Effect of Edible Flower Extracts on Antioxidative and Biological Activities (식용꽃 추출물이 항산화 및 세포의 생리활성에 미치는 영향 - 유채꽃, 칡꽃, 장미꽃을 중심으로 -)

  • In order to promote the value of the flowers as new agricultural products, we investigated the biological activities of rape, arrowroot, and rose extracts. Biological activities investigated included antioxidant activity and the effects on 3T3-L1 fibroblast cells. When each flower was extracted with methanol, the antioxidant index and electron donating activity of roses was the highest $(IC_{50}$ of rose extract was $17.6 \mu{g}/m\ell$). When 3T3-L1 fibroblast cells were treated with extracts made with hexane, ethyl acetate, and ether, the rape extracts had a cytotoxic effect on the cells. 12.2% of cells survived when treated with a 3mg/$m\ell$ ether extract while those treated with the same concentration of hexane and ethyl acetate had survival rates of 76.2% and 78.6% respectively. In contrast to rape, the ether extract of arrowroot and rose stimulated the growth of 3T3-L1 cells. The effect of rose extracts was much bigger than those of other extracts. Although every rose extract stimulated the growth of the 3T3-L 1 cells, the ether extract stimulated growth up to 168.6% compared to the control at the concentration of $0.3mg/m\ell$, and 148.3% at the concentration of $1mg/m\ell$. The toxicity on cells treated with $H_2 O_2$ of $450\mu{M}l$was decreased with the addition of rose extract. The survival rate after treatment with rose extract at the concentration of $100\mu{g}/m\ell$ was increased to 71% compared to the 32% survival rate of control. From these results, it can be concluded that the extracts of arrowroot and rose seem to stimulate cells, whereas the extract of rape has a cytotoxic effect. Biological activities of ether extract were the strongest compared to those of other extracts at the tested concentrations.

  • Rape (Brassica napus L.) plants are one of the major oilseed crops. The main components of rapeseed are oil (35 to 47%) and protein (15 to 32%). For the biodiesel production, the development of a new variety of rape plant with high biomass and/or oleic acid contents is required. In order to determine the optimum dose of gamma-ray irradiation, the rape seeds of cvs. Hanra (Hr), Youngsan (Ys), Tammi (Tm), and Tamra (Tr) were irradiated with a 100~4,000 Gy dose range of gamma-rays. Considering the growth factors, the optimum doses were determined to be within the range of 600~1,000 Gy for the selection of useful mutant lines. Six-hundred and eighty-eight (688) $M_2$ mutant lines were obtained from 600~1,000 Gy gamma-ray-irradiated $M_1$ plants through selfing. The growth characteristics, leaf shape, early flowering, and flower color were all investigated. The selected mutant numbers of early flowering, leaf shape, and flower color were 34, 52, and 3 from the four cultivars, respectively. These mutant lines will be used for the development of a new variety of rape plant with high biomass and oleic acid contents.

Isolation of Differentially Expressed Genes by Low Temperature Treatment in Winter Oilseed Rape (Brassica napus L. cv. Tammi)

  • To investigate flowering related genes in winter-type oilseed rape (Brassica napus L. cv. Tammi), differentially expressed genes were isolated from leaves of the plant after low temperature treatment which is requirements for floral induction. As a result of suppression subtractive hybridization (SSH), 288 clones were randomly selected from SSH library. Using reverse Northern blot analysis, 150 of 288 clones were identified to be differentially expressed. Out of these 150 clones, 45 clones showed very high identities with the known genes. Four clones showed very high identities over 90% with metallothionein-like gene that is related to flowering-induced genes. Of these 4 clones, the cDNA clone, rfs-13, revealed high identity with meotallothionein-like protein in Arabidopsis thaliana (98%) and Brassica compestris (89%). Furthermore, gene expressed in immature flower stages was confirmed by Northern blot analysis.

  • In Korea, Brassica species planted in summer remain in rosette and do not flower. Seed production of vernalized rape seedlings grown in summer was evaluated to establish method of generation shortening and genetically pure seed production without isolation of seed fields. When vernalized rape seedlings of different maturity groups were transplanted in summer, all plants bolted and flowered, and average days from sowing to bolting and flowering was 37 and 58. respectively. It took 85days for early maturity group, 88 days for medium maturity group, and 108 days for late maturity group to mature. Seed yield of vernalized plants grown in summer was 8.8 gr per plant and 53kg per l0a which was 23% of seed yield of rape planted in autumn. However, enough seeds were produced to test oil composition and cake and to plant for a next generation. If seedlings are raised, they can be transplanted in more than 1, 000 times of original seed field, indicating that genetically pure seed can be produced without caging seed fields with nets and isolation of seed fields since other Brassica species do not flower in summer.

