Three different strains of carotenoid accumulating XantlwphyUomyces dendrorhous mutants, JH1, JH2, and JH3, were isolated by NTG (N-methyl-N'-nitro-N-nitrosoguanidine) mutagenesis, which might potentially be useful for animal feed as well as for studies on the regulation and biosynthesis of astaxanthin. Mutants were selected based on the capability of growth and carotenoid production on the YM agar plate containing chemical inhibitor, $\beta$-ionone. Astaxanthin-overproducing mutant JH1 produced 4.032 mg astaxanthinlg dry cell weight, and this value was about 15-folds higher than that of wild-type. $\beta$-Carotene-overproducing mutant JH2 produced 0.273 mg $\beta$-carotene/g dry cell weight, and this was 4-folds increase from that of wild-type. In contrast, JH3 was a white-colored mutant that was unable to produce carotenoid pigment.

Red ginseng(RG, steamed and dried root of Panax ginseng C. A. Meyer, family Araliaceae) and fermented red ginseng(FRG, fermented red ginseng by yeast and lactic acid bacteria) are known to show different pharmacological effects by changed composition of saponins through fermentation. We examined the effects of RG and FRG on $\beta$-hexosaminidase secretion, ICAM-1 expression, the mitogen-induced proliferation of lymphocyte from mice in ex vivo systems and HaCaT cell(keratinocyte) proliferation to compare the anti-allergic and anti-inflammatory effects between both groups. RG groups showed inhibition of $\beta$-hexosaminidase secretion and ICAM-1 expression at $1{\mu}g/ml$, $10{\mu}g/ml$ and the same effects were observed at all concentrations in FRG groups. In our study, RG increased LPS-induced B cell proliferation at $1{\mu}g/ml$ and ConA-induced B cell proliferation at $100\;{\mu}g/ml$ but FRG decreased LPS- and ConA-induced lymphocytes at $100\;{\mu}g/ml$. We showed that FRG increased the proliferation of HaCaT at 1, $10{\mu}g/ml$ but not by RG. These findings suggest that RG and FRG might have anti-inflammatory and anti-allergic effects, which can be needed to proper clinical concentration to applied to various allergic diseases and inflammation.
This study was to investigate beneficial effects of microbial mixture on red pepper which was capable of promoting plant growth by solubilizing insoluble phosphate as well as protecting plants from pathogenic attack. Saccharomyces sp. L13 was isolated for phosphate solubilizing activity on aluminium phosphate, tricalcium phosphate, calcium hydrophosphate, and magnesium hydrophosphate. On the other hand, Bacillus sp. L32 was isolated for antagonistic activity against Phytophthora capsisi and Colletotrichum gloeosporioides, causing Phytophthora blight and Anthracnose disease in pepper, respectively. The strain L32 exhibited antagonistic activities both under dual culture assays and detached leaves assays. The each strain under the condition of mixed cultivation exhibited the same growth rates as one under pure cultivation. In greenhouse study, the mixed culture showed the both effect of plant growth promotion and reduction of disease symptom development against P. capsisi and C. gloeosporioides providing a potential as effective microbial agent for plant husbandry.

To maintain the quality of sweet persimmon during storage, the samples were treated with a combination of 50 ppm $ClO_2$ and 0.1% fumaric acid after harvest, packaged with low density polyethylene film, and stored at $1{\pm}1^{\circ}C$ for 35 days. The combined treatment reduced the populations of total aerobic bacteria, yeast and molds by 1.82 and 2.07 log CFU/g, respectively, compared to the control. During storage, hardness of all samples decreased, but total soluble solids and weight loss were not significantly different among treatments. In addition, high-quality dehydrated sweet persimmon was prepared using red algae extract as a dehydrating agent. The rehydration ratio and vitamin C content of red algae extract-treated sample were greater than those of hot-air dried sample. These results suggest that the combined treatment of $ClO_2$ and fumaric acid can be useful for maintaining microbiological safety of sweet persimmon during storage and dehydration of sweet persimmon slices using red algae extract is an efficient drying method for the preparation of high-quality dehydrated sweet persimmon.
Muscat Bailey A grapes, one of the major grape varieties in Korea, contain 18-20% (w/w) sugars, which is less than the amount required to make red wine. In the present study, fermentation of freeze-concentrated Muscat Bailey A grape juice to $24^{\circ}Brix$ was performed using several industrial wine yeasts, including Saccharomyces cerevisiae $OC_2$, S. cerevisiae Fermivin and S. cerevisiae W-3. During fermentation, changes in the levels of soluble solids, alcohols, and yeast viable counts were monitored. Wine quality parameters including organic acid, minor alcohol, sensory score, etc. were also analyzed. Alcohol contents reached maximal levels after 9 days of fermentation, and were 12.6% (v/v) when Fermivin was used and 13% (v/v) when strains $OC_2$ and W-3 were used. No major difference between strains was apparent, except that Fermivin fermentation was somewhat slower in terms of both alcohol production and sugar consumption. Similar levels of soluble solids and total phenols were measured in wines fermented by each of the three strains. The total acid level of W-3 wine was high, whereas the alcohol content of Fermivin wine was low. Although the wines were variable in terms of acetaldehyde and minor alcohol contents, the levels of these materials were much lower than the limits set by the Korean National Tax Service. Upon sensory evaluation, $OC_2$ wine obtained the highest scores in terms of color, flavor, and overall acceptance. However, the best taste score was awarded to the Fermivin wine, which had the same flavor score as $OC_2$ wine but a slightly lower overall acceptance score.
An improved procedure for the rapid purification of glucose-6-phosphate dehydrogenase from extracts of Saccharomyces cerevisiae was developed by using affinity chromatography. Among six affinty media tested, $NADP^+ -agarose$ and Affi-gel Blue were more effective than others (i.e., Affi-gel Red, AMP-agarose, ATP-agarose, and $NAD^+ -agarose$). Conditions to desorb the enzyme bound to the affinity media were examined to increase the purity as well as yield. The best result was obtained when the column was developed with a linear gradient of KCl (0-1.0M). In case of Affi-gel Blue, introduction of $NAD^+$ (15mM) washing step prior to the salt gradient was most effective to remove $NAD^+ -binding$ proteins. For a large scale preparation of G-6-P dehydrogenase higher recovery was obtained by Affi-gel Blue than $NADP^+ -agarose$, however, the purity of the enzyme was decreased by 10 times if the former was used as the affinity medium. The capacity of Affi-gel Blue for G-6-P dehydrogenase was found to be 5 times higher than that of $NADP^+ -agarose$. Furthermore Affi-gel Blue could be reused repeatedly and its preparation is relatively easier and less expensive than $NADP^+ -agarose$.
The red ginseng extract and its components were investigated for their activation effects on the growth of Saccharomyces cerevisiae IAM and Saccharomyces formosensis No. 396 IAM. Changes in the number of cells, alcohol production, $CO_2$ evolution, pH and the rate of sugar consumption and of fermentation were compared during growth at $30^{\circ}C$ for 120 hours. The addition of ethanol extract and saponins from red ginseng were found to exihibite a significant increase in all physiological activaties of yeast, and its maximum activites were obtained at 1.5% ethanol extract concentration. The physiological effects of panaxadiol and panaxatriol, two major groups of saponin, were also compared to those of crude saponin and found that the former showed a small increase in physiological changes. However the difference was not significant. The overall contents of ginsenosides of ethanol extract and crude saponin during fermentation were not significantly affected by the growth of roasts, except a small increase in ginsenoside $-Rg_2$ and decrease in -Rd.

