Carbohydrate, Lipid Inhibitory Activity and Antioxidant Activity of Extracts from Several Economic Resource Plants in Vitro

  • The objective of this study was determined to evaluate ${\alpha}$-amylase, ${\alpha}$-glucosidase, pancreatic lipase inhibition in vitro and DPPH radical scavenging activity of the several Korean resources plants. The ${\alpha}$-amylase inhibitory activity of Salicornia herbacea, Erythronium japonicum (flower) and Phragmites communis (root) in water extract showed relatively high 62.8%, 66.5% and 69.3%, respectively. The ${\alpha}$-amylase inhibitory activity of Citrus junos (pericarp) and Cornus officinalis in methanol extract was found to have an effect with 32.8% in Citrus junos (pericarp) and 60.9% in Cornus officinalis. Corylopsis coreana in both water and methanol extract had the highest ${\alpha}$-glucosidase inhibitory activity of 81.7% and 89.5%, while the extract of Portulaca oleracea, Ficus carica and Citrus junos was not measured ${\alpha}$-glucosidase inhibitory activity at given experiment concentration. Depending on the extraction solvent and the plant species, it was observed that there was a significant difference in ${\alpha}$-glucosidase inhibitory activity. The pancreatic lipase inhibitory activity showed relatively higher in the methanol extract than water extract except pericarp of Citrus junos. The DPPH radical scavenging activity of selected plants was much difference between measured plant species, and showed that the increase was proportional to the concentration. These results suggested that selected plants had the potent biological activity on carbohydrate, lipid Inhibitory activity and antioxidant activity, therefore these plant resources could be a good materials to develop medicinal preparations, nutraceuticals or health functional foods for diabetes or obesity.

Sensory Evaluation and Bioavailability of Red Ginseng Extract(Rg1, Rb1) by Complexation with ${\gamma}$-Cyclodextrin (${\gamma}$-cyclodextrin으로 포접한 홍삼추출물의 관능평가 및 Rg1, Rb1의 생체이용율)

  • In order to reduce the bitter taste and improve the bioavailability of red ginseng extract(RGE), inclusion complexes (RGE-CD) of the extract with ${\alpha}-,\;{\beta}-,\;{\gamma}$-cyclodextrin were prepared and studied for their sensory quality and bioavailability compared to RGE. By complexation, the bitter taste-reducing efficacies of ${\alpha}$-CD and ${\beta}$-CD were much lower than that of ${\gamma}$-CD. In comparative sensory analysis for the bitter taste, RGE-${\gamma}$-CD10, prepared using 10%(w/w) of ${\gamma}$-CD, showed a score of 1.93(decreased by about 78%) compared to RGE as the control. In addition, in sensory analysis for flavor, RGE-${\gamma}$-CD10showed a score of 5.60. Upon increasing the amount of ${\gamma}$-CD to 15%(w/w) and 20%(w/w), respectively, the bitter taste of RGE-${\gamma}$-CD was removed and the flavor of RGE disappeared(scores of 2.67 and 1.67, respectively). Therefore RGE-${\gamma}$-CD10 was chosen as an optimum. The same dosages of RGE and RGE-${\gamma}$-CD10 were orally administered to SD(Sprague-Dawley) rats on a saponin basis, and the plasma concentrations of ginsenoside Rg1 and Rb1 were measured over time to estimate the average AUC(area under the plasma concentration versus time curve) of the ginsenosides. After the oral administration, there were no significant differences in the AUC values of the RGE and RGE-${\gamma}$-CD 10 groups for ginsenoside Rg1. However, AUC values for ginsenoside Rb1 were $25.8{\mu}g{\cdot}hr/mL$ in the RGE group and $81.5{\mu}g{\cdot}hr/mL$ in the RGE-${\gamma}$-CD 10 group, respectively. Therefore, the bioavailability of ginsenoside Rb1 in the RGE-${\gamma}$-CD 10 group was significantly higher by up to 315% compared with that in the RGE group(p = 0.0029). These results show that the bitter taste of RGE can be simultaneously removed by the complexation of RGE and ${\gamma}$-CD(RGE-${\gamma}$-CD) along with increased bioavailability.

Growth Characteristics of Candida parapsilosis Isolated from Deteriorated Ginseng Extract (인삼추출물로부터 분리한 Candida parapsilosis의 생육특성)

  • We investigated the growth parameters of Candida parapsilosis isolated from spoiled red ginseng extract. The optimum pH range for C. parapsilosis was 6.0, whereas the minimum and maximum pH values that permitted growth were 3.0 and 11.0, respectively. For cells grown in PG medium plus 0 and 60% sucrose, the optimum water activity(Aw) values were 0.98 and 0.97, respectively. The optimum temperature for C. parapsilosis were 3$0^{\circ}C$ at an Aw of 0.90 in 4.8% potato dextrose broth with 18% sucrose (PGS). Cations inhibiting the growth of C parapsilosis were L $i^{+}$ $Ca^{2+}$, $Mg^{+2}$, $K^{+}$ in decreasing order, while anions were S $O_4$$^{2-}$, N $O_3$$^{[-10]}$ , C $l^{[-10]}$ .TEX> .

  • To evaluate the quality of commercial dehydrated tea-products, the relationships between particle sizes, densities, moisture absorption & desorption and color appearance were studied by using Hunter-lab tristimulus colorimeter and spectrophotometer. Among the tea-products was held no significant relation between particle sizes and color appearance but red ginseng extract powder (RGEP) was included L, a and b values when was reduced particle size. appearance color of tea-products indicated red-orange color, L, a and b values were ranged 32.7 to 48.0, 4.0 to 10.0 and 5.6 to 18.0, respectively, densities of tea-products ranged 0.232 to 0.898 g/ml and increased L values, Hunter's a/b ratio values was included in 0.61 to 0.90. Color stability in this products was well agreed with decrease of total color difference value ($\Delta$E) and chromaticity difference value ($\Delta$C) of the Hunter-lab color data.

