinase 활성저해 및 세포 내 tyrosinase 발현을 억제시킴으로서 멜라닌 합성을 저해하는 것으로 판단된다. 이와 같은 결과로 미루어 볼 때 루꼴라 추출물은 피부미백 소재 등 피부개선 효과를 지닌 기능성 화장품에 활용하기 위한 매우 효과적인 재료가 될 수 있다고 판단되며 향후 주름개선 효과 등 추가적인 실험을 통하여 화장품 소재 개발 응용 가능성을 타진할 것이다.

Beneficial effects of fermented black ginseng and its ginsenoside 20(S)-Rg3 against cisplatin-induced nephrotoxicity in LLC-PK1 cells

  • Background: Nephrotoxicity is a common side effect of medications. Panax ginseng is one of the best-known herbal medicines, and its individual constituents enhance renal function. Identification of its efficacy and mechanisms of action against drug-induced nephrotoxicity, as well as the specific constituents mediating this effect, have recently emerged as an interesting research area focusing on the kidney protective efficacy of P. ginseng. Methods: The present study investigated the kidney protective effect of fermented black ginseng (FBG) and its active component ginsenoside 20(S)-Rg3 against cisplatin (chemotherapy drug)-induced damage in pig kidney (LLC-PK1) cells. It focused on assessing the role of mitogen-activated protein kinases as important mechanistic elements in kidney protection. Results: The reduced cell viability induced by cisplatin was significantly recovered with FBG extract and ginsenoside 20(S)-Rg3 dose-dependently. The cisplatin-induced elevated protein levels of phosphorylated c-Jun N-terminal kinase (JNK), p53, and cleaved caspase-3 were decreased after cotreatment with FBG extract or ginsenoside 20(S)-Rg3. The elevated percentage of apoptotic LLC-PK1 cells induced by cisplatin treatment was significantly abrogated by cotreatment with FBG and the ginsenoside 20(S)-Rg3. Conclusion: FBG and its major ginsenoside 20(S)-Rg3, ameliorated cisplatin-induced nephrotoxicity in LLC-PK1 cells by blocking the JNKep53ecaspase-3 signaling cascade.

산화 스트레스에 대한 신선초 녹즙과 돌미나리 녹즙의 보호효과 (Protective Effect of Angelica keiskei Juice and Oenanthe javanica DC Juice on Oxidative Stress)

  • 본 연구에서는 신선초 녹즙과 돌미나리 녹즙을 다양한 실험을 통하여 산화방지 활성을 측정하였다. DPPH 라디칼, NO, $O_2{^-}$, ${\cdot}OH$ 라디칼 소거능 실험을 이용하여 유기농 및 일반농 신선초 녹즙과 돌미나리 녹즙의 산화방지 활성을 측정해 본 결과, 모든 시료군에서 높은 라디칼 소거 활성이 나타났다. 또한, 산화적 스트레스에 민감한 LLC-$PK_1$ 세포를 이용하여 유기농 및 일반농 신선초 녹즙과 돌미나리 녹즙의 산화적 스트레스 개선 효과를 살펴본 결과, 신선초 녹즙과 돌미나리 녹즙은 AAPH, SNP, 파이로갈롤과 SIN-1에 의해 유발된 산화적 스트레스(NO, $O_2{^-}$$ONOO^-$)에 대한 세포 생존율을 증가시키고, 지질과산화를 억제시켜 라디칼에 의한 산화적 스트레스에 대한 개선 효과가 뛰어난 것으로 사료된다. 이상의 결과로부터 신선초 녹즙과 돌미나리 녹즙은 우수한 산화방지 활성과 지질과산화 개선 효과를 나타내어 신선초 녹즙과 돌미나리 녹즙으로부터 산화방지 영양소를 충분히 섭취하는 것이 산화적 손상과 관련된 질병을 예방하는 좋은 방법이 될 수 있을 것이라 사료된다.

흑삼의 프로사포게닌 추출물이 Streptozotocin으로 유도된 당뇨 쥐에 대한 항당뇨 효과 및 신장보호 효과 (The Anti-Diabetic Effects and Nephroprotective Effect of Black Ginseng Prosapogenin Extract in Streptozotocin-Induced Mice)

  • Background: This study examined the hypoglycemic and kidney protective effect of black ginseng in streptozotocin-induced diabetic mice. Methods and Results: Diabetes was induced by treating mice with streptozotocin (STZ) for four weeks. In vivo studies were performed in order to investigate the hypoglycemic effect of the black ginseng prosapogenin (GBG05-FF) extract. The body weight and blood glucose level were measured. Moreover, after the mice were sacrificed, the kidneys were isolated and histological changes were observed with hematoxylin and eosin staining. Blood urea nitrogen and creatinine levels were also measured. The results showed that administration of black ginseng increased body weight. Compared to blood glucose levels in STZ mice, blood glucose levels were reduced by 48% in STZ mice supplemented with 300 mg/kg of black ginseng, and by 69% in STZ mice supplemented with 900 mg/kg. Furthermore, histopathological examination of STZ mouse kidneys revealed, changes in the kidneys, epithelial cell damages, inflammatory cell infiltration and glomerulus hypertrophy. However, a significant reduction of glomerular water droplets (indicative of glomerulus hypertrophy) was observed in the kidneys of STZ mice supplemented with black ginseng extract. Conclusions: These results suggest that black prosapogenin (GBG05-FF) ginseng extract has a significant hypoglycemic effect and can be used as an anti-diabetic substance and renal protective agents as part of dietary supplements or novel drugs.

