Rabies causes the highest mortality of all viral diseases in the world unless the victim has been protected either by active immunization or post-exposure immunoprophylaxis. Infected stray dogs, raccoons, skunks, foxes and bats are the demonstrated carriers of most cases of rabies. It is difficult to diagnose a rabid animal in the field unless characteristic clinical signs are evident. However, this study used a commercial fast check kit comprised of immunochromatographic test (ICT) strips (ICTS) to diagnose rabies infection in clinically suspected samples obtained from a wildebeest. A 10-year old male wildebeest (approximate weight, 150 kg) died at Bangabandhu Sheikh Mujib Safari (BSMS) Park, Cox's Bazar, Bangladesh with a clinical history of severe excitation and abundant oral secretions. A gross pathological examination revealed no specific lesions indicating any fatal diseases. The entire brain was collected within 6 hours of death, and the brain sample was tested using the ICT strips following the manufacturer's directions. The rabies viral antibody was detected within the brain stem and medulla of the brain tissue of the dead wildebeest.
This study was conducted to investigate the prevalence rate for rabies antibody (PRRA) of dogs near the Pukhansan National Park and in some other districts in Seoul Metropolitan city. From April to July 2000, a total of 414 serum samples were taken from dogs for breeding (92), pet dogs (162), and unclaimed/stray dogs (162). Rabies virus antibodies were detected by neutralizing peroxidase-linked assay (NPLA). Of 414 sera tested, 145 (35%) were positive to rabies virus antibody. PRRA in Pukhansan National Park area and in the other districts of Seoul city were 34.8% and 35.4%, respectively. There was no significant difference in the prevalence rate between these two districts. PRRA in pet dogs, unclaimed/stray dogs, and dogs for breeding were 39.5%, 35%, and 27.2% respectively. PRRA in dogs from residential areas, apartments, animal hospitals, and farms were 32.5%, 60%, 35.3%, and 26.7% respectively. Especially, the dogs reared in apartments had a significantly higher seroprevalence (60%) than those in residential or farm areas. PRRA in less than 1 year, 1~<2 years, 2~<3 years, and over 3 years old dogs were 14.7%, 40.4%, 38.4%, and 53% respectively, and overall 35% in the dog population. In addition, we found that dogs less than 1 year of age had lower antibody prevalence than those over 1 year of age. It was concluded that enhancement of vaccination is important in the prevention of the rabies, and that rabies vaccines should not be less supplied than the population of the dog.
During the period of 2 years from 1993 to 1994, epidemiological investigation were carried out each time the outbreak of rabies suspected cases were informed and all specimens were transported to the Kangwon-do Veterinary Service Lab. At the Lab., autopsy and laboratory examinations were conducted as well. The results obtained as follows 1. The dogs and cattles were confirmed to be infected with rabies virus by routine histopathological findings, indirect flulrescent antibody technique, and mouth inoculation test. 2. The total numbers of rabies occured in dogs and cattles were 9, 5 heads, respectively. The regional distribution of total of 14 heads were 9 heads in Chulwon, 2 heads in Hwachun, 1 head Yangku, 2 heads in Inje and the highest number of outbreaks were orrured in Chulwon. 3. Most of dogs showed furious type and dumb type found in only one dog. 4. Cattles showed the symtoms of uneasiness feeling, excitement, disturbance howling, the change of voice, sexual accelaration, foreign body swatting and attacked to human and ani-mal and found to be died after two or four days. 5. In the seasonal outbreaks of rabies, 8 heads of the highest incidence were occured in win-ter followed by 4 heads in autumn, 1 head in spring and 1 head in summer. 6. The first outbreaks of rabies was reported in Chulwon and then transmitted into Hwachun, Yangku and Inje in turn. 7 The infectious source was supposed to be wild animals such as laccon dog, badger etc. living around the D. Objectives: The aim of this study was to predict rabies protective behaviors (RPB) based on protection motivation theory (PMT) among fourth-grade students at schools in Chonburi Province, Thailand. Methods: This cross-sectional study was conducted from December 2020 to February 2021. A multistage sampling technique was used for sample selection. The questionnaire was divided into socio-demographic data and questions related to PMT and RPB. Descriptive statistical analysis was conducted using the EpiData program and inferential statistics, and the results were tested using the partial least squares model with a significance level of less than 5%. Results: In total, 287 subjects were included, of whom 62.4% were girls and 40.4% reported that YouTube was their favorite media platform. Most participants had good perceived vulnerability, response efficacy, and self efficacy levels related to rabies (43.9, 68.6, and 73.2%, respectively). However, 54.5% had only fair perceived severity levels related to rabies. Significant positive correlations were found between RPB and the PMT constructs related to rabies (β, 0.298; p<0.001), and the school variable (S4) was also a predictor of RPB (β, -0.228; p<0.001). Among the PMT constructs, self efficacy was the strongest predictor of RPB (β, 0.741; p<0.001). Conclusions: PMT is a useful framework for predicting RPB. Future RPB or prevention/protection intervention studies based on PMT should focus on improving self efficacy and response efficacy, with a particular focus on teaching students not to intervene with fighting animals. The most influential PMT constructs can be used for designing tools and implementing and evaluating future educational interventions to prevent rabies in children.
