Identification of Ku70/Ku80 as ADD1/SREBP1c Interacting Proteins

  • In vertebrates, multisubunit cofactors regulate gene expression through interacting with cell-type- and gene-specific DNA-binding proteins in a chromatin-selective manner. ADD1/SREBP1c regulates fatty acid metabolism and insulin-dependent gene expression through binding to SRE and E-box motif with dual DNA binding specificity. Although its transcriptional and post-translational regulation has been extensively studied, its regulation by interacting proteins is not well understood. To identify cellular proteins that associate with nuclear form of ADD1/SEBP1c, we employed the GST pull-down system with Hela cell nuclei extract. In this study, we demonstrated that Ku proteins interact specifically with ADD1/SREP1c protein. GST pull-down combined with peptide sequencing analysis revealed that Ku80 binds to ADD1/SREBP1c in vitro. Additionally, western blot analysis showed that Ku70, a heterodimerizing partner of Ku80, also associates with ADD1/SREBP1c. Furthermore, co-transfection of Ku70/Ku80 with ADD1/SREBP1c enhanced the transcriptional activity of ADD1/SREBP1c. Taken together, these results suggest that the Ku proteins might be involved in the lipogenic and/or adipogenic gene expression through interacting with ADD1/SREBP1c.

작업형태 및 최대 수의적 수축에 따른 등척성 근지구력에 관한 연구 (The Study of Isometric Endurance Time by Task Type and Maximum Voluntary Contraction)

  • This study was performed to investigate the isometric endurance time as percentages of maximum voluntary contraction. Electromyogram(EMG) and Borg's CR-I0 value were measured by push-pull-up-down tasks for 10 healthy males. The normalized EMG value and the MPF(mean power frequency) were used to estimate the muscle recruitment pattern and the development of muscle fatigue. The subjects exerted and maintained 5 levels of %MVC(maximum voluntary contraction) in $90^{\circ}$ shoulder flexion/ 180oelbow extension at sitting posture. The up-task showed the lower endurance time and higher Borg's CR-I0 value than the other task types. Comparing Rohmert's curve with the endurance time of task types. Rohmert's curve overestimated the endurance time of up-task and underestimated the endurance time of push-pull-down tasks. The normalized EMG value showed that muscles recruitment patterns were different from task types. The 4 muscles(biceps brachii muscle, tricep brachii muscle. middle deltoid muscle. trapezius muscle) recruitment patterns of up-task were higher than those of other tasks. The MPF value decreased with the endurance time, and the shift of MPF at up-task was larger than that of the other task types.

Interactions among Measles Virus Hemagglutinin, Fusion Protein and Cell Receptor Signaling Lymphocyte Activation Molecule (SLAM) Indicating a New Fusion-trimer Model

  • For measles viruses, fusion on the cell membrane is an important initial step in the entry into the infected cells. The recent research indicated that hemagglutinin firstly leads the conformational changes in the fusion protein then co-mediates the membrane fusion. In the work, we use the co-immunoprecipitation and pull-down techniques to identify the interactions among fusion protein, hemagglutinin and signaling lymphocyte activation molecule (SLAM), which reveal that the three proteins can form a functional complex to mediate the SLAM-dependent fusion. Moreover, under the confocal microscope, fusion protein and hemagglutinin protein can show the cocapping mediated by the SLAM. So fusion protein not only is involved in the fusion but also might directly interact with the SLAM to be a new fusion-trimer model, which might account for the infection mechanism of measles virus.

Isolation of Proteins that Specifically Interact with the ATPase Domain of Mammalian ER Chaperone, BiP

  • BiP, immunoglobulin binding protein, is an ER homologue of Hsp70. However, unlit other Hsp70 proteins, regulatory protein(s) for BiP has not been identified. Here, we demo strafed the presence of potential regulatory proteins for BiP using a pull -down assay. Since BiP can bind any unfolded protein, only the ATPase domain of BiP was used for the pull -down assay in order to minimize nonspecific binding. The ATPase domain was cloned to produce recombinant protein, which was then conjugated to CNBr-activated agarose. The structural conformation and ATP hydrolysis activity of the recombinant ATPase domain were similar to those of the native protein, light proteins from metabolically labeled mouse plasmacytoma cells specifically bound to the recombinant ATPase protein. The binding of these proteins was inhibited by excess amounts of free ATPase protein, and was dependent on the presence of ATP. These proteins were eluted by ADP. Of these proteins, Grp170 and BiP where identified. while the other were not identified as known ER proteins, from Western blot analyses. The presence of the ATPase-binding proteins for BiP was first demonstrated in this study, and our data suggest similar regulatory machinery for BiP may exist in the ER, as found in prokaryotes and other cellular compartments.

In vitro Interaction of Recombinantly Expressed Kringle 5 (rK5) with Ras Guanine Nucleotide Dissociation Stimulator-like Factor (Rgl2)

  • Kringle 5 (K5), located outside of angiostain (K1-4) in human plasminogen, displays more potent antiangiogenic activity on endothelial cell proliferation than angiostatin itself. Using a yeast two-hybrid system in vivo, we have recently identified Rgl2 (guanine nucleotide dissociation stimulator (RalGDS)-like factor 2) as a binding protein of human K5. In order to confirm in vitro protein interaction between K5 and Rgl2, we developed bacterial recombinant expression systems for them. K5 and Rgl2 proteins were expressed in high yields and purified into pure forms with His tags and GST fusion, respectively. GST-pull down experiments clearly demonstrated that K5 interacts specifically with Rgl2 in vitro. These results indicate that Rgl2 functions as a receptor protein for K5 in vitro as well as in vivo, leading to anti-angiogenesis through regulating Ras signaling pathways.

