There are recognized three new species of Polysiphonia sensu lato at a 10-20 m water depth from Jeju Island, Korea. We described the morphology of three new species and analyzed rbcL sequences to infer phylogenetic relationships among Polysiphonia sensu lato. Polysiphonia amplacapilli sp. nov. is characterized by an epiphytic, discoid holdfast, 5-7 pericentral cells, large trichoblasts, lateral branches connecting with trichoblasts, only slightly corticated in the basal parts, cut-off rhizoids and a spiral arrangement of tetrasporangia. Polysiphonia morroides sp. nov. is characterized by saxicolous, cut-off rhizoids, four pericentral cells, ecorticate, spiral arrangement of tetrasporangia, and it was collected at the fast-flowing water habitat in 20-25 m depth. Polysiphonia sabulosia sp. nov. is characterized by ecorticate, 5-6 pericentral cells, a single filamentous trichoblast, lateral branches connecting with the trichoblasts, cut-off rhizoids, and spiral arrangement of tetrasporangia, and is growing in the sandy locations. The phylogenetic analysis of rbcL sequences demonstrated that three new species are clearly distinguished from other species of Polysiphonia sensu lato.
In this study, we report genetic characterization of Orobanche cumana, the causal agent of sunflower wilting in Serbia. The genetic diversity of this parasitic plant in Serbia was not studied before. Random amplified polymorphic DNA (RAPD) markers and partial rbcL gene sequences analysis were used to characterize the O. cumana populations at the molecular level. While phylogenetic analyses of RAPD-PCR amplicons were performed using unweighted pair-group Method analyses, rbcL gene sequences were analyzed using neigbor joining method and minimum spanning tree. Molecular analyses of RAPD-PCR analysis revealed high genetic diversity of O. cumana populations which indicated high adaptive potential of this parasitic weed in Serbia. Further analyses of rbcL gene using minimum spanning tree revealed clear differences among diverse sections of Orobanche genus. Although this molecular marker lacked the resolution to display intrapopulation diversity it could be a useful tool for understanding the evolution of this parasitic plant. Our results suggested that O. cumana has great genetic potential which can lead to differentiation of more virulent races which is important for determining crop breeding strategies for their control.
Nuclear SSU and ITS1 rDNA and plastid rbcL sequences were determined to identify the seven samples of Porphyra cultivated by means of natural seeding on the southwest coast of Korea and analyzed to access the phylogenetic relationships of them with the natural populations of P. tenera and P. yezoensis from Korea and Japan. SSU, rbcL and ITS1 data from 18, 21 and 31 samples, respectively, including previously published sequences were investigated in the study. Results from our individual and combined data indicated that the seven samples were all P. yezoensis and the entities except one from Muan 2 aquafarm strongly grouped together with the natural populations of P. yezoensis from the south and the west coast of Korea. The sample from Muan 2 seems to be derived from a strain of P. yezoensis introduced from Japan by Porphyra farmers, based on DNA sequence data.
The filamentous freshwater red alga Compsopogon caeruleus(Compsopogonophyceae, Compsopogonaceae, Rhodophyta) occurs in tropical and subtropical regions of worldwide. This species has been reported from Asia, America, Africa, Europe and Oceania, and the worldwide distribution of Compsopogon caeruleus is in variable water habitats. Several morphospecies of the genus Compsopogon had been recorded, but recent molecular phylogenetic analyses with worldwide sampling identified a monospecific genus, C. caeruleus. In the present study, we first report a freshwater red alga Compsopogon caeruleus from Korea. We identified Compsopogon caeruleus in an urban river in Yongin City, and analyzed its morphological and genetic characteristics. Nuclear 18S rDNA, plastidal rbcL gene and mitochondrial cox1 gene sequences isolated from Korean Compsopogon caeruleus showed high sequences similarity with Compsopogon caeruleus from worldwide (98.6-100% (18S rDNA), 99-100% (rbcL) and 97.7-100% (cox1)). These sequences similarities support the identification of a red alga found in Korea as Compsopogon caeruleus. This new geographical report provides the useful information for understanding the distribution and habitat range of Compsopogon caeruleus especially concerning urban freshwater environments.
Objectives : Due to the morphological similarity of in the roots of herbal medicine, the official herbal medicine is very difficult to authenticate between the original plants of Patriniae Radix and two adulterant Patrinia species. Therefore, we introduced DNA barcode analysis to establish a powerful tool for the authentication of Patriniae Radix from its adulterants. Methods : To analyze DNA barcode regions, genomic DNA was extracted from twenty-nine specimens of Patrinia scabiosaefolia, Patrinia villosa, Patrinia saniculifolia, and Patrinia rupestris, and internal transcribed spacer 2(ITS2), matK and rbcL genes were amplified. For identification of species specific sequences, a comparative analysis was performed by the ClastalW based on entire sequences of ITS2, matK and rbcL genes, respectively. Results : In comparison of three DNA barcode sequences, we identified 22, 22, and 12 species-specific nucleotides enough to distinguish each four species from ITS2, matK and rbcL gene, respectively. The sequence differences at the corresponding positions were available genetic marker nucleotides to discriminate the correct species among analyzed four species. These results indicated that comparative analysis of ITS2, matK and rbcL genes were useful genetic markers to authenticate Patriniae Radix. Conclusions : The marker nucleotides enough to distinguish P. scabiosaefolia, P. villosa, P. saniculifolia, and P. rupestris, were obtained at 22 SNP marker nucleotides from ITS2 and matK DNA barcode sequences, but they were confirmed at 12 SNP marker nucleotides from rbcL. These differences could be used to authenticate Patriniae Radix from its adulterants as well as discriminating each four species.
