This study was carried out to confirm the phylogenetic relationships in Korean Rhus species. Sequences from internal transcribed spacers (ITS) of nuclear ribosomal DNA and rbcL gene of chloroplast DNA were determined. Cotinus coggygria was selected as outgroup because it is closest allied with Rhus in Anacardiaceae. Also, ingroup was limited as six Korean Rhus species. ITS 1 sequences in six species of Rhus and one species of Cotinus ranged from 246 to 253 bp and ITS 2 sequences from 234 to 244 bp. Concerning the G+C content of the studied taxa, ITS 1 sequences ranged from 58.0 to 68.13% and ITS 2 from 59.75 to 68.46%. On the other hand, rbcL sequences were same size in the all species examined by 1,428 bp. G+C contents of rbcL sequences were ranged from 43.56 to 43.77% which means there are nearly no different from interspecies each other. Phylogenetic tree strongly supports the colse relationships between R. succedanea and R. sylvestris. Rhus javanica and Cotinus coggygria were also closely allied with each other in ITS and rbcL trees. Therefore, R. javanica was regarded as most primitive species among the Korean Rhus species. ITS 1 region of nuclear ribosomal DNA was suggested as very useful taxonomical marker for genus Rhus.

Morphology and molecular phylogeny of a red algal species, Hypnea flexicaulis that is recently described from Japan, were investigated based on 23 collections from Korea (21), Taiwan (1), and the Philippines (1). Hypnea flexicaulis has percurrent axes with flexuous, antler-like branches which have wide branching angles, and abaxially curved ultimate branchlets. In order to study DNA divergence and phylogenetic relationships of the species, we determined plastid rbcL and mitochondrial cox1 sequences from the 23 collections. All 21 specimens from five different locations in Korea were almost identical to H. flexicaulis from Japan in rbcL sequences. Although there was a difference of three to five base pairs (bp) between samples from Korea and the Philippines or between the Philippines and Taiwan, Bayesian analyses of the rbcL data showed that all specimens from Korea, Japan, the Philippines, and Taiwan were strongly monophyletic. However, it is interesting that specimens from the Philippines differed by 31-34 base pairs in mitochondrial cox1 gene from those of materials from Korea and Taiwan, which differed by one to seven bp in rbcL between them. Although H. boergesenii is different from H. flexicaulis in having many antler-like branchlets, both appeared as sisters in all analyses of the rbcL data. This is the first report of H. flexicaulis from Korea based on morphology, rbcL, and cox1 gene sequences.

