Quantitative Analysis of Bioactive Marker Compounds from Cinnamomi Ramulus and Cinnamomi Cortex by HPLC-UV

  • In this study, quantitative and pattern recognition analysis for the quality evaluation of Cinnamomi Ramulus and Cinnamomi Cortex using HPLC/UV was developed. For quantitative analysis, three major bioactive compounds were determined. The separation conditions employed for HPLC/UV were optimized using an ODS $C_{18}$ column ($250{\times}4.6$ mm, 5 ${\mu}m$) with gradient conditions of acetonitrile and water as the mobile phase, at a flow rate of 1.0 mL/min and a detection wavelength of 265 nm. This method was fully validated with respect to linearity, accuracy, precision, recovery, and robustness. The HPLC/UV method was applied successfully to the quantification of three major compounds in the extract of Cinnamomi Ramulus and Cinnamomi Cortex. The HPLC analytical method for pattern recognition analysis was validated by repeated analysis of thirty eight Cinnamomi Ramulus and thirty five Cinnamomi Cortex samples. The results indicate that the established HPLC/UV method is suitable for quantitative analysis.

  • GCSB-5 preparation is a purified extract from a mixture six herbal medicines (Acanthopanacis Cortex, Achyranthis Radix, Saposhnikoviae Radix, Cibotii Rhizoma, Glycine Semen Nigra, Eucommiae Cortex) that have been widely used in traditional medicine to treat various bone disorders. This study was carried out to obtain the HPLC analysis method that can be used to establish quantitative analysis of Glycine Semen Nigra and Eucommiae Cortex for standardization of GCSB-5 preparation. HPLC analysis methods for the simultaneous determination of genistin (Glycine Semen Nigra) and geniposide (Eucommiae Cortex) were established for the quality control of herbal medicinal raw material and preparation. And validation of HPLC analysis methods were conformed for verification of HPLC methods by check to specificity, linearity, intra-day precision, inter-day precision and accuracy following ICH guideline. As the result of quantitative analysis, the contents of genistin and geniposide in the raw material of GCSB-5 preparation were 0.0426-0.0427 mg/g and 0.431-0.432 mg/g. And GCSB-5 preparation contained genistin of 0.0202-0.0203 mg/capsule and geniposide of 0.211-0.212 mg/capsule, respectively.

  • In this study, quantitative and pattern recognition analysis for the quality evaluation of Magnoliae Flos using HPLC/UV was developed. For quantitative analysis, eleven major bioactive lignan compounds were determined. The separation conditions employed for HPLC/UV were optimized using ODS $C_{18}$ column ($250{\times}4.6\;mm$, $5\;{\mu}m$) with isocratic elution of acetonitrile and water with 1% acetic acid as the mobile phase at a flow rate of 1.0 mL/min and a detection wavelength of 278 nm. These methods were fully validated with respect to the linearity, accuracy, precision, recovery, and robustness. The HPLC/UV method was applied successfully to the quantification of eleven major compounds in the extract of Magnoliae Flos. The HPLC analytical method for pattern recognition analysis was validated by repeated analysis of twenty one reference samples corresponding to seven different species of Magnoliae Flos and nine samples purchased from market. The results indicate that the established HPLC/UV method is suitable for the quantitative analysis and quality control of multi-components in Magnoliae Flos.

The separation conditions employed for HPLC/UV were optimized using ODS $C_{18}$ column ($250{\times}4.6mm$, $5{\mu}M$) with isocratic elution of acetonitrile and water with 0.1% phosphoric acid as the mobile phase at a flow rate of 1.0 mL/min and a detection wavelength of 323 nm. These methods were fully validated with respect to the linearity, accuracy, precision, recovery, and robustness. The HPLC/UV method was applied successfully to the quantification of three major compounds in the extract of Cimicifugae Rhizoma. The HPLC analytical method for pattern recognition analysis was validated by repeated analysis of twelve reference samples corresponding to five different species of Cimicifugae Rhizoma and seventeen samples purchased from markets. The results indicate that the established HPLC/UV method is suitable for the quantitative analysis and quality control of multi-components in Cimicifugae Rhizoma.

  • The purpose of this study is to suggest the methodology of $CO_2$ Emission Factor Verification and Quantitative Assessment in Ethylene Product Processes. At first, this study compare the IPCC (Intergovernmental Panel on Climate Change) 1996 Guideline and 2006 Guideline. And analyse methodology for estimating $CO_2$ emission and $CO_2$ emission factor in Ethylene product process. Also analyse cases of estimating $CO_2$ emission factor based on material balance. Methodology of $CO_2$ Emission Factor Verification and Quantitative Assessment are following the categories proposed by GIR (Greenhouse Gas Inventory and Research Center). There are total 12 factors in 8 categories and give 5 or 10 points according to their importance. Also this study suggests necessary data of document to meet the conditions. The result would help estimate accuracy Greenhouse Gas Inventory. Also contribute to establish policy on environmental assessment, air conservation, etc.

