Antihyperglycemia Effect of Medicinal Plants Mixture in Streptozotocin-Induced Diabetic Rats (Streptozotocin 유발 당뇨쥐에서 항당뇨 생약 복합물의 혈당강하 효과)

  • This study was performed to investigate the hypoglycemic effect of single and repeated oral administration of medicinal herbal mixture (AD) in streptozotocin (STZ) induced diabetic rats. Angelica decursiva, Lycium chinense and Adenophora triphylla var. japonica Hara were selected by oral glucose tolerance test (OGTT) and mixed for AD mixture. In an oral glucose tolerance test, the AD inhibited the increase in blood glucose levels at 1 hr and 2 hr and decreased incremental glycemic response area under the curve. In a single administration of AD1 (100 mg/kg) and AD2 (500 mg/kg), significant reductions by 5.3% and 12.3% were observed in fasting blood glucose level for 4 hours. During the 1 month of the experimental period, AD1 and AD2 was given to the STZ induced diabetic rats. At 4th week, the fasting blood glucose levels of AD1 and AD2 caused a fall of 25.5% and 37.9%, respectively. In addition, the body weights were decreased by 7.7% (AD1) and 1.7% (AD2), respectively, compared with diabetic control (DC, decreasing of 10.2%). This study suggests that AD could be potentially useful for fasting and post-prandial hyperglycemia treatment and all these effects concluded to the use of this plant extract to manage diabetes mellitus.

Effect of Green Tea Products on Bone Metabolism Marker in Ovariectomized Rats with High Cholesterol Diet Intake (고콜레스테롤 식이를 섭취한 난소절제 흰쥐에서 녹차가공품이 골 대사 지표에 미치는 효과)

  • This study was designed to evaluate the effect of green tea products (GTP) on bone metabolism marker in ovariectomized (OVX) rats fed high cholesterol diet. Forty Sprague-Dawley female rats, 10 weeks of age ($279{\pm}2g$), were divided into 4 groups and fed on the experimental diets for 6 weeks: sham operated control (Sham-C) and OVX-control (OVX-C) groups treated high cholesterol diet. OVX-GTP 5% (OVX-G5) and OVX-GTP 20% (OVX-G20) groups were treated with high cholesterol diet containing 5% GTP and 20% GTP, respectively. Food efficient ratio was significantly (p<0.05) lower in OVX-G20 than in the other OVX groups. Bone mineral density of femur was not significantly different among the experimental groups in the order of Sham-C>OVX-G5 and OVX-G20>OVX-C. Alkaline phosphatase activities on serum was lower in the GTP supplement groups than in the OVX-C. Estradiol levels of serum were higher in the GTP supplement groups than in the OVX-C. Osteocalcin levels of serum was the lowest in the OVX-G20. Deoxypyridinoline crosslink values of urine, indicator of bone absorption, was the lowest in the OVX-G20 group. The GTP supplemented groups had a lower bone resorption ratio than in the OVX-C group. From the above results, these findings suggest the possibility of using GTP as a functional food materials related to bone metabolism in menopause.

Effects of Puerariae Flos on Antioxidative Activities and Lipid Levels in Hyperlipidemic Sprague-Dawley Rats (갈화(Puerariae Flos) 추출물이 고지혈 유발 흰쥐의 항산화활성 및 지질농도에 미치는 영향)

  • This study was carried out to investigate the effects of Puerariae Flos extracts on antioxidative potential, free radical generation and the lipid levels in rats. Sprague-Dawley rats were divided into 2 diet groups: AIN-76 diet (control group) and modified AIN-76 diet (cholesterol 0.5%) with 0.5% Puerariae Flos extracts for 4 weeks. Body weight and feed efficiency ratios from both groups were not significantly different. Antioxidative potentials significantly increased in the group fed Puerariae Flos extracts compared to control group (p<0.05). However, there was no difference in free radical generation. The weight of organs, such as heart, kidney, liver, and spleen, in rats were not different in both groups. The ratio HDL cholesterol to total cholesterol in the Puerariae Flos group was significantly higher than in the control group, while the other serum lipid parameters (total cholesterol, HDL cholesterol, triglyceride, and phospholipids) were not different between the two groups. These results imply that supplementation of Puerariae Flos extracts may beneficially contribute to improve antioxidant potential and to decrease the lipid levels in the blood.

