Plant Regeneration from Seed-derived Callus in Kentucky Bluegrass(Poa pratensis L.) (켄터키 블루그래스의 종자유래의 캘러스로부터 식물체 재분화)

  • Plant regeneration from seed-derived callus of Kentucky bluegrass(Poa pratensis L. cv. Kenblue) was investigated. Callus induced on the medium supplemented with 2 mg/L 2,4-D and 0.2 mg/L BAP showed highest frequency of plant regeneration on the regeneration medium supplemented with 1 mg/L NAA and 5 mg/L kinetin. Callus induced in the dark condition showed higher regenerability than that induced in the dim light. MS medium was better than N6 and B5 medium in enhancing plant regeneration. Maltose was superior to sucrose in plant regeneration as carbon source in the medium.

Efficient plant regeneration from immature embryo cultures of Jatropha curcas, a biodiesel plant

  • Jatropha curcas L. (Physic nut) is a commercially important non-edible oil seed crop known for its use as an alternate source of biodiesel. In order to investigate the morphogenic potential of immature embryo, explants from four developmental stages were cultured on medium supplemented with combinations of auxins and cytokinins. It was found that the size of embryo is critical for the establishment of callus. Immature embryos (1.1-1.5 cm) obtained from the fruits 6 weeks after pollination showed a good response of morphogenic callus induction (85.7%) and subsequent plant regeneration (70%) with the maximum number of plantlets (4.7/explant) on Murashige and Skoog's (MS) medium supplemented with IBA (0.5 $mg\;l^{-1}$) and BA (1.0 $mg\;l^{-1}$). The above medium when supplemented with growth adjuvants such as 100 $mg\;l^{-1}$ casein hydrolysate + 200 $mg\;l^{-1}$ L-glutamine + 8.0 $mg\;l^{-1}$ $CuSO_4$ resulted in an even higher frequency of callus induction (100%). Plant regeneration (90%) with the maximum number of plantlets (10/explant) was achieved on MS medium supplemented with 500 $mg\;l^{-1}$ polyvinyl pyrrolidone + 30 $mg\;l^{-1}$ citric acid + 1 $mg\;l^{-1}$ BA + 0.5 $mg\;l^{-1}$ Kn + 0.25 $mg\;l^{-1}$ IBA. It was observed that plantlet regeneration could occur either through organogenesis of morphogenic callus or via multiplication of pre-existing meristem in immature embryos. The age of immature embryos and addition of a combination of growth adjuvants to the culture medium appear to be critical for obtaining high regeneration rates. Well-developed shoots rooted on half-halfstrength MS medium supplemented with 0.5 $mg\;l^{-1}$ IBA and 342 $mg\;l^{-1}$ trehalose. The rooted plants after acclimatization were successfully transferred to the field in different agro-climatic zones in India. This protocol has been successfully evaluated on five elite lines of J. curcas.

  • An efficient plant regeneration system has been developed for figleaf gourd (Cucurbita ficifolia Bouch$\'{e}$), which is exclusively used as a rootstock for cucumber. The protocol is based on results obtained from a series of culture experiments involving different parts of the cotyledons and various media. The culture of cotyledon explants was critical for the enhancement of shoot regeneration frequency. The lower parts of the cotyledon excised at the plumule base were found to display a markedly enhanced production of adventitious shoots compared to other cotyledon regions. Culture in silver nitrate-supplemented Murashige and Skoog (MS) medium was not beneficial for shoot regeneration and suppressed root regeneration. Efficient shoot regeneration was obtained on MS medium containing 1.0 $mg\;l^{-1}$ zeatin and 0.1 $mg\;l^{-1}$ indole-3-acetic acid. Regenerated shoots successfully elongated and rooted in medium containing 0.1 $mg\;l^{-1}$ 1-naphthalene-acetic acid after 10-15 days of subculturing. The plantlets were satisfactorily acclimatized in a greenhouse and grew into normal plants without any morphological alterations.

  • The optimal conditions for the protoplast formation and regeneration of Streptomyces mitakaensis have been investigated. S. mitakaensis cells were converted to protoplast by treating with 0.1 mg/$m\ell$ of lysozyme in phosphate-tris buffer (pH 7.2) to the cells grown at the late logarithmic growth phase in the GBYN medium (gycerol 20g, beef extract 5g, yeast extract 5g, NaCl 5g in 1 liter of distilled water) contained 0.5% glycine. Cell regeneration from protoplast was accomplished in 10 days post inoculation on the R2 regeneration agar medium and at 3 days post inoculation on the H2 regeneration liquid medium. The efficiency of the regeneration was 0.l% in 3 days at 35$^{\circ}C$.

Factors for high frequency plant regeneration in tissue cultures of Indian mustard (Brassica juncea L.)