Despite continued studies on red algal flora in Korea, the taxonomy of the tiny ceramiaceous algae has received little attention. We report for the first time Griffithsia okiensis from Korea on the basis of morphology and molecular data. The species is small in thalli height (0.3-1.5 cm), and in diameter of vegetative cells (50-500 μm), and the ratio of cell length/breadth is 2-3 times. It has two carpogonial branches from the supporting cell of procarp. We generated psbA and rbcL sequences from ten specimens of G. okiensis isolated from Korea and Japan and from one G. japonica species isolated Japan. Eight specimens of G. okiensis from Korea were almost identical in both psbA and rbcL regions, nevertheless they differed from Japanese specimens by 4 ucleotides in psbA and 7 in rbcL. In all analyses of psbA, rbcL, and psbA + rbcL data sets, G. okiensis was determined to be a different species from G. japonica isolated from Japan, although both species showed a sister relationship. For all that extensive collection trips, we found no evidence for the occurrence of G. japonica in Korea.
Polysiphonia pacifica is rhodomelaceous red algal species that includes five varieties in Pacific Ocean: P. pacifica var. delicatula, P. pacifica var. distans, P. pacifica var. determinata, P. pacifica var. disticha, and P. pacifica var. gracilis. We here report morphology and phylogeny of P. pacifica to confirm the relationships among previously described varieties as a loan of type specimens from US and to assess phylogenetic relationships of closely related species using plastid protein-coding rbcL gene. Polysiphonia pacifica is distinguished by having creeping filaments attached by unicellular rhizoids not cut off by cross walls, four pericentral cells, ecorticate, trichoblasts rare, ultimate branchlets attenuate at the tip but not pungent, and tetrasporangia in long straight series in the ultimate branchlets. The protein-coding plastid rbcL gene sequence data show that P. pacifica is distinctly different from the superficially similar species, P. morrowii and P. stricta. However, the rbcL sequences of P. pacifica var. pacifica and var. disticha are identical though they have morphological variation.
지모 (Anemarrhena asphodeloides)는 탁월한 해열작용과 진정작용을 갖는 한약재로 한국, 중국, 일본에서 널리 이용되어 왔다. 본 연구에서는 먼저 국내 연구소에서 형태학적 분류 결과 지모로 확인된 3종의 식물체를 수집하여 엽록체 DNA의 trnL-F 염기서열을 분석하였다. 분석 결과, 국내 연구기관에서 보관중인 지모 식물체들이 모두 동일한 trnL-F의 염기서열을 보여서, 형태학적 분류와 계통유전학적 분류가 동일함을 확인하였다. 최초로 얻어진 지모 trnL-F 염기서열은 NCBI database에 등록하였다. 다음으로 국내 한약재 시장과 중국 한약재 시장에서 유통 중인 지모 한약재를 다량 구입하여 trnL-F의 염기서열을 분석하였다. 그 결과, 유통 중인 지모 한약재들이 모두 기원식물과 동일한 TrnL-F의 염기서열을 보여서 지모 약재의 경우 진품이 유통되고 있음을 알 수 있었다. TrnL-F의 염기서열로 계통수를 작성한 결과 지모는 아스파라거스목 (Asparagales), 용설란과 (agavaceae)에 속한 것으로 보여 졌다. 엽록체 rbcL 유전자 염기서열로 얻은 계통수와 비교한 결과 trnL-F 계통수와 rbcL 계통수가 비슷한 결과를 보여주어서 분자유전학적 분류에 두 유전자가 상호보완적으로 이용될 수 있음을 확인하였다.
A marine algal species was collected from the southern coast of Korea. This alga is characterized by a large and cartilaginous thallus, a dichotomous branching pattern, compressed axes, rare proliferations near the apex, and cystocarps protruded toward surface. In a phylogenetic tree based on rbcL sequences, this species nests in the same clade as Neodilsea yendoana as a terminal clade of the genus Neodilsea. The genetic distance between both sequences within the clade was calculated as 0.2% considered to be in the intraspecific range within the genus. Based on the morphological and molecular data obtained in the present study, this Korean species is identified as N. yendoana. This is the first record of the genus Neodilsea and N. yendoana in Korea.
A marine brown alga was collected from Geoje, located on the southern coast of Korea. This species shares the generic features of Dictyota and is morphologically characterized by the combined features of erect or prostrate thallus with smooth margin, dichotomous to subdichotomous branches not twisted at the terminal part, absence of surface proliferations, 1-3 cell layered cortex and unilayered medulla, and lack of involucrum in sporangia subtended by a single stalk cell. In a phylogenetic tree based on rbcL sequences, this species nests in the same clade with Dictyota bartayresiana. The genetic distance between both sequences within the clade was calculated as 1.2%, considered to be within the intra-species range for the genus. Based on these morphological and molecular analyses, this Korean alga is identified as D. bartayresiana described originally from the West Indies. This is the first record of D. bartayresiana in Korea.
