  • The effect of thiol compounds on development and intracellular glutathione(GSH) concentrations of bovine embryos produced by in vitro maturation and in vitro fertilization(IVM/IVF) was examined in CRlaa medium with or without $\beta$-mercaptoethanol(0, 10, 25 and 50$\mu$MME) and cysteamine(0, 25, 50 and 75 $\mu$M). Numbers of cells comprising blastocysts were also counted using double fluorescence stain and the total glutathione levels(oxidized and reduced form) of morula and blastocyst embryos were than measured by an enzymatic method. Following routine IVM/IVF procedures oocytes and zygotes were cultured for 40 to 44h in CRlaa medium. Then 2 to 8-cell embyos had cumulus cell removed and were allotted randomly to the experimental medium. In Experiment 1, the proportion of embryos developing to and beyond morulae stages in 0, 10, 25 and 50 $\mu$M $\beta$-ME was 42.9%, 50.0%, 53.7% and 65.6%, respectively. Fifty $\mu$M $\beta$-ME group was significantly higher than those of any other groups (P<0.05). In Experiment 2, the percentages of embryos developed beyond morulae stages in 0, 25, 50 and 75 $\mu$M cysteamine was 42.9%, 40.4%, 60.0% and 59.2%, respectively. Fifty and 75$\mu$M cysteamine groups were significantly higher than in 0 and 25 $\mu$M cysteamine groups, but all of culture medium containing cysteamine(52.6%) was not significantly difference in control group(42.9%). In Experiment 3, the intracellular GSH concentrations of morulae and blastocyst embryos in 0 and 50 $\mu$M $\beta$-ME was 42.4 pM and 44.9 pM, 49.5 pM and 67.8 pM, respectively. Morulae embryos were not difference, but blastocyst embryos were significantly difference between treatments(P<0.05). In Experiment 4, the intracellular GSH concentrations of morulae in CRlaa with or without cysteamine were 39.8 pM and 45.6 pM, and blastocysts were 59.3 pM and 66.8 pM, respectively. Cell numbers of blastocysts were similar to in all experimental groups. These experiments indicate that thiol compounds can increase the proportion of embryos that developing to and beyond morulae stage and the intracellular GSH concentrations.

Contribution of Arginine 13 to the Catalytic Activity of Human Class Pi Glutathione Transferase P1-1

  • Arg13 is a conserved active-site residue in all known Pi class glutathione S-transferases (GSTs) and in most Alpha class GSTs. To evaluate its contribution to substrate binding and catalysis of this residue, three mutants (R13A, R13K, and R13L) were expressed in Escherichia coli and purified by GSH affinity chromatography. The substitutions of Arg13 significantly affected GSH-conjugation activity, while scarcely affecting glutathione peroxidase or steroid isomerase activities. Mutation of Arg13 into Ala largely reduced the GSH-conjugation activity by approximately 85 - 95%, whereas substitutions by Lys and Leu barely affected activity. These results suggest that, in the GSH-conjugation activity of hGST P1-1, the contribution of Arg13 toward catalytic activity is highly dependent on substrate specificities and the size of the side chain at position 13. From the kinetic parameters, introduction of larger side chains at position 13 results in stronger affinity (Leu > Lys, Arg > Ala) towards GSH. The substitutions of Arg13 with alanine and leucine significantly affected $k_{cat}$, whereas substitution with Lys was similar to that of the wild type, indicating the significance of a positively charged residue at position 13. From the plots of log ($k_{cat}/{K_m}^{CDNB}$) against pH, the $pK_a$ values of the thiol group of GSH bound in R13A, R13K, and R13L were estimated to be 1.8, 1.4, and 1.8 pK units higher than the $pK_a$ value of the wild-type enzyme, demonstrating the contribution of the Arg13 guanidinium group to the electrostatic field in the active site. From these results, we suggest that contribution of Arg13 in substrate binding is highly dependent on the nature of the electrophilic substrates, while in the catalytic mechanism, it stabilizes the GSH thiolate through hydrogen bonding.

  • It has been known that quercetin is one of bioflavonid compounds and has anti-tumor effect by suppressing tumor growth in vitro and in vivo, including multiple biological effects by antioxidant and effective anti-inflammatory agent. The present study investigated whether quercetin can enhance antioxidant enzyme activity (glutathione proxidase: GPX, superoxide dismutase: SOD, catalase: CAT) and regulate the intracellular reactive oxygen intermediate levels on the B16F10 murine melanoma cells in the presensece of vitamin E, L-ascorbic acid (vitamin C) and reduced glutathione (GSH). 도라지 부탄올 추출물의 항산화 및 nitric oxide 생성 저해 효과 (Inhibitory Effect of Extracts of Platycodon grandiflorum (the Ballon Flower) on Oxidation and Nitric Oxide Production)

