Evaluation of Nutrients during Rack and Bag Culture or Suspended Culture of Pacific Oyster Crassostrea gigas (부유망식과 수하식 양성방법에 따른 참굴(Crassostrea gigas)의 영양상태)

  • The mineral contents, and free amino acid, and fatty acid composition of rack and bag or suspended cultured oysters Crassostrea gigas were compared. The moisture content did not change much from 82.8% to 86.3% in the different culture methods. The glycogen level was low 4.7% during rack and bag culture but high 7.2% during suspended culture. The sodium level was the highest during suspended culture, while copper and zinc level were highest during rack and bag culture. The taurine level was 1,385-1,375 mg/100 g in oyster cultured using the rack and bag method, but that in the market sample was lowest at 1,015.8 mg/100 g. Hydroxyproline, glutamic acid, proline, glycine, and alanine were detected at high levels in the experimental oysters. The polyunsaturated fatty acid content was similar among samples. The docosahexaenoic acid level was 14% with rack and bag culture and 12% with suspended culture, whereas the eicosapentaenoic acid level was 15% with rack and bag culture and 20% with suspended culture. The atherogenic index (AI), thrombogenicity index (TI), and hypocholesterolemic/ hypercholesterolemic (h/H) ratio were important factors in the nutritional evaluation. The AI and TI values were 0.5-0.2, and the h/H ratio were 2.0 and 2.4 with the rack and bag and suspended culture, respectively.

Study on Optimal Condition for Oyster Rack Culture in terms of tidal exposure and rack height in Wando Coast, Korea

  • We investigated the growth performance of oysters (initial shell height $57.5{\pm}8.5$ mm) under differing conditions of tidal exposure time and culture rack height in an experiment that commenced in April, 2011. Significant differences were observed in shell height from June 2011, in total weight from August, and in meat weight from September. Fatness tended to decrease during the experimental period, but was not significantly different at the end of the experiment. Significant differences in survival rates were mainly observed from June to August. After September, further changes were not observed in any experimental treatment group. The greatest growth potential ($L_{\infty}$) and survival rate were observed at a sea level of approximately 116 cm. The results indicate that in the study area the use of oyster culture conditions involving 1 or 2 h of tidal exposure and 60 - 70 cm rack height could result in oysters reaching the favored commercial half shell size within 14 months, with > 80% survival.

Study on natural short-necked clam, Ruditapes philippinarum, stocks in Yeongheung coast of Wando Island, Korea (완도 영흥지선 연안의 천연 바지락 자원에 대한 연구)

  • In order to understand the effect of introduction of oyster rack culture on natural short-necked clam, Ruditapes philippinarum, stock, we investigated the status of natural clam stock prior to introduction of oyster rack culture at Yeongheung Coast of Wando Island. The growth equation of the clam was estimated as: $L_t=61.46{\cdot}(1-e^{-0.172(t+0.155)})$ from ring radius composition of shell. Instantaneous coefficient of total mortality and natural mortality were calculated as: 2.4087/year and 0.478/year, respectively. The age at first capture was estimated to be 2.55 year. The total biomass was estimated to 3.23 ton in the bed (0.8 ha). Applied by these parameters, the annual recruit biomass and the current yield per recruit (Y/R) was corresponded to $114.7individuals/m^2$ and $92.0g/m^2$, respectively. Maximum sustainable yield (MSY) was estimated 0.77 ton/year which was close to annual catches, 0.74 ton/year, in the area.

Freshness and Antioxidant Activities in Pacific Oyster Crassostrea gigas Using Rack-and-Bag Culture or Suspended Culture Methods (부유망식과 수하식 양성방법에 따른 참굴(Crassostrea gigas)의 선도와 항산화활성)

  • The nucleotides and their related compounds, including ATP (adenosine triphosphate), ADP (adenosine diphosphate), AMP (adenosine monophosphate), IMP (inosine monophosphate), HxR (inosine) and Hx (hypoxanthine), were nearly identical in oysters Crassostrea gigas from the two culture methods. The K-value was lower than the threshold value such as 11.2-12.1. Although oysters have low amount of IMP, it was detected in this experiment. DPPH radical scavenging activity did not vary significantly with sample amounts (100, 300, and $500{\mu}g/mL$). DPPH (1,1-diphenyl-2-picryl-hydrazyl) radical scavenging activity was 76.0-80.7% compare with the ascorbic acid standard. Superoxide anion scavenging activity reached 49.3% in the rack-and-bag culture sample at $500{\mu}g/mL$. However, the reducing power and $Fe^{2+}$ chelating activity were very low compared with their respective standard. The oyster culture methods did not affect oyster quality in terms of antioxidant activities.

Estimation of primary production of the waters around rack oyster farm at Wando, Korea

  • To establish a comprehensive management strategy, as part of the optimization of cultural practice for an oyster rack culture system, we used a numerical model to estimate the primary production in the waters on the eastern coast of Wando island, South Korea. The estimated primary production ranged from 17.12 to $1052.55mgC\;m^{-2}day^{-1}$ ($204.22{\pm}224.75mgC\;m^{-2}day^{-1}$ in average). Except for the times of peak phytoplankton blooms, the estimated primary production (PP) was consistently under $200mgC\;m^{-2}day^{-1}$, which is more similar to the value of PP measured off the western coast of South Korea than the southern coast. No clear relationship was observed between nitrogen content and rainfall with the exception of heavy rainfall events, indicating that precipitation might not be the main source of nutrients in these waters. No clear influence was observed from Doam tidal discharge, located 24 km north from these waters due to main tide comes in this area from the channel between Gunwe-myeon in Wando island and Pukpyeong-myeon in Haenam-gun. Because of the shallow water depth and strong tidal current, resuspension of sediments, which causes an input of nitrogen into the system, could be easily caused by even mild wind and the infrequent passing of ships. Microscopic examination of the phytoplankton composition showed additional contribution of benthic species such as Paralia sulcata into the waters, which increase the productivity of oyster farms in the waters. The availability of nitrate and phosphate for primary production was temporarily limited throughout most of the spring and autumn blooming season.

