Interspecific Hybridization of the Bumblebees Bombus ignitus and B. terrestris

  • The large bumblebee, Bombus terrestris, indigenous to Europe and used extensively for high-value crop pollination, has been artificially introduced in several parts of the world. Here we show the interspecific hybridization between bumblebee species, B. terrestris and B. ignitus, under laboratory conditions. The mating and oviposition percentages of the interspecific hybridization of a B. terrestris queen with a B. ignitus male were higher than those of the intraspecific mating of B. ignitus. Furthermore, the competitive copulation experiment indicated that the mating of B. ignitus males with B. terrestris queens was 1.8-fold more frequent than with B. ignitus queens. The interspecific hybridization of a B. ignitus queen with a B. terrestris male produced either B. ignitus workers or the B. ignitus male phenotype, and the hybridization of a B. terrestris queen with a B. ignitus male produced B. terrestris males. Genetic tests using a portion of the mitochondrial COI gene for the parent and hybrid phenotypes indicated that mitochondrial DNA in the interspecific hybridization was maternally inherited. Our results indicated that interspecific hybridization occurred between B. ignitus and B. terrestris, which suggests that the hybridization will have a negative impact of competition and genetic pollution of native bumblebees.

Availability of Sugar Solutions for Colony Development and Progeny-Queen Production of the European Bumblebee, Bombus terrestris

  • Bumblebees are widely used to pollinate various crops, especially tomato, in greenhouses and fields. Here, we investigated whether different sugar solutions have any effects on the oviposition and colony development of Bombus terrestris queens. The types of sugar solution used were a white sugar solution, brown sugar solution, dark brown sugar solution, and prepared sugar solution; all solution contained with a 40% sugar concentration and 0.3% sorbic acid. Among these sugar solutions, B. terrestris queens reared on the white sugar solution exhibited the best results; the rates of oviposition, colony foundation and progeny-queen production were $75.0{\pm}11.0$, $33.0{\pm}9.6$ and $21.7{\pm}7.3$, respectively, which corresponded to 1.1-33.0-fold increases over the results of other sugar solutions. Queens reared on the prepared sugar solution did not found any colonies. The death rate within one month was lowest for the queens reared on the white sugar solution at $26.1{\pm}10.9%$, which was 1.2-2.0-fold lower than the rate for the other sugar solutions. Furthermore, the numbers of progeny produced by queens reared on the white sugar solution, $131.4{\pm}38.8$ workers and $51.0{\pm}40.6$ queens, were also higher, corresponding to 1.1-1.2-fold increases compared to queens raised on the other sugar solutions. Therefore, the white sugar solution was the favorable sugar solution for the egg-laying and colony developmental characteristics of B. terrestris queens.

[ $CO_{2}$]-Narcosis Time Favorable for Colony Development in the Bumblebee Queen, Bombus terrestris

  • As a means for year-round rearing of bumblebee, CO$_{2}$ -narcosis time favorable for colony development was identified in Bombus terrestris. CO$_{2}$ -narcosis time divided into five classes: 11 days of adult emergence (A-11), the day of adult emergence (A-0), late pupal stage (LP), middle pupal sta ge (MP), and early pupal stage (EP). In egg-laying characteristics, the oviposition rate of LP, A-11 and A-0 was over 76.0$\%$, but that of MP and EP was less than 61.1 $\%$. At the same time, the days needed to first oviposition shortened to 9.8 ­10.5 days in A-11, A-O and LP, comparing to 13.7 -16.1 days in MP and EP. The rate of colony foundation, progeny-queen produced and period of colony foundation of A-11 were the best results in among those at different CO$_{2}$-treatment time. The number of worker produced was 109.2 -110.5 in A-H, LP and A-H, comparing to 82.0 - 86.8 in MP and EP. Also, the number of progeny-queen produced of A-H, A-O and LP was 36.1, 41.0 and 71.3, respectively, which corresponded to 1.5 - 3.1 fold higher than MP and EP. Taken these together, CO$_{2}$-narcosis time favorable for colony development was determined to be 11 days of adult emergence. Also, the day of adult emergence and late pupal stage showed a positive effect on the oviposition and colony development in CO$_{2}$-narcosis time.

Tobacco Promotion and Availability in School Neighborhoods in India: a Cross-sectional Study of their Impact on Adolescent Tobacco Use

  • Background: Adolescent tobacco use is a major public health problem. However, there is little information about the impact of tobacco advertising and availability near schools on adolescent tobacco use in India. Methods: The various tobacco products and brands available in outlets within 100 meters of two high schools in an Indian town were identified. A stratified random sample of 172 participants from these two schools completed a questionnaire on tobacco use and socioeconomic status. Results: Eighteen outlets selling tobacco products were identified. In the two schools the current use of smoked and smokeless tobacco was 9.1% and 17.4% respectively. School location and low socio-economic status of adolescents were associated with tobacco awareness of advertisements (p=0.001) and the receipt of a free sample (p= 0.032). Advertisements on billboards, posters and the receipt of a free tobacco sample were significant factors (p=0.031, p=0.016, p=0.017 respectively) in current tobacco use. Conclusion: In this study a significant proportion of adolescents used tobacco. Tobacco-promotion activities (advertising, the receipt of a free sample), school location and economic status were found to be associated with adolescent tobacco initiation. The local environment should be included in the prevention of adolescent tobacco initiation.

