노니 과실에 함유된 항산화물의 추출 공정 (Extraction Procedures for Free Radical Scavenging Activity from Noni Fruit (Morinda citrifolia))

  • In this study, we investigated maximum yield of radical scavenging activity (RSA) using each of various extraction solvents and procedure from noni friut (Morinda citrifolia). Noni fruits were either sun-dried (FS) or hot-air dried (FO) at $60^{\circ}C$ after steam blanching. For optimum aqueous extraction, noni fruits should be sun dried and autoclave extraction time should not be over 30 min to produce extraction with high RSA with low cost. In case of 50% ethanol extraction, reflux extracts of FS and FO resulted in $IC_{50}$ of 1.92 mg/mL and 3.06 mg/mL at 8 hr. When $IC_{50}$ values were lower than 5 mg/mL, coefficient of correlation was 0.71 indicating that 71% of the phenolic antioxidants in noni fruits were accounted for the activity by scavenging free DPPH. However, coefficient of correlation significantly decreased to 0.63 over $IC_{50}$ values of 5 mg/mL. Autoclave extraction contained chlorogenic acid of $14.69 \;{\mu}g/mL$ and scopoletin of $3.86 \;{\mu}g/mL$. Reflux extraction showed all three compounds, chlorogenic acid ($26.19 \;{\mu}g/mL$), quercetin ($19.59 \;{\mu}g/mL$), and scopoletin ($17.4 \;{\mu}g/mL$). Therefore, the result of this study indicated that the potential antioxidant activities and functional values were obtained significantly with reflux extraction from noni fruit.

  • Performance of a continuous cocurrent supercritical fluid extraction column for processing of anhydrous milk fat (AMF) was determined. The extract loading increased and the extraction yield decreased as the superficial velocity of AMF increased. The maximum solubility of AMF in $CO_2$ at 4$0^{\circ}C$/3,500 psig was 0.0195g/g. The increase of the carbon dioxide density enhanced the solubility of milk fat and decreased the cholesterol concentration in the extract. Three operation conditions, such as recycle , reflux, and temperature gradient of extraction column, were tested. More short-chain fatty acvids were extracted by the reflux operation . Longchain fatty acids and thehighest ratio fo long-chain unsaturated to saturated fatty acids were also obtained in the raffinate phase with refluex.

Antioxidant Properties of Water and Aqueous Ethanol Extracts and Their Crude Saponin Fractions from a Far-eastern Sea Cucumber, Stichopus japonicus

  • Water and 70% ethanol extracts obtained from a sea cucumber (Stichopus japonicus) body wall by heat reflux or pressurized solvent extraction showed 2,2-diphenyl-1-picrylhydrazyl ($DPPH^{\cdot}$) and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) radical cation ($ABTS^{{\cdot}+}$) scavenging activities comparable to those of fruits and vegetables. The highest activities were observed for the water extract from heat reflux extraction. Crude saponins exhibited higher radical scavenging activities than the soluble matters in the extracts. However, they were responsible for only about 3 to 15% of the scavenging activities of the extracts. Total phenolic contents showed a significant correlation with $DPPH^{\cdot}$ scavenging activities, suggesting a significant contribution of phenolic constituents to the antioxidant properties of the extracts. However, total flavonoid contents showed little correlation with the radical scavenging activities. The results suggest that the water or 70% ethanol extract obtained from sea cucumber body wall by simple heat reflux extraction could provide considerable antioxidant benefits.

