Dexamethasone 투여(投與)가 토끼 virus성(性) 간염(肝炎) 유기(誘起)에 미치는 영향(影響) (Effect of dexamethasone administration on the induction of viral hepatitis in native rabbit)

  • In order to know the effect of dexamethasone on the induction of the rabbit viral hepatitis, the pathological changes were observed in the native rabbits, 2 to 6 months old in age, that were injected by dexamethasone and liver emulsion of Angora rabbit naturally infected with viral hepatitis. The results were summarized as follows: The native rabbits injected by dexamethasone and liver emulsion were infected with viral hepatitis and died between 2 and 7 days after inoculation. Clinical signs and gross lesions were very similar to those of Angora rabbit naturally occurred, In microscopical findings, the hepatic lesions were characterized by peripheral necrosis of the lobules, and peripheral necrosis of the lobules with fatty changes of hepatic cells was occurred in a few cases. Perivascular lymphocytic infiltration in the central nervous system was observed in some cases, The lesions of the other organs were very similar to those of Angora rabbit naturally occurred. On the other hand, the native rabbits that were injected by only liver emulsion of Angora rabbit naturally infected with viral hepatitis were not infected with the disease except very few cases.

  • Dichloroisoproterenol(DCI) i; one of the well known ${\beta}$-adrenergic receptor blocking agents. According to Moran and Perkins, DCI has sympathomimetic like action in relatively low concentrations. Fleming and Hawkins confirmed that DCI acts upon the receptors concerned with positive chronotropic and inotropic actions in the heart. Vogins reported that DCI, in concentration of $5{\times}10^{-8}$ to $5{\times}10^{-6}g/ml$, had properties of sympathomimetic amine causing positive inotropic and chronotropic actions in normal rat atria. And James and Nadeau found that DCI had not only adrenergic blocking effect in moderate and higher concentrations, but it also blocked the effect on the sinus node by vagal stimulation and of directly administered acetylcholine in higher concentrations. As stated above by many authors, DCI has complicated actions according to its concentrations. Our aim at the present experiments was to study the effects of DCI to the action of ouabain and acetylcholine upon the excised rabbit atria, as well as to the action of barium chloride and acetylcholine upon the excised rabbit intestine. In addition, Pan ax Ginseng is widely used as tonics in oriental nations, its pharmacological action, however, has not been clearly established. So we atempted to investigate the effects of the water extract of Panax Ginseng to the action of ouabain and DCI upon both atria and intestine. The results obtained were as follows. 1) DCI has a negative inotropic effect on the excised rabbit atria at concentration of $10^{-5}$ and a positive inotropic effect at concentration of $10^{-6}$. 2) DCI (at concentration of $10^{-6}$) potentiates the positive inotropic effect of ouabain upon the excised rabbit atria. 3) DCI antagonizes the action of acetylcholine upon the excised rabbit atria. 4) The water extract of Panax Ginseng, at concentration of $10^{-3}$, decreases the contractile force of rabbit atria, and tends to slightly increase that of rabbit atria at $10^{-4}$. 5) The water extract of Panax Ginseng exhibits a synergistic action with ouabain on the contractile force of rabbit atria. 6) DCI, in concentrations of $10^{-7}{\sim}10^{-6}$, depresses the tone and amplitude of contraction of the excised rabbit intestine. The depression of the intestinal tone markedly appears in pretreatment with reserpine 2mg/kg 24 hours. 7) DCI antagonizes the contractile effect of barium chloride on the excised rabbit atria. 8) DCI has no significant influence on the action of acetylcholine upon the excised rabbit intestine. 9) The series of those evidences indicates that DCI has a sympathomimetic-like action and more over a relaxing action directly on the excised rabbit intestine. 10) The water extract of Panax Ginseng in concentrations of $10^{-4}{\sim}10^{-3}$, has transient depression of the intestinal tone, but later gradually recovers its normal motility: 11) The water extract of Panax Ginseng has a synergistic action with ouabain on the intestinal contractility.

  • 1. Nepetae Herba water extract (NHW) produced a fall of blood pressure in the rabbit. 2. The depressor effect of NHW was inhibited by atropine, but was not affected by propranolol, hexamethonium and diphenhydramine. 3. NHW showed pressor response in the rabbit treated with atropine. 4. The pressor response in the atropine treated rabbit was inhibited by phentolamine and guanethidine.

택사의 혈압강하작용에 관한 연구 (A Study on the Hypotensive Action of Alismatis Water Extract)

  • Effects of Alismatis water extract (AE) on the blood pressure were investigated in the rabbit and the dog. AE, when administered into the vein of rabbit and dog, into the lateral ventricle of rbbit, produced fall of blood pressure. The depressor response of the rabbits to intravenous AE was abolished by treatment with atropine, but not with chlorisondamine weakened the depressor effect of AE. Intravenous AE in this rabbit produced secondary elevation of the blood pressure. AE potentiated the pressor response of the rabbit to norepinephrine and tyramine, but nor to angiotensin, acetylcholine, and DMPP.

