Optimization of Hydroxyl Radical Scavenging Activity of Exopolysaccharides from Inonotus obliquus in Submerged Fermentation Using Response Surface Methodology

  • The objectives of this study were to investigate the effect of fermentation medium on the hydroxyl radical scavenging activity of exopolysaccharides from Inonotus obliquus by response surface methodology (RSM). A two-level fractional factorial design was used to evaluate the effect of different components of the medium. Corn flour, peptone, and $KH_2PO_4$ were important factors significantly affecting hydroxyl radical scavenging activity. These selected variables were subsequently optimized using path of steepest ascent (descent), a central composite design, and response surface analysis. The optimal medium composition was (% w/v): corn flour 5.30, peptone 0.32, $KH_2PO_4$ 0.26, $MgSO_4$ 0.02, and $CaCl_2$ 0.01. Under the optimal condition, the hydroxyl radical scavenging rate (49.4%) was much higher than that using either basal fermentation medium (10.2%) and single variable optimization of fermentation medium (35.5%). The main monosaccharides components of the RSM optimized polysaccharides are rhamnose, arabinose, xylose, mannose, glucose, and galactose with molar proportion at 1.45%, 3.63%, 2.17%, 15.94%, 50.00%, and 26.81%.

Roasting and Cryogenic Grinding Enhance the Antioxidant Property of Sword Beans (Canavalia gladiata)

  • The objective of this study was to optimize the conditions for enhancing the antioxidant properties of sword bean (Canavalia gladiata) as a coffee substitute in two processing methods, roasting and grinding. The optimum conditions for removing off-flavor of the bean and maximizing functionality and efficiency were light roasting and cryogenic grinding (< 53 ㎛). In these conditions, extraction yield was 16.75%, total phenolic content (TPC) was 69.82 ± 0.35 mg gallic acid equivalents/g, and total flavonoid content (TFC) was 168.81 ± 1.64 mg quercetin equivalents/100 g. The antioxidant properties were 77.58 ± 0.27% for DPPH radical scavenging activity and 58.02 ± 0.76 mg Trolox equivalents/g for ABTS radical scavenging activity. The values for TFC and ABTS radical scavenging activity were significantly higher (p < 0.05) than in other conditions, and TPC and DPPH radical scavenging activity were second highest in lightly roasted beans, following raw beans. HS-SPME/GC-MS analysis confirmed that the amino acids and carbohydrates, which are the main components of sword bean, were condensed into other volatile flavor compounds, such as derivatives of furan, pyrazine, and pyrrole during roasting. Roasted and cryogenically ground (cryo-ground) sword beans showed higher functionality in terms of TFC, DPPH, and ABTS radical scavenging activities compared to those of coffee. Overall results showed that light roasting and cryogenic grinding are the most suitable processing conditions for enhancing the bioactivity of sword beans.

흑양파로부터 항산화 활성 물질인 3,4-Dihydroxybenzoic acid의 분리 및 동정 (Isolation and Identification of Antioxidative Compounds 3,4-Dihydroxybenzoic acid from Black Onion)

  • 양파의 매운 맛과 냄새를 감소시켜 섭취가 용이하도록 제조된 흑양파가 항산화 활성이 있다고 보고되어있어서 흑양파로부터 항산화 활성물질을 구명하고자 하였다. 흑양파 MeOH 추출물을 용매분획하여 얻어진 분획물들 중 EtOAc층이 높은 DPPH와 $ABTS^+$ radical scavenging 활성이 있었다. 그래서 EtOAc층을 silica gel과 Sephadex LH-20 등의 column chromatography를 이용하여 2종의 항산화 활성물질을 분리하였다. 단리한 이들 화합물을 대상으로 ESI-MS 및 NMR 분석을 통하여 3,4-dihydroxybenzoic acid (1)와 quercetin (2)로 각각 동정하였다. 이 화합물들은 $ABTS^+$ 및 DPPH radical scavenging 활성이 있었으며, 화합물 2는 화합물 1에 비해 더 높은 radical scavenging 활성이 있었다.

