강원도 고성 뒤배재 현무암은 산출상태, 구성광물, 주성분원소 및 미량원소의 특징에 의해 하부 현무암과 상부 현무암으로 구분된다. 하부 현무암은 집괴암의 특징을 보이고, 맨틀 및 지각기원의 포획암과 감람석, 휘석, 사장석 포획광물이 있다. 상부 현무암은 주상절리로 산출되며, 맨틀 포획암이 상대적으로 더 많고, 포획광물로는 감람석이 우세하다. 현무암의 주성분원소 및 미량원소 분석 결과 상부 현무암은 하부 현무암에 비하여 초생 마그마에 가까운 조성을 보인다. 원시맨틀로 표준화한 미량원소 및 희토류원소의 부화 및 결핍 패턴은 두 현무암이 매우 유사하나, 하부 현무암이 상부 현무암 보다 LREE의 부화 정도가 더 크다. 뒤배재 화산체의 하부 현무암은 약 0.8-1.2%, 상부 현무암은 약 3.7-4.0%의 동일한 맨틀 물질인 석류석 페리도타이트의 배취 용융으로 형성되었다. 그리고 하부 현무암내의 포획된 화강암, 포획광물의 반응연 및 사장석, 석영 등의 포획광물은 하부 현무암을 형성한 마그마와 지각물질과의 동화 가능성을 지시한다.
혼합되지 않는 두 용액 사이의 계면(interface between two immiscible electrolyte solutions, ITIES)에서의 전하 이동 반응에 대한 전기화학적 연구는 이온 검출용 센서, 바이오센서, 생체막 모델링, 약물 전달 반응, 상전이 촉매반응, 연료 생성, 태양에너지 전환 등을 포함한 다양한 연구 분야에 적용이 가능하기 때문에 크게 주목받고 있다. 특히 ITIES에서의 이온 전이 반응을 이용하여 이온물질 및 생물질 등을 검출할 수 있는 센서로 개발하기 위해 불안정한 ITIES의 한 쪽 액체층을 젤(gel)화하여 안정화하고, 마이크로 계면 형성을 통해 전압강하를 최소화 시키는 등의 연구가 활발하게 이루어졌다. 본 총설에서는 ITIES 계면에서의 이온 전이 반응을 이용하여 개발된 다양한 센서의 원리와 응용 및 발전 가능성에 대해 다루고자 한다. ITIES 계면을 (i) 보편적인 액체/액체 계면형, (ii) 마이크로피펫 팁형, (iii) 고분자 박막에 형성된 단일 마이크로홀 또는 마이크로홀 어래이형 및 (iv) 실리콘 기판에 제작된 마이크로홀 어래이형으로 분류하고, 이들 계면에서의 직접적인 이온 전이 반응과 보조 이온 전이 반응을 활용하여 수질 환경 오염의 원인이 되는 이온 및 농약 성분을 선택적으로 검출할 수 있는 이온 선택성 센서와 생물질을 분석할 수 있는 바이오센서 개발 연구에 대해 초점을 두고 소개하려 한다.
A systematic study was conducted to identify and isolate a serologically pertinent antigen with high specific activity and low cross reactivity from Cysticercus parenchymal antigen. Differential centrifugation of the homogenate yield three particulate and one soluble fractions ; the $480{\times}G$ pellets($CyL_2$), the $7650{\times}G$ pellet($CyL_3$), the $100000{\times}G$ pellet($CyL_4$), and $100000{\times}G$ supernatant($CyL_6$). We compared antigenicity of these antigens to that or cystic fluid antigens($CyF_1$), saline extract of cystic wall($CyL_1$), and n-butanol treated $GyL_4$ antigen ($CyL_6$) based on SDS-PAGE and immunoblot techniques. The data obtained were as follows : 1) The ratio of O.D. value of ELISA against cysticercosis positive pool sera to that of negative pool sera was highest when using $CyF_1$ as antigen. However the ratio was relatively low in case of $CyL_{3.4}$ and $CyL_5$. 2) We have noted in previous paper that most strong antigenic activities are present in 63Kd band with low cross reactivities. An effective serologic reagent must contain components that are recognized by most infected sera. 63Kd band met this criteria and could be considered as a reliable band for the diagnosis of cysticercosis. As far as 63Kd band concern, $CyL_5$ showed most strong activities without disturbance of cross reaction by EITB in spite of low applicability to microplate ELISA. 3) $CyL_5$ could detect the serum antibody of cysticercosis even in very low titers, around cut-off values of microplate ELISA, by immunoblot. It also could detect the cross reactivities of Echinococcus species, which showed high absorbance value in micro plate ELISA and some sparganosis cases. Further purification of this antigen will be able to represents a antigen that can be used in the diagnosis of cysticercosis.
