  • A combination of computer codes such as LATREP, HWR, AXAV and CITATION is utilized in an attempt to analyze the nuclear design characteristics of the CANDU-PHWR of the Wolsung Unit 1. The major nuclear properties to be computed are tile lattice properties of CANDU fuel channel and the core channel power distribution. The computed results are compared with the PSR documentation for the Wolsung reactor. The observed discrepancies between our computation and the PSB values are discussed in terms of incomplete information on the description of the core configuration in the PSR and the different calculation methods.

Isolation and Identification of Photosynthetic Bacterium Useful for Wastewater Treatment

  • For wastewater treatment and utilization of the biomass, a photosynthetic bacterium was isolated based on its cell growth rate, cell mass, and assimilating ability of organic acids. The isolate was a Gram-negative rod-shaped bacterium that contained a single polar flagellum and formed a lamellar intracytoplasmic membrane (ICM) system, including bacteriochlorophyll $\alpha$. The major isoprenoid quinone component was identified as ubiquinone Q-10, and the fatty acid composition was characterized as to contain relatively large amount of C-16:0 (18.74%) and C-18:1 (59.23%). Based on its morphology, phototrophic properties, quinone component, and fatty acid composition, the isolate appeared to be closely related to the Rhodopseudomonas subgroup of purple nonsulfur bacteria. A phylogenetic analysis of the isolate using its 16S rRNA gene sequence data also supported the phenotypic findings, and classified the isolate closely related to Rhodopseudomonas palustris. Accordingly, the nomenclature of the isolate was proposed as Rhodopseudomonas palustris KUGB306. A bench-scale photosynthetic bacteria (PSB) reactor using the isolate was designed and operated for the treatment of soybean curd wastewater.

Complete Genome Sequences of Crepidiastrum denticulatum (Asteraceae)

  • The genus Crepidiastrum (Asteraceae), containing ca. 20 species, is mainly distributed in Asia. Crepidiastrum denticulatum, an edible plant that commonly call "e-go-deulppae-gi" in Korean, distributes in Korea, Japan, and China. The complete chloroplast (cp) genome sequences of C. denticulatum was characterized from MiSeq2000 (Illumina Co.) pair-end sequencing data. The cp genome of C. denticulatum has a total sequence length of 152,689 bp and show a typical quadripartite structure. It consists of the large single copy (LSC: 84,022 bp), small single copy (SSC: 18,519 bp), separated by a pair of inverted repeats (IRs: 25,074 bp) and contains 110 unique genes and 18 genes duplicated in the IR regions. Our comparative analysis identified three cpDNA regions (matK, rbcL, and psbA-trnH) from three Crepidiastrum species, which may be useful for molecular identification of each species, and providing a guideline for its clear confirming about dried medical herb.

Comparison of Common Enrichment Methods for Recovery of Yersinia Enterocolitica from Artificially Inoculated Swine Feed Samples

  • Five different enrichment methods were studied to find an optimal method to recover Yersinia enterocolitica from swine feed samples. When the recovery of Y. enterocolitica GER-C (serotype O:3) strain was studied at 1000 CFU/g feed, phosphate-buffered saline (PBS) enrichment at $4^{\circ}C$ and PBS plus sorbitol and bile salts (PSB) enrichment at $4^{\circ}C$ and $21^{\circ}C$ were not effective (< 22%). In contrast, both irgasan-ticarcillin-potassium chlorate (ITC) and tryptic soy broth plus polymyxin B sulfate and novobiocin (TSBPN) enrichment methods showed a full recovery (100%) at 100-1000 CFU/g feed. At 10 CFU/g feed, both ITC and TSBPN methods still recovered the strain (> 50%). In recovery of ATCC 9610 (serotype O:8) strain, TSBPN method was more sensitive than any other methods (P < 0.05) at 1000 CFU/g feed. Using TSBPN method, the strain was still recovered at 100 CFU/g feed, but not at 10 CFU/g feed. With its sensitivity and relatively simple recipe, TSBPN was most desirable method to recover Y. enterocolitica from swine feed samples.

