In vivo와 in vitro에서 김치 및 김치재료의 피브린 분해활성 (The Fibrinolytic Activity of Kimchi and its Ingredients in vivo and in vitro)

  • Fibrionolytic activity(FA) of kimchi in rat and FAs of water and methanol extracts of kimchi ingredient were determined using fibrin plate method to see where the active principles for FA are present in kimchi. Nine Sprague-Dawley per each group were fed diet containing 3, 5 or 10% of freeze dried kimchi for 6 weeks. The FA of plasma obtained from 10% kimchi group showed stronger FA than that of control. FAs of 3 and 5% kimchi group were not noticeable. FA from methanol extract of kimchi was approximately 6 times higher than that from water extract assuming that the active material may be present in a fat soluble fraction. From water extract of kimchi ingredients, dropwort, green onion, radish and baechu showed FA in descending order. And from methanol extracts, red pepper powder, radish, green onion and baechu showed FA in decending order. The Fa of methanol extract of red pepper powder was the most noticeable among samples.

갯무 추출물의 스코폴라민 유도 기억력 저하 모델에서의 뇌신경 보호 효과 (Neuroprotective Effect of Wild Radish Extract on Scopolamine Induced Memory Impairment)

  • Raphanus sativus var. hortensis f. raphanistroides Makino (Korean wild radish [WR]) are root vegetables belonging to the Brassicaceae family. These radish species mostly grow in sea areas in Asia, where they have been traditionally used as a medicinal food to treat various diseases. To investigate the effect of WR on neuronal cell death in SH-SY5Y cells, beta-amyloid was used to develop the cell death model. WR attenuated neuronal cell death in SH-SY5Y and regulated the mitogen-activated protein kinase (MAPK) signaling. WR extract also inhibited acetylcholinesterase inhibitor activity. Additionally, the WR treatment group ameliorated the behavior of the memory-impaired mice in a scopolamine-induced mouse model. In the behavior test, WR treated mice showed shorter escape latency and swimming distance and improved the platform-crossing number and the swimming time within the target quadrant. Furthermore, WR prevented histological loss of neurons in hippocampal CA1 regions induced by scopolamine. This study shows that WR can prevent memory impairment which may be a crucial way for the prevention and treatment of memory dysfunction and neuronal cell death.

Effects of Korean Radish on DSS-Induced Ulcerative Colitis in Mice

  • The present study aimed to investigate the comparative evaluation of pharmacological efficacy between sulfasalazine alone and combination with herbal medicine on dextran sodium sulfate (DSS)-induced UC in mice. Balb/c mice received 5% DSS in drinking water for 7 days to induce colitis. Animals were divided into five groups (n = 9): group I-normal group, group II-DSS control group, group III-DSS + sulfasalazine (30 mg/kg), group IV-DSS + sulfasalazine (60 mg/kg), group V-DSS + sulfasalazine (30 mg/kg) + Radish Extract mixture (30 mg /kg) (SRE). DSS-treated mice developed symptoms similar to those of human UC, such as severe bloody diarrhea and weight loss. SRE supplementation, as well as sulfasalazine, suppressed colonic length and mucosal inflammatory infiltration. In addition, SRE treatment significantly reduced the expression of pro-inflammatory signaling moleculesthrough suppression both mitogen-activated protein kinases(MAPK) and nuclear factor-kappa B ($NF-{\kappa}B$) signaling pathways, and prevented the apoptosis of colon. Moreover, SRE administration significantly led to the up-regulation of anti-oxidant enzyme including SOD and Catalase. This is the first report that Radish extract mixture combined with sulfasalazine protects against experimental UC via the inhibition of both inflammation and apoptosis, very similar to the standard-of-care sulfasalazine.

