Paraquat 유도 산화적 스트레스에 대한 홍삼 사포닌의 항산화 효과 (Antioxidative Effects of Red Ginseng Saponins on Paraquat-induced Oxidative Stress)

  • 본 연구에서는 PQ의 독성과 홍삼 성분의 항산화 활성성분을 알아보기 위해 생쥐를 이용하여 PQ 투여 후 총 사포닌, PPD, PPT성분을 1, 3, 7일간 경구 투여한 후, 간 조직에서의 SOD, CAT GPx의 활성과 GSH, MDA 및 과산화수소 함량등을 측정 비교함으로써 홍삼의 항산화활성 성분이 PQ 독성에 대한 회복 작용 그리고 지질과산화와의 상호 관련성을 검토하였다. 항산화 효소인 SOD, CAT GPx활성도는 전반적으로 PPD계가 높았고, SOD 활성은 PPD 투석군에서 전일에 걸쳐 가장 높은 활성을 보였다. 과산화수소의 함량은 PQ 투여군 기준으로 PPD 투여군에서 가장 낮은 결과를 보였다. 자유 라디칼에 의해 생성된 지질과산화의 최종산물인 MDA의 함량은 사포닌 투여군에서 유의성 있게 감소하였으며, 특히 PPD에서 다른 투여군들에 비해 현저한 함량 감소를 보였다. 이와 같이 사포닌의 항산화효과는 SOD, CAT및 GPx와 같은 항산화 효소의 직접적인 작용과 홍삼의 특정 성분들이 생체 내에서 내인성 항산화 물질의 합성능력을 강화시킴으로서 산화적 손상에 대한 회복작용을 향상시키는 결과로 생각된다.

  • One kind of fungus was isolated and identified from comtaminated red-ginseng in order to give fundermental data for improving hygienic quality of ginseng product. The isolated strain was identified as Aspergillus sp. Hyphae of the strain had septum structrue. The strain showed vesicle and sterigmata structure which were typical characteristics of Aspergillus species. The growth of the strain was slightly inhibited by sodium benzoate and potassium sorbate at a concentration of 0.05%. The strain showed no growth at 4.0% potassium sorbate. The isolated stain Aspergillus sp. showed no significant degradation in the presence of red-ginseng saponins.

  • From in vivo and in vitro experiments using Korean Red Ginseng and its main components. ginsenoside saponins, it was found that ginesenoside may exerts its anti-thrombotic action by decreasing $TXA_2$ formation in platelets and increasing $PGI_2$ formation in vascular walls. This may certainly contribute to prevention and development of atherosclerosis and thrombosis by administration of Korean Red Ginsengs.

인삼청량음료 제조에 관한 연구 (Studies on the Manufacturing of Soft Ginseng Drink Part I. Purification procedure of raw ginseng extracts)

  • Red ginseng tails were extracted with ethanol solutions over a range of concentrations and temperature conditions. Investigations were carried out to study the effects of treatments on yields, soluble solids, saponin and precipitate occured in red ginseng extract beverage during storage. It was found that: (1) Higher concentration of ethanol at low temperature resulted in less yield of crude extract (2) The amount of precipitate in the non-purified extract beverage were less with decrease in ethanol concentration used (3) The treatment for purification of extracts and storage of purified extract at 37$^{\circ}C$ for 6 months had no effect on HPLC chromatogram pattern of saponins (4) The amount of purified extract decreased by purification treatment and more decrease was found as the temperature and concentration of ethanol increased. For Preparation of red ginseng extract beverage, the treatment of extracts with ethanol at low temperature was found to be more effective to minimize precipitation in tile beverage.

장기저장 홍삼의 품질안정성 (Quality Stability of Red Ginseng Stored for Long Periods)

  • Samples of red ginseng, which had been manufactured and packaged by the Korean Monopoly Corporation, were stored at ambient temperatures and humidities ($12{\sim}28^{\circ}$ and $55{\sim}68$ percent) for one to nine years to examine their overall quality stability. The proximate compositions, contents of 50% ethanol and water extracts of the samples and the TLC and HPLC patterns of ginsenosides in the samples remained almost unchanged in all cases. The lipids and fatty acids in the samples, which are otherwise susceptible to oxidation, were stable judged on the basis of the changes of the TLC and GLC patterns of the lipids and fatty acids. It was also found that polyunsaturated fatty acids such as linoleic(C18:2) and linolenic and(C18:3) present in the samples had been very stable during the long storage periods. It, therefore, seems that the autoxidations of the lipid and fatty acids of red ginseng were prevented by antioxidative compounds which will be progressively formed in red ginseng through non-enzymatic browning reactions during manufacturing process and long-term storage.

