Effect of the Various Recovery Diets on the Body Composition of Protein-depleted Rat (각종 회복식이가 무단백식이를 섭취한 흰쥐의 체구성 성분에 미치는 영향)

  • The changes of the total body composition, internal organs, skeletal muscles, and epididymal fat pad in rats fed protein depletion diet and 4 different recovery diets were examined. Seventy-eight male Sprague-Dawley rats weighing $212{\pm}2$ gr were used, and the results were as follows : A) After the 2 weeks of protein depletion, body weight decreased about 20% from the initial weight. It was mainey due to the total body lipid reduction. Among various organ weights, liver and spleen reduced $35{\sim}58%$, kidney and heart reduced $18{\sim}30%$, and muscles reduced $2{\sim}13%$, while brain, epididymal fat pad were not changed significantly. In regarding protein and lipid contents of these tissues, protein in liver, lipid in muscle, and both in spleen were markedly reduced. B) With the 2 weeks of feeding recovery diets, the increases of body weight were different among 4 Groups. High-fat group gained at the highest level (67%), and high-CHO group the lowest (30%). Total body composition (%) cf the standard and high-protein groups recovered to the level of 0 day protein depletion, while protein in the high-fat group and water in the high-CHO group decreased, and fat in these 2 groups increased. Weights of organs and muscles of the high-protein and high-fat groups were similar to the standard group and tllose of the high-CHO group were lower than the standard group. Composition of organs and muscle in the high-protein group was similar to the standard group, while the N contents of the high-fat and high-CHO groups were lower an the lipid content of the high-fat group was higher than the standard group. The weight and lipid content of epididymal fat pad were the highest in the high-fat group.

  • Purpose : Phospholipase D (PLD) catalyzes the hydrolysis of phosphatidylcholine to phosphatidic acid (PA) and choline. Recently, PLD has been drawing much attentions and considered to be associated with cancer Process since it is involved in cellular signal transduction. In this experiment, oleate-PLD activities were measured in various tissues of the living rats after whole body irradiation. Materials and Methods : The reaction mixture for the PLD assay contained $0.1\;\muCi\;1,2-di[1-^{14}C]palmitoyl$ phosphatidylcholine 0.5mM phosphatidylcholine, 5mM sodium oleate, $0.2\%$ taurodeoxycholate, 50mM HEPES buffer(pH 6.5), 10mM $CaCl_2$, and 25mM KF. phosphatidic acid, the reaction product, was separated by TLC and its radioactivity was measured with a scintillation counter. The whole body irradiation was given to the female Wistar rats via Cobalt 60 Teletherapy with field size of 10cmx loom and an exposure of 2.7Gy per minute to the total doses of 10Gy and 25Gy. Results : Among the tissues examined, PLD activity in lung was the highest one and was followed by kidney, skeletal muscle, brain, spleen, bone marrow, thymus, and liver. Upon irradiation, alteration of PLD activity was observed in thymus, spleen, lung, and bone marrow. Especially PLD activities of the spleen and thymus revealed the highest sensitivity toward $\gamma-rar$ with more than two times amplification in their activities In contrast, the PLD activity of bone marrow appears to be reduced to nearly $30\%$. Irradiation effect was hardly detected in liver which showed the lowest PLD activity. Conclusion : The PLD activities affected most sensitively by the whole-body irradiation seem to be associated with organs involved in immunity and hematopoiesis. This observation s1ron91y indicates that the PLD is closely related to the physiological function of these organs, Furthermore, radiation stress could offer an important means to explore the phenomena covering from cell Proliferation to cell death on these organs.

Inhibitory Effects of ${\gamma}$-Aminobutyric Acid on the Contractility of Isolated Rat Vas Deferens (흰쥐의 적출 정관 수축성에 대한 ${\gamma}$-Aminobutyric Acid의 억제작용)

