Studies on the Components of Sokokju, and Commercial Yakju (소국주(小麴酒)와 시판약주(市販藥酒)의 성분(成分)에 관(關)한 연구(硏究))

  • The components of several kinds of Sokokju (old Korean cleared rice wine) prerared in the laboratory by two steps mashing and commercial Yakju (Korean cleared rice wine) were evaluated. The Sokokju mashed with half steamed waxy rice in tile primary fermentation yielded higher contents of alcobol and extract. Especially considerable amount of pyruvate was contained in it. The commercial Yakju sample contained abundantly tartarate and citrate while dominant organic acids in the fermented mashes of Sokokju were lactate, fumarate and succinate. The taste characteristics of the Sokokju brewed with half steamed waxy rice were most acceptable to the sensory panel.

Effects of Supplemention of Rehmannia radix on Performance and Physiological Status in Broiler Chicks (건지황 첨가가 육계의 성장과 생리적 변화에 미치는 영향)

  • The present study was conducted to investigate the effects of dietary supplemental dry powder of Rehmannia radix (RR) root on the growth performance and physiological status of broiler chicks. The treatments consisted of corn-soybean meal control diet, and RR 0.5, 1, and 2% diets. A total of 160 Arbor Acre male broiler chicks were randomly allotted to 16 pens ; four pens per treatment and 10 birds per pen. The growth performance, feed intake and carcass weight did not show any differences among the treatment groups, but feed conversion was significantly lower (P< 0.05) in RR 1% group than that of the control group. The muscle color was lighter (P < 0.05) in RR lof group than those of the other treatment groups. The blood concentrations of protein, glutamic-oxaloacetic-transaminase (GOT) and glutamic- pyruvic-transaminase (GPT) were not different (P > 0.05) among the treatment groups, but the blood total cholesterol content was significantly decreased (P < 0.05) in RR 2% group than those the other treatment groups. The blood concentration of sugar was lower (P <0.05) in RR 2% group than those of the other treatment groups, and hemoglobin concentration was decreased (P <0.05) by addition RR. Crude fat content of thigh muscle was higher in RR 1% group than those of the other treatment groups, and bone hardness of broiler chicks fed RR was higher (P < 0.05) than that of the control group. The saturated fatty acid content of the thigh muscle tende to be high in control and RR 0.5% groups, and unsaturated fatty acid content tende to be high in RR 1 and 2% groups than those of the other treatment groups

Effect of IVM Medium and Protein Source on In Vitro Maturation of Canine Oocytes

  • This study was conducted to determine the ability of nuclear development of canine oocytes depend on the kind of maturation media and addition of serum sources. Ovaries were collected from a bitches at various stages of estrus cycle by an ovariohysterectomy. Oocytes were collected of cumulus oocytes complexes after slicing of ovaries with blade. The maturation medium was containing 0.6 mM/ml cysteine, 0.2 mM pyruvic acid, 20 ng/ml $E_2$ and 1 $\mu g/ml$ rbST Exp. 1, the oocytes were matured in four different maturation medium as follows: 1) TCM-199, 2) DMEM, 3) NCSU37 and 4) modified-NCSU37 with 10% FBS. Exp. 2: the oocytes were matured in mNCSU37 supplemented with different protein sources (10% FBS, 10% EDS, 0.3% BSA and 0.1% PVA) to select the optimal one. Oocytes were matured in a humidified atmosphere containing 5% $CO_2$ at $39{\circ}C$ for 72 hrs. The maturation rate were analyzed by Duncan's multiple range test using General Linear Models procedure in SAS. The rates of meiotic resumption to MI-MII depend on different culture media were achieved with TCM-199 (5.2%), DMEM (5.0%), NCSU37 (7.2%) and m-NCSU37 (5.9%), respectively. The rates of meiotic resumption to MI-MII according to addition of protein source were 10% FBS (13.3%), 10% EDS (25.0%), 0.3% BSA (25.0%) and 0.1% PVA (15.4%), respectively. In conclusion, the results obtained showed that in vitro maturation media and protein supplement to m-NCSU37 culture medium tested did not promote the final steps of IVM in canine oocytes.