Objectives : Araliae Continentalis Radix and Angelicae Pubescentis Radix have been used as the same medicinal name Korean and Chinese traditional medicines, respectively. The authentic Araliae Continentalis Radix is described only the root of Aralia continentalis in the Korean Pharmarcopoeia. However, the dried root of Angelica biserrata, Levisticum officinale, or Heracleum moellendorffii also has been distributed adulterants of Araliae Continentalis Radix. To develop a reliable method for identifying Araliae Continentalis Radix from adulterants, we carried out the analyses of universal DNA barcode sequences.Methods : Four plants species were collected from different habitate and nucleotide sequences of matK and rbcL were analyzed. The species-specific sequences and phylogenetic relationship were estimated using entire sequences of two DNA barcodes, respectively.Results : In comparative analysis of matK sequences, we were identified 104 positions of marker nucleotide for Ar. continentalis, 3 for An. biserrata, 4 for L. officinale and 8 for H. moellendorffii enough to distinguish individual species, respectively. Furthermore, we obtained marker nucleotides in rbcL at 42 positions for Ar. continentalis, 5 for An. biserrata and 2 for H. moellendorffii, but not for L. officinale. The phylogenetic tree of matK and rbcL were showed that all samples were clustered into four groups constituting homogeneous clades within the species.Conclusions : We confirmed that species-specific marker nucleotides of matK sequence provides distinct genetic information enough to identify four species. Therefore, we suggest that matK gene is useful DNA barcode for discriminating authentic Araliae Continentalis Radix from inauthentic adulterants.
The taxonomic distinctiveness and cosmopolitan distributions of the red algae $Gelidium$$crinale$ and $G.$$pusillum$ remain unclear. Both species were first described in Devon in southwestern England; namely in Ilfracome for $G.$$crinale$ and Sidmouth for $G.$$pusillum$. We analyzed mitochondrial $cox$1 and plastid $rbc$L sequences from specimens collected in East Asia, Australia, Europe and North America. In all phylogenetic analyses of $cox$1 and $rbc$L sequences, $G.$$crinale$ was distinct from congeners of the genus. The analyses also revealed a sister relationship with the $G.$$coulteri$ and $G.$$capense$ clade. Nineteen $cox$1 haplotypes were identified for $G.$$crinale$, and they were likely geographically structured. Despite the distinctiveness in both $cox$1 and $rbc$L datasets, the sister relationship of $G.$$pusillum$ in the genus was not resolved. Our $cox$1 and $rbc$L datasets indicate that $G.$$crinale$ is a cosmopolitan species, found in East Asia, Australia, Europe and North America, while the distribution of $G.$$pusillum$ is restricted to Europe and Atlantic North America. Our results suggest that infraspecific classification of $G.$$pusillum$ may be abandoned.
The brown algal family Chordariaceae sensu lato is a focus of taxonomy because recent studies suggest a broad concept of the family, including genera formerly classified in the Dictyosiphonales. Using morphology, plastid rbcL and nrDNA ITS sequences, we evaluated relationships of the monotyic genus Soranthera (S. ulvoidea), which has been classified in the Punctariaceae. The species occurs in Bering Sea and Aleutian Islands, Alaska to Baja California. Thalli are globose to lobed, hollow, 3-5 cm in diameter, and covered with evenly distributed sori. However, two forms within the species are recognized: f. ulvoidea for globose forms and f. difformis for lobed forms. Plastid rbcL and nuclear ITS region sequences were newly determined in samples of S. ulvoidea from the Pacific coast of the North America. We found little variations in the ITS sequences among samples of S. ulvoidea from five different locations and in the rbcL region from two different locations. These results do not support previous classification of f. ulvoidea and f. difformis within the species. All analyses of our rbcL sequence dataset show that Soranthera was placed in the Chordariaceae s.l., but more related to Botrytella than Punctaria and Asperococcus.

Objectives : Pinelliae Tuber has been used as a typical unauthentic herbal medicines. Due to the morphological similarity between Pinelliae Tuber and adulterants, the correct authentication is very difficult. Therefore, we introduced DNA barcode to establish a powerful tool for the authentication of Pinelliae Tuner from adulterants. Methods : To obtain DNA barcode regions, genomic DNA was extracted from nineteen specimens of Pinellia ternata, Pinellia pedatisecta, Pinellia tripartita, and Typhonium flagelliforme, and matK and rbcL genes were amplified. For identification of species specific sequences and analysis phylogenetic relationship, a comparative analysis were performed by the ClastalW and UPGMA based on entire sequences of matK and rbcL genes, respectively. Results : In comparison of two DNA barcode sequences, we elucidated the phylogenetic relationship showing distinct four groups depending on species and identified 40 and 20 species specific nucleotides enough to distinguish each species from matK and rbcL gene, respectively. The sequence differences at the corresponding positions were avaliable genetic marker nulceotides to discriminate the correct species among analyzed four species. These results indicated that phylogentic and comparative analysis of matK and rbcL genes are useful genetic markers to authenticate Pinelliae Tubers. Conclusions : The marker nucleotides enough to distinguish P. ternata, P. tripatrita, P. peditisecta, and T. flagelliform, were observed at 40 positions in matK gene and 20 positions in rbcL gene sequence, respectively. These differences can be used to authenticate Pinelliae Tuber from adulterants as well as discriminate each four species.
Although Pterocladiella is a well-known agar-producing red algal genus, its taxonomy in Korea appears to be confused. In the present study, we demonstrate the occurrence of Pterocladiella nana and P. tenuis, as well as P. capillacea in Korea on the basis of morphological and plastid rbcL sequence comparisons. P. capillacea commonly occurs along the coasts of Korea and has regular pinnate branches with abundant second-order branches, growing up to 10 cm in length. P. nana occurs in the intertidal zone of Jeju and is distinguished by the small size of its thalli (up to 5 cm) and regular branches with up to third-order branches. P. tenuis is distributed mostly subtidally on the southern coast of Jeju and has irregular branches with rarely second-order branches, growing up to 19 cm in length. We determined rbcL sequences from 19 specimens (15 from Korea and four from France) and downloaded 28 sequences from GenBank. Analyses of all 47 rbcL sequences revealed that each of three species was consistently resolved. P. capillacea and P. tenuis always formed a sister clade with P. nana at the base. Given that 12 rbcL haplotypes from 28 specimens of P. capillacea have been found to date, analysis of a fast-evolving gene from across the range of the species should highlight its genetic diversity.