Sanitary assessment of the oyster rack culture waters in Wando, Korea (완도군 군외면 연안 수평망식 참굴 (Crassostrea gigas) 양식해역의 세균학적 수질 평가)

  • To introduce an oyster rack culture in tidal flat of Wando-gun, Korea, we investigated sanitary conditions from 2012 April to 2013 March in oyster growing waters and adjacent area in Wando. Average seawater temperature and salinity ranged $8.6-28.0^{\circ}C$ and 26.2-33.6 psu, respectively. The coliform group and fecal coliform of seawater ranged < 1.8-13.0 MPN/100 mL each. Only one of 60 samples was exceeded in the European criteria of fecal coliform. Influence of adjunct basin's pollution load on cultural waters was negligible, which half radius of dilution was not above 300 m. According to our observation, however, landing of oyster should be banned for at least 5 days after the heavy rainfall. Our findings meet the sanitary seawater criteria for domestic use and export of shellfish and classified as 'Class A' in European Community regulations for bivalve culture.

Saikosaponin Contents in Bupleurum falcatum Root Produced by Tissue Culture (조직배양(組織培養)한 시호근(柴胡根)의 Saikosaponin 함량(含量))

  • The study for the quantitative analysis of saikosaponin of Bupleurum falcatum root produced by tissue culture was attempted. The optimal concentration of 2,4-D was 0.1 mg/l for inducing the callus. The induction and growth of the adventitious root from the callus was obtained in the suspension culture containing MS basal medium supplemented with no plant growth regulators. The results of the quantitative analysis of saikosaponins by high performance liquid chromatography (HPLC) showed that the content of saikosaponin a and c was high in the one-year-old root from somatic embryos and that of saikosaponin d was remarkably high in three-months-old adventitious root from callus.

Comparisons of Growth and Mortality of the tidal flat Oyster Crassostrea gigas by the Net Bag Rack Culture System in Two Districts in Western Korea (한국 서해안 2개 갯벌지역 수평망식 굴, Crassostrea gigas의 성장과 폐사 비교 연구)

  • We investigated environmental characteristics of the tidal flat oyster aquafarms to clarifyeffects of the tidal flat environmental factors on growth and mortality of the tidal flat oyster Crassostrea gigas by the Net Bag Rack Culture System in two districts in Western Korea. In this study, we have carried out the basic environmental investigation on growth of the single tidal flat, water quality and sediments, etc. in Taean and Seosan districts, Choongcheongnam-do, where the single tidal flat oyster aquaculture have been performed by the net bag rack culture system. In June 2011 when the final survey carried out at the two districts in western Korea, the mortality in Seosan district was higher than that in Tae-an district. Judging from the results of growth and mortality of the single tidal flat oysters investigated at two sites of Jinsan-ri in Taean and Chang-ri in Seosan districts, two results of Taean district showed higher growth and lower mortality than those of Seosan district. It is assumed that the proper acceptable density possible for growth of the tidal flat seeds and the secure of economics, in case of plastic cultivate net with the size of $50{\times}80cm$, are about 200 individuals. In this study, It was clarified that selection of the suitable sites, the input into the net bag of high quality oyster seeds and selective dispersion in the optimal density of the single tidal flat oyster have an effect on growth and mortality of the tidal flat oyster.

  • PMO-P4, being cultivated as "Sanghwang" in Korea, was proved to be P. baumii based on ITS (internal transcribed spacer) sequencing and RFLP (Restriction Fragment Length Polymorphism) patterns along with some Phellinus species including P. linteus. The similaraty of ITS sequencing between PMO-P4 and other Phellinus species was given the range of 48.6%~72.2%, showing the highest homology from P. linteus and the lowest from P. gilvus.

Effects of PCB Congeners in Rodent Neuronal Cells in Culture

  • We attempted to analyze the mechanism of polychlorinated biphenyl (PCB)-induced neurotoxicity and identify the target molecules in the neuronal cells for PCBs.Since the developing neuron is particularly sensitive to PCB-induced neurotoxicity, we isolated cerebellar granule cells derived from 7-day old Sprague Dawley (SD) rats and grew cells in culture for additional 7 days to mimic PND-14 conditions. Only non-coplanar PCBs at a high dose showed a significant increase of total protein kinase C (PKC) activity at phobol 12,13-dibutyrate ([$^3M$]PDBu) binding assay, indicating that non-coplanar PCBs are more neuroactive than coplanar PCBs in neuronal cells. PKC isozymes were immunoblotted with the selected monoclonal antibodies. PKC-${\alpha}$, ${\delta}$, and ε were activated with non-coplanar PCB exposure. Receptor for activated C kinase-1 (RACK-1), anchoring protein for activated PKC, was more induced with exposure to coplanar PCBs than non-coplanar PCBs. Reverse transcription PCR (RT-PCR) analysis showed induction of neurogranin (RC-3) and growth associated protein-43 (GAP-43) mRNA with non-coplanar PCBs. The results indicate that these factors may be useful biomarkers for differentiating non-coplanar PCBs from coplanar PCBs. The present study demonstrated that non-coplanar PCBs are more neuroactive congeners than coplanar PCBs.
