  • When fish meat is washed for the processing of surimi, about 50ff of lipid in the fish meat is removed from the fish meat to the effluent. The removed lipid was easily recovered by centrifugation or filteration of wastewater washed fish meat. Then, the recovered lipid was utilized as a material of mayonnaise sauce processing. The major fatty acids in the recovered lipids are $C_{16:0},\;C_{18:0},\;C_{16:1},\;C_{20:5},\;and\;C_{22:6}$ Polyenoic fatty acids were composed of $33.6\%$ to total fatty acids. When the recovered lipid was substituted for soybean oil in processing of mayonnaise sauce, the maximum percentage of substitution ratio presumed to be $30\%$ according to viscosity, color difference, and emulsion stability evalution for the substituted ones.

  • The effect of temperature and ethanol concentration on lipids yield and free fatty acids were investigated during 1-5 times of 8 hours extraction. The crude lipids recovered was generally increased as the ethanol concentration increased while it was decreased at higher extraction temperature. Fatty acids composition in free fatty acids showed linoleum and plasmatic acids to be the major fatty acids, accounting 60% of 16 fatty acids identified. Fatty acids pattern was little affected by the changes in ethanol concentration while increase in temperature resulted significant changes in fatty acids composition.

  • Supercritical carbon dioxide ($SCO_2$) extraction was used to recover concentrated proteins and to remove lipids and odor causing compounds from anchovy. Engraulis japonicus $SCO_2$ was used as the solvent for extraction, which was performed in a semi-batch flow reactor. The experimental conditions used were pressure, 15-25 MPa; temperature, $40-60^{\circ}C$ and sample size, 500 ${\mu}m$. The proteins obtained under these conditions performed well in a sensory evaluation; moreover, effective lipids and odor removal was achieved. The stability and characteristics of the proteins recovered with different solvents were also evaluated. The samples were sterilized by processing with $SCO_2$. Escherichia coli was not detected after storage for several days. The sensory characteristics were found to be superior to those of a sample produced by hexane extraction. Thus, the protein concentrate was obtained at $60^{\circ}C$ and 25 MPa was deemed valuable as a foodstuff.

  • A lot of water soluble proteins and lipids are released from minced mackerel meat and lost into the washing waste during the leaching process of Kamaboko or surimi manufacture. The removed proteins and lipids are not only an edible things but also a big burden for treating the wastewater. In order to recover the proteins from the effluent and to use as food stuff, the "pH-shifting" treatment, a modified isoelectric point precipitation method, was tried. This method is based on a myogen-aggregation phenomenon, which occurs when a solution of sarcoplasmic proteins is acidified or alkalified beyond the critical pH zone of 2∼3 or 12∼13 respectively and then neutralized. The maximum amount of precipitation was obtained by shifting the pH of the wastewater from original pH to isoelectric point (pH 4) or alkali pH 12 and then changing to neutral pH. The precipitates were easily collected by filteration or centrifuging at 10,000rpm. The oils which were only floating in the washing wastewater are easily recovered by seperating with oil separator after pouring. The recovered proteins were slightly denaturated during this pH shifting precipitation process, while the composition of amino acids was good balance as a food.

  • The rats were fed with 12% ethanol with and/or without 0. l% ginseng saponin instead of water for 6 days, and the acetaldehyde level of liver and serum, and [$NAD^+$]/ [NADH] and [$NADP^+$]/[NADPH] ratios of the liver were investigated. Acetaldehyde level of ethanol fed group (control) in liver and serum was much higher than not-ethanol fed group (normal), but that of ginseng saponin containing ethanol fed group (test) was only slightly higher than that of normal group. Decrease of [$NAD^+$] / (NADH) ratio of test group was also much greater than that of control group. Distribution of the radioactivity in hepatic lipids after the [l-$^{l4}C$]-ethanol feeding intraperitonealy was investigated 30 minutes later. It was found that total radioactivity of the hepatic lipids of test group was much lower than that of control group. Analysis of individual lipids such as phospholipids, cholesterol, fatty acid and triglycerides showed that the depression of phospholipid biosynthesis and increase of fatty acid and triglycerides caused by ethanol feeding were significantly recovered by the co-feeding of ginseng saponin.

  • Mackerel water-soluble protein fraction produced by washing the mackerel meat were concentrated by isoelectric point shifting precipitation process, and the concentrates were utilized as the alternative feed of fish meal. In the 1st aquaculture diet experiment for Israel common carp, the feed conversion ratio decreased in proportion to the rise in the percentage of the recovered protein containing a residual lipid, which was added to the fish meal. It was supposed that the low feed efficiency was because of lipid oxidation in the recovered protein fraction. In addition, 2nd aquaculture diet experiment for Israel common carp was conducted after removing the oxidized lipid in the recovered protein fish meal. When a portion of the fish meal was substituted by the recovered protein devoid of the residual lipid, the feed conversion ratio increased in proportion to the amount of the substitute being added to the fish meal. Therefore, the recovered protein fraction of the mackerel washing wastewater from mackerel processing factory could be used as the alternative feed of fish meal.