선캠브리아기의 지리산 편마암 복합체는 초기의 백립암상에서 후기의 각섬암상 또는 녹색편암상까지 후퇴변성작용을 받았으며, 편마암 복합체 내 관입한 하동 회장암체는 이 중 후기 변성작용의 영향을 받았다. 회장암체 내 주요 유색광물은 각섬석으로 이를 통해 변성조건 및 변성상에 관한 추정이 가능하다. 회장암체 내 각섬석은 개방니콜 하에서 주로 청녹색 내지 녹색을 나타내고, 반자형 내지 타형의 형태를 보인다. 일부 갈색의 각섬석 주변부로 청록색으로 변하는 누대조직(zonal texture)이 존재하고, 사방휘석 내에는 각섬석으로 변하는 반응부가 존재해 후퇴변성 내지 변질 양상을 보인다. 회장암을 구성하는 각섬석은 크기 및 산상에 따라 기질을 이루는 미정질의 각섬석[I 그룹 : 페로혼블렌드(ferrohornblende)]과 1 mm 이상의 크기를 보이며, 주로 불투명광물 내지 사장석과의 경계에서 산출되는 각섬석[II 그룹 : 페로파가사이트(ferropargasite)]으로 구분된다. 화학분석결과, 각섬석 내 $Al^{vi}$의 함량을 통해 회장암체의 변성압력이 5 kbar 이하의 저압의 조건이었음이 인지되는 한편, 각섬석 내 Ti 함량범위 및 각섬석+사장석+석류석+흑운모+녹니석의 광물조합을 통해 회장암체의 변성상이 각섬암상에 속하는 것으로 판단된다.
경남 산청지방에서 쏘가리를 양식하던 어장에서 대량폐사가 발생하였다. 주요 증상으로 전신 피부에서 궤양이 진행되었고 조직학적으로는 주요 실질 장기에서 세균의 집락과 주변부위로 염증 및 괴사가 관찰되었다. 분리된 세균을 API 20E 및 API 20NE를 이용하여 생화학적 검사를 실시한 결과 Aeromonas salmonicida subsp. salmonicida (typical A. salmonicida)로 진단할 수 있었다. 그러나 atypical A. salmonicida와 typical A. salmonicida로 구분할 수 있는 PCR 기법을 실시한 결과, 본 세균은 atypical A. salmonicida임을 확인할 수 있었다. 본 연구는 우리나라 쏘가리에서 A. salmonicida로 인한 발병의 최초 보고임과 동시에, A. salmonicida의 정확한 진단을 위해선 생화학적 검사가 아닌 PCR 검사를 이용한 진단을 실시해야 함을 제시하였다.
독도에 자생하고 있는 식물의 뿌리로부터 내생진균의 분리를 시도하여, 동도로 부터 참억새, 돌피, 쇠무릎과 서도에서 갯별꽃 및 율무쑥등을 연구재료로 사용하였다. 총 21종의 내생진균을 분리하였고, universal primers ITS-1과 ITS-4를 사용하여 ITS 영역을 PCR로 증폭하여 동정하였다. 염기서열 분석결과, 동도에 자생하는 참억새에서는 Penicillium 속 균주가 75%, Aspergillus 균주가 25%, 분리되었고, 돌피에서는 Penicillium 속 균주가 55%, Aspergillus 속 균주가 30%, Zygorhynchus 속 균주가 15%가 분리되었으며, 쇠무릎에서는 Penicillium 속 균주가 50%, Aspergillus 속 균주가 12%, Gibberella 속 균주가 13%, Talaromyces 속 균주가 9%, Umbelopsis 속 균주가 8% 분리되었다. 서도에 자생하는 갯별꽃에서는 Penicillium 속 균주가 76%, Pestalotiopsis 속 균주가 24% 분리되었고, 율무쑥에서는 Penicillium 속 균주가 81%, Mucor 속 균주가 19% 분리되었다. Bioassay결과, 돌피에서 분리된 Ec-3-1균주가 식물생장촉진활성을 나타내었다. 그리고 독도의 5종류 식물에서 Penicillium 속의 내생진균이 가장 많이 존재함을 알 수 있었다.
연산오계는 오래전부터 건강기능 증진 및 치료 효능이 높은 것으로 알려져 왔다. 최근 건강 기능식품 소재로 기능성 펩타이드 효능이 알려짐에 따라, 연산오계 내장 부산물로 부터 올리고 펩타이드 최적 생산 공정 및 생성물 특성에 대하여 연구를 수행하였다. 최적 효소가수 분해 공정 표면반응 분석을 이용하여 수행하였다. 최적 공정 조건을 확립하기 위해서 온도 (40, 50, $60^{\circ}C$), pH (pH 6.0, 7.0, 8.0), 효소 (1, 2, 3%) 범위에서 수행을 하였다. 생성물에 대한 가수분해도, 아미노산, 분자량 분포를 분석하였다. 효소 가수분해 최적 온도는 $58^{\circ}C$, pH 7.5, 효소의 농도는 3% 이었다. 최적 조건에서 2 시간 효소 가수분해를 한 결과 75-80% 이었다. 구성 아미노산의 총 함량은 386.15 mg/100 g 이었고 유리 아미노산 총량은 155.26 mg/100 g. 분자량를 Maldi-TOF 으로 분석을 한 결과 90% 이상이 300-1,000 Da 분포를 보여주었다.