추출조건이 노니 부정근에 함유된 rubiadin의 추출 효율에 미치는 영향 (Effects of Extraction Condition on Extraction Efficiency of Rubiadin in Adventitious Roots of Noni (Morinda citrifolia))

  • 노니($Morinda$ $citrifolia$) 부정근의 주요 성분인 rubiadin은 간 보호 효과가 있어 제약산업에서 높은 가치가 있다. 노니 부정근의 주요 유효성분인 rubiadin의 효율적인 추출조건을 규명하고자 용매 종류, 물과 메탄올의 비율(물, 20%, 40%, 60%, 80%, 100%), 추출시간 및 추출방법을 달리하여 추출하였다. 노니 부정근에서의 rubiadin의 함량분석은 HPLC 이용 분석조건을 확립하였으며, C-18 컬럼을 사용하여 280nm에서 메탄올과 물로 농도구배를 주어 분석하였다. 용매별 추출효율은 메탄올(0.08%) > 에탄올(0.05%) > 아세토니트릴(0.03%) > 아세톤(0.02%) 및 메틸렌클로라이드(0.02%) 순으로 증가하였다. 메탄올에 물의 혼합 비율을 달리하여 초음파 추출기로 1시간 동안 추출한 결과 60% 메탄올(0.21%) > 80% 메탄올(0.13%) > 100% 메탄올(0.07%) 및 40% 메탄올(0.07%) 순으로 효과적이었으며, 환류냉각 추출기로 2시간 동안 추출한 결과 60% 메탄올(0.21%) > 40% 메탄올(0.17%) > 80% 메탄올(0.14%) 용매처리구 순으로 효율이 좋았다. 추출방법 및 추출시간에 따른 rubiadin의 추출 효율을 비교하기 위해 환류냉각추출 및 초음파추출, 진탕추출법으로 추출하였다. 추출방법 및 추출시간에 따른 rubiadin의 추출효율은 초음파추출 및 진탕추출에서 각각 8 및 24시간 동안 추출했을 때 효율적이었다.

산양산삼약침의 추출법 별 성분 및 항산화 활성 비교 (Comparison of Index Compounds Content and Antioxidative Activity of Wild Ginseng Pharmacopuncture by Extraction Methods)

  • Objectives: Wild ginseng pharmacopuncture is widely used in oriental medicine. However, there is no standard method for efficiently extracting the active ingredient. In this study, in order to determine an efficient extraction method, wild ginseng was extracted by the distillation and 70% ethanol reflux methods, respectively. In comparing each extract, the index compounds were analyzed, and antioxidant activity was measured. Methods: The index compounds, ginsenoside Rg1 and ginsenoside Rb1, were detected using high performance liquid chromatography (HPLC). Antioxidative activities of total phenolic compounds, DPPH (${\alpha}$, ${\alpha}$-diphenyl-${\beta}$-picrylhydrazyl), ABTS (2,2'-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid)) and FRAP (ferric reducing antioxidant power) were measured to compare their bioactivities. Since saponin is known to be hemolytic, the hemolytic activity of each extract was compared. Results: The index compounds were analyzed. Nothing was detected in the wild ginseng distilled extracts (WGDE). In the wild ginseng 70% ethanol reflux extracts (WGEE), ginsenoside Rg1 was 3.66 mg/g, and ginsenoside Rb1 was 16.70 mg/g. WGEE showed higher levels than WGDE in all antioxidative activities. In the hemolytic test, the extracts showed almost no toxicity, but WGEE showed lower toxicity than WGDE. Conclusions: In this study, it was concluded that WGEE is more advantageous than WGDE in the detection of index compounds and bioactivity. However, additional studies of additional extraction methods and other bioactivity tests are needed.

작약(Paeonia lactiflora Pall.)에서 paeoniflorin 추출방법 및 HPLC 분석조건 (Extraction Methods and HPLC Analysis Conditions of Paeoniflorin in Peony, Paeonia lactiflora Pall.)