붉나무(Rhus chinensis Mill) 에탄올 추출물의 항균 및 항산화 활성에 대한 연구 (A Study on Antimicrobial and Antioxidant Activity of Ethanol Extract of Rhus chinensis Mill)

  • In this study, the Rhus chinensis Mill was divided into bark and inner bark. The antimicrobial activity and the antioxidative activity of the extracts were investigated by using the organic solvent fractions after the extraction and concentration with ethanol. This study showed the possibility of functional materials such as raw materials for cosmetics and food supplements. This study was the antimicrobial activity and antioxidant activity of five microorganisms (S. aureus, E. coli, B. subtilis, P. aeruginosa, and P. ovale). The following conclusions were obtained. First, the antimicrobial activity of B. subtilis was found to be high in the ethanol extract of. Second, DPPH scavenging activity was 86.4% free radical scavenging activity at 2.5 mg/mL bark part and 61.9% free radical scavenging activity at 2.5 mg/mL in inner Rhus chinensis Mill bark part. The ABTS scavenging activity was 79.2% free radical scavenging at 1 mg/mL bark fraction and 63% free radical scavenging activity at 1 mg/mL in inner bark, and bark showed higher antioxidant activity than inner bark. These results suggested that the antimicrobial activity and antioxidant activity of Rhus chinensis Mill extract can be used as a natural material. Specific and diverse physiological activity studies are expected in the future.

상지(桑枝)와 상백피(桑白皮)의 항산화효과 비교 연구 (Comparative Study on Antioxidative Effects of Mori Ramulus and Mori Cortex)

  • In recent year, We are concerned about anti-aging, disease-prevention, longevity, so many methods are used in solving this problem. And Those are related with antioxidative ability. Recently, We heard that Mori Cortex was known to reduce the hypertension and was helpful in promoting health, and Mori Ramulus was effective against obesity, etc. So, This study was performed to investigate the antioxidative effect of hot-water extracts of Mori Cortex and Mori Ramulus used for 3 methods, those are DPPH radical scavenging activity, Nitric oxide(NO) radical scavenging activity and Bovine serum albumin(BSA). And we compared Mori Cortex and Mori Ramulus on Antioxidative Effects. The results of this study were as follows: We measured levels of DPPH radical scavenging activity and Nitric oxide(NO) radical scavenging activity. And we obtained results that Mori Ramulus was most effective with the concentration of 5 $mg/m{\ell}$, and Mori Cortex was most effective with the concentration of 2.5 $mg/m{\ell}$, And we examined the antioxidative effects of Mori Ramulus and Mori Cortex with $CU^{2+}/H_20_2$-induced Bovine serum albumin(BSA). And we obtained that antioxidative ability was increased after 1.25 $mg/m{\ell}$ and that was most effective with the concentration of 5 $mg/m{\ell}$ on both of them. And antioxidative ability of Mori Cortex was better than Mori Ramulus(p<0.05). So I guess that hot-water extracts of Mori Ramulus and Mori Cortex have effects on antioxidative ability, but Mori Cortex is better than Mori Ramulus on antioxidation. Hereafter we need differential experimental methods of antioxidative effect on both of them.

Phytotoxic Effect, DPPH Radical Scavenging Activity and Chlorogenic Acid Level of Methanol Extracts from Aerial Parts of Several Korean Salad Plants

  • A series of aqueous or methanol extracts from four different Korean salad plants were assayed to determine their allelopathy and antioxidant activity. The extracts applied on filter paper in a Petri-dish bioassay significantly inhibited root growth of against alfalfa (Medicago sativa) seedlings. Leaf extracts from 40 g dry tissue $L^{-1}$ of Aster yomena was most phytotoxic to alfalfa root growth, and followed by that of Cirsium japonicum, Taraxacum officinale, and Ixeris dentate. Methanol extracts of plants dose-dependently increased DPPH free radical scavenging activity in vitro. Antioxidant activity of methanol extracts from the same plant species was investigated, and the result showed high DPPH free radical scavenging activity in Cirsium japonicum, Aster yomena, and Ixeris dentate, however, in Taraxacum officinale was least activity. By means of HPLC analysis, chlorogenic acid, p-coumaric acid, and total phenolics with 7.68, 17.47 and 18.64 mg, $100g^{-1}$, respectively, showed the highest amounts in methanol extracts from Cirsium japonicum leaves. These results suggest that Compositae salad plants contain water-soluble substances with allelopathic potential as well as antioxidant activity.