Effects of Aqueous Azadirachta indica Extract on Hepatotoxicity in Rats (수용성 님추출물이 랫드의 간 독성에 미치는 영향)

  • BACKGROUND: Azadirachta indica Extract(AIE) containing azadirachtin as active ingredient have been used worldwide as environment-friendly organic material having pest control properties. However, the extracts prepared with different solvent and from different plant site is very diverse and have different toxicity. METHODS AND RESULTS: In this study, the four week repeated oral dose toxicity test of aqueous AIE in Sprague-Dawley rats was carried out to investigate the toxic effect of liver, main toxicity target organ of AIE. The male and female rats were divided into 4 groups, respectively; control(0 g/Kg bw), low-dose group(0.5 g/Kg bw), middle-dose(1.0 g/Kg bw) and high-dose group(2.0 g/Kg bw). As a results, relative liver weight increased with dose-dependent of AIE(p<0.05). Serum LDH in all AIE-treated groups were significantly lower than the control in male rats(p<0.05). However, serum GOT and GPT were significantly increased in all male AIE-treated groups in male rats(p<0.05) and, in particular, increase of serum GPT in dose-dependent manner raise the possibility of liver damage. Even through serum GLU was increased significantly in high-dose group in male rats compared to control, there were no significant differences of urinary GLU among all groups(p<0.05). In addition, histopathological examination of the liver did not reveal any lesions in all AIE-treated groups. CONCLUSION: In conclusion, 4 weeks of the repeated oral administration of AIE 2.0 g/Kg to rats has resulted no toxic response in liver. Therefore, AIE was no indicated to have any toxic effect in the SD rats, when it was orally administrated below the dosage 2.0 g/Kg/day for 4weeks.

Laminin-1 Expression in Bone Marrow Stromal Cells of Cyclophosphamide-treated Rat (Cyclophosphamide가 흰쥐 골수의 기질세포에서 Laminin-1의 발현에 미치는 영향)

  • The purpose of the present study is to investigate whether stromal cells supporting specific microenvironment for hematopoiesis of bone marrow are affected by toxicants and therapeutic drugs such as antibiotics and anticancer drugs and whether laminin-1 is associated with such effects. SD rats were intraperitoneally injected with 75 mg/kg of cyclophosphamide which is widely used to treat infant's solid tumor, leukemia and myeloma and sacrificed after 3 days, 1 week, 3 weeks or 5 weeks of injection. The bone marrow extracted and paraffin-sectioned was analyzed using immunohistochemical staining. A part of tissues was subjected to electron microscopy following reaction with rabbit anti-laminin antibody, biotinylated goat anti-rabbit IgG conjugated with 12 nm gold particles, and staining with uranyl acetate. 1. The bone marrow tissue at day 3 post injection with cyclophosphamide displayed dilated venous sinus, partial necrotic death, and decreased number of hematopoietic cells. Laminin-1 was intensively stained in the reticular and adipose tissues. 2. Up to 5 weeks post injection, laminin-1 was stained at a low level in the stromal tissue of bone marrow and the number of hematopoietic cell was increased. 3. Deposition of the gold particle which represents laminin-1 expression was observed at the highest level in the stromal cells of bone marrow obtained 3 days after injection, and decreased after 1 to 5 weeks. These results suggest that stromal cells which play a role in supporting microenvironment for bone marrow hematopoiesis augment induction of laminin-1 expression and activation upon administration of cyclophosphamide.

Ultrastructure of the Integument of Capillaria hepatica (syn. Calodium hepatica) (간모세선충(Capillaria hepatica) 표피의 미세구조)