  • To know the characteristics of flowering in rape, flower numbers, flowering speed, flowering period, pod numbers, pod formation period, and oil content were examined by individual plant and by each of branches per plant. The results are; 1) plant had in average 1, 400 to 1, 500 flowers, 2) completion of flowering per plant took 30 days, 3) pod numbers per plant was ranged from 560 to 630, 4) plant had 18 to 20 primary branches, 5) five days were needed for pod formation, 6) no difference in flowering speed by the position of branches, 7) flowering period was shortened by 0.5 days per branch by going to upper part, 8) oil content of seeds was significantly higher on the upper and lower part than on the middle part branches.

Molecular Cloning of Two Genes Encoding Cinnamate 4-Hydroxylase (C4H) from Oilseed Rape (Brassica napus)

  • Cinnamate 4-hydroxylase (C4H) is a key enzyme of phenylpropanoid pathway, which synthesizes numerous secondary metabolites to participate in development and adaption. Two C4H isoforms, the 2192-bp BnC4H-1 and 2108-bp BnC4H-2, were cloned from oilseed rape (Brassica napus). They both have two introns and a 1518-bp open reading frame encoding a 505-amino-acid polypeptide. BnC4H-1 is 57.73 kDa with an isoelectric point of 9.11, while 57.75 kDa and 9.13 for BnC4H-2. They share only 80.6% identities on nucleotide level but 96.6% identities and 98.4% positives on protein level. Showing highest homologies to Arabidopsis thaliana C4H, they possess a conserved p450 domain and all P450-featured motifs, and are identical to typical C4Hs at substrate-recognition sites and active site residues. They are most probably associated with endoplasmic reticulum by one or both of the N- and C-terminal transmembrane helices. Phosphorylation may be a necessary post-translational modification. Their secondary structures are dominated by alpha helices and random coils. Most helices locate in the central region, while extended strands mainly distribute before and after this region. Southern blot indicated about 9 or more C4H paralogs in B. napus. In hypocotyl, cotyledon, stem, flower, bud, young- and middle-stage seed, they are co-dominantly expressed. In root and old seed, BnC4H-2 is dominant over BnC4H-1, with a reverse trend in leaf and pericarp. Paralogous C4H numbers in Brassicaceae genomes and possible roles of conserved motifs in 5' UTR and the 2nd intron are discussed.

Production of Haploids from Proton Ion and Gamma-Ray Irradiation Treated $M_2$ Generation of Isolated Microspores in Brassica napus L. ssp. oleifera (앙성자 및 감마선을 처리한 유채 $M_2$ 세대의 소포자로부터 반수체 배발생)

  • This experiment was carried out to investigate the effect of proton ion and gamma-ray irradiation on microspore culture of the flower buds of $M_2$ generation in winter type of Brassica napus L. ssp. oleifera. The seeds of three rape varieties, 'Halla', 'Naehan' and 'Tammi' were pretreated with proton ion and gamma-ray 400 Gy and 600 Gy, respectively. When microspore culture techniques were used, embryogenesis was increased in some varieties by proton ion and gamma-ray irradiation treated flower buds of $M_2$ generation than control. In genotypes 'Naehan' showed the highest embryo production frequency, but 'Tammi' showed lowest embryo production frequency. Some of the embryoids developed directly into plantlets, whereas others developed abnormally multilobe. Plants were regenerated and successfully acclimatized in pots.