Effect of Ovary Transport Temperature on Survivability and Maturation Rate of Canine Oocytes

  • This study examined the viability of canine oocytes following storage at 4 or $38{\circ}C$ for 5 hrs. The ovaries were collected from domestic dog following ovariohysterectomy at a local veterinary clinics and transported to laboratory In two different transport temperature at 4 or $38{\circ}C$ within 5 hrs. The cumulus oocyte complexes (COCs) were recovered after slicing with blade. In Exp. 1, the oocytes collected were matured in DMEM supplemented with l0% FBS, 0.6 mM/mlcysteine, 0.2 mM Pyruvic acid, 20 ng/ml $E_2$ and 1 $\mu g/ml$ rbST at humidified atmosphere containing 5% $CO_2 38{\circ}C$ for 24 or 48 hrs to analysis of survivability. In Exp 2, to assess nuclear development at 38{\circ}C$ group, the oocytes were matured in maturation medium for 24, 48 or 96 hrs. Survivability was judged by a morphological appearance and PI staining. Survivability rates were analyzed by General Linear Models procedure in SAS. The survival rates at 4{\circ}C$ ovary transport group showed significantly lower than at 38{\circ}C$ group (0 vs 72.9% in 48 hrs and 13.2 vs 77 8% in 24 hrs; P<0.05). The nuclear development of oocytes to MI to MII stages at 24, 48 and 96 hrs was 8.3% (6/72), 8.9% (9/101), and 9.5% (8/84). These results showed that the canine oocytes were remarkably sensitive to a low temperature and did not increase nuclear development rate depend on maturation time to 96 hrs.

Changes of Component on Green Discolored Garlic (녹변된 마늘의 성분 변화)

  • Effects of temperature, discoloration, and conditioning on garlic compositions before and after storage were examined. Proximate composition, soluble solids, pH, organic acids, and total sugars of all samples were not significantly different. Sugar contents were sucrose > fructose > glucose, and included such fructooligosaccharide as 1-kestose, 1-nystose, and 1-F-fructosyl nystose. Allicin contents in control, green discolored, and conditioned garlics were 647,9, 613.8, and 648.0 mg%, respectively. Thirty-two flavor compounds were identified by GC/MSD, including 11 sulfide compounds, 5 acids, and 3 aldehydes. Contents of sulfur compounds, including disulfide(di-propenyl), 2-thiophene methanethiol, thiophene, 3,4 dihydro-3-vinyl-1,2-dithiin, and 2-vinyl-4H-1,3-dithin, and water-soluble pectins in low temperature-stored garlic were higher than those in control, and decreased by conditioning or inducing green discoloration. These results show sulfur compounds and water-soluble pectins accumulated in low temperature stored-garlic, and are then converted into green pigments by alliinase in disintegrated garlic tissues.

EFFECTS OF ACTIVATED CARBON ON GROWTH, RUMINAL CHARACTERISTICS, BLOOD PROFILES AND FEED DIGESTIBILITY IN SHEEP

  • This study was carried out to investigate the effects of activated carbon (AC) on growth, ruminal charateristics, blood profiles and feed digestibility in sheep, using roughage-based or concentrate-based diets. Twelve Suffolk breed of sheep of similar age and weight were distributed into 4 groups in a $2{\times}2$ factorial design. Two groups were fed a roughage-based diet with (R + AC) and without AC (R - AC), while the other two were fed a concentrate-based diet with (C + AC) and without AC (C - AC), respectively. The addition of 0.3% AC was based on dry matter of feed offered to animals. The incorporation of AC in roughage and concentrate based diets had no marked effects on feed intake, daily gain and feed conversion of the animals within experimental diets. The results obtained might be due to the low level of AC added in the diet. The animal on both concentrate-based diets were higher than the roughage-based diets in terms of daily gain and feed conversion ratio. However, it was observed that the animals provided with AC in the concentrate-based diet did not suffer from diarrhea and easily adjusted to high concentrate feeding. Further, the pH value for all diets before feeding was noted to be similar. After feeding, however, pH was shown to be higher in R + AC (p < 0.05) than in C + AC diet. Rumen protozoa number was decreased after feeding for both + AC diets, but in C - AC diet it was higher than in the roughage-based diets. For ammonia-nitrogen, C - AC was found to be higher than C + AC diet and the roughage-based diets before feeding. Total volatile fatty acid concentration, propionate and valerate molar ratios for both diets and time of collection were not affected. However, acetate, butyrate and valerate molar ratios were observed to be affected by diets and time of collections. The diets with AC increased (p < 0.05) before feeding for acetate molar ratio, but not different within diet, however, the roughage diets were found to be higher (p < 0.05) in acetate than the concentrate diet. In the blood parameters, the glutamic pyruvic transaminase (GPT), red and white blood cell (RBC, WBC) counts and packed cell volume (PCV) did not differ within and among the diets. Likewise, the WBC differential count in both diets with either - AC or + AC were similar in trend. However, lymphocyte count was noted to be increased in R + AC than the R - AC diet. The addition of AC in both diets did not affect nutrient digestibilities within diets.

