Tylosin Production by Streptomyces fradiae Using Raw Cornmeal in Airlift Bioreactor

  • Using a 50-1 airlift bioreactor, for the effective production of tylosin from Streptomyces fradiae TM-224 using raw cornmeal as the energy source, various environmental factors were studied in flask cultures. The maximum tylosin concentration was obtained at $32^{\circ}C$ and pH between 7.0 and 7.5. When seed was inoculated after 24 h of culture, the maximum tylosin concentration, 5.7 g/l, was obtained after 4 days of culture. Various concentrations of raw cornmeal were tested to investigate the optimum initial concentration for the tylosin production. An initial raw cornmeal concentration of 80 g/l gave the highest tylosin concentration, 5.8 g/l, after 5 days of culture. Of the various nitrogen sources, soybean meal and fish meal were found to be the most effective for the production of tylosin. In particular, with the optimal mixing ratio, 12 g/l of soybean meal to 14 g/l of fish meal, 7.2 g/l of tylosin was obtained after 5 days of culture. To compare raw cornmeal and glucose for the production of tylosin in the 50-1 airlift bioreactor for 10 days, fed-batch cultures were carried out under the optimum culture conditions. When raw com meal was used as the energy source, the tylosin production increased with increasing culture time. The maximum tylosin concentration after 10 days of culture was 13.5 g/l, with a product yield from raw cornmeal of 0.123 g/g of consumed carbon source, which was about 7.2 times higher than that obtained when glucose was used as the carbon source.

어패육에서의 Vibrio vulnificus의 증식에 관한 연구 (The Growth of Vibrio vulnificus in Meat Homogenates of Fish and Shellfish)

  • 방어, 참돔, 피조개, 굴 4종의 어패육에 V. vulnificus M-8을 접종하여 저장온도와 시간에 따른 균수의 변화를 시험한 결과는 다음과 같다. 1. V. vulnificus의 저온저항성은 $-20^{\circ}C$$1.5\%$ 식염-인산완충희석수에서는 32시간에 거의 사멸하였으며, 방어육 균질액에서는 인산완충희석수에서 보다 감소가 완만하였고 72시간 이후에도 균이 검출되었다. 그리고 $4^{\circ}C$ 저장에서도 균의 감소가 현저하였다. 따라서 이 균의 저온저항성은 약하지만 단시간 냉동 및 냉장에 의한 균의 사멸은 어렵다는 것을 알 수 있었다. 2. 어패육에 접종하여 $30^{\circ}C$에 저장한 시료중의 V. vulnificus의 증식은 일반적으로 16시간만에 최대균수에 달하였으나 피조개의 경우는 12시간으로 약간 빨랐으며 특히 방어육에 있어서는 초기에 감소하는 특징과 더불어 균의 증식속도가 완만하였다. 그리고 가열처리육보다 생육에서의 균의 증식이 빠르게 나타났다.

  • This study aimed to examine the establishment of the fish-paste processing industrial cluster in Busan. In order to successfully promote the fish-paste processing industrial cluster in Busan, first of all, it is required to implement various fish-paste processing related infra-structures and foundations. In addition to this, the agreements among fish-paste processing related organizations and businesses are inevitable to secure plans of inter-working with fisheries policies, to make efficient conditions for the stable supply of raw materials, and to pursue various supports from central and local governments for a viable development of fish-paste processing business companies in Busan.

배합사료와 생사료로 사육한 넙치의 체조성 및 육질 비교 평가 (Comparative Evaluation of Proximate Composition and Muscle Quality according to the Olive Flounder (Paralichthys olivaceus) fed Extruded Pellets and Raw Fish-Based Moist Pellet)

  • This study was conducted to evaluate the quality characteristics (proximate composition, fatty acids, amino acids and sensory properties) of whole body and dorsal muscle in olive flounder, which is switch size from juvenile to growing fed extruded pellets (EP) and raw fish-based moist pellet (MP) for 9 months. Sample was collected at 5 and 9 months to compare with those of EP and MP. Whole body crude protein of fish fed EP at 9 months was significantly higher than that of fish fed MP (P<0.05). Linolenic acid(18:3) and n-6/n-3 ratio in dorsal muscle of fish fed EP were significantly higher than those of fish fed MP (P<0.05). No significant differences were observed in the results of the flavor, color, taste and texture of the muscle of the fish fed EP compared to those fed MP (P>0.05). Based on the experimental results, we concluded that olive flounder cultured with EP is not inferior to those fed MP for quality characteristics of whole body and muscle.

