Validation of a HPLC Method for Determination of Pentoxifylline in Human Serum and Its Application to Single-dose Pharmacokinetics (인체혈청 중 펜톡시필린의 정량을 위한 HPLC 분석법의 점증 및 단일용량 투여에 의한 약물동태)

  • A simple HPLC method was employed for the determination of pentoxifylline in human serum. After addition of internal standard (IS, 50 uL of 3 ug/mL chloramphenicol methanol solution) into the serum sample, the drug and IS were extracted by dichloromethane. Following a 1-min vortex-mixing and a 15-min centrifugation at 3500 게m, the organic phase was transferred and evaporated to dryness under a vacuum. The residue was reconstituted with 120 ${\mu}L$ of mobile phase and 50 ${\mu}L$ was injected into C18 column with a mobile phase composed of 0.034 M phosphoric acid adjusted to pH 4 with 10 M NaOH and acetonitrile (75:25, v/v). The samples were detected using an ultraviolet detector at 273 nm. The method was simple, specific and validated with a limit of 10 ng/mL. Intra- and inter-day precision and accuracy were acceptable for all quality control samples including the lower limit of quantification. The applicability of this method was evaluated by analysis of human serum after oral administration of a single 400 mg dose to 8 healthy subjects. The pharmacokinetic parameters for pentoxifylline in human subjects were calculated using WinNonlin program. As a result, $AUC_{t},\;C_{max},\;T_{max}$ and $t_{1/2}$ were $962.28{\pm}645.69\;ng{\cdot}/mL$, $132.82{\pm}42.05$ ng/mL, $2.06{\pm}2.68$ hr and $8.74{\pm}4.38$ hr, respectively. Based on the results, this validated method appears to be useful fur the pharmnacokinetic study of pentoxifylline in humans.

  • An enzyme ${\beta}$-galactosidase or ${\beta}$-galactohydrolase [EC3.2.1.23], commonly called lactase, mediates galacto-oligosaccharide (GOS) synthesis under conditions of high substrate concentrations. Also, lactase hydrolyzes ${\beta}$($1{\rightarrow}4$) lactose into glucose and galactose, the latter is successively transferred to free lactose to make various oligosaccharides via transgalactosylation. GOS is non-digestible to human digestive enzymes and has been used as a functional prebiotics. Among the 24 lactic acid bacteria (LAB) strains used, Lactobacillus paracasei YSM0308 was selected based on its exhibition of the highest ${\beta}$-galactoside hydrolysis activity, and the crude lactase was prepared for examination of reaction conditions to affect the GOS synthesis. Lactase activity was measured with a spectrophotometer using ONPG (o-nitropheyl ${\beta}$-D-galactopyranoside) method. Lactase activity was not detected in the culture supernatant and was mostly present in the cell pellet after centrifugation. Activity of the crude lactase preparation ranges from102 to 1,053 units/mL, with the highest activity determined for L. paracasei YSM0308. Optimal conditions for GOS synthesis are as follows: concentration of whey powder, pH, temperature, and time were 30%, pH 6.5-7.0, $30^{\circ}C$, and 4 h, respectively. The final GOS concentration was 19.41% (w/v) by the crude YSM0308 lactase, which was obtained from strain YSM0308 grown in the 10% (w/v) reconstituted whey-based medium.

  • The concept of probability in current school mathematics has been dealt with in the classic viewpoint (mathematical probability) and part of the frequency viewpoint and axiomatic viewpoint have been introduced. However, since the exact understanding of the probability concepts is not possible only with the classic viewpoint, we need to research further on methods to complement classic viewpoint and emphasize various aspects of probability concepts (Lee, Kyung Hwa, 1996). Therefore, this study is to find out optimal computer simulation plans in teaching statistical probability. For the purpose, it examines how the nature of mathematical knowledge may be changed when statistical probability is taught with a use of computer simulation based on the Theory of Didactical Situation presented by Brousseau(1997). Next, it identifies how probability curriculum should be reconstituted for introducing statistical probability through computer simulation. Finally, it develops specific teaching materials that introduce statistical probability using computer simulation based on the results obtained.