Co-inoculation of Burkholderia cepacia and Alcaligenes aquatilis enhances plant growth of maize (Zea mays) under green house and field condition

  • The synergistic effect on phosphate solubilization of single- and co-inoculation of two phosphate solubilizing bacteria, Burkholderia cepacia (C1) and Alcaligenes aquatilis (H6), was assessed in liquid medium and maize plants. Co-inoculation of two strains was found to release the highest content of soluble phosphorus (309.66 ?g/mL) into the medium, followed by single inoculation of B. cepacia (305.49 ?g/mL) and A. aquatilis strain (282.38 ?g/mL). Based on a plant growth promotion bioassay, co-inoculated maize seedlings showed significant increases in shoot height (75%), shoot fresh weight (93.10%), shoot dry weight (84.99%), root maximum length (55.95%), root fresh weight (66.66%), root dry weight (275%), and maximum leaf length (81.53%), compared to the uninoculated control. In a field experiment, co-inoculated maize seedlings showed significant increases in cob length (136.92%), number of grain/cob (46.68%), and grain weight (67.46%) over control. In addition, single inoculation of maize seedlings also showed improved result over control. However, there was no significant difference between single inoculation of either bacterial strains and co-inoculation of these two bacterial strains in terms of phosphate solubilization index, phosphorous release, pH of the media, and plant growth parameters. Thus, single inoculation and co-inoculation of these bacteria could be used as biofertilizer for improving maize growth and yield.

Taxonomy and phylogeny of the genus Cryptomonas (Cryptophyceae, Cryptophyta) from Korea

  • The genus Cryptomonas is easily recognized by having two flagella, green brownish color, and a swaying behavior. They have relatively simple morphology, and limited diagnostic characters, which present a major difficulty in differentiating between species of the genus. To understand species delineation and phylogenetic relationships among Cryptomonas species, the nuclear-encoded internal transcribed spacer 2 (ITS2), partial large subunit (LSU) and small subunit ribosomal DNA (rDNA), and chloroplast-encoded psbA and LSU rDNA sequences were determined and used for phylogenetic analyses, using Bayesian and maximum likelihood methods. In addition, nuclear-encoded ITS2 sequences were predicted to secondary structures, and were used to determine nine species and four unidentified species from 47 strains. Sequences of helix I, II, and IIIb in ITS2 secondary structure were very useful for the identification of Cryptomonas species. However, the helix IV was the most variable region across species in alignment. The phylogenetic tree showed that fourteen species were monophyletic. However, some strains of C. obovata had chloroplasts with pyrenoid while others were without pyrenoid, which used as a key character in few species. Therefore, classification systems depending solely on morphological characters are inadequate, and require the use of molecular data.

Assessment of the macroalgal diversity of Kuwait by using the Germling Emergence Method

  • Cryptic stages of diverse macroalgae present in natural substrata, "the bank of microscopic forms", were isolated into clonal cultures and identified based on both morphological characteristics and DNA barcoding. Approximately 120 clonal isolates from 308 natural substratum samples were collected from the entire coastline of Kuwait. Amongst these isolates, 77 (64%) were identified through DNA barcoding using the nuclear ribosomal small subunit, RuBisCO spacer (ITS2, tufa, rbcL, psaA, and psbA) and sequencing. Twenty-six isolates (34%) were identified in the division Chlorophyta, 18 (23%) as Phaeophyceae, and 33 (43%) as Rhodophyta. For all DNA sequences in this study, species-level cut off applied was ≥98% homology which depend entirely on the markers used. Three putative new records of Chlorophyta new for the Arabian Gulf were made: Cladophora laetevirens (Dillwyn) Kützing, Ulva torta (Mertens) Trevisan and Ulvella leptochaete (Huber) R. Nielsen, C. J. O'Kelly & B. Wysor in Nielsen, while Cladophora gracilis Kützing and Ulva ohnoi M. Hiraoka & S. Shimada are new records for Kuwait. For Phaeophyceae, Ectocarpus subulatus Kützing and Elachista stellaris Areschoug were new records for the Gulf and Kuwait. In the Rhodophyta, Acrochaetium secundatum (Lyngbye) Nägeli in Nägeli & Cramer, Ceramium affine Setchell & N. L. Gardner, Gelidium pusillum var. pakistanicum Afaq-Husain & Shameel and Dasya caraibica Børgesen are new records for the Gulf and Kuwait, while the red alga Stylonema alsidii (Zanardini) K. Drew is a new record for Kuwait. Several isolates identified corresponded to genera not previously reported in Kuwait and / or the Arabian Gulf, such as Porphyrostromium Trevisan, a new genus from the Bangiales, and two unidentified species for the Planophilaceae Škaloud & Leliaert. The isolates cultivated from substrata enhance understanding of the marine macroalgal diversity in the region and confirmed that the Germling Emergence Method is suitable for determining the actual diversity of a given study area through isolation from cryptic life-history phases.