  • 본 연구에서는 도라지 분말을 유기용매로 추출한 후 도라지의 추출물과 분획물들에 의한 세포 내 활성산소종 및 glutathione (GSH)를 측정하여 항산화효과를 검토하였고 NO 생성 저해 효과를 알아보았다. 세포 내 활성산소종 생성억제 실험에서 건조 도라지의 A+M 및 MeOH 추출물과 추출물을 n-hexane, 85% aq. MeOH, n-BuOH, water로 다시 추출하여 얻어진 각각의 분획물들을 농도별로 HT1080 세포에 처리하였을 때 A+M과 MeOH 추출물 모두 측정시간 120분 동안 $500\;{\mu}M$ $H_2O_2$만을 처리한 control군에 비해 세포 내 활성산소종을 크게 억제시켰으며, MeOH 추출물에 의한 항산화 효과가 더 높게 나타났다. 또한, 각 분획물들 중 n-BuOH 분획물이 다른 분획물들에 비해 우수한 항산화 활성을 보였다. GSH 농도 측정 실험에서 A+M 및 85% aq. MeOH 분획물를 처리했을 때 GSH 함량이 증가하였다. NO 생성 저해 실험에서는 A+M 및 MeOH 추출물이 0.01 및 0.05 mg/mL의 농도에서 NO 생성을 억제하는 것으로 나타났으며, A+M 추출물에 의한 저해 효과가 높았다. 각 분획물들은 모두 control보다 낮은 NO 생성량을 나타내었으며, 특히 85% aq. MeOH 및 n-BuOH 분획물은 0.05 mg/mL 농도에서 blank에 가까운 NO 생성 억제율을 나타냈다. 이상의 연구결과로부터 n-BuOH 분획물에 의한 세포 내 활성산소종 생성 억제 효과 및 NO 생성 저해 효과가 우수함을 알 수 있었으며, 향후 분획물의 분리 정제를 통한 새로운 기능성 물질의 개발이 필요할 것으로 사료된다.

Biochemical Characterization of Transgenic Tobacco Plants Expressing a Human Dehydroascorbate Reductase Gene

  • Dehydroascorbate (DHA) reductase (DHAR, EC 1.8.5.1) catalyzes the reduction of DHA to reduced ascorbate (AsA) using glutathione (GSH) as the electron donor in order to maintain an appropriate level of ascorbate in plant cells. To analyze the physiological role of DHAR in environmental stress adaptation, we developed transgenic tobacco (Nicotiana tabacum cv. Xanthi) plants that express a human DHAR gene isolated from the human fetal liver cDNA library in the chloroplasts. We also investigated the DHAR activity, levels of ascorbate, and GSH. Two transgenic plants were successfully developed by Agrobacterium-mediated transformation and were confirmed by PCR and Southern blot analysis. DHAR activity and AsA content in mature leaves of transgenic plants were approximately 1.41 and 1.95 times higher than in the non-transgenic (NT) plants, respectively In addition, the content of oxidized glutathione (GSSG) in transgenic plants was approximately 2.95 times higher than in the NT plants. The ratios of AsA to DHA and GSSG to GSH were changed by overexpression of DHAR, as expected, even though the total content of ascorbate and glutathione was not significantly changed. When tobacco leaf discs were subjected to methyl viologen at $5\;{\mu}M$, $T_0$ transgenic plants showed about a 50% reduction in membrane damage compared to the NT plants.

Protective Role of Selenium and High Dose Vitamin E against Cisplatin - Induced Nephrotoxicty in Rats

  • Background: Cisplatin (CDDP) is one of the most active cytotoxic agents in the treatment of cancer. We investigated the effect of selenium (Se) with high dose vitamin E (VE) administration to prevent CDDP-induced nephrotoxicity in rats. Materials and Methods: In this study, 40 female Wistar rats were randomly divided into five equal groups. The first group, which served as the control, was administered physiological saline (2.5 cc/day, 5 days) intraperitoneally (IP), while group A was administered cisplatin (6 mg/kg BW/ single dose) plus physiological saline IP. Groups B, C, D received IP five doses of Se (1.5 mg/kg BW), and a high dose of VE (1000 mg/kg BW) (Se-VE) in combination before, simultaneously, and after CDDP, respectively. The rats were sacrificed five days after CDDP administration. Plasma malondialdehide (MDA), glutathione peroxidase (GSH-Px), reduced glutathione (GSH), catalase, urea, creatinine levels, renal histopathological changes were measured. Results: The histopathological injury score, plasma levels of MDA, urea, creatinine were found to increase in group A compared to the control (p<0.05), while plasma levels of GSH-Px, GSH and catalase decreased (p<0.05). In contrast, plasma levels of MDA decreased (p<0.05) in groups B, C, D, which were treated with Se- VE, whereas levels of GSH-Px, GSH were found to increase only for group D (p<0.05). Plasma urea, creatinine levels improved in the treatment groups compared to group A (p<0.001). Histopathological changes caused by CDDP were also significantly improved after Se-VE treatment (p<0.05). Conclusions: Oxidative stress increases with CDDP-induced nephrotoxicity in rats. Se-VE supplementation might thus play a role in the prevention of CDDP-induced nephrotoxicity in patients.