The pepsinolytic hydrolysate from Johnius belengerii frame inhibited LPS-stimulated production of pro-inflammatory mediators via the inactivating of JNK and NF-κB pathways in RAW 264.7 macrophages

  • The objective of this study was to investigate the anti-inflammatory effects of the pepsinolytic hydrolysate from the fish frame, Johnius belengerii, on lipopolysaccharide (LPS)-stimulated RAW 264.7 macrophages. The J. belengerii frame hydrolysate (JFH) significantly suppressed nitric oxide (NO) secretion on LPS-stimulated RAW264.7 macrophages. Moreover, the JFH markedly inhibited the levels of protein and mRNA expression of inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2). Furthermore, the LPS-stimulated mRNA expression of pro-inflammatory cytokines, including tumor necrosis factor $(TNF)-{\alpha}$, interleukin $(IL)-1{\beta}$, and IL-6 was downregulated when cells were cultured with the JFH. The JFH significantly reduced the phosphorylation of c-Jun N-terminal kinase (JNK) and the translocation of nuclear factor-kappa B ($NF-{\kappa}B$) in macrophages. As the result, the JFH has the potential anti-inflammatory activity via blocking the JNK and $NF-{\kappa}B$ signal pathways.

Anisakiasis에 대한 임상적 고찰 (Clinical Review of Acute Gastric Anisakiasis)

  • 7 cases of Acute Gastric Anisakiasis have been reviewed at the Department of Internal Medicine. Yeungnam University Hospital from March, 1983 to December 1986. 1. Of 7 cases, the ratio of male and female was 3:4 and the age 30s occupied most (5 cases) the 50s' followed (2 cases). 2. Chief complains were in order of epigastric pain (6 cases), vomiting (3 cases), Nause (2 cases), Ulticaria (1 case) & epigastric fulling sensation (1 case). 3. In all 7 cases, the causative fish is Astroconcer myriaster. 4. The onset of symptoms was observed within 5 to 7 hours after eating the causative raw fish. 5. Gastrofiberscopic exam was undergo within 18 hours since onset of symptoms. 6. The gastrofiberscopic findings were gastric mucosal edema and redness in all 7 cases. One case also had mucosal erosion. The involved sites of the stomach were in frequency of order, the body (5 cases), the antrum (2 cases). 7. The symptoms of the patients relieved spontaneously without medical treatment after the extraction of the larva with biopsy forcep during gastrofiberscopy. 8. In Korea, many people take raw fish and there may be many patients of Anisakiasis so if much concern and careful observation are paid to whom, with severe cramping epigastric pain after taking of raw marine fish in about 10 hours, we could find out and confirm more patients suffering from Anisakiasis by immediate endoscopic gastrofiberscopy.

Comparative analysis of nutritional values of riverine and marine hilsa (Tenualosa ilisha; Hamilton, 1882)

  • A study was performed to analyze the biochemical composition (moisture, protein, fat, ash, salt value, iron, calcium and phosphorus) of raw and salted hilsa. Pure (with less than 1% impurities) and clean dry salt was used (fish weight : salt weight = 3 : 1) for salting the hilsa. The nutrients values of the hilsa from two different regions were significantly (p < 0.05) varied. The biochemical compositions were also different before and after the processing of the hilsa. Riverine hilsa contains relatively more moisture ($57.79{\pm}0.51%$) and protein ($15.65{\pm}0.50%$) than marine hilsa. Fat ($16.39{\pm}0.51%$) and salt ($1.80{\pm}0.14%$) contents are higher in marine hilsa; whereas the ash ($7.88{\pm}0.35%$) content was higher in the riverine hilsa. Minerals like iron ($4.92{\pm}0.32mg/100g$) and calcium ($480.02{\pm}6.73mg/100g$) remain in large amounts in the marine hilsa, but the phosphorus ($112.36{\pm}4.40mg/100g$) content remains at a high level in the riverine hilsa. In addition, the protein (raw condition, $18.54{\pm}0.46%$, riverine; $17.12{\pm}0.42%$, marine and salted condition, $32.54{\pm}0.5%$, riverine; $27.31{\pm}0.48%$, marine) and fat (raw condition, $15.41{\pm}0.46%$, riverine; $19.07{\pm}0.51%$, marine and salted condition, $11.58{\pm}0.39%$, riverine; $13.6{\pm}0.55%$, marine) contents were higher in the abdominal region of the riverine and marine hilsa both in the raw and salted conditions than in the head and caudal region.

