PIV와 수치해석을 이용한 분지관내 맥동유동의 가시화 (Flow Visualization of Pulsatile Flow in a Branching Tube using the PIV System and Numerical Analysis)

  • The objective of the present study is to visualize the pulsatile flow fields by using three-dimensional computer simulation and the PIV system. A closed flow loop system was built for the steady and unsteady experiments. The Harvard pulsatile pump was used to generate the pulsatile pressure and velocity waveforms. Conifer powder as the tracing particles was added to water to visualize the flow field. Two consecutive particle images were captured by a CCD camera for the image processing. The cross-correlation method in combination with the moving searching area algorithm was applied for the image processing of the flow visualization. The pulsatile flow fields were visualized effectively by the PIV system in conjunction with the applied algorithm. The range validation and the area interpolation methods were used to obtain the final velocity vectors with high accuracy. The finite volume predictions were used to analyze three-dimensional flow patterns in the bifurcation model. The results of the PIV experiment and the computer simulation are in good agreement and the results show the recirculation zones and formation of the paired secondary flow distal to the apex of the bifurcated model. The results also show that the branch flow is pushed strongly to the inner wall due to the inertial force effect and helical motions are generated as the flow proceeds toward the outer wall.

  • The objective of the present study was to visualize the pulsatile flow field in a branching model by using the high-resolution PIV system. A bifurcated flow system was built for the experiments in the pulsatile flow. Harvard pulsatile pump was used to generate the pulsatile velocity waveforms. Conifer powder as the tracing particles was added to water to visualize the flow fields. Two consecutive particle images at several cross sections of the flow filed were captured by the CCD cameras ($1K{\ast}1K$ and $640{\ast}480$). The results after the image processing clearly showed the recirculation zones and the formation of the paired secondary flows from the distal to the apex in the bifurcated model. The results also indicated that the flow velocities in the inner wall moved faster than those in the outer wall due to the inertial force effects and the helical motions generated in the branch flows as the flow proceeded toward the outer wall. While the PIV images from the $1K{\ast}1K$ camera were closer to the simulation results thantheimagesfromthe640${\ast}$480camera,bothresultsofthePIVexperimentsusingthetwocamerasgenerallyagreed quitewellwiththeresultsfromthenumericalsimulation.

Neuroendocrine Control of Gonadotropin Secretion during the Menstrual Cycle

  • Two modalities of gonadotropin secretion, pulsatile gonadotropin and preovulatory gonadotropin surge, have been identified in the mammals. Pulsatile gonadotropin secretion is modulated by the pulsatile pattern of GnRH release and complex ovarian steroid feedback actions. The neural mechansim that regulates the pulsatile release of GnRH in the hypothalamus is called "GnRH pulse generator". Ovarian steroids, estradiol and progesterone, appear to exert thier feedback effects both directly on the pituitary to modulate gonadotropin release and on a hypothalamic site to modulate GnRH release; estradiol primarily affects the amplitude while progesterone decreases the frequency of the pulsatile GnRH. Steroid hormones are known to affect catecholamine transmission in brain. MBH-POA is richly innervated by NE systems and close apposition of NE terminals and GnRH cell bodies occurs in the MBH as well as in the POA. NE normally facilitates pulsatile LH release by acting through ${\alpha}-receptor$ mechanism. However, precise nature of facilitative role of NE transmission in maintaining pulsatile LH has not been clearly understood. Close apposition of DA and GnRH terminals in ME might permit DA to influence GnRH release. Action of DA transmission probably is mediated by axo-axonic contacts between GnRH and DA fibers in the ME. Dopamine transmission does not normally regulate pulsatile LH release, but under certain conditions, increased DA transmission inhibit LH pulse. Endogenous opioid acts to suppress the secretion of GnRH into hypophysial portal circulation, thereby inhibiting gonadotropin secretion. However, an interaction between endogenenous opioid peptides and gonadotropin release is a complex one which involves ovarian hormones as well. LH secretion appears to be most suppressed by endogenenous opioids during the luteal phase, at a time of elevated progesterone secretion. The arcuate nucleus contains not only cell bodies for GnRH and ${\beta}-endorphin$ but also a dense aborization of fibers suggesting that GnRH release is changed by the interactions between GnRH and ${\beta}-endorphin$ cell bodies within the arcuate nucleus. The frequency and amplitude of pulsatile LH release seem to be increased during the preovulatory gonadotropin surge. Estradiol exerts positive feedback action on the hypothalamo-pituitary axis to trigger preovulatory LH surge. GnRH is also crucial hormonal stimulus for preovulatory LH surge. It is unlikely, however, that increased secretion of GnRH during the preovulatory gonadotropin surge represents an obligatory neural signal for generation of the LH discharge in primates including human. Modulation of preovulatory LH surge by catecholamines has been studied almost exclusively in rats. NE and E may be involved in distinct way to accumulate GnRH in the MBH and its release into the hypophysial portal system during the critical period for LH surge on proestrus in rats. However, the mechanisms whereby augmented adrenergic transmission may facilitate the formation and accumulation of GnRH in the ME-ARC nerve terminals before the LH surge have not been clearly understood.