  • 경북 영천군 고경면 작약 재배농가의 정식 4년차인 의성품종(Paeonia lactiflora Pall.)을 1993년 6월 17일 수확하고, 굵기가 27mm 이상인 뿌리를 선별하여 paeoniflorin 추출방법 및 HPLC 분석조건에 대한 조사결과를 요약하면 다음과 같다. 1. 초음파추출 및 환류추출법에 의한 작약근 추출물의 HPLC chromatogram은 상호간의 차이가 없었으며, 두 추출법 모두에서 paeoniflorin retention time이 10.2분대에 peak가 존재하였다. 2. 환류추출법에 의한 paeoniflorin 추출은 60min., 1회로 충분하였다. 3. 초음파추출에서 추출시간이 1hr.∼2hr. 까지의 paeoniflorin함량은 동일시간의 환류추출보다 각각 0.28%, 0.32% 낮게 평가되었고 3hr.∼4hr.의 추출에서는 두 방법간에 차이가 없었다. 4. 초음파 추출법은 분석작업이 용이함으로 다수계통의 상대적 비교시에는 환류추출법보다 효과적이다. 5. 작약추출물 중 paeoniflorin, albiflorin을 HPLC를 이용하여 분석할 때 높은 감도를 보이는 가장 효과적인 UV Detector 파장은 각각 240nm, 254nm이고, 동시정량시에는 254nm가 비교적 안정적이었다.

Application of the Microwave-assisted Process to the Fast Extraction of Isoflavone from the Waste Residue of the Soybeans

  • Due to the importance of isoflavone content in soybean extracts, the microwave-assisted process (MAP) was compared to the conventional extraction methods. For comparison of the three methods, all extraction parameters (solvent, sample to solvent ratio, temperature, etc.) were kept the same; the microwave extractor was operated at 187.5 W with an emission frequency of 2450 MHz under atmospheric pressure conditions and the extractions were carried out at 75 ${^{\circ}C}$ for 3 min while the conventional reflux was at 75 ${^{\circ}C}$ for 3 h. Total yield and crude isoflavone content were determined by ultraviolet spectrophotometric and compared with the three methods. Results indicated that the MAP was comparable to the conventional method in its capability to extract target compounds without causing any degradation; in addition it dramatically reduced the extraction time from 3 h to a few minutes, suggesting that it can be an alternative technique to the time-consuming conventional reflux method.

열수추출조건이 동결건조 오미자의 추출 및 항산화 특성에 미치는 영향 (Effect of Reflux Conditions on Extraction Properties and Antioxidant Activity of Freeze Dried-Schisandra chinensis)

  • 오미자 유효성분의 식품 소재화를 위한 기초 자료를 얻기 위해 물과 에탄올을 이용하여 에탄올 농도(%), 추출온도($^{\circ}C$), 추출시간(h)을 달리한 추출물의 이화학적 특성과 항산화 작용을 확인하였다. 환원당 함량은 추출온도가 증가할수록 증가하였고 에탄올 농도는 50%, 추출시간은 4시간 추출하였을 때 높은 함량을 나타내었다. 산도는 에탄올 농도 50%, 추출시간 8시간, 추출온도 $100^{\circ}C$일 때 가장 높은 적정 산도를 나타내었다. 색도는 명도($L^*$)는 추출온도, 추출시간이 증가할수록 감소하는 것을 알 수 있었고, 오미자의 주 색상인 적색도($a^*$)는 에탄올 농도, 추출온도, 추출시간이 증가할수록 감소하는 경향을 보였다. 총 폴리페놀 함량은 50% 에탄올 추출물에서 가장 높은 함량을 나타내었고 추출시간과 온도가 증가할수록 유의적으로 증가하였다. 이와 같은 경향은 DPPH 및 ABTS 라디칼 소거능에서도 유사한 경향을 나타내었다. 각 추출조건별 실험 인자들의 상관성을 분석한 결과, 에탄올 농도는 총 페놀 및 ABTS 항산화능과 매우 높은 상관성을 나타내었다. 추출시간과 온도에 따른 총 페놀함량은 DPPH 및 ABTS 항산화능, 환원당 함량, 적색도, 적정산도에서 강한 상관관계를 나타내었고, 각 실험 항목 사이에서도 모두 높은 상관성을 나타내었다. 이상의 결과로 볼 때 오미자의 최적 추출조건은 용매는 50% 에탄올, 추출시간 4-6시간, 추출온도는 $60-80^{\circ}C$의 조건이 가장 바람직하다고 사료된다.