Effect of Antioxidants added perilla Oil Diet on Serum and Tissue in Rats (항산화물첨가 들깨기름식이가 흰쥐의 혈청과 조직에 미치는 영향)

  • This study was done to determine the effect of antioxidants added perilla oil diet on the content of cholesterol, vitamin E, and lipid peroxide in serum and tissue of rats. Four groups of experimental diets, such none added perilla oil diet, ascorbic acid added perilla oil diet, vitamin E added perilla oil diet, EDTA added perilla oil diet were fed ad libitum to the 4 weeks white male rats of Sprague-Dawley strain. The results obtained are summarized as follow: 1) The body weight gain in all experimental diet groups was higher than the control gorup and EDTA added diet group was lower than the other experimental diet group, while food intake in vitamin E added diet group was the highest and vitamin C added diet group was the lowest in the control group. 2) Total cholesterol levels in serum of all experimental diet groups were lower than that of the control group and especially the level of total cholesterol in none added diet group and vitamin C added diet group were significantly lower than that of control group. 3) HDL-cholesterol levels of all experimental diet groups were lower than that of the control group and especially none added diet group was significantly lower than that of control group. 4) The activities of glutamic oxaloacetic transaminase (GOT) in serum of all experimental diet group except EDTA added diet group were higher than that of the control group and especially none added diet group was significanly higher than that of the control group. The activites of glutamic pyruvic transaminase (GPT) in serum of all experimental diet groups except vitamin C added group were higher than that of control group. 5) Vitamin E levels in serum of none added diet group and vitamin C added diet group were lower than that of the control group and vitamin E added diet group and EDTA added diet group were higher than that of the control group. 6) Vitamin E levels in liver of all experimental diet groups were higher than that of control group and especially none added diet group and vitamin E added diet group were significantly higher than that of the control group. 7) Lipid peroxide in serum of all experimental diet group were lower than that of control group and especially EDTA added diet group. 8) Lipid peroxide in liver and spleen of all experimental diet groups were higher than that of the control group and lipid peroxide in kidney of all experimental diet groups except EDTA added diet group were higher than that of the control group. Four these results, as vitamin C, vitamin E and EDTA added diets have an effect to lipid peroxide by antioxidants, it could be suggested that perilla oil diet has required to add antioxidant because it has not sufficient vitamin E for antioxidant and intake and overtake level of perilla oil diet should be studied to go ahead.

Effects of Gami-Cheongpyesagan-Tang on Body Fat in High Fat Diet-Fed Obese Mice (가미청폐사간탕(加味淸肺瀉肝湯)이 고지방식이 비만생쥐의 체지방에 미치는 영향)

