  • Salmonella enterica serovars Gallinarum(SG, causative agent of fowl typhoid) and S Pullorum(SP, causative agent of pullorum disease) are very important bacterial pathogens in poultry industry. They share some common antigenic properties though the characteristics of outbreaks are quite different. To discriminate between SG and SP, we developed two rapid diagnostic methods, allele-specific real-time PCR and 3'-tailed PCR over 2 single nucleotide polymorphisms ($237^{th}\;and\;598^{th}$). In both methods, $237^{th}$ allele was found to be a good target for differential diagnosis, while $598^{th}$ allele produced some non-specific reactions.

  • An agglutinability between naturally infected positive chicken serum of pullorum disease and hyperimmumized rabbit antiserum was compared. And the following results were obtained and summarized. 1. On the agglutinability, Salmonella pullorum antigen which irradiated gamma-ray was more excellent than another both formalized and heated antigen. 2. Time of judgemented as positive titer in the tube agglutination test to the naturally infected positive chicken serum was it most suitable for 12 hours at $37^{\circ}C$. 3. Agglutination titer of positive immune chicken serum against gamma-ray irradiate Salmonella pullorum were as 320~640x.

Experimental Pathogenesis of Pullorum Disease with the Local Isolate of Salmonella enterica serovar. enterica subspecies Pullorum in Pullets in Bangladesh

  • The research work was carried out to study the pathogenesis covering the clinical signs, gross and histopathological lesions in different organs, and reisolation and identification of the organisms after experimental infection with the local isolate of Salmonella enterica serovar. enterica subspecies (S.) Pullorum at different time interval of the experiment during the period February 2006 to December 2006. One hundred pullets (seronegative to S. Pullorum of 12 weeks age were purchased and divided into 5 (A, B, C, D and E) groups and each group consisted of 20 birds. Four groups (A, B, C and D) were infected orally with a dose of $10^6\;CFU$, $10^7\;CFU$, $2{\times}10^7\;CFU$, $10^8\;CFU$ of S. Pullorum, respectively, and one group (E) was treated as uninfected control. The used methods were necropsy and histopathology, culture of bacteria, staining and biochemical test of Salmonella. Five birds from each group were randomly selected and sacrificed $1^{st}$ week, $2^{nd}$, $3^{rd}$ and $4^{th}$ weeks of post infection (PI). From all the groups, the bacteriological samples (crop, liver, lung, heart, spleen, bile duodenum, ceca and blood) were collected with pre enriched in buffered peptone water in sterile poly bags. Liver, lungs, heart, spleen, intestine, etc. were collected in 10% buffered-formalin for histopathological examination. No clinical signs, gross and histopathological lesions were found in control group and no S. Pullorum was reisolated. Clinical sign of experimentally infected with S. Pullorum in pullets were loss of appetite (100%), slight depression (75%), ruffled feathers (85%), diarrhea (60%) and loss of weight (100%) in chickens. The feed intake and body weight at different weeks after PI differed significantly (p<0.01) among the groups. Grossly, the highest recorded lesion was button-like ulcer in the ceca (80%) and the lowest was white nodules in lungs (1.25%). S. Pullorum were reisolated from crop (91.25%), liver (91.25%), lung (83.75%), heart (71.25%), spleen (87.75%), bile (33.25%), duodenum (92.50%), ceca (97.50%) and from different group of infection (61.25%). The highest microscopic findings were intestinal and cecal mucosa and submucosa exhibited infiltration of mononuclear cells and congestion (96.25%), and the lowest finding was nodule formation in the lungs (3.75%). The pattern of the disease production by local isolate of S. Pullorum in Bangladesh is almost similar with other isolates in different countries.

  • Salmonella enterica serovar Gallinarum is the causative agent of fowl typhoid (FT) and Salmonella enterica serovar Pullorum is pullorum disease (PD), a severe systemic disease of chick and it has the same antigenic fomula, the close relation but distinct pathogen. The traditional bacteriologic and serologic methods routinely used but tedious, time consuming. some of biochemical differences are helpful in differentiating the two organisms, however variation in the characteristics of some strains can be observed. During 2006 to 2008, there was isolated 30 strains. The biochemical characteristics of S. Gallinarum was nonmotile, fermentation of dulcitol, maltose but positive arginine (6.6%), lysine (83.3%) and arabinose (20.0%). The antimicrobial susceptibility test showed 100% sensitive to amikacin, ampicillin, amoxicillin/clavulanic acid and florfenicol, but resistant to penicillin (100%) and erythromycin (60.0%). This PCR method can be applied in the diagnosis between S. Gallinarum and S. Pullorum.

  • Prevalence of major legal communicable diseases in chickens and ducks, which had occurred in Jeonbuk province from year 2004 to 2008. Total 283 farms 1,419,244 chickens and ducks have been affected by avian diseases. Specifically, fowl typhoid (FT) occurred in 92 farms 416,600 chickens, Marek's disease (MD) in 45 farms 145,563, duck virus hepatitis (DVH) in 31 farms 199,200, infectious bursal disease (IBD) in 27 farms 113,220, infectious bronchitis (IB) in 27 farms 280,300, low pathogenic avian influenza (LPAI) in 26 farms 78,495, avian mycoplasmosis in 16 farms 103,774, Newcastle disease (ND) occurred in 11 farms 61,052, avian encephalomyelitis (AE) in 7 farms 21,000, Pullorum disease (PD) occurred in 1 farm 40. According to total analysis about major legal communicable diseases, 1 species of first-class legal communicable diseases have occurred, 3 species of second-class and 6 species of third-class all adding up to 10 species. In the first-class diseases, Newcastle disease have occurred. Pullorum and fowl typhoid, duck virus hepatitis in the second-class have occurred and as third-class diseases, Marek's disease, Infectious bursal disease, Infectious bronchitis, avian mycoplasmosis, avian encephalomyelitis, low pathogenic avian influenza have occurred.