Milk yield and kit development of four breeds of rabbit in Ibadan, Nigeria

  • Background: Rabbit breeding with high performance imported ones would be of benefit for genetic diversity and improvement of performance in domestic rabbit breeds. The rearing of more productive rabbit breeds could be pathway to improve the productivity and reduce the production cost. Maternal nutritional status exert a great influence on reproductive functions of does, which may expand from conception, through gestation and parturition and development of kits to puberty. Methods: Four breeds of rabbit were evaluated for their parturition, weaning and pubertal differences among the rabbit population in Ibadan, Nigeria. The breed consist of Fauve De Bourgogne (FDB), Chinchilla (CHA), British Spot (BS) and New Zealand White (NZW) rabbits. A total of 60 bucks and 360 does consisting of 15 bucks and 90 does per breed were mated in 6 mating cycles, three each of natural mating and artificial insemination. All does were synchronized for estrus with 20 IU pregnant mare serum gonadotropin 48 h prior to mating. The does after parturition were assessed for milk yield (g) and kit survival rate (%) till weaning, weight changes of kits from birth to puberty. At puberty, the pubertal age (days) and weight (g) of the offspring were assessed. Result: Results obtained reveals that British Spot doe had highest milk yield among the breeds which significantly increased growth of kit and weight at weaning in British Spot rabbits. Survival rates of Chinchilla kits were significantly (p < 0.05) higher than Fauve de Bourgogne, British Spot and New Zealand White kits. Puberty attainment of the rabbits indicates that British spot does and Fauve de Bourgogne bucks are early maturing. Conclusion: Chinchilla shows high kit survivability and British spot has highest milk yield among the four breeds of rabbit.

Controlled Partial Skin Thickness Burns: Rabbit Ear as a 2nd Degree Burn Wound Model for Studies of Topical Therapy

  • This study was designed to prepare an animal model for partial thickness bum wound which can be employed for testing topical therapy. We first evaluated whether rabbit ear and mouse back skin wound model could differentiate the wound healing process in terms of degree of re epithelialization, required days for complete wound closure, presence of scarring. $2^{nd}$ degree wet bum were prepared on mouse back skin and rabbit ear by applying 5 mL hot water($85{\pm}0.1^{\circ}C$) for 7 see followed by 5 mL ice-cold 0.5% acrynol solution for cooling and disinfecting the inflicted area. After removing the dead epidermis layer at 24 hr, tested dressings were applied for specified time and wound progression was investigated. In mouse model, wound contraction was the primary wound closing mechanism, which is quite different from human wound healing process. In rabbit ear model, epidermal regeneration was the major wound healing process rather than wound contraction and the difference in wound healing property among tested dressings could be clearly demonstrated. A rabbit ear model could differentiate the wound progression among open, occluded and epidermal growth factor(EGF) treated wound. Four sites of circular wound(diameter: 1 cm) on the anterior part of rabbit ear could be employed for the comparative wound healing study. For obtaining reproducible bum wound, degree of bum depth and bum sites should be carefully controlled in addition, employing rabbits of same strain and weight. The result suggests that rabbit ear could be employed as a reliable and human-resembled wound model.

  • A 2-year-old female lop-eared rabbit (Oryctolagus cuniculus) was presented to the veterinary clinic at a zoo with pruritus, alopecia, and crusting of the ear. Examination of skin scrapes revealed an infestation with the rabbit ear mite, Psoroptes cuniculi. Weekly subcutaneous ivermectin injection over a three-week period resulted in remission of the clinical signs and improvement of the overall conditions of the rabbit.

Expression, Purification, and Characterization of Prothrombin Kringle 2

  • Previously, we reported that the prothrombin kringle 2 (fragment 2), induced by LPS administration into rabbit, inhibited bFGF-stimulated BCE cell growth (Lee et al., 1998). In this study, we cloned and overexpressed the kringle 2 domain of rabbit and human prothrombin as a fusion protein with the pelB leader sequence in E. coli using the T7 promoter. The fusion protein was cleaved during translocation into the peri plasmic space, and cleaved recombinant protein was readily isolated from whole cell lysate by DEAE-Sepharose and Sephacryl S-200 gel filtration chromatography. Both the recombinant rabbit and human prothrombin kringle 2 showed very similar biochemical and functional characteristics to the rabbit prothrombin kringle 2 purified from rabbit serum, in terms of abnormal electrophoretic migration and endothelial cell growth inhibitory activity.

  • Effect of Jeo Ryong-Tang water Extract on Renal Function in Rabbit and Dog. In order to investigate the Pharmacological action of Jeo Ryoung-Tang on renal function, this study was performed in rabbit and dog, making use of it's water extract. Jeo Ryoung-Tang water extract (JRWE), when given into ear vein of rabbits, produced diuresis in a small dose, but antidiuresis in a large dose. Diuretic action of JRWE accompanied the increase of glomerular filtration rate (GFR), renal plasma flow (RPF) and amounts of $Na^+$ in exdreated in urin, but fractional excretion of filtered $Na^+$ was not changed. JRWE, when injected into proleg's vein of dog, produced diuresis, At this time, changes of renal function were similar to that of diuresis in rabbit. JRWE, when infused into a renal artery of dog, exhibited the diuresis in both kidney. It is thought that JRWE, when given into vein of rabbit or dog, induces the diuresis, and the mechanism of it's diuresis is the increase of renal plasma flow through secondary action by some endogenous humoural substance.

  • In this study, we compared skin response between Hartley guinea pig and New Zealand white rabbit. New Zealand white rabbit was treated by a glycolic acid (0, 8, 24, 40, 56 mg/$cm^2$) and UVB (0, 0.4, 3.0 J/$cm^2$) for 14 days. Skin irritation by glycolic acid and UVB were increased in dose and time-dependent manners, and the combination treatment of UVB increased glycolic acid-induced skin irritation. Comparison the skin irritation index between guinea pig and rabbit showed that guinea pig was much more sensitive to glycolic acid and UVB. This study indicated that selection of reliable species of animal could be considered in chemical-induced skin irritation study.