  • An efficient system for high frequency plant regeneration was established through investigating various factors such as plant growth regulator combinations, explant types and ages, and addition of $AgNO_3$ influenced on shoot regeneration in Brassica juncea L. cv. BARI sarisha-10. Murashige and Skoog (MS) medium supplemented with 0.1 mg/L NAA (1-naphthaleneacetic acid) and 1 mg/L BA (6-benzyladenine) showed the maximum shoot regeneration frequency (56.67%) among the different combinations of NAA and BA. Explant type, explant age, and addition of $AgNO_3$ also significantly affected shoot regeneration. Of the four type of explants (cotyledon, hypocotyl, root, and leaf explants)- cotyledon explants produced the highest shoot regeneration frequency and hypocotyls explants produced the highest number of shoots per explant, whereas root explants did not produce any shoot. The cotyledonary explants from Four-day-old seedlings showed the maximum shoot regeneration frequency and number of shoots per explant. Shoot regeneration frequency increased significantly by adding $AgNO_3$ to the medium. Two mg/L $AgNO_3$ appeared to be the best for shoot regeneration with the highest shoot regeneration frequency (86.67%) and number of shoots per explant (7.5 shoots). Considerable variation in shoot regeneration from cotyledonay explants was observed within the B. juncea L. genotypes. The shoot regeneration frequency ranged from 47.78% for cv. Shambol to 91.11% for cv. Rai-5. In terms of the number of shoots produced per explant, B. juncea L. cv. Daulot showed the maximum efficiency. MS medium supplemented with 0.1 mg/L NAA showed the highest frequency of rooting. The regenerated plantlets were transferred to pot soil and grown to maturity in the greenhouse. All plants were fertile and morphologically identical with the source plants.

  • In order to optimize the conditions of callus induction and plant regeneration in seed culture of Italian ryegrass, the effects of basic medium and carbon sources on seed culturability and genotypic difference of plant regenerability were investigated. MS medium was better than N6 and B5 medium in enhancing callus growth and plant regeneration. Sucrose was superior to maltose in plant regeneration as carbon source in the medium. The genotypic difference in plant regenerability was obvious among four cultivars of Italian ryegrass tested. 'Rio' and 'Jeanne' showed to have higher regenerability with the frequency of 38% and 56%, respectively.

  • Apical meristems tissue were cultured on LS medium with different zeatin and NAA concentrations to compare their potential to regenerate shoots, roots and bulblet formation. Shoot regeneration from apical meristem was effective on LS medium added with zeatin 0.1, 0.5, 1.0 mg/L alone treatment or zeatin 0.5 mg/L and NAA 1.0 mg/L combination treatment. A high frequency of root regeneration was obtained on LS medium supplemented with zeatin 0.5 mg/ and NAA 1.0 combination treatment. Linsmaier and Skoog(LS) medium with NAA 3.0 mg/L and zeatin 1.0 mg/L combination treatment gave the best results for normal bulblet formation#KW=Allium wakegi ; plant regeneration ; bulblet formation

  • conditions for callus induction and plant regeneratin from seeds of lawngrass (Zoysia japonica Steud.) were confirmed in this study. MS (Murashige and Skoog) medium containg 2,4-D 3 or 5mg/l was used for callus induction, and MS medium with different volumes of BA, NAA and kinetin hormones was used to regenerate the plants from induced calli. MS basic medium containing agar with no hormones or kinetin 1.0mg/l and MS basic medium containing gelite and NAA 1.0mg/l were higher for green callus induction. MS medium containing agar and kinetin 1.0mg/l ws highest degree of efficiency for plant regeneration.

  • This study was conducted to establish mass propagation system from the axillary bud culture of chrysanthemum zawadskii H. which was used as material of medicinal plants. Shoot egeneration was better on MS medium with NAA and BA. The optimum concentraions of growth regulator for shoot regeneration differed depending on accessionsof C. Zawadskii. Shoot regeneration in Keungucheolcho was better on MS Medium with NAA 0.01mg/1 and BA 0.1mg/1 while Hyangrobonggucheocho was better with NAA 0.1mg/1and BA 0.3mg/1. Addition of NAA into medium was effective for induction of root from shoots regenerated. Shoot multiplcation was more effective when 10mg/1 spermine was added into medium than when other polyamines were treated ino medium . Randomly and specifically amplified polymorphic DAC banding patterns based on polymerase chain reaction (PCR) analysis were used to assess the genetic variation of plants regenerated from in vitro culture.

The Spheroplast Formation and Regeneration of Pseudomonas spp. (Pseudomonas spp.의 원형질체 생성과 재생)

  • The optimal conditions for the formation and the regeneration of Pseudomonas spheroplast were measured. Pseudomonas spp. cells were transformed to spheroplasts from 99.0% to 99.9% by treatment of $100{\mu}g/ml$ lysozyme and 10mM EDTA at room temperature. The optimal pH for the spheroplast formation was pH 8.0. Magnecium chloride, calcium chloride and streptomycin were effective on the stabilization of Pseudomonas spheroplast, while $Mg^+\;and\;Na^+$ ions were effective on the formation of Pseudonomas spheroplast. Rich Regeneration Medium was used for the regeneration of Pseudonomas spheroplast. To improve regeneration frequency, Bovin Serum Albumine and cationic ions were added to the spheroplast dilution beffer and regeneration environment respectively. Treatment of 20mM calcium chloride in ehr Rich Regeneration Medium could improve the yield of regenerants as much as 28-fold. Treatment of 1% Bovin Serum Albumine in the spgeroplast formation and dilution buffer increased the yield of regenerants to 10-fold. Also, the regeneration frequency was improved to 14-fold shen Rich Regeneration Meidum containing 0.5% Gelatin was used for regeneration as well as 1% Bovin Serum Albumine.
