상엽 유래 퀘세틴과 아스트라갈린의 항염증에 대한 상승효과 (Synergic Effect of Quercetin and Astragalin from Mulberry Leaves on Ani-inflammation)

  • The leaf of mulberry (Morus alba L) has long been used in Oriental medicine for the prevention or treatment of several diseases. However, little is known about the inhibitory effects of a single compound isolated from the mulberry leaves on inflammatory response. We are isolate a single compound of quercetin (3,3',4',5,7-pentahydroxyflavone) and astragalin (kaempferol-3-O-glucopyranoside) from the mulberry leaves, and then investigate the anti-inflammatory effects of quercetin, astragalin or quercetin plus astragalin in lipopolysaccharide (LPS)-stimulated murine peritoneal macrophages. Each compound suppressed the production of inflammatory mediators (NO, $PGE_2$ and IL-6) in LPS-stimulated murine peritoneal macrophages in a dose-dependent manner. Especially, the cotreatment of quercetin (2.5 ${\mu}M$) and astragalin (2.5 ${\mu}M$) markedly suppressed the production and the expression of inflammatory mediators. These suppressive effects were synergistically increased by their combination. These results suggest that the combination of quercetin and astragalin from the mulberry leaves may be useful for therapeutic drugs against inflammatory immune diseases, probably by suppressing the production of inflammatory mediators.

국내산 참나무속 수종 잎의 추출성분 (Chemical Constituents of Domestic Quercus spp. Leaves)

  • 국내산 주요 참나무류인 신갈나무, 상수리나무, 떡갈나무, 졸참나무, 갈참나무 및 상수리나무 잎의 추출성분의 구조를 규명하고 수종 상호간의 성분의 특성 및 상호 연관성 등을 조사하였다. 신갈나무에서는 gallic acid, (+)-catechin, (-)-epicatechin, (+)-gallocatechin, kaempferol, a stragalin, astragalin-6"-O-gallate, isoquercirin, isoquercitrin-6"-O-gallate 및 myricetin 등 10종의 화합물이 단리되었고 상수리나무에서는 gallic acid, kaempferol과 quercetin이 떡갈나무에서는 gallic acid, (+)-catechin, (-)-epicatechin, (+)-galocatechin, (-)-epigallocatechin, kaempferol, quercetin, guajaverin 및 tamarixin 등 9종의 화합물이 단리되었으며 갈참나무에서는 gallic acid, caffeic acid, astragalin, quercetin과 isoquercitrin이 졸참나무에서는 gallic acid, (+)-catechin, (-)-epicatechin, kaempferol, quercitrin, isoquercitrin 및 myricetin 등 7종의 화합물이 단리되었고 굴참나무에서는 gallic acid, (+)-catechin, astragalin, astagalin-6"-O-gallate와 isoquercitrin 등 5종의 화합물이 단리되었다. Gallic aicd는 모든 수종에서 단리되어 참나무류 잎 추출성분의 지표 화합물로 이용될 수 있다.

  • Sevral flavonoids were isolated from the ethanol extract of Carthami flos which has been used in treatment of uterin congestion and also as analgesic and antiinflammatory. They were elucidated as kaempferol, quercetin, 6-hydroxy kaempferol, kaempferol 3-glucoside (Astragalin), quercetin 3-glucoside (isoquercitrin), quercetin 7-glucoside (quercimeritrin), kaempferol 3-rutinoside and quercetin 3-rutinoside (rutin). The structures of the isolated compounds were established by spectroscopic and chemical methods.

두충나무잎의 생리활성 Flavonoid 분석 (Flavonoid Analysis from the Leaves of Eucommia ulmoides)

  • 기호음료 및 약용으로 사용되는 두충나무잎의 품질 평가를 위한 HPLC로서 함유된 flavonoid 화합물을 정성, 정량 분석하였다. 즉 두충나무잎의 n-BuOH 분획으로 부터 astragalin, isoquercitrin, quercetin $3-O-{\beta}-D-xylopyranosyl(1-2)-{\beta}-D-glucopyranoside$의 3종 flavonoid 성분들을 분리하였으며, 이들 화합물은 LPLC의 THF-dioxane-MeOH-HOAc-5% $H_3PO_4-H_2O$(145 : 125 : 50 : 20 : 2 : 658) 혼합용매에서 양호한 분리능을 나타내었다. MeOH 엑스와 n-BuOH 분획 중에 astragaline의 함량은 각각 0.09%(w/w), 0.46%(w/w), isoquercitrin은 0.08%(w/w), 0.48(w/w), quercetin $3-O-{\beta}-D-xylopyranosyl(1-2)-{\beta}-D-glucopyranoside$은 0.40%(w/w), 1.22%(w/w) 함유되어있다.

  • Seven flavonoids were isolated from the aerial parts of Lonicera japonica. Their structures were characterized as hydnocarpin 1, quercetin 2, ochnaflavone 3, ochnaflavone 4'-O-methylether 4, astragalin 5, isoquercitrin 6, and rhoifolin 7 by chemical and spectroscopic evidences.

  • A chemical examination of the aerial parts of Euphorbia pekinensis $R_{UPRECHT}$. (Euphorbiaceae) has led to the isolation of seven hydrolyzable tannins and ten fl avonoid glycosides. The former ones have been identified as gallic acid, methylgallate, 3-O-galloyl shikimic acid, 1,3,4,6-tetra-O-galloyl-${\beta}-_D$-glucose, 1,2,3,4,6-penta-O-galloyl-${\beta}-_D$-glucose, corilagin, geraniin and the latter ones as isoquercitrin, quercitrin, astragalin, afzelin, prunin, rutin, kaempferol-3-O-rutinoside, quercetin-3-O-(2"-O-galloyl)-${\beta}-_D$-glucoside and quercetin-3-O-(2"-O-galloyl)-${\alpha}-_L$-rhamnoside on the basis of chemical and spectroscopic evidence.

산마늘(Allium victorialis)의 추출성분 (Extractives from the Allium victorials)

  • 산마늘(Allium victorials var. platyphyllum)을 채취하여 건조시킨 후 분쇄하여 1.5 kg을 acetone-$H_2O$ (7:3, v/v)로 추출하고 헥산, 메틸렌클로라이드, 에틸아세테이트, 부탄올 및 물 가용부로 분획하여 동결건조하였다. 에틸아세테이트 가용부를 Sephadex LH-20 및 silica gel로 충전한 칼럼에서 다양한 용출용매로 사용하여 칼럼크로마토그래피를 실시하였다. 단리된 화합물들은 박층크로마토그래피(TLC)로 확인한 후 $^1H-$, $^{13}C$-NMR, HMBC 등의 스펙트럼을 사용하여 정확한 구조를 구명하였고 El-MS로써 분자량을 측정하였다. 산마늘의 에틸아세테이트 가용부에는 astragalin (화합물 1), kaempferol (화합물 2), quercetin (회합물 3), ferulic acid (화합물 4)가 분리되었다.

  • Five compounds were isolated from the whole plant of Arabis glabra (Cruciferae) through repeated silica gel and Sephadex LH-20 column chromatography. Their chemical structure were elucidated as salicylic acid, 2,5-dihydroxybenzoic acid, astragalin, rutin, and $quercetin-3,7-O-{\beta}-D-diglucopyranoside$ by spectroscopic analysis.
