Production and Regeneration of Lactobacillus bulgaricus Protoplasts

  • Conditions for the production and regeneration in Lactobacillus bulgaricus protoplasts were investigated. Protoplasts of L bulgaricus strains were obtained by treatment with mutanolysin and lysozyme together in a protoplast forming buffer containing 0.02 M N-2-hydroxyethylpiperazine-N'-2-ethanesulfonic acid (HEPES) (pH 7.0) and 0.5 M sucrose. High protoplast yield was obtained from cells cultured in the de Man, Rogosa and Sharpe(MRS) medium at the middle to late logarithmic growth phase. Regeneration was efficiently accomplished with a complex medium containing 1% sucrose, 20 mM $MgCl_2$, 5% gelatin, and 0.5% bovine serum albumin. The frequency of regeneration of protoplasts was 10~20% after 5 days of incubation at $30^{\circ}C$.

  • In an effort to optimize tissue culture responses of tall fescue, the effects of basic medium and carbon sources on seed culturability and genotypic difference of plant regenerability were investigated. The MS basal medium was superior to $N_{5}$ 6/ and $B_{medium}$ in enhancing callus growth and plant regeneration. To determine the effect of carbon sources on plant regeneration, the seeds were cultured on medium with 30 g/L sucrose and maltose, respectively. Medium supplemented with 30 g/L sucrose resulted in regeneration of shoots from 50% of the calli. The genotypic difference in plant regenerability was obvious among five cultivars of tall fescue tested. \\`KY31\\` and \\`Hokuryo\\` showed to have higher regenerability with the frequency of 33% and 51%, respectively.

  • The optimal conditions for the production and regeneration of L. helveticus protoplasts were examined. The protoplast formation of L. helveticus was most efficient obtained when the cells grown to mid and late logarithmic phase in MRS medium were used. The maximum number of protoplasts was obtained when lysozyme and mutanolysin were used to lysis the cell wall in 20mM HEPES buffer (pH 7.0) containing 1M sucrose. Regeneration was accomplished with a complex medium containing 10% sucrose, 10mM MaCl2, 20mM CaCl2, 5% gelatin and 0.5% bovine serum albumin. The regeneration frequency of the protoplasts was 10-20% after 5 days of incubation at 30C.

Efficient Plantlet Regeneration via Callus Formation from Leaf Segment of Lilium Oriental Hybrid 'Casa Blanca'

  • Callus induction from a leaf explant has been achieved in Lilium Oriental hybrid 'Casa Blanca'. The highest frequency of callus induction was obtained on MS medium supplemented with 0.5 mg/L BA and 2.0 mg/L NAA after 2 months of culture. The cultures maintained continuously without change in color and type of callus when they cultured in the dark. Plantlet regeneration with a high frequency was achieved from induced calli on the same medium. A number of shoots are formed from one cluster of callus, and bulblets developed into intact plantlets after transfer to hormone-free MS medium. No phenotypic variations were observed among regenerants. Enhancement in plantlet regeneration via callus formation would be expected to facilitate the efficiency of transformation of this Oriental hybrid 'Casa Blanca'.

  • Recombinant chimeric plasmid constructed with Xba I digested pUBl10 and -pE194 was transformed by polyethylene glycol induced protoplast transformation system into Bacillus subtilis BR 151 on the mannitol regeneration media, and two genes of antibiotics resistance were expressed simultaneously in the transfromant. Transformation frequency of the recombinant plasmid was 6.5 $\times$ 10$^{-5}$ on the mannitol regeneration agar plate containing neomycin and erythromycin. The replication of recombinant plasmid in the recipient cells was confirmed by the alkaline extraction method and agarose gel electrophoresis.

Analysis of Composition and Diversity of Natural Regeneration of Woody Species in Jebel El Gerrie Dry Land Forest East of Blue Nile State, Sudan