하고초 추출물의 streptozotocin 유발 당뇨 랫트 사구체 손상 개선 효과 (Protective Effects of Prunella Vulgaris on Glomerular Injury in Streptozotocin-Induced Diabetic Rats)

  • Prunella vulgaris, well-known traditional medicinal plant, is used for the cure of abscess, scrofula, hypertension and urinary diseases. Diabetic nephropathy is the most common cause of end-stage renal disease. The pathological characteristics of diabetic nephropathy are glomerular and tubular basement membrane thickening. The aim of the present study was to evaluate the effect of Prunella vulgaris, on diabetic glomerular injury in streptozotocin-induced diabetes rats. Diabetes mellitus was induced by a single intraperitoneal injection of streptozotocin (STZ; 45 mg/kg) and confirmed by random glucose level higher than ${\leq}300mg/dL$. The experimental rats were divided into five groups: control group (Male SD rats), STZ group (Male SD rats injected STZ), Aminoguanidine group (Male SD rats injected STZ + AG 100 mg/kg/day), Low dose group (Male SD rats injected STZ + APV 100 mg/kg/day), High dose group (Male SD rats injected STZ + APV 300 mg/kg/day). AG or APVs were administered once a day for 8 weeks. Body weight and food/water intake were measured every four weeks. At the end of study, the kidneys were collected and cut into pieces for immunohistochemistry and western blot analysis. Our study showed that body weight and water/food intake were no significant differences between untreated STZ-induced diabetic rat and APV treated-STZ rat. However, phosphorylation of receptor-regulated Smads (Smad3) was significantly decreased in APV treated-STZ rat as compared with the diabetic group. In addition, APV was improved nephrin level in kidney tissue. Therefore, we suggest that APV has a protective effect against STZ-induced diabetic glomerular injury.

상황버섯 및 카레를 첨가한 잡곡밥 추출물의 LLC-PK1 세포에서의 산화적 스트레스 보호 효과 (Protective Effects of Phellinus linteus and Curry-Added Cooked Mixed Grain Rice Extracts on Oxidative Stress-Induced LLC-PK1 Cell Damage)

  • 잡곡밥 메탄올 추출물이 가지는 산화적 스트레스 개선 효과를 확인하기 위하여 $H_2O_2$로 유도된 산화적 스트레스에 대한 LLC-PK1 세포의 보호 효과를 조사하였다. $250{\mu}M$$H_2O_2$에 의한 산화적 스트레스가 유발된 LLC-PK1 세포에 시료추출물을 처리한 결과, WR, TMR, GR 순으로 $H_2O_2$ 처리군에 비해 세포 생존율이 증가하였다(P<0.05). 특히 GRS와 GRK에서 정상군에 가까운 세포 생존율을 나타내어 $H_2O_2$에 의한 산화적 스트레스로 유발된 세포 손상에 대한 잡곡밥 추출물의 보호 효과를 확인할 수 있었다. 세포 내 활성산소(ROS)의 수준과 세포 내 지질과산화물질(MDA)의 생성 억제 효과 역시 WR, TMR, GR, GRK, GRS 순으로 증가하는 것을 관찰하였다. $H_2O_2$로 인하여 세포 내 항산화 효소인 SOD, CAT와 GSH-px 등의 활성이 감소된 LLC-PK1 세포에 잡곡밥 추출물을 처리했을 때 이들 효소의 활성이 증가했으며 특히 GRK, GRS군에서 현저하게 증가되었다. LLC-PK1 세포에서 $H_2O_2$에 의해 발생하는 산화적 스트레스에 대한 보호 효과를 측정한 결과 잡곡밥 추출물은 세포 손상을 억제하고 세포 내 활성산소(ROS)의 수준과 지질과산화물질(MDA)의 생성을 억제하며, 세포 내 항산화 효소의 활성을 증가시키는 효과를 가지는 것으로 보인다. 또한 항산화 효소유전자 발현에서도 위와 비슷한 효과를 나타내었다. 이상의 결과로 잡곡밥 메탄올 추출물, 특히 기능성 물질인 카레가루와 상황버섯 추출물이 첨가된 잡곡밥은 LLC-PK1 세포에 대한 보호 작용과 in vitro에서의 항산화 효과가 높은 것으로 확인되었다.