Red Wine Prevents Brain Oxidative Stress and Nephropathy in Streptozotocin-induced Diabetic Rats

  • We have studied the effects of red wine on brain oxidative stress and nephropathy in streptozotocin (STZ)-induced diabetic rats. Diabetes was induced in Wistar rats with a single intraperitonally injection of STZ (50 mg/kg). Two weeks before and four weeks after injection, red wine was given orally in both normal and diabetic rats. Blood samples were taken from the neck vascular trunk in order to determine the glucose, triglycerides, total cholesterol, HDL-cholesterol (HDL-c), atherogenic index (AI), total protein, blood urea nitrogen (BUN), creatinine, insulin, lipid peroxidation products, reduced glutathione (GSH) and superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) activities. As well, we estimated the lipid peroxidtion, GSH and SOD, GSH-Px and catalase activities in brain and renal homogenates, and the excretion of albumin, proteins and glucose in urine over 24 h period. The administration of STZ caused significant increases in levels of glycosuria, proteinuria, albuminuria, glycemia, total cholesterol and AI, as well as in lipid peroxidation products in the brain, plasma and kidney, whereas it decreased the GSH content and SOD, GSH-Px and catalase activities. Treatment with red wine significantly prevented the changes induced by STZ. These data suggested that red wine has a protective effect against brain oxidative stress, diabetic nephropathy and diabetes induced by STZ, as well as it protects against hypercholesterolemia and atherogenic risk.

도토리추출물이 흰쥐의 체내 항산화효소계에 미치는 영향 (Effect of Acron Extracts on the Antioxidative Enzyme System)

  • This study was conducted to investigate the effect of acorn extracts on the antioxidative enzyme system. Male Sprague-Dawley rats(110$\pm$10g) were fed on containing normal and high fat diets. They were orally administrated (0.02g/100g B.W) of acorn ethylacetate-extract and water-extract at the same time once a day, respectively. Net weight gain and feed efficiency ratio were increased in high fat diet groups and decreased by acorn extracts administration. The effect of acorn extracts on hepatic glutathione peroxidase(GSH-Px), glutathione S-transferase(GST) and catalase activities did not show significancy in normal fat diet groups. GST and catalase activities and lipid peroxidation content(LPO) were significantly increased in high fat diet groups and this increment were decreased by acorn extracts administration. However GSH-Px activity and GSH content were decreased in high fat diet groups but increased by acorn extracts administration. The activities of lactate dehydrogenase(LDH), alkaline phosphatase(ALP) and aminotransferase in serum were significantly increased in high fat diet group but these increment reduced in acorn extracts administration groups. These results indicate that acorn extracts could improve the liver function and prevent the metabolic diseases by hyperlipidemia.

3-(4′-hydroxyl-3′, 5′-dimethoxyphenyl) Propionic Acid Suppresses NO Production and Elevates GSH Levels in Murine Macrophages

  • Previous studies have shown that kimchi and kimchi-derived 3-(4'-hydroxyl-3', 5'-dimethoxyphenyl) propionic acid have anti-oxidative and hypolipidemic effects in rats and rabbits. This study was designed to investigate whether chemically synthesized 3-(4'-hydroxyl-3', 5' -dimethoxyphenyl) propionic acid (HDMPPA) may ameliorate oxidative stress through the regulation of nuclear factor KB (NFkB) activation in lipopolysaccharide (LPS)-stimulated RAW 264.7 murine macrophage cells. Treatment of RAW 264.7 cells with 400 uM of HDMPPA significantly reduced LPS-stimulated nitric oxide (NO) production. Treatments with HDMPPA at 100 uM to 400 uM concentrations significantly elevated glutathione (GSH) level. However, cell viability and thiobarbituric acid-reactive substances (TBARS) concentrations were not affected by the concentrations of HDMPPA used. The specific DNA binding activities of NFKB, a transcription factor which is sensitive to oxidative stress, were not down-regulated by HDMPPA treatments. These results suggest that HDMPPA may have weak anti-oxidative activity against LPS challenge by scavenging NO and stimulating GSH production.

Hepatoprotective Activities of Rubus coreanus Depends on the Degree of Ripening

  • Rubus coreanus, commonly known as 'red raspberry' is used as a traditional oriental medicine in Korea for the management of diseases such as impotence, spermatorrhea and athsma, and for allergies, in combination with other herbal preparations, in many centuries. We undertook a comparison of the hepatoprotective effect of ethanol extracts of the unripe (UREx) and ripe (RREx) R. coreanus extract against acetaminophen (AAP) induced hepatotoxicity in rats. UREx reduced the elevated alanine aminotransferase (ALP), aspartate aminotransferase (AST), total bilirubin (TB), alkaline phosphatase (AP), lipid peroxide and nitric oxide content which had been increased by AAP administration. UREx also increased the cellular glutathione (GSH) content and induced the glutathione-S-transferase (GST), glutathione peroxidase (GSH-Px) content which had been decreased by AAP. RREx did not exhibit strong hepatoprotective effect or antioxidant activity under the same conditions. The experimental results show that the degree of the ripening of R. coreanus affects the hepatoprotective activity in the AAP-intoxicated rats. These findings of a protective mechanism are supportive evidence for the utility of unripened R. coreanus in traditional medicine for liver ailments.