Effects of added sugars and organic acid on the quality of reconstituted tobacco leaves (당과 산류가 담배 판상엽의 품질에 미치는 영향)

  • (A series of cigarettes made from) RTL containing different leveles of added sugars (2.0. 5.0 and 8.0%) and acids(0.4, 0.8 and 1.2%) have been examined. Compared to the control cigarette there was reduced in the delivery of tar and nicotine in smoke on 5.0% addition of glucose there was a signification reduced the delivery of nicotine the addition of glucose or fructose decreased the delivery of nicotine tar and co. However, tar in smoke was an increased in the delivery of tar and nicotine was observed, when sucrose and lactose were the added sugars. The smoking taste by the addition of glucose and fructose on R.T.L. was found to be milder than by the others sugars. Tar and nicotine deliveries showed a decreasing trend with the addition of organic acid and tartaric acid among the organic acid described above had greater effect than the others acids 1.2% addition of tartaric acid gave some 11-31% reduction in tar and nicotine delivery and a fall of smoke pH from 4.49 was found. Smoking taste by the addition of lactic acid and tartaric acid had milder than the others and heat treatment for 10 minutes at 13$0^{\circ}C$ gave also better on smoking: taste. The addition of mixture of sugars and organic acid (glucose 5% + lactic acid 0.6% + tartaric acid 0.6%) and then the successive heat treatment for 10 minutes at 13$0^{\circ}C$ improved greatly the Quality of R.T.L. and nicotine were reduced to 11-28% and pH was changed from 5.40 to 3.89.

Effects of Salicylate on the Activity of Isoperoxidase $A_3$ from Tobacco Callus (Salicylate가 담배 미분화세포 isoperoxidase $A_3$의 활성에 미치는 영향)

  • Salicylate is involved in the induction of pathogen-related proteins and plant defense response. The effects of salicylate on the activity isoperoxidase $A_3$ from tobacco callus (Nicotiana tabacum L.) and the protection against the enzyme inactivation by salicylate in the presence of $Fe^{2+}$ were examined. About 20% and 85% activity losses of peroxidase occurred at 0.48 mM and 0.6 mM salicylate, respectively, showing that isoperoxidase $A_3$ was inactivated by salicylate. The inactivation occurred depending on pH and showed noncompetitive inhibition mode. Moreover, inactivation of the enzyme by salicylate was completely protected in the presence of $Fe^{2+}$. Apoperoxidase without heme moiety was constructed and the effects of various metal ions on the recovery of enzyme activities were investigated. More than 80% of the activity was reconstituted by the addition of $Fe^{2+}$ or hemin. However, the enzyme activity was not recovered by $Cu^{2+},\;Zn^{2+},\;Co^{2+},\;or\;Mn^{2+}$.

Centrifuge Model Tests on Characteristics in Forced Replacement Method for Soft Ocean Ground to Build Coastal Structures (해안구조물 축조를 위한 해양연약지반의 강제치환 특성에 관한 원심모형실험)

  • This paper shows theresults of centrifuge model experiments to investigate the behavior of a replacement method in dredged and reclaimed ground. For this experimental work, centrifuge model tests were carried out to investigate the behavior of a replacement method in soft clay ground. Basic soil property tests were performed to find the mechanical properties of clay soil sampled from the southern coast of Korea, which was used for the ground material in the centrifuge model tests. The reconstituted clay ground of the model was prepared by applying reconsolidntion pressure in a 1 g condition with a specially built model container. Centrifuge model tests were carried out under the artificially accelerated gravitational level of 50 g. Replacement material of lead with a certain degree of angularity was used and placed until the settlement of the replacement material embankment reached a state of equilibrium. Vertical displacement of the replacement material was monitored during tests. The depth and shape of the replacement, especially the slope of the penetrated material and the water content of the clay ground were measured after finishing tests. Model tests for investigating the stability of an embankment after backfilling were also performed to simulate the behavior of a dike treated with replacement and backfilled with sandy material. As a result of the centrifuge model test, the behavior of the replacement, the mechanism of the replacement material being penetrated into clay ground, and the depth of the replacement were evaluated.