  • Objectives: This study was designed to investigate the effect of Gami-cheongpyesagan-tang extract (GCST) on high fat diet-induced obesity in rats. Methods: The mice were divided into six groups; normal diet control, high fat diet control (HFD), HFD+GCST administrated group (100, 200, and 400 mg/kg) and olistat-admistrated group. Obesity was induced by high fat diet (45%) for 7 weeks in mice, and GCST was administrated orally every day for 7 weeks. The body weight, food intake, and serological markers such as total cholesterol, triglyceride, lipid contents, leptin, adiponectin and glutamic oxaloacetic transaminase/glutamic pyruvic transaminase were measured in mice. The mRNA expression of obese-associating genes such as sterol regulatory element-binding protein (SREBP)-1c, fatty acid synthase (FAS), stearoyl-CaP desaturase (SCD-1), peroxisome proliferator-activated receptor $(PPAR)-{\alpha}$, COA oxidase (ACO), and carnitine palmitoyltransferase ($CPT-1{\alpha}$) was analyzed by reverse transcription polymerase chain reaction. Results: The administration of GCST at 400 mg/kg, significantly reduced the increase of body weight and food intake as well as food efficiency compared to HFD group. GCST decreased the serum levels of triglyceride, total cholesterol, low-density lipoprotein-cholesterol, leptin in HFD control group and inhibited lipid accumulation in liver and adipose tissues, but did not increase high-density lipoprotein-cholesterol. In the liver tissues of GCST administrated HFD group, the mRNA levels of SREBP-1c, FAS and SCD-1 were decreased and the mRNA levels of $PPAR-{\alpha}$, ACO, and $CPT-1{\alpha}$ were increased. Conclusions: These results indicate that GCST could improve high fat diet induced obesity through inhibiting the hyperlipidemia in fatty Liver. It suggest that GCST may be used clinically for declining the accumultion of body fat with hyperlipidemia.

Oleoresin Content and Functional Properties of Fresh Onion by Microwave-Assisted Extraction (마이크로웨이브 추출공정에 의한 양파의 올레오레진 함량 및 기능적 특성)

  • Fresh onions were extracted by two methods of conventional extraction(CE) and microwave assisted extraction(MAE) under different extraction conditions. Onion extracts obtained by CE and MAE were examined in oleoresin yield and physiological activities. The optimal extraction time of MAE was about 5 minutes, whereas that of CE was 2 hours. Therefore extraction time was shortened drastically by MAE but there was no significance in oleolesin yield. The electron donating abilities also showed negligible difference between two extracts obtained by CE and MAE, and 45% level in CE and 50% level in MAE. The nitrite scavenging effect was diminished while pH was increasing, and showed a high elimination effect over 85% at pH 1.2. The tyrosinase inhibitory effect was 20% level in both CE and MAE when water was used and 40% level when ethyl alcohol was used. The angiotensin I converting enzyme inhibitory effect showed higher activity with 70% level in MAE than 60% level in CE. The pyruvic acid content was 6.8 and 6.4 moles per 1g of fresh onion by CE and MAE when water was used, and was 4.3 and 5.6 moles by CE and MAE when ethyl alcohol was used.

Quality Change of Garlic during Storage by Stem and Root Cutting Treatments (마늘의 줄기 밑 뿌리절단에 따른 저장 중 품질변화)

  • Physicochemical properties of garlic bulb during storage with different cutting treatments were estimated. Garlic harvested in early June was used in this experiments. Various physicochemical factors of garlic bulb such as weight loss rate, rotten rate, sprouting rate, moisture content, reducing sugar content and total pyruvate content were investigated. Bulbs with roots, bulb without roots and bulb with stem length of l cm, 3 cm, 5 cm were stored at 2$\^{C}$ for eight months. The weight loss increased sharply after 8 months for all treatments. Weight loss occurred most severely in treated garlics with bulbs with 5 cm stem length and progressed steadily at low rates in bulbs with 1 cm stem length. Also, less decay and internal sprouting were observed in bulbs with 1 cm stem length. Cutting treatments of roots were not significant for internal sprouting. Incidence of other chemical properties of bulbs, contents of total sugar, reducing sugar and pyruvic acid were not significant statistically. Moisture contents of treated garlics was remarkably reduced in longer leaving stems. As a result, top-clipped leaving stems fur long, seemed to have a beneficial effect on physicochemical properties of garlic stored at 2$\^{C}$ for 8 months possibly due to reduced weight loss, rotten decaying and sprouting rates. Utilization of such results in the processing industry can be effective.