This study was conducted to investigate the effects of antibiotic and yeast supplemental high energy diet on growth performance, blood characteristics and carcass trait in broilers. Total of four hundred-eighty broilers were randomly allocated into three treatments with eight replications for five weeks. Dietary treatments included 1) CON (control; basal diet), 2) HED (high energy diet) and 3) YD (HED; yeast added to HED instead of virginiamycin, Sacchromyces cerevisiae, $15{\times}10^{10}$). During whole period, weight gain had high tendency in HED treatment. However, there were not significant among treatments (P>0.05). Feed intake was higher in YD treatment than others. However, there were not significant among each treatments (P>0.05). Feed/Gain ratio was significantly lower in HED treatment than others (P<0.05). In blood characteristics, RBC, WBC and lymphocyte were not significant (P>0.05) among treatments. Liver weigh, LW/BW ratio, leg meat weigh, LMW/BW ratio, breast meat weigh, BMW/BW ratio, abdominal fat weigh and AF/BW ratio were not significant (P>0.05). However, body weight was improved (P<0.05) in HED treatment. In conclusion, this experiment is shown that HED treatment affects Feed/Gain ratio and body weight at final period in broilers.
Thermal acid hydrolysis pretreatment of Kappaphycus alvarezii was carried out with 12% (w/v) seaweed slurry and 180 mM H2SO4 at 140°C for 5 min. Utility of the thermotolerant yeast Kluyveromyces marxianus KCTC7150 was evaluated with respect to cell growth and ethanol fermentation at 40°C was close to optimal for enzymatic hydrolysis. This could lead to the integration of both the saccharification and fermentation processes. The levels of ethanol production by simultaneous saccharification and fermentation (SSF) with non-adapted and adapted K. marxianus KCTC7150 were 9.1 g/l with an ethanol yield (YEtOH) of 0.24 and 10.2 g/l with an ethanol yield (YEtOH) of 0.27 at 156 h, respectively. The two-phase SSF process was employed in this study to improve the efficiency of ethanol fermentation. Adapted K. marxianus KCTC7150 using the two-phase SSF process produced 13.5 g/l with an ethanol yield (YEtOH) of 0.35 at 96 h. Development of the two-phase SSF process could enhance the overall ethanol fermentation yields of the seaweed K. alvarezii.
The changes in pH, acid contents and microbial counts were investigated during fermentation of Baikkimchi, a kind of Kimchi without red pepper, and the major microbial groups were also isolated and identified. Immediately after the preparation of Baikkimchi(pH 6.15, acid contents 0.03%), its major microbial group was Gram negative rods, and was composed of Pseudomonas(55%), Enterobacter(40%) and Erwinia(5%). After 2 days of fermentation at 15$^{\circ}C$, the most predominant microbial group was changed to lactic acid bacteria. Lactic acid bacteria showed 1st, 2nd and 3rd stationary phase on its growth curve in 4, 12 and 50 days of fermentation, respectively. At the 2nd stationary phase of lactic acid bacteria(pH 3.51, acid contents 0.59%), the group was composed of Lactobacillus bavaricus(55%), Leuconostoc mesenteroides subsp. mesenteroides(42.5%) and Leuconostoc paramesenteroides(2.5%), while at the 3rd stationary phase(pH 3.40, acid contents 1.10%), that was Lactobacillus plantarum(65%) and Lactobacillus brevis(35%). The physiological and biochemical characteristics identified as Leuconostoc mesenteroides subsp. mesenteroides, Leuconostoc paramesenteroides, Lactobacillus plantarum and Lactobacillus brevis showed good agreement with the current taxonomic system, but those identified as Lactobacillus bavaricus showed some disagreements. The number of yeast was decreased wit the increase in the number of lactic acid bacteria. Yeast showed stationary phase in 30 days between the 2nd and 3rd stationary phase of lactic acid bacteria, and the group was composed of only gunus Saccharomyces.