  • This study is on the feasibility of use of glycoprotein in various areas such as cosmetics and food etc. by extracting, isolating and refining glycoprotein from carrots, red ginseng extract residue, sesame and pine needles using protease(pepsin) and by analyzing general characteristics and measuring various bioactivities. The results of analysis of nutritional composition showed protein contents of glycoprotein. In the analysis of constitutive amino acids, the ratio of contents of hydroxy proline and glycine, the characteristics of glycoproteins appeared similar and the contents of glutamic acid and aspartic acid appeared higher. As a result of measurement contents of total polyphenol and flavonoid, it showed that glycoprotein had more contents generally, and the effect of bioactivity of glycoprotein appeared higher although different kinds of glycoprotein showed a little DPPH radical and nitrite scavenging ability, total antioxidant capacity by ABTS, ACE inhibitory.

  • 생쥐 신장의 근위세뇨관과 원위세뇨관 세포의 미세구조에 미치는 염화 메틸수은의 독성에 대한 홍삼유출물의 첨독성적 영향을 전자현징경적으로 연구하였다. 염화 메틸수은 처리군의 근위세뇨관 세포에서는 대조군에서 보다 철세 융모가 다소 축소되고 불규칙한 배열을 하였다. Brush border의 인접 부위에서는 작은 경계들이 나타나고 세포질 중앙 부위에서는 치밀체를 함유한 큰 육포들이 관색되었다. Mitochondria는 상당히 팽대되고 기양막은 부분적으로 비후되었으며 다수의 ITsosome이 나타났다. 염화 메틸수은 처리군의 원위세뇨관 세롱에서는 불규칙한 세포 표면, 양사된 세포, 그리고 다수의 ribosome과 소수의 지방사이 나타났으며 mitochondria 는 팽대되고 기고막은 부분적으로 비후되었다. 염화 메틸수은-홍삼추출물 병행 처리군의 근위세뇨관 세포에서는 염화 메틸수은 처리군에서 보다 mitochondria의 팽대 정도가 감소되었으며 육포의 크기와 수도 상당히 감소되었다. 염화 메틸수은-홍모적출물 병행 처리군의 원위세뇨관 세롱에서는 세포 표면이 어느정도 규칙적이고 mitochondria의 정해와 기저막의 비후 정도가 감소되어 정상 세포와 거의 유사하였다.

  • 생쥐의 체장, 신장 및 혈청내 LDH 활성에 있어서 간장과 신장에서는 염화메틸수은 투여군과 염화 메틸수진-홍삼추출물 투여군의 총 LDH 활성치가 대조군에 비하여 감소되었으나 염화 메틸수은-홍삼추출물 투여군의 활성치는 염화메틸수기 투여군 보다 약간 감소되었다. 혈청에서는 염화 메틸수은 투여군의 LDH 활성치가 대조군에 비하여 2배로 크게 증가되었으며 염화 메틸수은홍삼추출물 투여군의 활성치는 대조군과 유사하였다. LDH isosyme의 전기영동상에 있어서 간장의 대조군과 염화 메틸수은-홍삼적출물 투여군에서는 5가지 LDH isoBym삐 모두 나타났으나 염화 메틸수진투여군에서는 LDH1 제외한 4가지 LDH isosyme 나타났다. 신장과 혈청에서는 3군 모두에서 5가지 LDH isozyme 나타났다.

  • Caffeic acid is a kind of phytochemicals occurred in coffee, which is worked as a carcinogenesis restrainer and antioxidants on the human body. In this study, as one type of caffeic acid derivatives, methoxypolyethylene glycol caffeate(MPC) was synthesized by the esterification between caffeic acid and methoxypolyethylene glycol with N,N'-dicyclohexyl carbodiimide, 4-dimethylamino pyridine. The synthesized product was confirmed by using FT-IR and $^1H$-NMR, thin-layer chromatography. And these compound was investigated as antioxidant, Tyrosinase hindrance and skin moisturizers. In the free radical scavenging study, antioxidant effect of MPC was averagely high than the red ginseng extract that be used as a natural antioxidants. The result of Tyrosinase anti-activity test was better than embryo bud of rice extract at low concentration. At the iNOS anti-activity tested by using Raw 264.7 cell, and confirmed anti-inflammatory function. For the MPC handled with sodium lauryl sulfate, was tested for the skin moisture content and skin moisture loss.

Study on Consumption of Health Functional Foods according to Climacteric Symptoms in Middle-aged Women (중년 여성에서 갱년기 증상 정도에 따른 건강기능식품 섭취실태 조사 연구)

  • The purpose of this study was to analyze consumption patterns of health functional foods (HFF) according to climacteric symptoms. Subjects were 450 middle-aged women divided into four groups by Kupperman's index. This study collected all information by self-administrated questionnaires. Higher Kupperman's index was associated with higher Menopause-specific quality of life questionnaire (MENQOL) (P<0.001) and current smoker ratio (P<0.01) as well as lower monthly income (P<0.05). Exactly 28.4% of subjects showed negative responses to effectiveness of HFF. The average number of HFF intake per person was 2.01. The main reason for consumption of HFF was treatment or prevention of disease (33.6%). The information source for buying HFF was family or relatives (40.0%). The main types of HFF were vitamins (28.2%), omega-3 fatty acid (18.6%) and red ginseng (17.3%). The intake ratio of individual approval type (Cynanchum extract and Angeli extract) was significantly higher in subjects with high Kupperman's index (P<0.001). In conclusion, these results can be used as basic data that reflect middle-aged women's needs for HFF intake according to climacteric symptoms.