Effects of White Radish (Raphanus sativus) Enzyme Extract on Hepatotoxicity

  • Raphanus sativus (Cruciferaceae), commonly known as radish is widely available throughout the world. From antiquity it has been used in folk medicine as a natural drug against many toxicants. The present study was designed to evaluate the hepatoprotective activity of radish (Raphanus sativus) enzyme extract (REE) in vitro and in vivo test. The $IC_{50}$ values of REE in human liver derived HepG2 cells was over 5,000 ${\mu}g/ml$ in tested maximum concentration. The effect of REE to protect tacrine-induced cytotoxicity in HepG2 cells was evaluated by MTT assay. REE showed their hepatoprotective activities on tacrine-induced cytotoxicity and the $EC_{50}$ value was 1,250 ${\mu}g/ml$. Silymarin, an antihepatotoxic agent used as a positive control exhibited 59.7% hepatoprotective activity at 100 ${\mu}g/ml$. Moreover, we tested the effect of REE on carbon tetrachloride ($CCl_4$)-induced liver toxicity in rats. REE at dose of 50 and 100 mg/kg and silymarin at dose of 50 mg/kg were orally administered to $CCl_4$-treated rats. The results showed that REE and silymarin significantly reduced the elevated levels of serum enzyme markers induced by $CCl_4$. The biochemical data were supported by evaluation with liver histopathology. These findings suggest that REE, can significantly diminish hepatic damage by toxic agent such as tacrine or $CCl_4$.

강화약쑥 수용성 추출물의 식물 타감효과 및 HPLC에 의한 타감물질 분석 연구 (Allelopathic Effect of Aqueous Extract of Ganghwa Mugwort (Artemisia spp.) Vegetables and HPLC Aanalysis of Allelochemicals)

  • This study was conducted to evaluate the allelopathic effect of aqueous extract of Ganghwa domestic mugwort (Artemisia spp.) on vegetables and its related allelo-chemicals. When the receptor vegetables, such as Chinese cabbage, lettuce, and red radish, were treated with aqueous extract obtained from Sajabalssuk (A. $sp^*I$), Ssajuarissuk (A. $sp^*II$) or Ssajarissuk (A. $sp^*III$), their germination rate, leaf number, plant height, and root length were restricted with increasing concentration of aqueous extract. Allelopathic effect was the highest in radish, than lettuce and Chinese cabbage in order. The growth of topplant were more inhibited then root growth observing in restriction of plant height, root length, and chlorophyll contents. The plant height, the root length of red radish were 53.3 and 61.2% and their fresh weights were 19.8 and 26.4% compared to those of controls, respectively. A. $sp^*III$ showed the highest allelopathic effect among the donor plants. In HPLC analysis, 7 phenol compounds were identified in A. $sp^*I$ and A. $sp^*II$, and, in A. $sp^*III$, and hydroxybenzoic acid and phenylacetic acid were further identified as allelochemicals. It is considered that their plant growths were variously inhibited by the amounts and types of allelochemicals in aqueous extracts. To increase the productivity of farm land after cultivation of mugwort, these results can be useful to select the following field crops.

무청 추출물의 생리활성 (Physiological Activity of Extracts from Radish (Raphanus sativus L.) Leaves)

  • 건조 무청을 물, 50% 에탄올 및 50% 메탄올로 추출한 물질의 생리활성 효과를 조사하였다. 건조 무청의 추출물 수율은 브랜칭하지 않은 건조 무청인 관동 무청(DRLK)은 $26.56{\sim}32.56%$이고, 미농단백 무청(DRLM)은 $34.34{\sim}36.90%$였다. 반면에 브랜칭한 무청군 수율은 $18.70{\sim}23.56%$로 브랜칭하지 않은 시료보다 수율이 낮았다. 총 폴리페놀 함량의 경우 브랜칭한 무청 시료는 $49.06{\sim}59.37mg/g$ 범위였고, 브랜칭을 하지 않은 시료는 $53.41{\sim}62.08mg/g$이었다. 시료에 따라서는 관동 무청시료(DRLK, BDRLK)보다 미농단백무 시료(DRLM, BDRLM)가 약간 더 높은 폴리페놀 함량을 나타내었다. 전자공여능은 브랜칭하지 않은 시료는 80% 이상의 활성을 보였고, 아질산염 소거작용은 $36.63{\sim}51.17%$ 범위의 활성을 보였으며, SOD 유사활성은 $38.53{\sim}45.38%$의범위의 활성을 보인 반면 브랜칭한 시료는 각각의 생리활성 측정에서 브랜칭하지 않은 시료보다 낮은 활성을 보였다. 전반적으로 관동 무청군이 미농단백 무청군보다 전자공여능, 아질산 소거능 및 SOD 유사활성에 있어서 약간 높은 활성을 보였고, 브랜칭하지 않은 무청이 높은 활성을 나타내었다.