Effect of Conidial Number and Nutrition on the Germination of Conidia in Colletotrichum dematium f. sp. capsicum Causing Red Pepper Anthracnose (분생포자수(分生胞子數) 및 영양상태(營養狀態)가 고추 탄저병균(炭疽病菌)(Colletotrichum dematium f. sp. capsicum)의 분생포자(分生胞子) 발아(發芽)에 미치는 영향(影響))

  • Exogenous factor and nutrients affecting for conidial germination of Colletetrichum dematium f. sp. capsicum causing red pepper anthracnose were studied by slide germination test. Optimum temperature of conidial germination was at $28^{\circ}C$, ranging 15 to $35^{\circ}C$. Optimum pH was at 5.5, ranging 4.5 to 8.0, and more than 90% of relative humidity (RH) was optimum. Poor conidial germination of the fungus was observed on sterile distilled water, but potato sucrose broth (PSB), red pepper fruit broth (RPFB), green pepper fruit broth (GPFB) and pepper leaf broth (PLB) furnished a satisfactory nutrients for conidial germination. Exogenous supply of carbon and nitrogen sources were essential for conidial germination, while potassium, phosphorous and sulfur were not evident as that for carbon and nitrogen. Soluble starch was the most suitable as a carbon source for conidial germination and followed by D-glucose, D-galactose and lactose in that order. Maximum germination was attained in the $1{\times}10^4$ conidia per ml. Germination was decreased with increment of conidial concentration, and in the density of $5{\times}10^4$ conida per ml, germination was nearly supipressed. It suggested existing a self-inhibitor. Non-washed conidia germinated more than washed conidia, and conidial germination was also gradually decreased by increasing conidial density.

Development of Primer Sets for the Detection of Polygonum multiflorum, Cynanchum wilfordii and C. auriculatum (유전자 마커를 이용한 하수오, 백수오 및 이엽우피소 종 판별법 개발)

  • The aim of this study was to develop rapid screening method for the identification of Chinese herbal medicine species with similar appearance, Polygonum multiflorum, Cynanchum wilfordii and C. auriculatum, by using genetic markers. As a genetic marker, psbA-trnH gene in chloroplast was selected due to differences in sequence among the three species. Species-specific primers were designed based on the sequences of the marker gene of P. multiflorum, C. wilfordii, and C. auriculatum, and the expected size of PCR products was 160, 147, and 119 bp, respectively. Under the developed conditions, cross-reaction was not detected among these three plant species. To confirm the efficiency of our species-specific primers, the optimized method was applied to a variety of processed products composed of mostly P. multiflorum and C. wilfordii, demonstrating that our method was a rapid and easy screening assay. Our findings suggest this screening method can be utilized to prevent the distribution of economically motivated adulteration food and to improve consumer's right.

칼슘락테이트가 반죽발효와 빵의 품질 및 저장성에 미치는 영향

  • 다슬기분말(PSB)과 그 회분(ASB)으로 제조한 칼슘락테이트(PCaL 및 ACaL)를 0.5%씩 첨가한 반죽의 발효와 빵의 품질 및 저장성에 미치는 영향을 조사하였다. 반죽의 pH는 4.85~4.98로 ACaL.PCaL.대조군의 순으로 나타났다. 반죽의 부피와 빵의 loaf volume index는 대조군이 높았고 ACaL 첨가군이 낮았으며, pH를 5.50으로 조정하여 제조한 반죽의 부피와 빵의 loaf volume은 대조군과 큰 차이를 보이지 않았다. PCaL 및 ACaL을 첨가한 빵의 Ca함량은 29.4~29.7 mg/100 g-f.w로 대조군의 13.0 mg/100 g-f.w에 비하여 높았으며, 첨가군의 미량 무기질로 Mg, Fe, Zn이 0.03~0.98 mg/100 g-f.w 범위로 검출되었다. 빵의 L$^{*}$ 값은 대조군과 실험군의 유의적인 차이가 없었으며, a*값, b*값은 PCaL 첨가군이 가장 높아 황갈색을 띄었다. 빵의 hardness, gumminess는 대조군$^{\circ}C$ 실온에 두면서 저장한 결과 대조군은 3일째부터, ACaL 첨가군은 5일째부터, PCaL 첨가군은 6일째부터 곰팡이가 번식하였다.