Bioactivity of Trifoliate Orange (Poncirus trifoliate) Seed Extracts

  • Trifoliate orange seed extracts (TSEs) were made using either distilled water (TW), ethanol (TE), or n-hexane (TH), to measure total polyphenol contents, DPPH and ABTS radical scavenging activities, and anti-complementary activity. The results showed that the total polyphenol content showed higher value at TE (235.24 ${\mu}g/mL$, p<0.05) than those of TW (132.65 ${\mu}g/mL$) and TH (165.44 ${\mu}g/mL$) at 10 mg/mL and TE exerted the highest DPPH radical scavenging activity (61.77%, p<0.05), which occurred in the following order: TE TW (56.87%)>TH (39.78%). The results of ABTS radical scavenging activity showed that TW (34.26%) and TE (31.81%) showed similar activities, which were higher than TH (12.74%, p<0.05). Anti-complementary activity of TE (61% at 500 ${\mu}g/mL$) showed a higher activity when compared with the positive control (60% at 1,000 ${\mu}g/mL$) polysaccharide-K (PSK), a known immuno-active polysaccharide from Coriolus versicolor. Consequently, among TSEs, TE is a byproduct from trifoliate orange and could be an important source of dietary polyphenolic antioxidant compounds and immunopotentiating activity, including complement activation.

Radical Scavenging Activities of Korean Traditional Rice Wine, Takju

  • The principal objective of this study was to assess the radical scavenging activities and total polyphenols, flavonoids, and flavonol contents of takju, a Korean traditional rice wine. The antioxidant properties of the wine and takju samples were evaluated using five distinct assays: specifically, 1,1-diphenyl-2-picrylhydrazyl (DPPH), 2,2-azino-di-(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS); hydroxyl; superoxide anion; and nitric oxide (NO) radical-scavenging activity assays. In this study, the takju evidenced strong scavenging activities against the hydroxyl, superoxide anion, and NO radicals. Furthermore, the total polyphenol contents of T-2 were similar to those previously observed in white wine (p<0.05). However, the flavonoids and flavonol contents of all takju samples were significantly (p<0.05) lower than that of white wine. The results of this study show that takju possesses a powerful radical scavenging activity against a variety of oxidative systems. The findings of this study also indicate that takju should be considered a useful antioxidant, and that their functional compound reduces oxidative stress.

Free Radical Scavenging Activity of Enzymatic Hydrolyzates of Hot Water Extract from the Shell of Reeve's Turtle (Chinemys reevesii)

  • The shell of Reeve's turtle has been used as a traditional folk medicine in Korea. We produced a hot water extract from Reeve's turtle shell according to the traditional medical practice. To release bioactive peptides, the hot water extract was enzymatically hydrolyzed with various proteases, and the free radical scavenging activity of the hydrolysate was investigated against 1,1-diphenyl-2-picryl-hydrazyl (DPPH), hydroxyl and peroxyl radicals. The free radical scavenging activity of the enzymatic hydrolysates varied from 1 to 79% depending on the enzymes, free radical species, and concentration. The $EC_{50}$ values demonstrated that the enzymatic hydrolysates of hot water extract from the shell of Reeve's turtle are potential antioxidants.

Screening for Antioxidative Activity of Pueraria Radix

  • The antioxidant activity of solvent extracts of leaves, trunk and root Pueraria Radix was determined by measuring the radical scavenging effect on l.l-diphenyl-2-picrylny drazyl(DPPH) radical. Extacts prepared from root showed radical scavenging effect on DPPH radical. But, extracts prepared from leaves and trunk did not show activity. The ethyl acetate ewtract of Pueraria Radix root showed radical scavenging activity at an IC 50 value of 75.7 $\mu\textrm{g}$/$m\ell$.