  • Capillaria hepatica is a parasitic nematode which causes hepatic capillariasis in rodents and other mammals, including man. Rat species of the genus Rattus are main primary host and rates of genus Rattus of up to 100% have been reported. Infection to reservoir and other mammalian hosts occur incidentally due to ingestion of water or food contaminated with C. hepatica embryonated eggs. The worms mature exclusively inside the liver, but they die and disassemble soon after egg spawning in rats. Dead worms and their eggs cause immune response of focal necrosis and inflammation within the liver. C. hepatica adult with a thin and long body is similar to capillary. The members of Order Trichurida are characterized by having a stichosome and the bacillary bands in front of the body. As already mentioned, the adult C. hepatica residesin the liver, where it deposits groups of eggs, and finally die in the encapsulated tissue of the liver. They produce eggs that elicit a marked granulomatous reaction that eventually destroy the worms. And the adult worms were mixed with eggs. So the complete isolation of the worm and observation of intact ultrastructure is very difficult. In this study, integument structure of C. hepatica isolated from the liver of mouse at 7 weeks after inoculation of embryonated eggs were observed with scanning and transmission electron microscopy. As a results, body length of isolated C. hepatica was about 99 mm. Cuticle, bacillary band and bacillary pore were obtained in the integument of worm. Bacillary pore across cuticular surface of the worm were observed. According to the existence of cap material, external forms of bacillary pore can be divided into three types such as flat, ingression, and ingression with the cap material type. The complete isolation of the worm and observation of ultrastructure of integument will provide the fundamental data which is important in the nematode research including C. hepatica.

Tributyltin Induces Adipogenesis and Apoptosis of Rat Thymic Epithelial Cells (Tributyltin에 의한 흰쥐 흉선 내 상피세포의 지방세포 유도와 세포자연사 증가)

  • Tributyltin (TBT) is one of endocrine disrupters which are known as having similar function to sex steroid hormone inducing apoptosis in various tissues of rodents. Recently, it has been reported that TBT induces apoptosis in thymus causing the decreased thymic function, but little is known about the mechanism. To elucidate the mechanism, three-week-old SD female rats were orally administrated with TBT 1, 10, and 25 mg per body weight (kg) and sesame oil as a control for 7 days. On day 8, the thymi were obtained and weighed, and then the number of thymocytes was counted. We also performed H&E staining, TUNEL assay, and Annexin V flow cytometric analysis to examine the apoptosis rates and the structure in the thymus. Next, we investigated the adipogenesis and apoptosis-related mRNA expression levels in the thymi by real-time PCR. The thymic weight and the number of thymocytes were decreased by TBT in a dose-dependent manner. As a result of the H&E staining, the boundary between cortical and medullary area was blurred in the thymi of TBT treated rats compared to those of controls. In the results of TUNEL assay and Annexin V flow cytometric analysis, apoptosis rates in the thymus were increased after TBT treatment. The expression levels of thymic epithelial cell marker genes such as EVA, KGF, AIRE, and IL-7 were significantly decreased in the thymi of TBT treated rats, but $PPAR{\gamma}$, aP2, PEPCK, and CD36 were significantly increased. The expression of $TNF{\alpha}$ and TNFR1 as apoptosis-related genes also was significantly increased after TBT treatment. The present study demonstrates that TBT can increase the expression of adipogenesis and apoptosis-related genes leading to apoptosis in the thymus. These results suggest that the increased adipogenesis of thymus by TBT exposure might induce apoptosis in the thymus resulting in a loss in thymic immune function.

Effect of Ethane 1,2-Dimethane Sulfonate(EDS) on the Expression of Pituitary Gonadotropin in Male Rats (수컷 흰쥐 뇌하수체의 생식소자극호르몬 발현에 미치는 Ethane 1,2-Dimethane Sulfonate(EDS)의 효과)

  • Ethane 1,2-dimethane sulfonate(EDS), a toxin which specifically kills Leydig cells(LC), has been widely used to prepare the reversible testosterone(T) depletion rat model. In the present study, we monitored the gene expression profiles of pituitary gonadotropins, LH and FSH, up to 7 weeks after EDS injection. Adult male Sprague-Dawley rats($300{\sim}350\;g$ B.W.) were injected with a single dose of EDS(75 mg/kg i.p.) and sacrificed on weeks 0, 1, 2, 3, 4, 5, 6 and 7. Total RNAs were purified from each pituitary, and the message levels of common alpha subunit($C{\alpha}$) of pituitary glycoprotein hormones, LH beta subunit($LH{\beta}$), FSH beta subunit($FSH{\beta}$) and GnRH receptor(GnRH-R) were evaluated by semi-quantitative RT-PCRs. The message levels of $C{\alpha}$ increased sharply during weeks 1-4, then return to the control level on week 5. The mRNA levels of $LH{\beta}$ were elevated after week 2, reached the peak at week 4, then declined to the control level after week 5. The message levels of $FSH{\beta}$ were elevated after week 2, reached the peak at week 3, then declined to the nadir at week 5. Similarly, the mRNA levels of GnRH-R were elevated after week 2, reached the peak at week 3, then gradually declined to the control level after week 5. The present study indicated that EDS treatment could induce reversible alterations in the transcriptional activities of gonadotropin subunits and GnRH-R in the anterior pituitary from male rats. EDS injection model might be useful to understand the mechanism of hormonal regulation of hypothalamus- pituitary neuroendocrine axis in male rats.