  • 인삼분말에 대한 전자선(electron beam)의 이용가능성을 연구하고자, 방사선 살균처리(2.5∼15 kGy)가 백삼 및 홍삼분말의 사포닌 함량 및 패턴, 페놀함량, 산성다당체 함량, amylase activity, 단백질 패턴 등에 미치는 영향을 감마선 조사와 비교하였다. 사포닌 함량과 패턴을 TLC 및 HPLC로 분석해 본 결과, 조사선량과 에너지원에 따른 변화는 확인되지 않았으며, 실온 저장 4개월 이후에도 안정된 경향을 보였다. 총페놀성분 및 산성다당체 함량은 홍삼이 백삼 보다 높은 함량을 보였으며, 산성다당체 함량은 조사선량의 증가로 높은 추출량을 보였다. 백삼분말의 amylase activity는 방사선조사 직후 증가하였으나, 4개월 이후에는 대조구 보다 낮은 간을 보였으며, 추출단백질의 전기영동 패턴은 15 kGy의 전자선 및 감마선구에서도 대조구와 차이가 없었다.

  • As a part of studies on the quality control of crude drug preparation drinks, ginseng saponins were identified by HPLC. Ginsenoside-Rb1 was determined quantitatively by HPLC. Ginsenoside MeOH/H2O(65:35:10, v/v) on Si-gel plate. Ginsenoside-Rb1 content determined by HPLC on Lichrosorbtract drinks was 57.5-70.4% compared to the content in the red ginseng extract.

  • Korean red ginseng has shown therapeutic effects for a number of disease conditions. However, little is known about the anti-inflammatory effect of Korean red ginseng saponin fraction (RGSF) in vitro and in vivo. Therefore, in this study, we showed that RGSF containing 20(S)-protopanaxadiol type saponins inhibited nitric oxide production and attenuated the release of tumor necrotic factor (TNF)-${\alpha}$, interleukin (IL)-6, granulocyte monocyte colony stimulating factor (GMCSF), and macrophage chemo-attractant protein-1 in lipopolysaccharide (LPS) stimulated murine macrophage RAW264.7 cells. Moreover, RGSF down-regulated the mRNA expressions of inducible nitric oxide synthase, cyclooxyginase-2, IL-$1{\beta}$, TNF-${\alpha}$, GMCSF, and IL-6. Furthermore, RGSF reduced the level of TNF-${\alpha}$ in the serum and protected mice against LPS mediated endotoxic shock. In conclusion, these results indicated that ginsenosides from RGSF and their metabolites could be potential sources of therapeutic agents against inflammation.

Further Evidence in Support of Psychotropic Action on Red Ginseng

  • Using an ethopharmacological technique, we demonstrated that saponin fraction from red ginseng root possessed a potent psychotropic actions on either intermale or maternal aggression models. A series of experiments clearly indicated that one of psychoactive ingredient is ginsenoside Rbl. Although a drug-induced debilitation of motor performance remains a possible cause of the antiaggressive effect of the drug, ginsenoside Rb 1 did not alter the locomotor activity of the mice during agonistic confrontations. Thus, one can eliminate the possibility that the psychoactive effect of ginsenoside Rbl might be concealed by a drugindulced impairment of motor performance. More recently, we developed a new model for copulatory disorder and introduced into the behavioral analysis of drug action. Male mice which has been housed individually from weaning for 5 weeks failed to manifest copulatory behavior when they encountered with the sexually-receptive females. Daily administration of crude ginseng saponin during isolation housing period prevented the development of copulatory disorder, whereas both ginsenoside Rbl and Rgl were ineffective. 인삼Saponin의 산가수분해물이 Epididymal Adipose Tissue의 지방대식에 미치는 영향 (Effect of Acid Hydrolyzates of Ginseng Saponins on Lipid Metabolism in Rat Epi didymal Adipose Tissue)

  • Studies were carried out to clarify the effect of ginsenoside-Rbl -Rbr and acid hydrolyzatps of ginsenoside-Rbl, -Rb2 (HRbl, HRbf) on lipolysis and lipogenesis induced by epinephrine, glucagon, ACTH (adrenocorticotrophic hormone), TSH (thyroid-stimulating hormone) and insulin in rat adipose tissilr. HRbl , HRb2 slightly inhibited lipolysis induced by epinephine. glucagon and TSH. ACTH-induced lipolysis in fat tissue slices was significantly inhibited by ginsenoside -Rbl, -Rb2, HRbl and HRb2, particulary HRb2. None of ginsenoside-Rbl, -Rb2, HRbl and HRb2 accelerated insulin-stimulated lipogenesis in fat calls. Among ginseng products, extract powder (freeze dried), extract powder (spray dried), red ginseng powder inhibited ACTH-induced lipolysis in adipose tissue slices, but red ginseng extract not affect them.
