  • This study investigated the effect of ovariectomy (OVX) on the rat bone fur long term (22 weeks). In previous researches, there were many studies for morphology of OVX-induced osteoporotic bones based on micro-Computed Tomography (micro-CT). However, there were few studies fpr detecting and tracking changes of mechanical characteristics in the lumbar vertebrae of OVX rat fur long-term. For this study, one female Sprague-Dawley rat was used: an OVX rat. The 4th lumbar of the OVX rat was utilized as a specimen. Morphological characteristics could be investigated fur the lumbar vertebrae in an OVX rat by using in-vivo Micro-CT. An OVX rat was scanned at week 0 (just before surgery), at week 4, at week 8, at week 16 and at week 22 after surgery. Micro finite element $({\mu}FE)$ analysis was used to investigate mechanical characteristics in the lumbar vertebrae for an OVX rat.

The Bioactivity of Natural Product in the Ovariectomized Rat

  • To investigate the deaging effects of introperitoneally injected Chondroitin Sulfate (CS) on various enzyme activity (AST, ALT, MDA (Malon dialdehyde), SOD (Superoxide dismutase), GPx (Glutathione peroxidases) and histophathology of liver tissue, ovariectomized rats were used. The antioxidative effects of chondroitin sulfate (100 mg/kg and 200 mg/kg body weight) were investigated at the antioxidative enzyme activities of liver homogenate fractions (liver total homogenate, mitochondrial, and microsomal fractions) and sera. In addition, the rat liver was histologically examined. Intraperitoneally injected CS, depend on dosage, indicated a protective effect against ovariectomy-inducted aging. Moreover, inflammation and cirrhosis in liver tissue of CS treated group were significantly decreased. Based on these results, intraperitoneally injected CS is a useful material to delay aging.

The Change of Bone Metabolism in Ovariectomized Rats : Analyses of MicroCT Scan and Biochemical Markers of Bone Turnover

  • Objective : The purpose of this study was to verify the appropriateness of ovariectomized rats as the osteoporosis animal model. Methods : Twelve female Sprague-Dawley rats underwent a sham operation (the sham group) or bilateral ovariectomy [the ovariectomy (OVX) group]. Eight weeks after operations, serum biochemical markers of bone turnover were analyzed; osteocalcin and alkaline phosphatase, which are sensitive biochemical markers of bone formation, and C-terminal telopeptide fragment of type I collagen C-terminus (CTX), which is a sensitive biochemical marker of bone resorption. Bone histomorphometric parameters and microarchitectural properties of 4th lumbar vertebrae were determined by micro-computed tomographic (CT) scan. Results : The OVX group showed on average 75.4% higher osteocalcin and 72.5% higher CTX levels than the sham group, indicating increased bone turnover. Micro-CT analysis showed significantly lower bone mineral density (BMD) (p=0.005) and cortical BMD (p=0.021) in the OVX group. Furthermore, the OVX group was found to have a significantly lower trabecular bone volume fraction (p=0.002). Conclusion : Our results showed that bone turnover was significantly increased and bone mass was significantly decreased 8 weeks after ovariectomy in rats. Thus, we propose that the ovariectomized rat model be considered a reproducible and reliable model of osteoporosis.

갈근 추출물이 에스트로겐 결핍 Rat의 비만관련 호르몬에 미치는 영향 (Effects of Pueraria lobata on Obesity Related Hormones in Rats with Estrogen Deficiency)

  • Objectives: This study was conducted to evaluate the effects of Pueraria lobata (PL) extract on obesity related hormones in rats with estrogen deficiency. Methods: The experiments were performed with the use of ovariectomized rats as estrogen-deficient obesity rat model. They were grouped Normal (sham operation group), Control (ovariectomy group), 50 mg/kg PL (ovariectomy group+50 mg/kg of PL), 100 mg/kg PL (ovariectomy group+100 mg/kg of PL), 200 mg/kg PL (ovariectomy group+200 mg/kg of PL). PL extract was orally administered for 8 weeks once a day. Body weights and serum levels of hormones, such as leptin, estradiol, cholecystokinin (CCK), ghrelin, and adiponectin were estimated by ELISA. Results: PL extract significantly decreased body weight, the serum levels of leptin in estrogen-deficient obesity rats. PL extract significantly increased the serum levels of estradiol and CCK. However, PL extract did not directly effect on the levels of ghrelin and adiponectin in estrogen-deficient obesity rats. Conclusions: It is concluded that PL extract reduced body weight, and regulate the hormones related to energy metabolism. PL extract decreased the serum levels of leptin. PL extract increased the serum levels of estradiol and CCK. These results indicate that PL might have potentials for treatment of obesity and complications during the menopause caused by estrogen-deficiency.