  • Characteristics of the pulsatile flow in a 3-dimensional elastic blood vessel are investigated to understand the blood flow phenomena in the human body arteries. In this study, a model for the elastic blood vessel is proposed. The finite volume prediction is used to analyse the pulsatile flow in the elastic blood vessel. Variations of the pressure, velocity and wall shear stress of the pulsatile flow in the elastic blood vessel are obtained. The magnitudes of the velocity waveforms in the elastic blood vessel model are larger than those in the rigid blood vessel model. The wall shear stresses on the elastic vessel vary with the blood vessel motions. Amplitude indices of the wall shear stress for blood in the elastic blood vessel are $4\sim5$ times larger than those of the Newtonian fluid. As the phase angle increased, point of the phase angle is are moved forward and the wall shear stresses are increased for blood and the Newtonian fluid.

Twenty-four-hour Variation of Plasma Leptin Concentration and Pulsatile Leptin Secretion in Cattle

  • We conducted this study to investigate 24 h leptin profiles and to ascertain whether leptin secretion occurs in a pulsatile manner in cattle. Plasma leptin concentrations were measured every 10 min for 24 h in five Holstein steers aged 10 months. Simultaneously, feeding behavior was recorded every 5 min during this experiment. In two of the five cattle, leptin showed diurnal rhythmicity, which could be described by a cosine, with peaks between 15:00 and 16:00 and nadirs at around midnight. Pulsatile leptin release was quantified by model-free Cluster analysis. Plasma leptin showed a pulsatile pattern in all cattle, with an average number of pulses at 15 peaks/24 h. The daily number of pulses was not related to total time spent eating, ruminating or chewing. However, when divided into six 4 h time intervals, time spent ruminating was positively related with pulse number (p=0.05) in cattle showing no diurnal plasma leptin variation. These results suggest that cattle may have unique diurnal variation and pulsatile patterns of plasma leptin, differing from those of monogastric animals.

  • The arterial stenosis related to the intimal thickening of the arterial wall is the main cause of many diseases in human cardiovascular system. Hemodynamic behavior of the blood flow is influenced by the presence of the arterial stenosis. In this study, effects of the pulsatile flow, caused by the periodic motion of the heart, on the blood flow and its interaction with the arterial stenosis are analyzed by the FEM-based computational fluid dynamics. As a result, it was found that the characteristics of the pulsatile flow in the artery with stenosis are quite different from those of the steady flow. And, the pulsatile flow condition affects the wall shear stress, which is one of the most important physiological parameters in the hemodynamics.

