Genetic analysis of rainbow trout populations in hatchery stocks is important in order to develop a strategy for their management. In this study, chromosome number and polymorphisms of diploid and artificially induced triploid rainbow trout are discribed. The relation-ship between chromosomal polymorphism and their economic values for the aquaculture industry are also discussed.
Experiments were carried out to examine early life history, growth and gonadal development of triploid rainbow trout to evaluate their suitability for commercial aquaculture. A high degree ($96.5\%$) of triploid fish was induced when the fertilized eggs were treated with heat shock. Mean embryonic development time of triploids was slightly shorter than diploids and survival rate of triploid at swim-up was $80.2\%$ relative to control. Erythrocytes and their nuclei of triploid were larger in both axes than those of diploid. Triploid rainbow trout showed improved growth relative of diploid after 1 year of age. Histological analysis of the gonads revealed sterility in triploid fish.
Astaxanthin is a valuable pigment source for many aquacultured species, including salmonoids, shrimp, sea bream, and ornamental species. Conjugated linoleic acid (CLA) and ascidian tunic extracts were mixed with the basal diet of rainbow trout to investigate their pigmentation effects. Synthetic Carophyll Pink and natural carotenoids that came from the tunic extracts were incorporated into muscle and skin tissues. The main carotenoids found in muscle after 8 weeks were canthaxanthin in CP12 (13.4%), and CP52 (17.2%), and astaxanthin in CP12 (58.5%), and CP52 (59.2%) in the Carophyll Pink group, while those in skin were canthaxanthin in CP14 (34.5%), and CP54 (29.2%), and astaxanthin in CP14 (32.0%), and CP54 (36.5%) in the ascidian tunic extract group. The total carotenoid content in skin (53.0-69.3 mg/kg) was greater than that in muscle (9.5-13.8 mg/kg).
We investigated the antioxidant and cholinesterase inhibitory activities of the aqueous extract of rainbow trout Oncorhynchus mykiss. The antioxidant activity of O. mykiss aqueous extract was determined by in vitro peroxynitrite scavenging activity and reducing power assays. The aqueous extract of O. mykiss showed potent peroxynitrite radical scavenging activity ($IC_{50}=0.12{\pm}0.001\;mg/mL$) and reducing power (absorbance=$0.47{\pm}0.001$) at the concentration of 1 mg/mL. The in vitro cholinesterase inhibitory activity of O. mykiss aqueous extract was examined using spectrophotometric analyses of acetyl- and butyrylcholinesterase. The aqueous extract of O. mykiss showed acetylcholinesterase inhibitory activity ($IC_{50}=1.61{\pm}0.13\;mg/mL$), but did not exhibit inhibitory activity against butyrylcholinesterase. These results suggest that O. mykiss possesses antioxidant and acetylcholinesterase inhibitory activities and provide scientific evidence for the health benefits of O. mykiss aqueous extract.
Effects of bisphenol-A (BPA) on vitellogenin (VTG) synthesis and estrogen-estrogen receptor $(E_2- ER)$ binding activity were examined in primary hepatocyte cultures of rainbow trout, Oncorhynchus mykiss. Hepatocytes were precultured for 2 days and then $estradiol-17\beta\;(E_2,\;2\times10^{-6}M)\;BPA\;(10^{-5}-10^{-8}M)$ and/or 4-hydroxy-tamoxifen $(4-OHT,\;10^{-6} M)$ were simultaneously added to the incubation medium. Hepatocytes were cultured for 5 more days and then spent medium was analyzed by SDS-PAGE for VTG production. The addition of BPA to the incubation medium had no effect on the viability of hepatocytes in the culture. On the other hand, BPA increased VTG production in a concentration-dependent way and a significant increment occurred at BPA concentrations greater than $10^{-6}$M. Although VTG was increased by the addition of $E_2\;(2\times10^{-6}\;M)\;or\;BPA\;(10^{-5}M)$, its were reduced by a simultaneous 4-OHT $(10^{-6}\;M)$ addition. BPA inhibited $E_2-human$ ER binding activity by $72\%$ at $10^{-5}$ M of BPA. These results suggested that BPA induced VTG synthesis by BPA-ER binding activity in the hepatocyte of rainbow trout.