Histomorphometric Analysis of the Spine and Femur in Ovariectomized Rats Using Micro-Computed Tomographic Scan

  • Objective : The purpose of this study was to evaluate the different patterns of bone loss between the lumbar spine and the femur after ovariectomy in rats. Methods : Twenty-four female Sprague-Dawley rats underwent a sham operation (the sham group) or bilateral ovariectomy (the ovariectomized group). Four and eight weeks after operation, six rats from each of the two groups were euthanized. Serum biochemical markers of bone turnover including osteocalcin and alkaline phosphatase (ALP), which are sensitive biochemical markers of bone formation, and the telopeptide fragment of type I collagen C-terminus (CTX), which is a sensitive biochemical marker of bone resorption, were analyzed. Bone histomorphometric parameters of the 4th lumbar vertebrae and femur were determined by micro-computed tomography. Results : Ovariectomized rats were found to have higher osteocalcin, ALP and CTX levels than sham controls. Additionally, 8 weeks after ovariectomy in the OVX group, serum levels of osteocalcin, ALP and CTX were significantly higher than those of 4 weeks after ovariectomy. Bone loss after ovariectomy was more extensive in the 4th lumbar spine compared to the femur. Bone loss in the 4th lumbar spine was mainly caused by trabecular thinning, but in the femur, it was mainly caused by trabecular elimination. Conclusion : The present study demonstrates different patterns of bone loss between the 4th lumbar spine and the femur in ovariectomized rats. Therefore, when considering animal models of osteoporosis, it is important that bone sites should be taken into account.

The Effect of Chondroitin Sulfate against Oxidative Stress and Atherosclerosis In Ovariectomized Rat

  • The surgically ovariectomized rat induces aging by reactive oxyyen species(ROS) generation. Free oxygen radicals have been proposed as important causative agents of aging. The purpose of this study was to investigate the effect of Chondroitin Sulfate(CS) to prevent ovariectomy(OVX) induced oxidative stress and atherosclerosis. 난소절제로 유도한 노화쥐에서 chondroitin sulfate에 의한 산화 스트레스의 감소효과 (Reduction of Oxidative Stress by Chondroitin Sulfate in the Ovariectomy-Induced Aging Rat)

  • 노화과정에 산소라디칼이 관여할 가능성을 난소를 절제한 횐 쥐를 동물모델로 하여 알아보았다. 난소를 절제한 쥐와 난소를 절제하지 않은 정상 쥐에서의 생리활성 및 항산화 효과를 알아보기 위하여 CS를 사용하였다. 난소를 절제한 횐 쥐에 각각 CS를 각 100 mg/kg과 200 mg/kg을 투여하고, 이것과 비교하기 위해 난소를 절제한 대조군과 난소를 절제하지 않은 정상군으로 하여 항산화 효과에 관한 실험방법으로 진행하였다. 노화 유도된 쥐의 각 조직에서 지질 과산화가 증가되었고 유리기 반응이 더 심하게 일어난 간 조직에서 노화 진행이 그만큼 촉진되었음을 알 수 있었다. 산소라디칼 반응이 항진된 원인을 규명코자 간 조직에서 항산화 효소의 활성을 조사한 결과, SOD와 GSH의 활성이 감소되었으며, 노화나 지질과산화가 심하게 진행되었던 간 조직에서 이들 항산화 효소의 활성감소가 심한 경향을 나타내었다. AST의 활성은 OVX 군이 Sham 군에 비해 2.1배 높게 나타났으며, ALT 활성은 크게 변화하지 않았다. 과산화지질한량은 OVX 군이 Sham 군에 비해 1.4배 높게 나타났으며, CS를 투여한 군에서는 31%-38%의 저해율을 보였다. 항산화 효소들은 Sham 군에 비해 OVX 군에서 낮게 나타났으며, 물질 투여 군에서는 정도의 차이는 있지만 대체로 증가하는 경향을 보였다. 이러한 결과에서 보듯이, 난소절제로 생성된 유리기는 물질 투여로 인한 항산화 효소의 증가를 가져오고 이는 결국 유리기로 손상된 막의 지질과산화 정도를 완화시켜 세포를 보호하는 효과를 가져왔다고 할 수 있다. 따라서 CS는 여러 가지 측면에서 난소절제로 인해 유도된 지질과산화 및 활성산소에 의한 공격으로부터 세포를 보호하고 회복시켜주는 효과가 있다고 사료되어진다.