  • Tetrandra is the root of Stephania tetrandra 5. Moore (family Menispermaceae), or of Aristolochia frangchi Wu (family Aristolochiaceae). It is a Differ-flavored and cold-property herb acting on the urinary bladder, kidney and spleen meridiands. Known biological effects of this herb are expelling wind to relieve pain and inducing diuresis to alleviate edema. This herb also has anti-inflammatory and anti-hypersensitivity actions. Recent studies have shown that Stephanniae Tetrandrae Radix has antimicrobial effects, namely, a protective effect on acute renal failure induce by gentamicin sulfate and a suppressive effect against clostridium perfringes. However, there is a lack of studies concerning the immunological activities of this herb. The present study was conducted to evaluate the immunological activities of Stephanniae Tetrandrae Radix on the regulatory mechanisms of cytokines and nitric oxide (NO) in Raw 264.7 cells. Cell viability was measured by MTT assay after the treatment of Stephanniae Tetrandrae Radix extract (STRE) and NO production was monitored by measuring the nitrite content in culture medium. COX-2 and iNOS were determined by immunoblot analysis, and levels of cytokine were analyzed by sandwich immunoassays. Results provided evidences that STRE inhibited the production of nitrite and nitrate (NO), inducible nitric oxide synthase (iNOS), cyclooxygenase-2 (COX-2) tumor necrosis $factor-{\alpha}\;(TNF-{\alpha})$, $interleukin-1{\beta}(IL-1{\beta})$ and interleukin-6 (IL-6) in Raw 264.7 cells activated with lipopolysaccharide (LPS). These findings showed that STRE could produce some anti-inflammatory effects which might play a role in adjunctive therapy in Gram-negative bacterial infections.

칼슘과 마그네슘이 조절된 해양심층수가 신장세포에서 항산화에 미치는 영향 (Effects of calcium and magnesium-balanced deep sea water on antioxidation in kidney cells)

  • 본 연구에서는 미네랄이 함유된 해양심층수(DSW)가 신장 기능에 미치는 항산화 효과를 세포 모델을 사용하여 확인하였다. DSW 샘플은 최적의 칼슘/마그네슘 비율을 결정하기 위한 목적으로, 각기 다른 칼슘 및 마그네슘 비율을 가진 4가지 샘플-미량 미네랄(TM), 고 마그네슘(HM), 고 마그네슘 저염(HMLS) 및 고 마그네슘 고 칼슘(HMHC)-로 준비되었다. 신장 세포주 HEK293를 2시간 동안 NaCl로 처리하여 ROS를 유도한 후, 마그네슘과 칼슘 등의 미네랄이 서로 다른 비율로 용해된 물로 처리하여 ROS 농도와 항산화 효소 활성 및 단백질을 측정하였다. 물 샘플 중 HMLS는 ROS에 대한 세포에 가장 많은 보호효과를 나타냈다. 세포 내 글루타티온 함량은 HMLS 그룹과 HMHC 그룹에서 가장 높았다. 반면, TM과 HMHC는 항산화 유전자의 mRNA 발현에서 대조군과 유사한 경향을 보였다. 이러한 결과는 DSW가 과도한 나트륨 섭취로 인한 신장의 산화 스트레스를 예방하는 데 도움이 될 수 있음을 시사한다. 또한 ROS 농도와 항산화 마커 측정 결과를 종합하여 볼 때 HMLS와 HMHC가 신장 세포 모델에서 우수한 항산화 효과를 가진 DSW 샘플이라고 판단할 수 있다.

Protective Effect of Phosphatidylcholine on Lipopolysaccharide-Induced Acute Inflammation in Multiple Organ Injury

  • Soybean polyunsaturated phosphatidylcholine (PC) is thought to exert anti-inflammatory activities and has potent effects in attenuating acute renal failure and liver dysfunction. The aim of this study was to investigate the effects of PC in protecting multiple organ injury (MOI) from lipopolysaccharide (LPS). Six groups of rats (N=8) were used in this study. Three groups acted as controls and received only saline, hydrocortisone (HC, 6 mg/kg, i.v.) or PC (600 mg/kg, i.p.) without LPS (15 mg/kg, i.p.) injections. Other 3 groups, as the test groups, were administered saline, HC or PC in the presence of LPS. Six hours after the LPS injection, blood and organs (lung, liver and kidney) were collected from each group to measure inflammatory cytokines and perform histopathology and myeloperoxidase (MPO) assessment. Serum cytokines (TNF-${\alpha}$, IL-6 and IL-10) and MPO activities were significantly increased, and significant histopathological changes in the organs were observed by LPS challenge. These findings were significantly attenuated by PC or HC. The treatment with PC or HC resulted in a significant attenuation on the increase in serum levels of TNF-${\alpha}$ and IL-6, pro-inflammatory cytokines, while neither PC nor HC significantly attenuated serum levels of IL-10, anti-inflammatory cytokine. In the organs, the enhanced infiltration of neutrophils and expression of ED2 positive macrophage were attenuated by PC or HC. Inductions of MPO activity were also significantly attenuated by PC or HC. From the findings, we suggest that PC may be a functional material for its use as an anti-inflammatory agent.