  • Production machines have been more important, due to quality level of vehicle motor core is getting higher. That is why, to improve assembly fit of tooling and to be emphasized how much moving down caused of deterioration of high speed press, it is also getting more important parts as solution of problems. To analyze how much move based on condition of movement as tooling and high speed press, and to measure how much impact to embossing height caused of changing movement down. As the result of investigation, in case of material thickness 0.5mm, there is highest pull and force power when emboss height is 0.45mm. If emboss height is less than 0.45mm, pull and force power is getting lower, if emboss height is higher than 0.45mm, it is impossible to make it forming caused of changed press movement, also it has been piercing.

E-business 웹사이트에서의 데이터 입력디자인에 관한 비교 연구 (A Comparative Study on Data Input Design of E-business Websites)

  • 본 연구를 통해 e-business 웹사이트에서 사용자의 입력에 사용되는 어느 정도 표준화된 인터페이스 디자인들을 실험을 통해 비교하고 최적의 사용법을 알아내었다. 풀다운 메뉴, 텍스트 입력 창, 리스트, 라디오 버튼 등의 입력 디자인(도구)들이 실험에 사용되었으며 이들은 호텔 객실의 예약 웹사이트 시뮬레이션을 통해 비교되었다. 실험 결과 사용자의 입력 선택 사양이 4가지 이상인 경우 전문가에겐 텍스트 입력 창이 입력시간을 줄여주고 일반 사용자에게는 풀다운 메뉴가 사용성 측면에서 효율적임을 알 수 있었다. 단지 두 가지의 선택 사양이 존재할 경우엔 만족도, 유연성, 단순성을 고려했을 땐 리스트가 우수하며 사용 편의성 면에선 라디오 버튼이 최적의 인터페이스로 나타났다. 연구 결과를 사용자의 데이터 입력이 필요한 인터렉티브한 웹사이트의 디자인에 적용할 경우 경제적 효율과 사용성을 증대시킬 것이다.

기피-유인을 이용한 바퀴의 효율적 방제 (Effective Control in Managing German Cockroach, Blattella germanica (Orthoptera: Blatellidae) Using a Push-Pull Strategy)

  • 제한된 주거공간에서 바퀴의 방제효과를 높이기 위하여 한쪽에 기피성분(push)을 처리하고 반대쪽에 일반적인 유인성분(pull)이 포함된 독먹이제(bait)를 처리함으로서 행동자극에 의한 기피-유인 방제효과를 검증하였다. 먼저 상형장치를 이용한 mini-field 시험에서, 바퀴는 기피제형이 처리된 장소로부터 벗어나서 유인 먹이가 처리된 곳으로 몰려들었으며, 처리된 유인먹이와 독먹이의 소비량은 기피-유인 효과에 의해 밀집된 곳의 먹이에서 높게 나타났다. 따라서 바퀴에 대하여 기피 효과가 우수한 식물정유를 선별 적용함으로서 살충효과를 높이기 위하여 Citrus 속의 grapefruit, lemon, lime, orange, 그리고 petitgrain 오일에 대한 기피-유인 살충효과를 평가하였다. 동시 처리된 기피-유인 처리는 유인 독먹이만 처리된 대조구에 비해 더 빠르고 높은 살충효과를 나타냈으며, Citrus 속 오일의 기피에 의한 살충효과는 grapefruit > lemon > lime > orange > petitgrain 오일 순으로 나타났다. 따라서 본 실험은 제한된 공간에서 기피제로서 Citrus속 오일과 기피-유인 방법을 이용하는 효율적인 방제법을 제시한다.

경영정보학의 학문적 위상에 관한 연구 (A Study on the Scientific Status of MIS)

  • The inability of the management information systems (MIS) field to progress as a scientific discipline has been attributed to the lack of systematic research and a cumulative tradition, an identity crisis, and the poverty of scientism. While research on the status of MIS is very important in order to enhance the field as a scientific discipline, few have investigated this issue. Following Thomas Kuhn's idea of paradigm, this paper studies other fields to investigate when they progressed, when they did not, and why. After research paradigm was broken down into technology-push and demand-pull types, a model on the science life cycle was developed in an effort to explain the path how a science has progressed. A test of this model in the fields of physics and chemistry with an old historial background reveals that the scientific progress in the area of demand-pull is more desirable if this progress turns out to be in the right direction. An application of the model to the MIS field shows that the research paradigm in this field is mainly of technology-push. In order to shift this paradigm toward the demand-pull area, this paper suggests the research on the relationship among MIS subfields and the adoption of appropriate reference disciplines.

심전계(ECG)의 제작설계

  • Laboratory information system (LIS) is a key tool to manage laboratory data in clinical pathology. Our department has developed an information system for routine hematology using down-sized computer system. We have used an IBM 486 compatible PC with 16MB main memory, 210 MB hard disk drive, 9 RS-232C port and 24 pin dot printer. The operating system and database management system were SCO UNIX and SCO foxbase, respectively. For program development, we used Xbase language provided by SCO foxbase. The C language was used for interface purpose. To make the system use friendly, pull-down menu was used. The system connected to our hospital information system via application program interface (API), so the information related to patient and request details is automatically transmitted to our computer. Our system interfaced with fwd complete blood count analyzers(Sysmex NE-8000 and Coulter STKS) for unidirectional data tansmission from analyzer to computer. The authors suggests that this system based on down-sized computer could provide a progressive approach to total LIS based on local area network, and the implemented system could serve as a model for other hospital's LIS for routine hematology.