  • GABA is an inhibitory neurotransmitter in central nervous system and produce sedative, antianxiety and muscle reaxing effects via $GABA_A$ receptor or $GABA_B$ receptor. Recently it is known that GABA is widely distributed throughout peripheral organs and may playa physiological role in certain organ. The vas deferens is innervated by species-difference. These study, therefore, was performed to investigate the mode and the mechanism of action of GABA on the norepiniphrine-, ATP- and electric stimulation-induced contraction of vas deferens of rat. Sprague-Dawley rats were sacrificed by cervical dislocation. The smooth muscle strips were isolated from the prostastic portion and were mounted in the isolated muscle bath. PSS in the bath was aerated with 95/5%-$O_2/CO_2$ at $33^{\circ}C$. Muscle tensions were measured by isometric tension transducer and were recorded by biological recording system. 1. GABA, muscimol, a $GAB_A$ agonist, and baclofen, a $GABA_B$ agonist inhibited the electric field stimulation(EFS, 0.2Hz, 1mSec, 80 V, monophasic square wave)-induced contraction with a rank order of potency of GABA greater than baclofen greater than muscimol. 2. The inhibitory effect of GABA was antagonized by delta aminovaleric acid(DAVA), a $GABA_B$ antagonist, but not by bicuculline, a $GABA_A$ mtagonist. 3. The inhibitory effect of baclofen was antagonized by DAVA, but the effect of muscimol was not antagonized by bicuculline. 4. Exogenous norepinephrine(NE) and ATP contracted muscle strip concentration dependently, but the effect of acetylcholine was negligible : and GABA did not affect the NE-and ATP-induced contractions. 5. GABA, baclofen and muscimol did not affect basal tone, and GABA did not affect the NE-and ATP-induced contractionsm 6. EFS-induced contraction was including 2 distinctable components. The first phasic component was inhibited by beta gamma-methylene ATP(mATP), a desensitizing agent of APT receptor and the second tonic component was reduced by pretreatment of reserpine(3 mg/Kg, IP). 7. GABA inhibited the EFS-induced contraction of reserpinized strips, but not the mATP-treated strips. These results suggest that in the prostatic portion of the rat vas deferens, adrenergic and purinergic neurotransmissions are exist, and GABA inhibits the release of ATP via presynaptic $GABA_B$ receptor on the excitatory neurons.

Evaluating The Radioprotective Effect of Avocado Peel Extracts upon Rat Exposed to 6 MV X-Ray (6 MV X-ray에 피폭된 흰쥐에 대한 아보카도 껍질 추출물의 방사선방호효과 평가)

  • In this study aims to investigate the radiation protection effect of avocado peel extracts on the Sprague-Dawely rats. 52 male rats were randomly classified into 4 groups. NC Group was a normal control group, PA Group was a group injected avocado peel extracts, IR Group was irradiated group, and lastly PA+IR Group was set as an irradiated group after injected of avocado peel extracts. Avocado peel extract was administered orally at 200 mg/kg once a day for 14 days before irradiation, and the radiation dose was systemically irradiated with 6 MV X-ray of 7 Gy. On the 4 and 21 days after irradiation, the experimental animals were sacrificed to evaluate the change in blood cell composition, spleen index, and histopathological evaluation of the liver and small intestine. As a result, the PA+IR Group showed a significantly greater recovery of lymphocytes(p<0.01), red blood cells(p<0.01), and platelets(p<0.05) than the IR Group. It was also confirmed that the activation of Superoxide Dismutase(SOD) was further increased. Histopathologically, observed that nuclei aggregation and cytoplasmic expansion were slightly reduced in the PA+IR Group in the liver. and the damage was significantly reduce(p<0.01) in the change of villi length due to damage to the small intestine cells. Based on the above results, avocado peel extract can be expected to act as a radiation protection agent that can reduce damage to blood cells and major organs caused by irradiation.

Effect of Cassia tora L. Powder Added-Diets on the Accumulation of Cadmium in Rat (결명자 첨가식이가 흰쥐의 체내 카드뮴 축적에 미치는 영향)