Objectives: The aim of this study was to confirm the dose-dependent skin moisturizing effects of dried pomegranate concentrate powder (PCP) and pomegranate concentrate solution (PCS) in ICR mice. Materials and methods: To observe the in vivo skin moisturizing effects and possible underlying mechanisms of PCP and PCS, oral PCP (100, 200, and 400 mg/kg) and PCS (1, 2, and 4 mL/kg) were administered to normal ICR mice. Changes in body weight, skin water content, and skin type I collagen and hyaluronan contents were measured. Additionally, the mRNA expression levels of hyaluronan synthase (Has) 1, 2, and 3, and collagen type I alpha (COL1A) 1 and 2 were determined in the dorsal skin of mice by real-time reverse transcription polymerase chain reaction (RT-PCR). Results: Significant and dose-dependent increases in dorsal skin water content and type I collagen and hyaluronan contents were seen in PCP and PCS-treated mice. Moreover, the mRNA levels of Has 1, 2, and 3, involved in hyaluronan synthesis, and of COL1A1 and COL1A2, involved in collagen synthesis, were significantly and dose-dependently upregulated in PCS- and PCP-treated mice. Conclusions: In this study, PCP and PCS led to favorable skin moisturizing effects as indicated by increased skin water content and the upregulation of hyaluronan and collagen synthesis enzymes in mice treated with PCS (4 mL/kg) and PCP (200 mg/kg).
Background: Dendritic cell (DC)-based vaccines are currently being evaluated as a novel strategy for tumor vaccination and immunotherapy. However, inducing long-term regression in established tumor-implanted mice is difficult. Here, we show that deoxypohophyllotoxin (DPT) induces maturation and activation of bone marrow-derived DCs via Toll-like receptor (TLR) 4 activation of MAPK and NF-${\kappa}B$. Methods: The phenotypic and functional maturation of DPT-treated DCs was assessed by flow cytometric analysis and cytokine production, respectively. DPT-treated DCs was also used for mixed leukocyte reaction to evaluate T cell-priming capacity and for tumor regression against melanoma. Results: DPT promoted the activation of $CD8^+$ T cells and the Th1 immune response by inducing IL-12 production in DCs. In a B16F10 melanoma-implanted mouse model, we demonstrated that DPT-treated DCs (DPT-DCs) enhance immune priming and regression of an established tumor in vivo. Furthermore, migration of DPT-DCs to the draining lymph nodes was induced via CCR7 upregulation. Mice that received DPT-DCs displayed enhanced antitumor therapeutic efficacy, which was associated with increased IFN-${\gamma}$ production and induction of cytotoxic T lymphocyte activity. Conclusion: These findings strongly suggest that the adjuvant effect of DPT in DC vaccination is associated with the polarization of T effector cells toward a Th1 phenotype and provides a potential therapeutic antitumor immunity.

Altered environmental gravity, including both hypo- and hypergravity, may result in space adaptation syndrome. To explore the characteristics of this adaptive plasticity, the expression of immediate early gene c-fos mRNA in the vestibular related tissues following an exposure to hypergravity stimulus was determined in rats. The animals were subjected to a force of 2 g (twice earth's gravity) for 1, 3, or 12 h, and were examined poststimulus at 0, 2, 6, 12, and 24 h. RT-PCR (reverse transcription polymerase chain reaction) and real-time quantitative RT-PCR were adopted to analyze temporal changes in the expression of c-fos mRNA. The hypergravity stimulus increased the expression of c-fos mRNA in the vestibular ganglion, medial vestibular nucleus, inferior vestibular nucleus, hippocampus, cerebellum, and cortex. The peak expression occurred at 0 h poststimulation in animals stimulated with hypergravity for 1 h, and at 6 h poststimulus in those stimulated for 3 h. In contrast, those stimulated for 12 h exhibited dual peaks at 0 and 12 h poststimulus. Bilateral labyrinthectomy markedly attenuated the degree of c-fos mRNA expression. Glutamate receptor antagonist also dramatically attenuated the degree of c-fos mRNA expression. These results indicate that expression of c-fos mRNA in response to hypergravity occurs in the vestibular related tissues of the central nervous system, in which peripheral vestibular receptors and glutamate receptors play an important role. The temporal pattern of c-fos mRNA expression depended on the duration of the hypergravity stimulus.