초상화 복식을 중심으로 본 퀸 엘리자베스 1세의 영화 의상에 관한 연구 (A Study of Queen Elizabeth I's Cinema Costumes Viewed from Her Portrait)

  • In 16th century, during renaissance time, Britain's queen Elizabeth I, as a means to express her authority, had an exaggerated clothing styles and was the fashion leader of the time. Thus, through a media of Cinema, tried to find out the characteristics and different aspects of the history of clothing styles, of how they changed, ascertained, restored, and reappeared in the modern time. The research was based on the consideration of Elizabeth I's 33 portraits and her clothing styles; and the research is also from analysing the movies from 1937 to 2007, a total of 9 Cinema and 102 clothes. Movie costumes' most important mission is representing the cinema's timely and spatial background. Thus, it is inevitable that ascertaining and restoring Elizabeth I's portrait should be done. However, the movie costume has to show her dramatic lifestyle, dignity, absolute authority, and other visual images. In addition, the costume has to contribute in making artistic effect and thus the creativity of a designer to satisfy all these needs should be added. Based on the findings from the research, the characteristics of Elizabeth I's Cinema costume are first, ascertaining and restoring portraits, second, mixture of silhouette and detail depending on time period, third, partial change on clothing style design, fourth, emphasis of ruff collar, fifth, magnification colors, sixth, avoiding excessive swelling and decoration, and applying modern style on Protective costume. Thus, based on the research, creative design in Cinema clothing, plays an important part in the history of clothing design. I hope this research could work as a foundation reference for clothing design's history.

Interspecific Hybridization of the Korean Native Bumblebee Bombus hypocrita sapporoensia and the European Bumblebee B. terrestris

  • The large bumblebee $Bombus$ $terrestris$, indigenous to Europe and adjacent areas and used extensively for high-value crop pollination, has been artificially introduced to several parts of the world. Here we show the occurrence of interspecific hybridization between the bumblebee species $B.$ $hypocrita$ $sapporoensia$ and $B.$ $terrestris$ under laboratory conditions. The mating and oviposition percentages resulting from the interspecific hybridization of a $B.$ $terrestris$ queen with a $B.$ $h.$ $sapporoensis$ male were higher than those resulting from the intraspecific mating of $B.$ $h.$ $sapporoensis$. Furthermore, a competitive copulation experiment indicated that the mating of $B.$ $h.$ $sapporoensis$ males with $B.$ $terrestris$ queens was 1.2-fold more frequent than the mating of these males with $B.$ $h.$ $sapporoensis$ queens. The interspecific hybridization of a $B.$ $terrestris$ queen with a $B.$ $h.$ $sapporoensis$ male produced either $B.$ $terrestris$ workers or the $B.$ $terrestris$ male phenotype, and the hybridization of a $B.$ $h.$ $sapporoensis$ queen with a $B.$ $terrestris$ male produced $B.$ $h.$ $sapporoensis$ males. Our results indicated that interspecific hybridization occurred between $B.$ $h.$ $sapporoensis$ and $B.$ $terrestris$. These results suggest that such hybridization will have a negative competitive impact and will cause genetic contamination of native bumblebees.

Chilling Temperature and Humidity to Break Diapause of the Bumblebee Queen Bombus terrestris

  • Bumblebees are widely used to pollinate various crops, especially tomato, in greenhouses and fields. An artificial hibernation is essential for year-round rearing of the bumblebee, which passes through one generation per year. Here, we investigated whether a chilling temperature and humidity affect artificial hibernation of the bumblebee queen Bombus terrestris. In chilling temperature regimes of $0^{\circ}C$, $2.5^{\circ}C$, $5^{\circ}C$, $7.5^{\circ}C$ or $12.5^{\circ}C$ under constant humidity >70%, the queens stored at $2.5^{\circ}C$ exhibited the highest rate of survival, which was 74.0% at one month, 67.0% at two months, 60.0% at three months, 46.0% at 4 months, 33.0% at 5 months, and 24.0% at 6 months. Rates of survival decreased at the following temperatures: $0^{\circ}C$, $5^{\circ}C$, $7.5^{\circ}C$ and $12.5^{\circ}C$. Colony developmental characteristics after diapause were 1.2- to 1.5-fold higher than those of queens stored at $5^{\circ}C$. In terms of chilling humidity, the queens hibernated at 70% under $2.5^{\circ}C$ exhibited the highest rate of survival, which was $93.3{\pm}3.4%$ at one month, $83.3{\pm}0.0%$ at two months, $76.7{\pm}0.0%$ at 3 months and $36.7{\pm}12.1%$ at 5 months. The rates of oviposition, colony foundation and progeny-queen production of queens hibernated at 70% were 80.8%, 30.8% and 30.8%, respectively. These values correspond to 1.7- to 3.3-fold increases in comparison to queens stored at 50% humidity. Therefore, $2.5^{\circ}C$ and 70% R.H. were the favorable chilling temperature and humidity conditions for diapause break of B. terrestris queens.

  • BQCV multi-point PCR was developed as a rapid multiplex detection method for BQCV, one of the viral pathogens of honeybees. It could detect BQCV specific genes qualitative as well as quantitative detection based on ultra-rapid PCR. Three primer pairs (RNA dependent RNA polymerase, capsid protein, 3C like protease) were specifically designed for accurate the detection and were optimized for minimizing the detection time and increasing the sensitivity. Our advanced diagnostic system have the accuracy by lowering the concern about the variation in the BQCV detection site. In addition, it should be an opportunity to identify mutations that are mixed with other viruses.