Hazard Analysis and Critical Control Points (HACCP) is a systematic preventative approach for food safety and is generally used in food industry to identify potential food safety hazards. Recently, the Korean Ministry for Food, Agriculture, Forestry and Fisheries planed to applicate HACCP programs as an effective approach to food safety and protecting public health at the aquaculture farm of rainbow trout, Oncorhynchus mykiss. For the purpose, it need to identify what kinds of hazards are in the aquaculture farm. In the present study, we established Sanitation Standard Operating Procedures (SSOP), which addresses sanitation conditions and practices in the aquaculture farm. SSOP is essential prerequisite programs of HACCP to reduce or eliminate the risk of the hazards being realized. We anticipate that the HACCP programs of aquaculture farm will contribute to supply foods in safety to consumers.

Products from aquaculture have sometimes been focused on the problems caused by the contamination of chemical agents as the use of chemical agents in aquaculture has been annually increased. The risk of contamination of products by chemical agents is greater in freshwater than in seawater. In order to evaluate the food safety of a fish grown in freshwater, we investigated the residues of antibiotics (tetracycline, oxolinic acid and ciplofloxacin) and malachite green in cultured rainbow trout, Oncorhynchus mykiss. Malachite green, which was prohibited in the application of aquaculture, was not detected in samples tested in this study. The residual content of tetracycline was determined to be less than the permissible amount, <0.2 mg/kg. The contents of ciplofloxacin was also less than the permissible amount, <0.1 mg/kg. However, in case of oxolinic acid, one of samples was only exhibited higher content than the permissible amount (<0.1 mg/kg). The results obtained in this study suggested that the control and regulation of chemical agents such as antibiotics was important to maintain a safe and worry-free seafood supply.
This study was examined chromosome structure of kidney, fin, blood cell, sexual organ that were differently examined chromosome of a good cell activity from organ, medium, staining methods, instruments with rainbow trout 100. And so we obtained following conclusion. 1. The difference from each organ and medium is that a good cell activity of fin, kidney, sexual organ were obtained in TC-199 medium and a good cell activity in TC-199 medium+PHA(5%). 2. In the difference by staining methods, G-banding and C-banding were analyzed by electron microscope or cytoscan. Among them, heterochromatin of centromere analyzed by C-banding. 3. The zygotene and the leptotene were analyzed in this study. 4. Karyotype, heterochromatin and each stage of cell division were clearly analyzed by cytoscan.

Effects of Al on vitellogenin (VTG) and VTG mRNA induction by estradiol-17 $\beta$($E_2$) were examined in primary hepatocyte culture of rainbow trout. Hepatocytes were precultured for 2 days and then E2 ($2{\times}10^{-6}$M) and Al ($10^{-6}-10^{-4}$M) were simultaneously added to the incubation medium. Hepatocytes were cultured for 5 more days. Media and hepatocytes were then analyzed by SDS-PAGE and Northern blotting for VTG and VTG mRNA, respectively. These metal had no appreciable effect on the viability of hepatocytes in culture. However, Al interfered with VTG production and VTG mRNA expression. Al reduced VTG production in a concentration-dependent way and a significant reduction accurred at Al concentrations greater than $5{\times}10^{-5}$M. VTG mRNA expression also decreased with a negative correlation with Al concentration (r=-0.98). These results suggest that Al inhibit VTG production at the transcriptional level to reduce VTG mRNA expression.
Pollution caused by fecal solids released from recirculating aquaculture systems (RAS) is a growing global concern requiring immediate attention. Thus, this study investigated the physicochemical characteristics of fecal solid feed from RASs used for eel and rainbow trout farming. The concentrations of proteins, lipids, crude ash, and moisture in eel fecal solids were 10.9%, 1.2%, 85.8%, and 72.2%, respectively, while those in rainbow trout fecal solid feed were 20.5%, 3.2%, 70.9%, and 87.0%, respectively. The control group was fed a mixture of sea cucumber and dried sea mud. The protein digestibilities of eel and rainbow trout fecal solids were 25.43% and 23.96%, respectively, while the respective lipid digestibilities were 35.62% and 36.80%, respectively.