Changes of Gangliosides Metabolism in Streptozotocin-Induced Diabetic Rats and Effect of Deer Antler (Streptozotocin 유발 당뇨병쥐 뇌에서 Gangliosides 대사 변화와 녹용의 효과)

  • In this study, we examined gangliosides from streptozotocin-induced diabetic rat brain. To obtain the diabetic rat brain, we sacrified the rat three days after injecting the streptozotocin into venus in tail. We measured blood glucose level according to Somogy-Nelson method and measured insulin level using $^{125}$ I-insulin RIA kit. The gangliosides were extracted according to Folch-Suzuki method from the rat brain. We also examined the effect of major lipid components extracted from deer antler on diabetic rat brain. The results showed that the major lipids components lowered both blood glucose and insulin level in normal rat. However only the blood glucose level in diabetic rat was lowered with major lipid components. In diabetic rat brain, gangliosides metabolism were changed. The amount of GMla was increased while GDla, GDlb, and GTlb were not synthesized. Furthermore, undefined ganglioside was found. In major lipid component-treated diabetic rat brain, the ganglioside metabolism proceeded as same as the normal rat. On the contrary, in bovine brain gangliosides-treated diabetic rat brain, the gangliosides metabolism was not recovered to normal one.

  • Recently, biohydrocarbons are gathering an interest as a new bioenergy due to the versatile applicability. In the present work, a process is proposed for the recovery of lipids from Recombinant E. coli MG1655 which provides longer chain fatty acids. After the growth of the recombinant E. coli, the cells were disrupted by high pressure homogenizer for obtaining intracellular lipids and the resulting solutions were centrifuged and extracted. For the efficient cell disruption with high pressure homogenizer, the pressure higher than 5,000 psi was required. In addition, under the conditions of applied pressure 5,000 to 20,000 psi, 1~3 pass homogenizing was enough for the more than 90% cell disruption. As organic solvents for extraction of lipid, hexane/isopropyl alcohol and ethyl acetate/ethanol systems showed excellent extracting power. With these solvent systems, the 60% lipid could be recovered. Moreover it was found that the extracted lipids contained long-chain fatty acids such as $C_{12}$, $C_{14}$, $C_{16}$ and $C_{18}$.

Effects of Oral Taurine Supplementation on Blood Antioxiant Enzyme Activities and Lipid Peroxidation in Healthy Female Adults (타우린보강이 건강한 성인영성의 혈중 항산화효소 활성과 지질과산화물 농도에 미치는 영향)

  • Effects of oral taurine supplementation (6g/day)for 2-4 weeks on activities of red blood cell(RBC)total superoxide dismutase (SOD) and plasma glutathione peroxidase(GSH-Px) and the level of malondialdehyde(MDA) were evaluated in healthy female adults (23.6$\pm$0.3 years old). Compared to the value for 0 week plasma GSH-Px activity of the subjects was significantly lower after 2 weeks of taurine supplementation(p<0.05) and recovered to the value similar to 0 week after 4 weeks of taurine supplementation. RBC total SOD activity tended to be decreased after 2 weeks of taurine supplmentation compared to the values for 0 week although the difference between the means of the two group was not statistically significant. Plasma MDA level was not significantly decreased by taurine supplementation most probably due to the fact that the subjects participated in the present study were healthy and their antioxidant defense system had been in the 'normal' range. Plasma MDA concentration was negatively correlated with plasma taurine concentration(r=-0.2003m p<0.05) but tended to be positively correlated with plasma cholesterol concentration(r=0.2465, p=0.0645) as expected Plasma GSH-Px activity was positively associated with the percentage of 22:0 (r=0.2892, p<0.05) or 20:4w6(r=0.2939, p<0.05). On the other hand plasma MDA concentration was positively correlated with the percentage of 20:5w3 in plasma total lipids(r=0.2635 p<0.05) and negatively correlated with $\Delta$5 desaturation index of w6 fatty acids(20:3w6⇒20:4w6) in plamsa total lipids(r=-0.2714, p<0.05) as well as in phospholipids(r=-0.2864, p<0.05). From these results protective effect of taurine supplementation against lipid peroxidation and antioxidant defense system in humans appears to be minimal when the subjects are in a relatively healthy state. Further studies concerning the antioxidant efficacy of taurine should be conducted in human subjects under various disease states related to oxidative stress such as diabetes and artheroxclerosis.

  • Purpose : Intravenous immunoglobulin(IVIG) has been used as an immunomodulatory treatment for several immune-mediated diseases. The early effect of high-dose IVIG on biochemical profiles including lipids and proteins was evaluated in patients with Kawasaki disease(KD). Methods : Twelve children with KD(nine boys) were treated with IVIG of 2 g/kg over 12 hours. Serial sera were collected from the patients four times : before IVIG treatment and two hours, 24 hours and seven days after IVIG treatment. The samples were frozen at $-20^{\circ}C$ before biochemical analysis. Results : A significant decrease in albumin concentration was found two hours h and 24 hours after IVIG treatment, but this recovered to the pretreatment level after seven days. Total cholesterol and triglyceride increased slightly after seven day. A significant decrease in HDL-cholesterol and C-reactive protein was seen two hours and 24 hours after IVIG treatment. Conclusion : High-dose IVIG affects immediate changes in protein profiles and HDL-cholesterol in KD. Changes in HDL-cholesterol induced by IVIG may be the result of changes in systemic protein metabolism.