Two different molecular markers, the plastid rbcL and mitochondrial cox1 genes, were used to define the taxonomic position of the northwest Pacific Ocean species currently named Gracilaria chorda. We analyzed both genes (1,222 bp for rbcL and 1,245 bp for cox1) from 18 specimens collected in Korea, Japan, and China. Phylogenetic reconstruction revealed that this organism should be classified in the genus Gracilariopsis, rather than in the Gracilaria. Thus, Gracilariopsis chorda (Holmes) Ohmi is the legitimate name for Gracilaria chorda Holmes. Within the species, the sequences differed by 8 bp (0.7%) in rbcL and 5 bp (0.4%) in cox1. Six haplotypes of cox1 tended to be geographically organized. Gp. chorda is characterized by coarse, elongate terete axes, short filiform branchlets usually at irregular intervals, an abrupt transition in cell size from medulla to cortex, cystocarps without tubular nutritive cells connecting the gonimoblast to the upper pericarp, and relatively large gonimoblast cells of the cystocarp in the specimens collected from Wando in southern Korea.

Accurately identifying species is the basis of all biological studies. There has been much confusion in the identification of Grateloupiacean species, which have finely pinnate gross morphology similar to Grateloupia filicina (the type species of the family). The objective of this study was to comprehensively investigate species identification and distribution of G. filicina-like species in the Northwest Pacific, based on the rbcL sequences. A total of 118 specimens from 78 sites in Korea and Japan were collected from 2001 to 2021 and analyzed for their rbcL sequences. Additional 341 sequences downloaded from the GenBank were included in our comprehensive dataset. Based on these sequences, we documented the nomenclatural history and geographical distribution of the species, and commented on the application of species name. G. asiatica was the most abundant G. filicina-like species in the Northwest Pacific, and its high degree of morphological variation caused many misidentifications. In particular, G. dalianensis, G. serra, and G. variata require reconsideration of their conspecificity with G. asiatica using more specimens from China. By contrast, G. oligoclora was presumed to be a heterotypic synonym of G. subpectinata. The occurrence of G. acuminata, G. ramosissima, and G. livida in Korea resulted from misidentifications with other species.

Actinotrichia is a calcified galaxauracean red algal genus with temperate and tropical distributions in the Indian and Pacific Oceans. Morphological characteristics, along with rbcL and cox1 sequences, were analyzed from specimens collected in the western Pacific and the Indian Oceans. Both rbcL and cox1 data confirmed the occurrence of A. fragilis, A. robusta, and Actinotrichia sp. in this region. The presence of A. fragilis was verified in tropical Indo-Pacific and temperate northeast Asian waters and was characterized by high genetic diversity. Although A. robusta commonly occurs in the East China Sea, we confirmed its presence on rocks and crustose algae in the subtidal zone of three islands in the Andaman Sea. Actinotrichia sp. was similar to A. calcea in morphology and distribution, but with sufficiently different sequences, thus, additional sampling over the range will enable a more realistic evaluation of its taxonomic status.