The Comparison of Quantitative Accuracy between Energy Window-Based and CT-Based Scatter Correction Method in SPECT/CT Images (SPECT/CT 영상에서 에너지창 기반 산란보정과 CT 기반 산란보정 방법의 정량적 정확성 비교)

  • In SPECT image, scatter count is the cause of quantitative count error and image quality degradation. This study is to evaluate the accuracy of CT based SC(CTSC) and energy window based SC(EWSC) as the comparison with existing Non SC. SPECT/CT images were obtained after filling air in order to acquire a reference image without the influence of scatter count inside the Triple line insert phantom setting hot rod(99mTc 74.0 MBq) in the middle and each SPECT/CT image was obtained each separately after filling water instead of air in order to derive the influence of scatter count under the same conditions. For EWSC, 9 sub-energy windows were set additionally in addition to main energy window(140 keV, 20%) and then, images were acquired at the same time and five types of EWSC including DPW(dual photo-peak window)10%, DEW(dual energy window)20%, TEW(triple energy window)10%, TEW5.0%, TEW2.5% were used. Under the condition without fluctuations in primary count, total count was measured by drawing volume of interest (VOI) in the images of the two conditions and then, the ratio of scatter count of total counts was calculated as percent scatter fraction(%SF) and the count error with image filled with water was evaluated with percent normalized mean-square error(%NMSE) based on the image filled with air. Based on the image filled with air, %SF of images filled with water to which each SC method was applied is non scatter correction(NSC) 37.44, DPW 27.41, DEW 21.84, TEW10% 19.60, TEW5% 17.02, TEW2.5% 14.68, CTSC 5.57 and the scatter counts were removed the most in CTSC and %NMSE is NSC 35.80, DPW 14.28, DEW 7.81, TEW10% 5.94, TEW5% 4.21, TEW2.5% 2.96, CTSC 0.35 and the error in CTSC was found to be the lowest. In SPECT/CT images, the application of each scatter correction method used in the experiment could improve the quantitative count error caused by the influence of scatter count. In particular, CTSC showed the lowest %NMSE(=0.35) compared to existing EWSC methods, enabling relatively accurate scatter correction.

Absolute Radiometric Calibration for KOMPSAT-3 AEISS and Cross Calibration Using Landsat-8 OLI

  • Radiometric calibration is a prerequisite to quantitative remote sensing, and its accuracy has a direct impact on the reliability and accuracy of the quantitative application of remotely sensed data. This paper presents absolute radiometric calibration of the KOMPSAT-3 (KOrea Multi Purpose SATellite-3) and cross calibration using the Landsat-8 OLI (Operational Land Imager). Absolute radiometric calibration was performed using a reflectance-based method. Correlations between TOA (Top Of Atmosphere) radiances and the spectral band responses of the KOMPSAT-3 sensors in Goheung, South Korea, were significant for multispectral bands. A cross calibration method based on the Landsat-8 OLI was also used to assess the two sensors using near simultaneous image pairs over the Libya-4 PICS (Pseudo Invariant Calibration Sites). The spectral profile of the target was obtained from EO-1 (Earth Observing-1) Hyperion data over the Libya-4 PICS to derive the SBAF (Spectral Band Adjustment Factor). The results revealed that the TOA radiance of the KOMPSAT-3 agree with Landsat-8 within 5.14% for all bands after applying the SBAF. The radiometric coefficient presented here appears to be a good standard for maintaining the optical quality of the KOMPSAT-3.

High-accuracy quantitative principle of a new compact digital PCR equipment: Lab On An Array

  • Digital PCR (dPCR) is the third-generation PCR that enables real-time absolute quantification without reference materials. Recently, global diagnosis companies have developed new dPCR equipment. In line with the development, the Lab On An Array (LOAA) dPCR analyzer (Optolane) was launched last year. The LOAA dPCR is a semiconductor chip-based separation PCR type equipment. The LOAA dPCR includes Micro Electro Mechanical System that can be injected by partitioning the target gene into 56 to 20,000 wells. The amount of target gene per wells is digitized to 0 or 1 as the number of well gradually increases to 20,000 wells because its principle follows Poisson distribution, which allows the LOAA dPCR to perform precise absolute quantification. LOAA determined region of interest first prior to dPCR operation. To exclude invalid wells for the quantification, the LOAA dPCR has applied various filtering methods using brightness, slope, baseline, and noise filters. As the coronavirus disease 2019 has now spread around the world, needs for diagnostic equipment of point of care testing (POCT) are increasing. The LOAA dPCR is expected to be suitable for POCT diagnosis due to its compact size and high accuracy. Here, we describe the quantitative principle of the LOAA dPCR and suggest that it can be applied to various fields.

Segmentation of the Glottis and Quantitative Measurement of the Vocal Cord Mucosal Morphology in the Laryngoscopic Image (후두 내시경 영상에서의 성문 분할 및 성대 점막 형태의 정량적 평가)

  • The purpose of this study is to compare and analyze Deep Learning (DL) and Digital Image Processing (DIP) techniques using the results of the glottis segmentation of the two methods followed by the quantification of the asymmetric degree of the vocal cord mucosa. The data consists of 40 normal and abnormal images. The DL model is based on Deeplab V3 architecture, and the Canny edge detector algorithm and morphological operations are used for the DIP technique. According to the segmentation results, the average accuracy of the DL model and the DIP was 97.5% and 94.7% respectively. The quantification results showed high correlation coefficients for both the DL experiment (r=0.8512, p<0.0001) and the DIP experiment (r=0.7784, p<0.0001). In the conclusion, the DL model showed relatively higher segmentation accuracy than the DIP. In this paper, we propose the clinical applicability of this technique applying the segmentation and asymmetric quantification algorithm to the glottal area in the laryngoscopic images.

  • In metal working, cold forging that has profit to satisfy dimension accuracy is using in various manufacturing products. Recently, most of the interest thing is precision forging of gear. Gear forging product is more strength than broaching gear, and it has many advantages with reduction of factory expenses. The reason of difficulty to improve accuracy of gear dimension compare to another products is the dimension accuracy is very high, approximately 10$\mu\textrm{m}$, and because die of involute teeth and elastic strain of forged tool differ from standard curve. This paper represent quantitative analysis of die and teeth of forged tool, namely difference of curves, with experiments and analyze the factor of dimension gap, finally, will design compensated involute curve.