  • The study aims to assess composition, diversity and population indices of natural regeneration of woody species in Jebel El Gerrie forest reserve, Blue Nile State, Sudan. We conducted field work between December 2018 and January 2019. We used random sampling to collect vegetation data in the forest where we made a total of 90 circular sample plots (radius 17.84 m) and distributed them proportionally to the area of each of the four density-based vegetation classes of the forest i.e. high density (C1), medium density (C2), low density (C3) and crop land (C4). In each sample plot we identified all regenerating tree species and counted their regeneration frequencies. We calculated ecological metrics of regeneration frequency, density, abundance, richness, evenness, diversity and importance value index (IVI) and drew abundance rank curve. Results revealed that out of fifteen mature tree species present, natural regeneration of 8 species, which belong to 6 families, was observed. The relatively most frequently naturally regenerating and abundant species were Anogeissus leiocarpa and Combretum hartmannianum. Richness, evenness and diversity of regenerating species were 1.33, 0.82 and 1.7, respectively. One-way ANOVA (α=0.05) of mean regeneration densities disclosed that there were significant differences (F3,86=16.77, p=0.000) between C2 & C3 (p=0.000) and C2 & C4 (p=0.000). While regeneration of seven tree species were absent, two, two and four species were of good, poor and fair regeneration status, respectively. A comparison of mean density of natural regeneration with that of parent trees reflects a poor regeneration status of the forest. The study provides empirical results on the regeneration status of species and signifies the need for management interventions for species conservation and restoration, maintenance of biodiversity and sustainable production.

Factors Affecting plant Regeneration in Unpollinated Ovary Culture of Rice (벼의 미수분 자방배양에 영향을 미치는 요인)

  • The optimal conditions for callus formation and plant regeneration were determined by the manipulation of culture method in unpollinated ovary culture of rice. The effect of cold pretreatment on callus formation and plant regeneration varied with duration of pretreatment. The maximum frequency (38.7%) of plant regeneration was obtained from the unpollinated ovary pretreated for 10 days at $12^{\circ}C$. The ability of callus formation and plant regeneration was higher on the medium with picloram (1 mg/L) than that of 2, 4-D (1 mg/L). However, the high concentration of picloram increased markedly the frequency of albino plant from unpollinated ovary-derived callus. Floral parts inoculated as a unit play important roles in callus formation and plant regeneration. Best result was obtained when ovary with partial cut glume, pedicel and secondary branch as a unit was cultured.

  • An efficient plant regeneration system has been developed for figleaf gourd (Cucurbita ficifolia Bouch$\'{e}$), which is exclusively used as a rootstock for cucumber. The protocol is based on results obtained from a series of culture experiments involving different parts of the cotyledons and various media. The culture of cotyledon explants was critical for the enhancement of shoot regeneration frequency. The lower parts of the cotyledon excised at the plumule base were found to display a markedly enhanced production of adventitious shoots compared to other cotyledon regions. Culture in silver nitrate-supplemented Murashige and Skoog (MS) medium was not beneficial for shoot regeneration and suppressed root regeneration. Efficient shoot regeneration was obtained on MS medium containing 1.0 $mg\;l^{-1}$ zeatin and 0.1 $mg\;l^{-1}$ indole-3-acetic acid. Regenerated shoots successfully elongated and rooted in medium containing 0.1 $mg\;l^{-1}$ 1-naphthalene-acetic acid after 10-15 days of subculturing. The plantlets were satisfactorily acclimatized in a greenhouse and grew into normal plants without any morphological alterations.

Effect of Copper on Callus Formation and Plant Regeneration in Seed Culture of Rice (벼 종자배양에서 Copper가 캘러스 형성 및 식물체 재분화에 미치는 영향)

  • This study was conducted to improve the regeneration efficiency from seed-derived calli of rice by optimizing the copper concentrations in the media. Mature seeds were cultured on MS medium supplemented with copper sulphate (0 to 5.0 mg/L) and 2 mg/L 2,4-D. Callus growth was influenced by the levels of copper sulphate containing with medium, The addition of copper sulphate (2.5 mg/L) in regeneration medium enhanced dramatically the ability of plant regeneration from seed-derived calli. The mean frequency of plant regeneration of 6 indica rices was 27.4% on medium containing copper sulphate, whereas that of the cultivars on copper-free medium was 2.4%. These results suggest that copper sulphate may have an important role in improving regeneration ability of indica rices.

  • in this work, we designed and characterized a 10Gb/s clock and regeneration circuit. The circuit was realized by integrating high-speed ICs and microwave circuits on alumina substrates. The quadri-correlation method was used for frequency and phase-locked loop. The frequency locking range was 150MHz and the rms jitter generated by the circuit was measured to be less than 1.0ps. The clock and data regeneration circuit was successfully applied to 10Gb/s optical receiver.