  • The purpose of this study is to investigate the effect of raw Cassia tora L. powder added-diets on reducing cadmium accumulation in rats. The experimental animals were Sprague-Dawley family(♂, 4 weeks) which was classified into normal group CN, compared group CS, Cd-added group Cl and groups C2, C3, C4 in which 0.5, 1.0 and 1.5% of the Cassia tora L. powder are added, respectively. The growth rate and food efficiency ratio, and the amounts of accumulated cadmium in rats for S weeks were measured and analyzed. The results are as follows; 1. The rates of weight gain decreased in the order of C3>C2>C4>Cn>Cs>Cl groups, and Cl group to which only cadmium water had been fed was the lowest among them. The correlation between groups Cl and C3 was significantly different at the 1% level. 2. Food efficiency ratio(FER) decreased in the order of C3>C2>Cs>Cn>C4>Cl, and the FERs of C3, C2, CS, CN and C4 are greater than that of Cl by 22.87, 19.59, 18.54, 14.20 and 13.17%, respectively. 3. As for the Cassia tora L. powder-added groups, the amounts of cadmium accumulated in organs and tissues, that is, the brain, heart, spleen, liver, lungs, testicles. kidney, femoral muscle and leg bones were 0.45 $\pm$ 0.04 to 0.83$\pm$0.04, 1.68$\pm$0.02 to 2.16$\pm$0.02, 3.26$\pm$0.05 to 4.62$\pm$0.27, 37.52$\pm$0.09 to 47.71$\pm$0.73, 1.07$\pm$0.10 to 1.66$\pm$0.04, 1.04$\pm$0.06 to 1.24$\pm$0.08, 36.79$\pm$0.20 to 39.61 $\pm$0.53, 0.87$\pm$0.02 to 1.00$\pm$0.02 and 0.65$\pm$0.17 to 1.27 $\pm$ 0.06 $\mu\textrm{g}$/g, respectively. 4. The accumulated Cd content for C4 was the lowest among Cassia tora L. powder-added groups. When the results for C4 are compared with those for Cl, it is observed that each cadmium content accumulated in the brain, heart spleen, liver, lungs, testicles, kidney. femoral muscle and leg bones is dropped by 49.03, 22.56, 36.02, 35.75, 41.75, 36.20, 37.00, 22.77 and 56.67 %, respectively. Effects of gamma-irradiation on the survival and development of Metagonimus yokogawai metacercariae in rats (감마선 조사가 요코가와흡충의 흰쥐 내 생존 및 발육에 미치는 영향)

  • To evaluate the feasibility of irradiation as a control measure for metagonimiasis, the metacercariae of Metagonimus yokogcwni were irradiated with gamma ray, either after isolation from the sweetfish (Plecoglossus cltivelis) or in situ of the fish, and their survival and development in rats were observed at 7 days post-infection. The radiation dose varied from 5 to 100 Gy for the metacercaria-irradiation group and from 5 to 500 Gy for fish-irradiation group. The results showed that the worm recovery rate from the irradiation groups decreased as the radiation dose was increased. Higher doses of radiation were required for the fish-irradiation group to obtain the same results as the metacercaria-irradiation group. The LD50 of the metacercaria-irradiation group was 4.5 Gy, whereas that of the fish-irradiation group 6.2 Gy A few number of worms which survived until 7 days in rats were severely retarded especially in the growth of their reproductive organs, j.e., complete or partial failure in the development of testes and formation of uterine eggs . The present study revealed that irradiation of sweetfish by 200 Gy is effective to control infectivity as well as development of M. vokogawai metacercariae in rats.

  • Background: Cassia tora L., an annual or perennial plant of the Fabaceae family, is traditional medicine with various biological activities, including anti-constipation and, anti-inflammation. Chemical compounds such as anthraquinone glycoside and naphthalene derivatives have been isolated from this plant. Cassia tora L. is a common contaminant of agricultural commodities, but is toxic to cattle and poultry. Methods and Results: To investigate the potential toxicity, Cassia tora L. aqueous extract (CO) was administered orally to rats for 26 weeks at 0 (control), 300, 1,500 and 3,000 mg/kg/day (n = 10 for male rats for each dose). The positive control comprised animals orally administered anthraquinone 100 mg/kg/day. There was no treatment-related mortality. An increase in the kidney weight was observed at 3,000 mg/kg/day of CO and anthraquinone 100 mg/kg/day. Macrophage infiltration in the colon was observed at CO 1,500 and 3,000 mg/kg/day and anthraquinone 100 mg/kg/day, but there were no significant toxicological changes in the incidence and severity of the finding. Conclusions: The oral no-observed-adverse-effect level (NOAEL) of CO was 3,000 mg/kg/day in male rats and no target organs were identified. In addition, 300 mg/kg was found to be the no-observed-effect level (NOEL) for systemic toxicity under the conditions of the study.

Gene Expression Analyses in Hypothalami of Immobilization-stressed and BoshimgeonbiTang-treated Mice Using cDNA Microarray (구속 스트레스 (immobilization stress)를 가한 rat의 hypothalamus에서의 유전자 발현 및 포심건비탕의 항스트레스 효과에 관한 cDNA microarray 분석)