Separation of Reducing Sugars from Rape Stalk by Acid Hydrolysis and Fabrication of Fuel Pellets from its Residues (산가수분해한 유채대로부터 유리당의 분리 및 이의 잔사로부터 펠릿의 제조)

  • This study was conducted to identify the potential of rape stalk as a raw material for biorefinery process of rape flower. At first, rape stalk (RS) was immersed in distilled water (DW), acetic acid (AA), oxalic acid (OA), sulfuric acid (SA) and sodium hydroxide (SH) solutions, and the content of reducing sugars liberated from immersed RS was analyzed. Glucose, xylose, arabinose and sucrose were detected varying with the immersion type. In particular, 1% AA-immersion of RS for 72 hr was the most effective conditions to liberate glucose from RS. Secondly, the RS residues were used for elementary analysis and fabrication of fuel pellets. In addition to the solution type, concentration of immersion solutions (0%, 1%, 2%) and immersion time (24, 72, 120 hr) were used as experimental factors. The contents of nitrogen, sulfur and chlorine reduced effectively through the immersion of RS in DW, AA and OA solutions. For properties of RS-based pellets, bulk density and higher heating value of RS-based pellets greatly increased with the immersion of RS, and the qualities were much higher than those of the A-grade pellet of the EN standards. Ash content decreased remarkably through the immersion of RS, and was satisfied with the A-grade pellet standard. Durability was negatively affected by the immersion of RS, and did not reached to B-grade of the EN standard. In conclusion, acid immersion of RS can be a pretreatment method for the production of fuel pellet and bioethanol, but use of the immersed RS for the production of high-quality pellets might be restricted due to low durability of immersed-RS pellets. Therefore, further studies, such as investigation of detailed immersion conditions, fabrication of mixed pellets with wooden materials and addition of binders, are needed to resolve the problems.

Monitoring of Veterinary Medicine Residues in Honey (벌꿀 중 동물용의약품 잔류량 모니터링)

  • This research was carried out to investigate residues of neomycin, streptomycin, dihydrostreptomycin, amitraz, 2,4-dimethylaniline (one of amitraz's metabolites), and coumaphos in honey in order to intensively control their use following the establishment of Korean maximum residue limits (MRLs) for veterinary drugs in honey in 2007. To monitor for residues, 110 honeys and food products with honey were collected and analyzed. The collected honeys included acasia, mixed flower, chestnut, rape flower, jujube, and native types. Neomycin, streptomycin, dihydrostreptomycin, oxytetracycline, and amitraz were not detected among samples. Coumaphos was found in the Korean acasia honey at 0.02 mg/kg, but its concentration was under the MRL (0.1 mg/kg) for coumaphos. According to the results, there were no violations of the Korean MRLs of veterinary drugs in honey.

IPA Study of Landscape Potentiality of Agricultural and Fishery Heritages - A Focus on Cheongsando - (농어업유산의 경관 잠재력 파악을 위한 IPA 연구 - 청산도를 중심으로 -)

  • The purpose of this study is to derive landscape characteristic elements of agricultural and fishery heritage and to classify landscape characteristic elements of Cheongsando designated as Korean agricultural and fishery heritage No.1 as well as a Globally Important Agricultural Heritage System (GIAHS). Additionally, this study was conducted to suggest priority for conservation and management of Cheongsando by grasping differences between factors which visitors deem as important and satisfying. To implement this study, the literature review was written, an on-site survey to derive landscape characteristic elements of Cheongsando and a survey on importance and satisfaction of landscape characteristic elements of Cheongsando were conducted, and reliability analysis, descriptive statistical analysis and IPA analysis were performed using SPSS 20.0. The results are as follows: The analysis showed the factors requiring persistent efforts in the first quadrant are the rape flower garden, the sea, green barley field, flat stone paddy field, breakwater and lighthouse, abalone farms, stone houses, thatched houses, a coastal road, the slow road, Stonewall Walkway of Seongseo Village, and residents' agricultural behaviors. The analysis showed the factors needing intensive management strategies in the second quadrant are the surrounding mountain area, dock, Docheong Harbor, vessels, fish market, Doksari stone wall, garish-roofed farm villages, excursion school to a slow island, pension and cafe, bus stop, shade trees, Raw Fish Street, the beach and the filming site. Analysis indicated that the factors needing management control in the third quadrant are the pine grove, the beach, tidal mudflat, the garlic fields, vinyl greenhouses, grain drying yard, sea mustard drying yard, heritage center, Choboon, Dangri exorcism, the market place, residents' fishery behaviors, residents' industrial behaviors, residents' ordinary behaviors, visitors' behaviors that visiting the dock, visitors' behaviors that walking the slow road, visitors' behaviors that eating and shopping for specialties, visitors' behaviors that experiencing agriculture and fishery. Excessive effort factors in the fourth quadrant were not derived.