  • This study concentrates on a new idea that is formulated as a creative spatialization in contemporary architecture and painting. It is stimulated from the classical baroque, which is expressed with the conceptual instruments, 'excessive', 'decorative', 'accidential' Suggested from that, this study tries to open the actual sense area of 'the baroque' by analysing the architectural ideas of wrenching and folding by Frank O. Gehry and in the same context by elucidating the dynamic deformalising of the figures by Francis Bacon. The redefined 'new baroque' operates in the chain of the transformable and flexible images that must be recognized as 'the real' in the sense of 'the most empirical' and are vulnerable to dispersal. The typical instrument of Gehry's 'new baroque architecture' is the enclosing of the space and its folding, and Bacon, whose painting can be called as the modern baroque, takes as the expressing methods a wrenching of the figures, a relieving of the colored background against the figure and a disappearing of figures into the background. These different ideas meet in the theoretical gravitation field of the philosophy of Deleuze, in which the reconstituted baroque conception means the tension between the folds, on expansion and contraction of which the variable space is dependent, Through the general principle of 'the new baroque', a free dispersal of power, to architect and painter is offered the possibility, not only to make the real world intimate to the real life, but also to produce from there a vivid new space. Finally it brings to enjoyer and viewer the possibility, to perceive the operating power in and around oneself.

Expression of Human Immunodeficiency Virus Type 1 Tat Proteins in Escherichia coli and Application to Study Tat Functions

  • The human immunodeficiency virus type 1 (HIV-1), transactivator of transcription (Tat), is one of the viral gene products that is essential for HIV-1 replication. The HIV-l Tat protein regulates transcription from an HIV-1 long terminal repeat (LTR) and affects the gene expression of cellular proteins during infection. In order to develop an expression system to overexpress and simply purify HIV-1 Tat proteins, the HIV-1 Tat coding sequences that contain one or two exons were amplified using PCR and cloned into a pET vector, which contains a consecutive stretch of six histidine residues at the amino-terminus. The reconstituted vectors were overexpressed in the E. coli strain and the soluble recombinant proteins were purified to be homogeneity in a single step by $Ni^{+2}-nitrilotriacetic$ acid Sepharose chromatography under nondenaturing conditions. Recombinant HIV-1 Tat proteins were shown to transactivate the HIV-1 LTR promoter in a dose-dependent manner when introduced into mammalian cells. In addition, treatment of human endothelial cells with purified Tat proteins resulted in a significant increase in the level of vascular cell adhesion molecule-1 (VCAM-1) expression. These results indicate that the recombinant HIV-1 Tat proteins are active in transactivating viral and cellular promoters. The expression and purification system described in this study will facilitate in characterizing the biological functions of the Tat proteins.

Content of Orotic Acid in Raw milk and Reduction of OA by Lactic Acid Bacteria during Fermentation (Orotic acid의 원유내 함량과 발효중 유산균에 의한 감소)

  • Changes in erotic acid (OA) contents of raw milk and during cultivation and storage at refrigerated temperature were determined. OA contents of raw and 10% reconstituted milk held at $121^{\circ}C$ for up to 15 min decreased by 17.6 and 22.4%, and those fermented at 40 and $37^{\circ}C$ for 72 hr with Lactobacillus helveticus 166 and Lactobacillus casei 955 decreased 37.8-43.2 and 41.8-76%, whereas when fermented at $40^{\circ}C$ for 72 hr with Streptococcus thermophilus ST-37, did not decrease significantly compared with fermentation by other lactobacilli. OA content did not change during storage at refrigeration temperature. OA reduction by Lactobacillus sp. was dramatically increased at the initiation of stationary phase during fermentation. OA reduction varied among different lactobacilli. These results show decrease in cultivation time, rapid cooling of yogurt, and proper selection of Lactobacillus sp. prevent OA content reduction.

Studies on Frozen Storage of Lactic Acid Bacteria (유산균의 동결보존에 관한 연구)

  • Three common species of lactic acid bacteria, Lactobacillus casei, Lactobacillus bulgaricus, and Streptococus lartis were tried to store in frozen state for various periods up to four months and their stabilities during the storage were evaluated. A needle of culture suspension was transferred to 10 $m\ell$ of 10% reconstituted skim milk and frozen immediately. After storage under said conditions the storage tubes were thawed and incubated under optimum growth temperatures for 48 hours after which plate counts and acidity determinations were made. The same incubation and analyses were repeated with organisms transferred from the previous culture tubes. No significant deterioration in both survival and acid producing activity were observed during the four month storage except that some decrease in acid forming by L. bulgaricus appeared after 60 days of storage. Among the three species tested L. casei was found to be most stable and the fast was reconfirmed with scale-upped fermentation experiments using the same organism.