  • The genetic effects of restraint stress challenge on HPA axis and the therapeutic effect of Boshimgeonbi-Tang on the stress were studied with cDNA microarray analyses on hypothalamus using an immobilization-stress mouse as stress model. Male CD-1 mice were restrained in a tightly fitted and ventilated vinyl holder for 2hours once a day, and this challenge was repeated for seven consecutive days. The body weights of the immobilization-stress mice were diminished about 25 percent degree as compared to normal ones. Seven days later, total RNA was extracted from the organs of the mouse, body-labeled with CyDye/sup TM/ fluorescence dyes (Amersham Bioscience Co., NJ), and then hybridized to cDNA microarray chip. Scanning and analyzing the array slides were carried out using GenePix 4000 series scanner and GenePix Pro/sup TM/ analyzing program, respectively. The expression profiles of 109 genes out of 6000 genes on the chip were significantly modulated in hypothalamus by the immobilization stress. Energy metabolism-, lipid metabolism-, apoptosis- and signal transduction-related genes were transcriptionally activated whereas DNA repair-, protein biosynthesis-, and structure integrity-related genes were down-regulated in hypothalamus. The 58 genes were up-regulated by the mRNA expression folds of 1.5 to 7.9. and the 51 genes were down-regulated by 1.5 - 3.5 fold. The 20 genes among them were selected to confirm the expression profiles by RT-PCR. The mRNA expression levels of Tnfrsf1a (apoptosis), Calm2 (cell cycle), Bag3 (apoptosis), Hspe1 (protein folding), Aatk (apoptosis), Dffa (apoptosis), Itgb1 (cell adhesion), Vcam1 (cell adhesion), Fkbp5 (protein folding), BDNF (neuron survival) were restored to the normal one by the treatment of Boshimgeonbi-Tang.

Effect of Chlorella on Metallothionein Synthesis and Binding Capacity of Cadmium in Cadmium Poisoned Rat Liver and Kidney

  • The rate of metallothionein synthesis on cadmium-poisoned rats reflects the level of toxicity, and also it reduces the toxicity which is caused by the uptake of cadmium. Chlorella supplementation in the diets of the cadmium-poisoned rats decreased the concentration of cadmium in blood and urine compared with the control group. Although the liver and kidneys of rats are major target organs of cadmium and coherence of metallothionein and cadmium, no previous study has determined the correlation between the rate of metallothionein synthesis in the liver and kidneys of rats and dietary supplementation of chlorella with cadmium uptake. This study analyzed total metallothionein level on the tissue of the liver and kidneys, the concentration of cadmium bound to the metallothionein, and the total concentration of cadmium on the tissue of the liver and kidneys after dietary supplementation with 1%, 5%, and 10% dried chlorella and 40 ppm of cadmium to 46 male SD rats (mean weight: $150\pm20\;g$) for 4 weeks. According to the data analysis of the total rate of metallothionein synthesis in the liver and kidneys, the group of SD rats on the supplementation with 1% chlorella and 40 ppm of cadmium showed a rate of $93.2\pm8.9\;ng/g$, a significant decrease of 58.8% compared to that of the control group of SD rats on the supplementation with cadmium only, which showed a rate of $227.3\pm32.5 ng/g$ (P=0.0001). In contrast, no significant difference was observed through the changing of chlorella concentrations between 5% and 10% chlorella supplementation with cadmium. The group supplemented with 1% or greater chlorella levels represented a greater decrease in the total cadmium concentration of the kidney and liver tissues, the amount of total metallothionein synthesis, the amount of metallothionein with binding to cadmium, and the concentration of free cadmium without binding to metallothionein. Consequently, the supplementation of 1% and 5% chlorella was effective in reducing the synthesis of metallothionein for cadmium uptake, but increased the rate of binding of cadmium to metallothionein.

Immunohistochemical Studies of Human Ribosomal Protein S3 (rpS3)

  • The human ribosomal protein S3 (rpS3) was expressed in E. coli using the pET-I5b vector and the monoclonal antibodies (mAbs) were produced and characterized. A total of five hybridoma cell lines were established and the antibodies recognized a single band of molecular weight of 33 kDa on immunoblot with purified rpS3. When the purified rpS3 was incubated with the mAbs, the UV endonuclease activity of rpS3 was inhibited up to a maximum of 49%. The binding affinity of mAbs to rpS3 determined by using a biosensor technology showed that they have similar binding affinities. Using the anti-rpS3 antibodies as probes, we investigated the cross-reactivities of various other mammalian brain tissues and cell lines, including human. The immunoreactive bands on Western blots appeared to be the same molecular mass of 33 kDa in all animal species tested. They also appear to be extensively cross-reactive among different organs in rat. These results demonstrated that only one type of immunologically similar rpS3 protein is present in all of the mammalian brain tissues including human. Furthermore, these antibodies were successfully applied in immunohistochemistry in order to detect rpS3 in the gerbil brain tissues. Among the various regions in the brain tissues, the rpS3 positive neurons were predominantly observed in the ependymal cells, hippocampus and substantia nigra pars compacta. The different distributions of rpS3 in brain tissues reply that rpS3 protein may play an important second function in the neuronal cells.