혈액모사유체의 미세협착 주변 맥동유동 시뮬레이션 (Numerical Simulation of Pulsatile Flows around Micro-Stenosis for Blood Analog Fluids)

  • Considering the role of viscosity in the hemorheology, the characteristics of non-Newtonian fluid are important in the pulsatile blood flows. Stenosis, with an abnormal narrowing of the vessel, contributes to block blood flows to downstream tissue and lead to plaque rupture. Therefore, systematic analysis of blood flow around stenosed vessels is crucial. In this study, non-Newtonian behaviors of blood analog fluids around the micro-stenosis with 60 % severity in diameter of $500{\mu}m$ was examined by using CFX under the pulsatile flow conditions with the period of 10 s. Viscosity information of two non-Newtonian fluids were obtained by fitting the value of normal blood and highly viscous blood. As the Newtonian fluid, the water at room temperature was used. During the pulsatile phase, wall shear stress (WSS) is highly oscillated. In addition, high viscous solution gives rise to increases the variation in the WSS around the micro-stenosis. Highly oscillating WSS enhance increasing tendency of plaque instability or rupture and damage of the tissue layer. These results, related to the influence on the damage to the endothelium or stenotic lesion, may help clinicians understand relevant mechanisms.

이중 박동성 인공심폐기의 동물 실험 (Application of the Pulsatile Cardiopulmonary Bypass in Animal Model)

  • 배경: 기존에 사용되고 있는 인공심폐기는 비박동성 펌프로 회로 내부에 형성되는 압력이 낮고 혈구 손상이 적어서 표준 인공 심폐기로 선호되고 있다. 그러나 많은 실험적 연구들을 보면 박동성 관류가 혈역학, 대사, 장기의 기능, 미세 순환에 대하여 비박동성 관류보다 유익함을 알 수 있다. 그러나 박동성 인공심폐기는 높은 회로 내부 압력과 혈구손상이 해결되어야 할 문제점으로 개발이 어려웠다. 이 연구의 목적은 국내에서 제작된 이중 박동성 인공심폐기의 안전성 및 유효성을 알아보는 것이다. 대상 및 방법: 6마리의 송아지를 대상으로 이중 박동성 인공 심폐기 실험을 하였으며, 완전체외순환을 시켰다. 체외순환은 상,하 대정맥과 대동맥 사이를 우회하였으며, 막형 산화기를 사용하여 혈액을 산화시켰다. 산화기는 좌우측 펌프 중간에 위치시켰다. 순환 시간은 4시간이었다. 동맥혈가스분석, 혈액검사, 혈장 유리 혈색소를 측정하였으며, 맥압과 회로 내부압력을 관찰하였다. 측정은 인공 심폐기 구동 전과 순환 1, 2, 3, 4시간째에 측정하였다. 결과: 동맥혈 가스분석상 pH, 이산화 탄소분압은 모두 정상 범위였으며, 동맥혈 산소 분압은 100 mmHg 이상의 충분한 산화를 보였다. 혈장 유리혈색소는 완전 체외순환하여 4시간이 경과하면서 15.87 $\pm$5.63 mg/㎗로 점차 증가소견을 보였으나 20 mg/㎗ 이내였다. 혈액검사소견상 뚜렷한 이상소견은 없었다. 순환전 수축기 혈압은 97.5$\pm$5.7 mmHg이었으며 시간 경과하여도 100 mmHg 이상의 혈압을 유지하였다. 확장기 혈압은 72.2$\pm$7.7 mmHg이었으며 시간 경과하여도 잘 유지되었다. 평균 혈압은 순환 전 83$\pm$9.2 mmHg이었으며, 시간 경과 시 증가된 소견을 보였다. 펌프의 유량은 3.3 L/min이상을 유지할 수 있었다. 완전체외순환 동안의 혈압의 변화를 보면 순환 전 심장과 유사한 동맥혈압을 보였다. 결론: 이중 박동 구조를 이용한 박동성 인공심폐기는 효과적인 박동성 혈류를 제공하면서 혈구 세포손상도 적었으며, 혈액학적 및 혈역학적인 면에서도 우수한 결과를 보였다. 따라서 모든 심장수술의 인공심폐기로 사용되어질 경우 말초장기에 생리적이고 안정적인 혈류를 공급할 수 있을 것으로 기대된다.