Antioxidant activity and phenolic acid content of Gynostemma pentaphyllum leaves according to extraction conditions

  • This study was intended to provide basic data for a health functional food study by exploring antioxidant activity of reflux extract according to the concentration of ethanol and the extract of ultrasonic waves extracted and reflux extracted under the same solvent conditions. In the same solvent condition, the reflux extract ($75.10{\pm}1.99mg$) showed a higher total phenol content than the ultrasonic wave extract ($51.74{\pm}2.28mg$). Flavonoids also had a higher reflux extract ($25.05{\pm}1.53mg$) than did ultrasonic extracts ($16.23{\pm}1.95mg$). Reflux extract according to ethanol concentration was found to have a higher phenol content than the 70% ethanol extract ($40.60{\pm}1.49mg$) in 60% ethanol extract. Flavonoid content was also similar to phenol content in reflux extract as determined by ethanol concentration from 60% ethanol ($25.05{\pm}1.53mg$) to 70% ethanol extract ($6.60{\pm}0.46mg$). In addition, the antioxidant activity (DPPH, TEAC, FRAP, ORAC) of the reflux extract in the same solvent conditions tended to be higher than that of ultrasonic extracts. Also, 60% ethanol extract had better antioxidant activity than 70% ethanol extract. However, an analysis of phenolic acid content through HPLC showed that the ultrasonic extract had a higher content in the same solvent condition than did the reflux extract. Not only the presence of phenolic acid, but also those of other compounds are believed to be attributed to the activity of antioxidants. Therefore, further studies are needed to clarify this phenomenon.

더덕 사포닌인 lancemasides의 최적 추출 방법 구명 (Determination the optimum extraction method for saponin lancemasides in Codonopsis lanceolata)

  • 본 연구는 더덕 사포닌의 최적 추출 방법을 구명하기 위해 수행되었다. 추출 방법에 따른 lancemasides 함량을 조사하기 위해 환류 추출(MeOH, BuOH), 열수 추출(Hot water), 초음파 추출(ultrasonic bath (40 kHz; continuous irradiation, interval irradiation), ultrasonicator (20 kHz))을 실시하였다. 그 결과, 총 lancemasides함량은 ultrasonic bath (MeOH; continuous irradiation) 추출에서 가장 높았고, ultrasonic bath (water; continuous irradiation) 추출>ultrasonic bath (MeOH; interval irradiation) 추출>ultrasonicator 추출>열수 추출>메탄올 환류 추출>부탄올 환류 추출 순으로 높았다. 따라서 더덕 사포닌의 최적 추출 방법은 MeOH을 이용한 초음파(ultrasonic bath, 40 kHz) 추출이었고, 추출 전 시료 건조 방법은 동결건조보다 음건으로 건조하는 것이 lancemasides 추출에 더 효과적이었다. 또한 초음파 추출 시 추출 시간은 2시간, 추출 온도는 $64^{\circ}C$에서 추출 하는 것이 효과적임을 확인할 수 있었다. 따라서 초음파(ultrasonic bath)추출은 기존에 주로 이용하던 환류 추출 및 분획을 실시했을 때 보다 많은 양의 사포닌(lancemasides)을 추출할 수 있었다. 그리고 추출 시간과 온도와 같은 추출 조건에 따라 aster saponin Hb가 lancemaside류로 성분 변화가 일어나는 것을 확인하였다. 특히 추출 온도가 증가함에 따라 aster saponin의 함량이 줄어들고, lancemaside A의 함량이 증가하는 것으로 보아 추출 온도가 사포닌 성분 변화에 영향을 주는 것으로 사료된다.