Effects of Ramie Leaves on Improvement of Lipid Metabolism and Antiobesity Effect in Rats Fed a High Fat/High Cholesterol Diet (모시잎이 고지방-고콜레스테롤 식이 흰쥐의 지질대사 개선 및 항비만 효과에 미치는 영향)

  • This study was performed to investigate the effects of ramie (Boehmeria nivea) leaf powder on improvements in lipid metabolism in serum, liver and adipose tissue, and the anti-obesity effect in rats fed a high fat/high cholesterol diet for 4 weeks to induce a hyperlipidemic and obese model rat. Male Sprague-Dawley rats weighting 210 g were divided into four groups; a normal diet group (N), a high fat/high cholesterol diet group (HFC), a high fat/high cholesterol diet with 5% ramie leaf powder group (HFC-RL), and a high fat/high cholesterol diet with 10% ramie leaf powder group (HFC-RH). The body weight gain increased with a high fat/high cholesterol diet, but gradually decreased in the ramie leaf powder fed groups compared with the HFC group. The liver index in the HFC group was highest among the four groups, although the difference was not significant compared with the ramie leaf powder fed groups. The adipose tissues weight in the HFC group was heavier than that of the N group, whereas those of groups fed ramie leaf powder decreased gradually. Alkaline phosphatase activity was not different between the HFC groups, but serum alkaline aminotransferase and lactate dehydrogenase activities decreased significantly after ramie leaf powder feeding. Levels of serum triglyceride, total cholesterol, LDL-cholesterol, atherogenic index and cardiac risk factors tended to decrease in the ramie leaf powder fed groups compared with the HFC group, whereas serum HDL-cholesterol level decreased in the HFC group and markedly increased in the ramie leaf powder fed groups. Levels of triglyceride and total cholesterol in the liver were significantly lower in the ramie leaf powder fed groups than in the HFC group. Levels of triglyceride and total cholesterol in adipose tissues were also lower in the ramie leaves powder fed groups than in the HFC group. The activities of heparinreleasable lipoprotein lipase (LPL) and total-extractable LPL in adipose tissues increased in the HFC group compared to that of the N group, but those of the ramie leaf powder fed groups decreased significantly. These results suggest that ramie leaf powder may improve lipid metabolism in serum, liver and adipose tissue and potentially reduce lipid storage.

Effects of Schizandra chinensis Extract on the Growth of Intestinal Bacteria Related with Obesity (오미자 추출물이 비만과 관련된 장내 세균의 생육에 미치는 영향)

  • This study was conducted to screen for plant food materials that improve human intestinal microflora, especially microorganisms associated with obesity. Among 30 tested plant food materials, the extract of Schizandra chinensis inactivated Eubacterium limosum, Bacteroides fragilis and Clostridium spp. Additionally, S. chinensis extract was also found to have a growth-promoting effect on Bifidobacterium spp.. The antimicrobial activity and antioxidant activity of the water extract did not decrease in respond to heating. Additionally, the water extract of S. chinensis did not show a toxic effect on the growth of Caco-2 cells. In vivo feeding tests were performed to investigate the influence of extract on the intestinal microflora in rats. Although the extract did not reduce obesity induced by a high fat diet, it led to significant increase in the population of Bifidobacterium spp. and a decrease in the population of Clostridium spp. in rats. Taken together, these results indicate that S. chinensis could be useful as a functional food component to control intestinal microbial flora.
