Evaluation of an Automated ELISA (VIDAS(R)) and Real-time PCR by Comparing with a Conventional Culture Method for the Detection of Salmonella spp. in Steamed Pork and Raw Broccoli Sprouts (편육과 브로콜리싹에서 Salmonella spp. 검출을 위한 배지법과 Real-time PCR 및 신속 검사키트(VIDAS(R))의 비교검증)

  • Salmonellosis is an important worldwide foodborne infectious disease that is transmitted by many food vehicles including raw and processed animal products and fresh produce. In this study, the effectiveness of automated ELISA ($VIDAS^{(R)}$) and realtime PCR in the detection of Salmonella spp. in steamed pork and raw broccoli sprouts was evaluated by comparing their results with those of a conventional culture method. Bulk samples (500 g) of steamed pork and raw broccoli sprouts were inoculated with various levels of Salmonella and divided into 20 samples (25 g each). All the samples, including the controls, were analyzed using a conventional culture method, $VIDAS^{(R)}$, and real-time PCR to detect the presence of Salmonella. In addition, the levels of background flora in the steamed pork and the raw broccoli sprouts were determined. In the steamed pork that contained less than 100 CFU/g of aerobic bacteria, all three methods detected low levels of Salmonella without a statistical difference in their performance. In the broccoli sprouts with high quantities of background flora (ca. $6.7{\times}10^7$ CFU/g), however, all three methods were unable to detect low levels of Salmonella, and real-time PCR and $VIDAS^{(R)}$ more sensitively detected Salmonella than the culture method, with significant statistical differences. In conclusion, $VIDAS^{(R)}$ and real-time PCR could be superior to conventional culture methods in detecting Salmonella in food with high levels of background flora.

  • Because worldwide interest in the health is increased, the real-time health monitoring system has been demanded to be more convenient non-contact and precise medical devices than conventional. Therefore we developed the blood pressure monitoring system using UWB(Ultra Wide Band) radio wave which contact to the human body through the radar and continuously collect a movement signal of the blood vessel. Then the collected data including pulse rate, systolic blood pressure, diastolic blood pressure is processed in real time. The system monitors and controls through a program-based embedded LCD(Liquid Crystal Display) using Qt GUI(Graphic User Interface) to be displayed in real time. We implement the system as a embedded system because of reducing the size of the limited resources. Existing PC GUI design mode is used relatively large memory, therefore it requires more CPU(Central Processing Unit) capacity and processing time.

  • This paper proposes IIKBAG(Intelligent Interlocking Knowledge Base Generator) which can build automatically the interlocking knowledge base utilized as the real-time interlocking strategy of the electronic interlocking system in order to enhance it's reliability and expansion. The IIKBAG consists of the inference engine and the knowledge base. The former has an auto-learning function which searches all the train routes for the given station model based on heuristic search technique while dynamically searching the model, and then generates automatically the interlocking patterns obtained from the interlocking relations of signal facilities on the routes. The latter is designed as the structure which the real-time expert system embedded on IS(Interlocking System) can use directly in order to enhances the reliability and accuracy. The IIKBAG is implemented in C computer language for the purpose of the build and interface of the station structure database. And, a typical station model is simulated to prove the validity of the proposed IIKBAG.

  • This paper proposed the real-time monitoring methodology of a traffic signal controller. The proposed methodology is based on the simulation technology, and it is necessary to construct a simulation model imitating the behavior of a traffic signal controller. By executing the simulation model, we can obtain the 'nominal system trajectory' of the traffic signal controller. On the other hand, an IoT(Internet of Things)-based monitoring device is implemented in a traffic signal controller. Through the monitoring device, it is possible to obtain the 'actual system trajectory'. By comparing the nominal system trajectory and the actual system trajectory, we can estimate the degree of deterioration of a traffic signal controller.

Development of Convenient Software for Online Shelf-life Decisions for Korean Prepared Side Dishes Based on Microbial Spoilage

  • User-friendly software was developed to determine the shelf-life of perishable Korean seasoned side dishes in real time based on growth models of spoilage and pathogenic microorganisms. In the program algorithm, the primary spoilage and fastest-growing pathogenic organisms are selected according to the product characteristics, and their growth is simulated based on the previously monitored or recorded temperature history. To predict the growth of spoilage organisms with confidence limits, kinetic models for aerobic bacteria or molds/yeasts from published works are used. Growth models of pathogenic bacteria were obtained from the literature or derived with regression of their growth rate data estimated from established software packages. These models are also used to check whether the risk of pathogenic bacterial growth exceeds that of food spoilage organisms. Many example simulations showed that the shelf-lives of the examined foods are predominantly limited by the growth of spoilage organism rather than by pathogenic bacterial growth.

  • This research suggested the new algorithm controlled by micro processor which is already stored by various PWM form of output voltage by using fundamental data of the carrier and signal. The determined PWM pattern is not concerned with the signal wave form and the new algorithm can obtain the desired pulse width by synchronous of carrier. The PWM wave can be controlled with real time by using extra hardware and digital software and to speed up program processing, the control signals to switch the power semi-conductor of three phase PWM inverter, simultaneously use the output signal by microprocessor and extra hardware, and control signal by software. In the end, this method was proved by applying to Three phase voltage source inverter.

  • Determination of the required reserve capacity has an important function in operation of power system and it is calculated based on the largest loss of supply. However, conventional method cannot be applied in future power system, because potential grid-connected distributed generator and abnormal temperature cause the large load imbalance. Therefore this paper address new framework for determining the optimal required reserve capacity taking into account the real time load imbalance. At first, we introduce the way of operating reserve resources which are the secondary, tertiary, Direct Load Control (DLC) and Load shedding reserves to make up the load imbalance. Then, the formulated problem can be solved by the Probabilistic Dynamic Programming (PDP) method. In case study, we divide two cases for comparing the cost function between the conventional method and the proposed method.

  • This paper discusses a method for the determination of frequency control reserve requirement with consideration of the interaction between ex-ante planning and real-time balancing. In proposed method, we consider the fact that the delivered energy for tertiary control reserve is determined based on required capacity for secondary control reserve and the expected amount of load errors. Uncertain load errors are derived by Brownian motion, an optimization method is suggested using a stochastic programming. In a short, we propose an interactive dependent method for determining secondary control reserve requirement based on the principle that it satisfies to minimize the total cost. As a result, this paper provides will analyze for an example model to demonstrate the capabilities of the method.

  • Using in silico approaches and RACE we cloned a full length trinucleotide (CAG) repeat-containing cDNA (cag-3). The cDNA is 2478 bp long and the deduced polypeptide consists of 140 amino acids of which 73 are glutamines. The genomic sequence spans approximately 79 kb on mouse chromosome 7 and the gene is composed of four exons. Standard and real-time PCR analyses of several mouse tissues showed that the gene is exclusively expressed in the brain and is not detected in embryonic stages. Within the brain, it is expressed throughout the forebrain region with predominant expression in the hypothalamus and olfactory bulb and very low levels in the mid- and hindbrain.