Importance: Rabies is a neglected tropical viral disease most often transmitted through the bite of an infected animal. Objective: This study assessed the level of knowledge, attitudes, and practices of the Shone Ttown community toward rabies. Methods: A survey-based cross-sectional study was conducted in Shone town, Ethiopia, from November 2022 to April 2023. Woreda was selected purposefully, while Kebeles and the study populations were selected by simple random sampling. Four hundred and sixteen respondents were interviewed using a semi-structured questionnaire. Results: All respondents had heard about rabies from different sources, with the majority hearing from informal sources (62%). Approximately 51.9%, 0.7%, and 47.4% of individuals were aware of saliva contact, rabid animal bites, and both as means of transmission, respectively. The survey showed that 64.4% of participants knew the 100% fatal nature of rabies once the clinical signs developed, and 35.6% did not. Approximately 51.4% of respondents agreed that killing stray dogs was an effective method for rabies prevention. In this study, 72.6% of the respondents had contact with pets, and 36.8% of the interviewees had vaccinated their dogs. Only the educational level (p = 0.03) was associated with knowledge of the transmission route. Age (p = 0.04) and educational level (p = 0.01) had a statistically significant association with knowledge of the risk of not vaccinating dogs. Conclusions and Relevance: A lack of formal education in the communities, low levels of education, and the majority of respondents acquiring their knowledge from unofficial sources are important contributors to the low levels of awareness.
Rabies virus (family Rhabdoviridae, genus Lyssavirus, RV) is the causative agent of rabies in mammals. We conducted a sero-epidemiological survey for RV using sera from South Korean stray and companion dogs in the present study. A total of 533 canine serum samples were collected between February 2006 and December 2007 and were screened for rabies immunity with a neutralizing peroxidase linked assay. Both companion (49.1%) and stray (60.1%) dogs demonstrated RV seropositivity. Regional RV antibody prevalence was measured in the Jeju (87.5%), Gyeonggi (62%), Gyeongsang (59.1%), Jeonra (42%), Chungcheong (37.9%), and Gangwon (30.4%) provinces. Prevalence increased with age but did not exceed 80% in any age group. Stray and companion dogs had RV antibody prevalence values of 26.7% and 23.7%, respectively. Seroprevalence was significantly associated with age $({\chi}2\;=\;9.46;\;p\;=\;0.024)$ for companion dogs, although this association was not evident in stray dogs. There were no significant differences in age between stray and companion dogs and no gender differences in RV seroprevalence. Our results suggested that a widespread and reinforced vaccination program must be applied to Korean dogs.
In June, 1957, a rabid wolf near Chinju, Korea attacked and bit 18 persons, and the resulting epidemiologic observations are summarized as follows: (1) Five of 18 persons bitten died of rabies. (2) Ten persons were exposed by bites on the head, of which 5 received Pasteur treatment, and 5 did not. The rabies mortality ratios for these 2 groups were 5:3, and 5:1 respectively. (3) Of 8 persons bitten on the extremities, only one of 5 not receiving Pasteur treatment died of rabies. (4) The incubation period lasted 13 to 17 days, and the clinical course 1 to 2 days.
The combination of rabies immunoglobulin (RIG) with a vaccine is currently effective against rabies infections, but improvements are needed. Genetic engineering antibody technology is an attractive approach for developing novel antibodies to replace RIG. In our previous study, a single-chain variable fragment, scFv57R, against rabies virus glycoprotein was constructed. However, its inherent weak stability and short half-life compared with the parent RIG may limit its diagnostic and therapeutic application. Therefore, an acidic tail of synuclein (ATS) derived from the C-terminal acidic tail of human alpha-synuclein protein was fused to the C-terminus of scFv57R in order to help it resist adverse stress and improve the stability and half-life. The tail showed no apparent effect on the preparation procedure and affinity of the protein, nor did it change the neutralizing potency in vitro. In the ELISA test of molecular stability, the ATS fusion form of the protein, scFv57R-ATS, showed an increase in thermal stability and longer half-life in serum than scFv57R. The protection against fatal rabies virus challenge improved after fusing the tail to the scFv, which may be attributed to the improved stability. Thus, the ATS fusion approach presented here is easily implemented and can be used as a new strategy to improve the stability and half-life of engineered antibody proteins for practical applications.