무청즙액을 이용한 녹엽단백질과 효모균체의 생산 (Production of Plant Protein Concentrate and Yeast Biomass from Radish Greens)

  • 무청즙액을 식물녹엽단백질의 생산과 효모의 균체생산을 위한 두가지 목적으로 이용하는 연구를 하였다. 무청즙액을 80-10$0^{\circ}C$로 가열하거나 pH를 조정하여 침전시켰을 때 침전물의 양은 모두 무청즙액 1리터당 10.0-16.5g(평균 13.5g)이었고 이 침전물의 조단백질 함량은 25-30%이었다. 단백질을 침전시켜 제거한 여액은 전당의 함량이 1.0%정도 되게 희석하여 효모 Candida utilis를 배양하였을 때 무청 여액원액 1리터당 최고 19.59의 효모균체를 생산할 수 있었다. 따라서 무청즙액 1리터로부터 생산할 수 있는 고단백질자원은 33g이었다.

  • Radish (Raphanus sativus L.) has long been recognized as a source of nutrients and phytochemicals. It is well known that the antioxidant properties of radish contributed to its popularity in pharmaceuticals. For this reason, the radish extract aided in the recovery and prevention from diseases. Hence, in this study, the sensory evaluation assessment was conducted following addition of radish oil to almond milk, oat milk, and soy milk (nondairy products) and Kefir, yogurt, and cow milk (dairy products) at different concentrations (containing 0.5% increments from 0% up to 2%). According to the results obtained in this study, all samples containing 0.5% radish oil scored higher on the sensory evaluation scale than the control. It is also very valuable as it is the first study to investigate sensory evaluation assessment by incorporating radish oil into almond milk, oat milk, and soy milk (nondairy products) and kefir, yogurt, and cow milk (dairy products). Furthermore, the results of this study can be used as a foundation for developing future products using radish oil. Additional research on addition of plant-based essential oil to various foods should be conducted.

Bioactivities of Sulfur Compounds in Cruciferous Vegetables

  • Cruciferous vegetables are rich in organosulfur compounds such as isothiocyanates and sulfides. While the isothiocyanates, corresponding to pungent principle, are generated from myrosinase-catalyzed hydrolysis of glucosinolates, the sulfides can be generated non-enzymatically. Recent studies provide evidences that some sulfur compounds in these vegetables show a chemopreventive action against carcinogenesis; while isothiocyanates such as sulforaphane induce phase 2 enzymes (glutathione S-transferase/quinone reductase), disulfides tends to elevate the level of phase 1 and 2 enzymes. Especially, sulforaphane rich in Cruciferae vegetables has been reported to express anticarcinogenic effect in some organs such as liver, kidney or intestine. When the level of sulfur compounds in Cruciferous and Alliaceous vegetables was determined by GC/MS (SIM), the richest in sulforaphane is broccoli followed by turnip, cabbage, radish, kale, cauliflower and Chinese cabbage. Meanwhile, the sulfides are predominant in Alliaceous vegetables such as onion. In related study, the administration of vegetable extract elevated the GST level by 1.5 fold for broccoli, 1.4 fold for radish, and 1.3 for onion. Thus, the vegetables frequently used in Korean dish contain relatively high amount of anticarcinogenic sulfur compounds. Moreover, the combination of broccoli and radish extracts elevated the GST induction up to 1.84 folds of control. In addition, the Kakdugi, fermented radish Kimchi was observed to show a comparable GST induction despite the decomposition of methylthio-3-butenylisothiocyanate (MTBI). Therefore, the combination of vegetables including broccoli, and fermented radish Kimchi would be useful as a functional food for chemoprevention.

미생물 생육에 대한 무 추출물의 억제 및 촉진 효과

  • Extracts active on the growth of food-born microorganisms were prepared from radish by using acetone, methanol and water as the solvents. Strains of bacteria and yeast, such as L. brevis, L. fermentum, L. faecalis, L. mesenteroides, B. subtilis, S. flexneri, S. aureus and C. albicans. Addition of 1 $m\ell$ of acetone extract exhibited the strongest growth effect on L. brevis. The water extracts turned out to stimulate growth of all Lactic acid bacteria. Especially, it was observed that the same material produced the highest stimulatory effect(by about 40-50 times) on L. faecalis and L. fermentum and L. mesenteroides. Also, it was proved to have a weak stimulate effect on C. albicans, S. flexneri and S. aureus. But, on the contrary to this, when applied to B. subtilis it showed growth stimulation by about 10 times.