수산부산물의 발생·이용 실태 평가 및 해양바이오 산업화 방안 (Current Status and Evaluation of Fisheries By-products: Major Options to Marine Bioindustrial Application)

  • Since the existing mass production and consumption systems are no longer sustainable, countries are pushing for policies to make fisheries by-products as resources in an eco-friendly manner, and international standards are also being strengthened to increase the value of by-products. In Korea, economic and environmental perceptions of the by-products are rapidly changing, such as realizing carbon neutrality and enhancing circular resources by Korean Sustainable Development Goals. Raw materials derived from the by-products have been steadily imported from 2018. In particular, the number of imports of fish collagen peptides was only 16 number of times in 2017, but was rapidly increased to 483 number of times in 2020. Simultaneously, the demand for raw materials and nutrients for health functional food derived from fish by-products, which did not exist statistically until 2017, started to arise from 2018, and in 2019, consumption of high-value-added raw materials for fish by-products increased by 45% compared to the previous year. However, limitations are in legal and biotechnical industry aspects while its value as a biomaterial is recognized in the by-products-related industry. In this study, therefore, the status of by-products for upcycling biomaterials was reported and provided a scientific basis for supporting governmental strategies. In order to fulfill with the principles of a sustainable circular economy, the factors on hinder the marine bio-industrialization of the by-products were derived and suggested directions and plans for development into a high-value added the by-products as the marine bio-industry by substituting imported raw materials to support the development.

넙치 실용배합사료개발을 위한 현장적용시험 (Commercial Scale Evaluation of Practical Extruded Pellet Feed for the Olive Flounder (Paralichthys olivaceus))

  • This study was conducted to evaluate the effects of extruded pellet (EP) diets, as compared to a raw fish moist pellet (MP) diet for olive flounder, Paralichthys olivaceus, grown in commercial-scale aquaculture for 1 year. Four diets with duplication per diet were formulated for this experiment: two experimental EP diets (EP1 and EP2), one commercial EP diet (CEP), and a raw fish MP diet (MP). The MP diet consisted of 80% frozen horse mackerel and 20% commercial binder meal. Fish weighing $30.1{\pm}0.1 g$ ($mean{\pm}SD$) were distributed randomly to each aquarium as a group of 2,600 fish. Weight gain (WG) and feed efficiency ratio (FER) of fish fed EP2 and MP were higher (P<0.05) than those of fish fed CEP, while those of fish fed EP1 did not differ (P>0.05) from those fed EP2 and MP. However, fish fed the MP diet had a higher survival rate than fish fed the other diets. Fish fed EP2 had higher serum, phospholipids and total protein levels, and lower levels of serum glutamic-oxaloacetic transaminase (GOT), glutamic-pyruvic transaminase (GPT), and total cholesterol than fish fed MP (P<0.05). Dorsal muscle and liver proteins and lipid of fish fed EP1 were higher (P<0.05) than those of fish fed CEP, while those of fish fed EP1 and MP did not differ from those of fish fed EP2 (P>0.05). These results strongly suggest that EP1 could be developed to replace MP for grow-out stage production of olive flounder without adverse effects on growth performance.

시판 넙치사료의 평가와 모이스트펠렛사료중 냉동어류를 분말배합사료로 대치 (Evaluation of Commercial Diets and Replacement of Raw Fish with Formulated Diets in Moist Pellet for Olive Flounder (Paralichthys olivaceus))

  • 6종의 시판 넙치 사료 $(Cl\~C6)$를 실험실에서 만든 배합사료 (S)와 비교하여 그 품질을 평가하고 넙치 사육에서 모이스트펠릿 제조시 냉동 어류를 배합 사료로 완전 대치할 수 있는지를 연구하기 위하여 두 가지 실험을 실시하였다. 실험 1에서는 넘치 (42g) 20마리를 넣은 수조 3개를 S 및 $Cl\~C6$에 각각 배치하여 수온 $17^{\circ}C$에서 4주간 실험 사료를 먹였다. 실험 2에서는 넙치 (78g) 400마리를 넣은 수조 3개를 각각 $100\%$ 실험실 배합 육성 사료 (G) $50\%\;G+50\%$ 냉동 고등어 ($70\%$ 수분)$50\%\;C6+50\%$ 법동 고등어 구에 배치하여 6주간 실험 사료를 먹였다. 실험 1에서 S를 먹인 넙치의 체중 증가율은 C2나 C3를 먹인 넙치의 체중 증가율 보다 현저히 (P< 0.05) 높았으며 (4주간 한마리 당 각각 26.7 vs 15.4와 17.5g). S와 기타 시판 넙치 사료간에는 유의차가 없었다(P> 0.05). 사료 효율은 C2 사료만이 S에 비해 현저히 (P< 0.03) 낮았다. 실험 2에서는 실험실 배합 사료 $100\%,\;50\%$ G 혹은 $50\%\;C6+$ 냉동 고등어로 된 모이스트펠릿 사료를 먹인 넙치간에는 체중 증가나 사료 효율에서 유의차 (P> 0.05)가 없었다. 이 연구 결과는 시판 넙치 사료는 실험실 배합 사료와 비교할 때 개선의 여지가 있음을 말해 주고, 냉동 어류를 사용하지 않고도 $100\%$ 배합 사료만으로 넙치사육이 가능함을 암시해 준다.