목 적 : 본 연구는 국립중앙의료원 단일기관에서 경험한 공수병 예방접종 및 교상 후 조치 현황에 대하여 기술하고 최근의 국외 및 국내 공수병 예방조치 지침과 비교하여 적절한 교상 후 조치가 이루어지고 있는지를 알아보았다. 방 법 : 2006년 11월부터 2011년 12월까지 공수병 예방접종 및 교상 후 조치를 위해 국립중앙의료원에 내원한 만 18세 이하 41명을 대상으로 하여 의무기록을 검토하여 나이, 성별, 상처부위, 물린 동물 종류, 물린 지역, 발생일로부터 내원일까지의 기간, 공수병 예방접종 및 면역글로불린 접종 실시 여부 등을 조사하였다. 결 과 : 연구기간 중 공수병 예방접종 및 교상 후 조치를 위해 본원에 방문한 만 18세 이하 소아는 41명이었고 교상 전 예방접종을 위해 내원한 소아 11명, 교상 후 예방조치를 위해 내원한 소아 30명이었다. 연령분포를 보면 6세 이후(83.3%)가 많았다. 물린 동물은 개에 의해 가장 많이 발생하여 전체 교상환자 30명 중 27명(90%)을 차지했다. 물린 부위는 다리(9명, 30%), 손(8명, 26.7%)에 많이 물렸다. 교상부위가 미파악된 경우는 전체 교상환자 30명 중 7명(23.3%)이었다. 전체 교상환자 중에서 동물에 대한 광견병 백신 접종을 받았는지 안 받았는지를 알 수 있는 경우는 30명중 10명이었고 이중 4명이 광견병 백신을 접종받은 동물에 의해 교상을 당하였다. 동물의 광견병 백신접종 여부에 따라 교상 후 치료형태를 나누어 보면 광견병 백신을 접종 받은 동물에 의한 교상환자 4명 중 1명(25%)이 상처소독 등의 1차적인 응급조치만 행해졌고 나머지 3명(75%)은 교상 후 조치(교상 후 조치 완료)를 받았다. 반면에 야생동물을 포함하여 광견병 백신접종을 받지 않았거나 백신접종 이력이 불명확한 동물에 의한 교상환자 26명중 9명(34.6%)이 응급조치만 하였으며 나머지 17명(65.4%)은 교상 후 조치 미완료 또는 교상 후 조치를 완료하였다. 결 론 : 이러한 결과는 야생동물을 포함하여 광견병 백신접종을 받지 않았거나 광견병 백신접종 이력이 불명확한 동물에 의한 교상환자에게 교상 후 조치를 실시하는 경우가 상대적으로 적었다는 것을 의미한다. 앞으로 국내에서 공수병 발생을 억제하기 위해서는 먼저 공수병 예방 및 관리 지침에 따른 교상 후 조치에 대한 올바른 이해가 필요하며 교상환자, 해당동물 및 교상지역에 관한 정보를 정확히 기록해야한다.
We characterized the proteomes of murine N2a cells following infection with three rabies virus (RV) strains, characterized by distinct virulence phenotypes (i.e., virulent BD06, fixed CVS-11, and attenuated SRV9 strains), and identified 35 changes to protein expression using two-dimensional gel electrophoresis in whole-cell lysates. The annotated functions of these proteins are involved in various cytoskeletal, signal transduction, stress response, and metabolic processes. Specifically, a-enolase, prx-4, vimentin, cytokine-induced apoptosis inhibitor 1 (CIAPIN1) and prx-6 were significantly up-regulated, whereas Trx like-1 and galectin-1 were down-regulated following infection of N2a cells with all three rabies virus strains. However, comparing expressions of all 35 proteins affected between BD06-, CVS-11-, and SRV9-infected cells, specific changes in expression were also observed. The up-regulation of vimentin, CIAPIN1, prx-4, and 14-3-3 ${\theta}/{\delta}$, and down-regulation of NDPK-B and HSP-1 with CVS and SRV9 infection were ${\geq}2$ times greater than with BD06. Meanwhile, Zfp12 protein, splicing factor, and arginine/serine-rich 1 were unaltered in the cells infected with BD06 and CVS-11, but were up-regulated in the group infected with SRV9. The proteomic alterations described here may suggest that these changes to protein expression correlate with the rabies virus' adaptability and virulence in N2a cells, and hence provides new clues as to the response of N2a host cells to rabies virus infections, and may also aid in uncovering new pathways in these cells that are involved in rabies infections. Further characterization of the functions of the affected proteins may contribute to our understanding of the mechanisms of RV infection and pathogenesis.