성숙한 쥐의 대퇴골과 요추의 조성과 골격관련 인자에 대한 난소절제와 칼슘섭취 효과 (Effect of Ovariectomy and Calcium Intake on Femur and Lumbar Compositions and Bone Related Factors in Adult Rats)

  • This study explored the effect of calcium levels and/or ovariectomy on bone composition and its related factors using the female Sprague-Dawley rats which achieved peak bone mass in normal(0.5%) calcium intake during growth period. The rats were randomly divided into six groups and fed 0.1%, 0.5% and 1.5% calcium diets for 8 weeks after ovariectomized and sham operation. The results indicated that body weight gain was higher in ovariectomized groups than sham groups, regardless of dietary calcium levels and food intakes. Serum Ca and P concentrations were normal level regardless of dietary calcium levels and operation. Estrogen concentration was decreased in ovariectomized rat groups. Serum alkaline phosphatase activity and urinary hydroxyproline were increased in ovariectomized groups. When rats were fed normal Ca diet during growing period, weight, length and breaking force of femur were not significantly different in all groups but increased the same level. Generally, lipid contents in lumbar and femur were lower in low calcium groups and this effect was more pronounced in femur. In case of femur, the significant decrease in Ca contents of bone was observed in a relatively short period of feeding low Ca diet, even though it appeared th desirable peak bone mass had achieved through the growing period by supplementing the adequate amount of calcium. The marked decrease of estrogen levels after ovariectomy did not seem to influence greatly bone parameters measured except bone wet weight. Therefore, this study indicated that lower intake of Ca might be more important determinating factor against osteoporosis than postmenopausal state stimulated by ovariectomy in female rats. With normal or high intake of Ca it appears possible to prevent bone loss in postmenopausal period, and this might apply only in case of achieving peak bone mass in the growing with the adequate intake of calcium. Therapeutic Advantages of Treatment of High-Dose Curcumin in the Ovariectomized Rat

  • Objective : Although curcumin has a protective effect on bone remodeling, appropriate therapeutic concentrations of curcumin are not well known as therapeutic drugs for osteoporosis. The purpose of this study was to compare the bone sparing effect of treatment of low-dose and high-dose curcumin after ovariectomy in rats. Methods : Forty female Sprague-Dawley rats underwent either a sham operation (the sham group) or bilateral ovariectomy (OVX). The ovariectomized animals were randomly distributed among three groups; untreated OVX group, low-dose (10 mg/kg) curcumin administered group, and high-dose (50 mg/kg) curcumin group. At 4 and 8 weeks after surgery, serum biochemical markers of bone turnover were analyzed. Bone histomorphometric parameters of the 4th lumbar vertebrae were determined by micro-computed tomography (CT). In addition, mechanical strength was determined by a three-point bending test. Results : High-dose curcumin group showed significantly lower osteocalcin, alkaline phosphatase, and the telopeptide fragment of type I collagen C-terminus concentration at 4 and 8 weeks compared with the untreated OVX group as well as low-dose curcumin group. In the analyses of micro-CT scans of 4th lumbar vertebrae, the high-dose curcumin treated group showed a significant increase in bone mineral densities (p=0.028) and cortical bone mineral densities (p=0.036) compared with the low-dose curcumin treated group. Only high-dose curcumin treated group had a significant increase of mechanical strength compared with the untreated OVX group (p=0.015). Conclusion : The present study results demonstrat that a high-dose curcumin has therapeutic advantages over a low-dose curcumin of an antiresorptive effect on bone remodeling and improving bone mechanical strength.