Specific DNA fragment analysis of Salmonella pullorum and S gallinarum by subtraction PCR (RDA method(Subtraction PCR) 기법을 이용한 닭의 Salmonella pullorum과 S gallinarum의 specific DNA fragment 분리 연구)

  • Pullorum disease and Fowl typhoid are kind of poultry specific disease for poultry. The peculiar character of these poultry specific diseases is that it can be infected by transmitting vertically and horizontally, also it is hard to be discovered by clinical sign, and pathology or immunology. So, to develop the PCR method which distinguishes these two genetically similar diseases of separated the specific DNA fragment from each strain and use it for differential diagnosis by subtraction PCR method. Standard strain of S gallinarum and S pullorum, and field isolation strain were verified by biochemistry, It confirmed existence of plasmid by using the PFGE. Then, Isolated DNA from it and used it as materials for the experiment. After cutting genomic DNA of two strains by using Sau 3Al, It ligated primer to tester DNA for PCR amplification and separated specific DNA fragment bacteria with method of subtraction PCR. And, It confirmed that it is a piece of unique DNA in every bacteria using base sequence of separated DNA fragment. 1. The six specific DNA fragment were separated from the DNA of S gallinarum and S pullorum by the subtraction PCR method. 2. In the result of comparison after setting base sequence of each fragment, each separated base sequence of DNA fragment they did not correspond to each other 3. As the result of each DNA fragment is derived from the each strain of DNA, and there was no homology of genomic DNA level in mutual. 4. The fragment originated in plasmid and includes S pullorum did not separate. 5. In the result of searching base sequence in Genebank, it partially shows homology in Salmonella enterica, S typhimurium, S dublin, Escherichia coli, Shigella flexneri, Yersinia pestis, Klebsiella pneumoniae. 6. Primer design by S gallinarum DNA 2, 3 fragment used PCR, They are positive reaction in only S gallinarum at 276, 367 bp position.

  • This survey was performed to investigate avian salmonellosis from broiler farms in Namwon area for 4 years. The whole samples from farms were 541 and chicken samples were 294. Out of 294, 83 cases were identified with avian salmonellosis for 4 years by various serological and biochemical tests. There was no outbreak of pullorum disease, but fowl typhoid occurred in 36 farms. The outbreak of avian salmonellosis occurred continuously regardless of seasons throughout the year. Avian salmonellosis mostly happened within 10 days of age which were 52 cases(63%). The selection of susceptible antibiotics has changed from Quinoline group in 2000 to amoxacillin + clavulanic acid(AmC).

  • A number of chemotherapeutic agents, namely antibiotics, sulfonamides and nitrofuran derivatives have been used, as a fred additive, for the purposes of growth improvement of chicken, increase of feed efficiency, decrease of animal mortality, and disease prevention. In these experiments, furamizole which is one of nitrofuran derivatives and feed additive was test, in field, its antibacterial activity against Mycoplasma gallisepticum causing air sac disease, its effect on pullorum disease control, its effect on the lowering the mortality of chicken and finally the increase of feed efficiency. Throughout the studies, furamizole, in concentration of 0.025% in feed fed to baby chicken continuously resulted as following: 1. Tested chicken showed no avian mycoplasma infection compared to 3.7% outbreak in control chicken. 2. Tested chicken showed a low degree of outbreak of pullorum disease. However, its outbreak was much more surpresed compared to that of control chicken. 3. Total mortality rate of 5.5% and 30.8% were obtained in test and control chicken respectively. 4. Feed efficiency were 2.83 and 2.97 in test and control chicken respectively.

Determination of sample size to serological surveillance plan for pullorum disease and fowl typhoid (추백리-가금티푸스의 혈청학적 모니터링 계획수립을 위한 표본크기)

  • The objective of this study was to determine appropriate sample size that simulated different assumptions for diagnostic test characteristics and true prevalences when designing serological surveillance plan for pullorum disease and fowl typhoid in domestic poultry production. The number of flocks and total number of chickens to be sampled was obtained to provide 95% confidence of detecting at least one infected flock, taking imperfect diagnostic tests into account. Due to lack of reliable data, within infected flock prevalence (WFP) was assumed to follow minimum 1%, most likely 5% and maximum 9% and true flock prevalence of 0.1%, 0.5% and 1% in order. Sensitivity were modeled using the Pert distribution: minimum 75%, most likely 80% and maximum 90% for plate agglutination test and 80%, 85%, and 90% for ELISA test. Similarly, the specificity was modeled 85%, 90%, 95% for plate agglutination test and 90%, 95%, 99% for ELISA test. In accordance with the current regulation, flock-level test characteristics calculated assuming that 30 samples are taken from per flock. The model showed that the current 112,000 annual number of testing plan which is based on random selection of flocks is far beyond the sample size estimated in this study. The sample size was further reduced with increased sensitivity and specificity of the test and decreased WFP. The effect of increasing samples per flock on total sample size to be sampled and optimal combination of sensitivity and specificity of the test for the purpose of the surveillance is discussed regarding cost.
