Effects of Hepatocyte Growth Factor on the PSA Signaling Pathway of U-251-MG Cells (U-251-MG 세포에서 PSA 경로에 작용하는 Hepatocyte Growth Factor의 효과)

  • Hepatocyte growth factor (HGF) and its receptor play an important role in the formation and progression of glioma. In this study, I investigated the ability of HGF to recover of the PSA siRNA-suppressed cell proliferation, migration and invasion in U-251-MG cells. PSA siRNA-transfected U-251-MG cells showed the reduction of the proliferation, migration and invasion with compared to control. Treatment of HGF on the PSA siRNA-transfected U-251-MG cells recovered the ability of proliferation, migration and invasion. These data suggest that PSA and HGF may use unique and parallel signaling cascade leading to the proliferative, migrative and invasive phenotype of U-251-MG cells. I also showed that PSA cooperated with HGF to a migrative and invasive phenotype via the increased secretion of matrix metalloproteinase-2 (MMP-2) and MMP-9.

The Effect of Plantaginis Semen Aquacupuncture on CCl4 Induced Hepatoxicity in Rats (CCl4로 유발(誘發)한 백서(白鼠) 간손상(肝損傷)에 미치는 차전자약침(車前子藥鍼)의 영향(影響))

  • Objective : This study was undertaken to determine if Plantaginis Semen Aquacupuncture(PSA) has a protective effect against $CCl_4$ induced Hepatoxicity in Rats. Methods : The experimental group were divided into Normal group(untreated group), Control group, PSA group. Rats were administered orally $CCl_4$(0.1 ml/kg) for 4 days. In experiments for PSA effect, rats received 0.1 ml of PSA extraction in both sides of corresponding $G{\bar{a}}nsh{\bar{u}}$(BL18) of human body for 3 days after treated $CCl_4$. Variation of weight and biochemical assays(GOT, GPT, LDH, ALP, total cholesterol, triglyceride, albumin) were performed. Results : In Control group, $CCl_4$ increased serum GOT, GPT, LDH, ALP and albumin and decreased weight, total cholesterol and triglyceride. PSA significantly decreased serum GOT, GPT, LDH and increased total cholesterol as compared with Control group. Conclusion : These results indicate that PSA could be used in prevention and treatment of hepatoxicity. However, precise mechanisms of PSA protection remain to be determined.

  • Paper industry discharges many by-products and quantity of PSA (Paper sludge ash) has been increased. In this study, hydration water was added to PSA for use as cement admixture. PSA with added water was mixed with anhydrite and this mixture was used as cement substitute. Physical properties of PSA cement were changed by contents of PSA, but PSA cement containing PSA less than 10% had similar properties to those of OPC. Compressive strength of PSA cement mortar had a certain relationship with $Ca(OH)_2$ content. Compressive strength at 3 days increased, as $Ca(OH)_2$ content increased. However, the strength at 28 days increased, as $Ca(OH)_2$ content decreased.

The Relationship Between Levels of Serum Testosterone and Prostate-specific Antigen in Healthy Men: An Integrative Review (성인 남성의 혈청 테스토스테론 농도와 전립선 특이항원의 관계에 대한 통합적 고찰)

  • Purpose: The purpose of this study was to review primary research exploring the correlations between the levels of serum testosterone and the prostate-specific antigen (PSA) in healthy men without prostate diseases. Methods: An integrative review was conducted using the Whittemore & Knafle (2005) framework. The keywords, 'testosterone & prostate-specific antigen', 'testosterone & PSA' and 'healthy men' were used to search peer-reviewed publications in six databases. Among 1,959 searched articles, eleven articles were selected after excluding articles that do not meet inclusion criteria. Literature quality was moderate (Level 3). Results: As a result of this study, it was confirmed through the nine articles that healthy adult men showed no significant correlation between the serum testosterone and the PSA. Conversely, two articles presented that the serum PSA correlate positively with the testosterone. In particular, it is inferred that the effect of the serum testosterone and the PSA secreted into a 24-hour circadian rhythm with different amplitudes and slopes would have had great influence. However, it does not consider the factors affecting the testosterone and the PSA, such as race, liver disease, and BMI, so there is insufficient empirical data to clearly explain the relationship between the testosterone and the PSA. Conclusion: The correlation between the serum testosterone and the PSA in healthy adult men is insignificant in relation to the circadian rhythm of the testosterone and the PSA secretion. It is that a large-scale study including various influencing factors using new biochemical indicators such as pro PSA be conducted in the future.

Remaining and emerging issues pertaining to the human reliability analysis of domestic nuclear power plants

  • Probabilistic safety assessments (PSA) have been used for several decades to visualize the risk level of commercial nuclear power plants (NPPs). Since the role of a human reliability analysis (HRA) is to provide human error probabilities for safety critical tasks to support PSA, PSA quality is strongly affected by HRA quality. Therefore, it is important to understand the underlying limitations or problems of HRA techniques. For this reason, this study conducted a survey among 14 subject matter experts who represent the HRA community of domestic Korean NPPs. As a result, five significant HRA issues were identified: (1) providing a technical basis for the K-HRA (Korean HRA) method, and developing dedicated HRA methods applicable to (2) diverse external events to support Level 1 PSA, (3) digital environments, (4) mobile equipment, and (5) severe accident management guideline tasks to support Level 2 PSA. In addition, an HRA method to support multi-unit PSA was emphasized because it plays an important role in the evaluation of site risk, which is one of the hottest current issues. It is believed that creating such a catalog of prioritized issues will be a good indication of research direction to improve HRA and therefore PSA quality.

  • This paper describes the development process, the innovative techniques used and insights gained from the latest integrated, full scope, multistate Level 2 PSA analysis conducted at the Leibstadt Nuclear Power Plant (KKL), Switzerland. KKL is a modern single-unit General Electric Boiling Water Reactor (BWR/6) with Mark III Containment, and a power output of $3600MW_{th}/1200MW_e$, the highest among the five operating reactors in Switzerland. A Level 2 Probabilistic Safety Assessment (PSA) analyses accident phenomena in nuclear power plants, identifies ways in which radioactive releases from plants can occur and estimates release pathways, magnitude and frequency. This paper attempts to give an overview of the advanced modeling techniques that have been developed and implemented for the recent KKL Level 2 PSA update, with the aim of systematizing the analysis and modeling processes, as well as complying with the relatively prescriptive Swiss requirements for PSA. The analysis provides significant insights into the absolute and relative importances of risk contributors and accident prevention and mitigation measures. Thanks to several newly developed techniques and an integrated approach, the KKL Level 2 PSA report exhibits a high degree of reviewability and maintainability, and transparently highlights the most important risk contributors to Large Early Release Frequency (LERF) with respect to initiating events, components, operator actions or seismic component failure probabilities (fragilities).

Practical modeling and quantification of a single-top fire events probabilistic safety assessment model

  • In general, an internal fire events probabilistic safety assessment (PSA) model is quantified by modifying the pre-existing internal event PSA model. Because many pieces of equipment or cables can be damaged by a fire, a single fire event can lead to multiple internal events PSA initiating events (IEs). Consequently, when the fire events PSA model is quantified, inappropriate minimal cut sets (MCSs), such as duplicate MCSs, may be generated. This paper shows that single quantification of a hypothetical single-top fire event PSA model may generate the following four types of inappropriate MCSs: duplicate MCSs, MCSs subsumed by other MCSs, nonsense MCSs, and MCSs with over-counted fire frequencies. Among the inappropriate MCSs, the nonsense MCSs should be addressed first because they can interfere with the right interpretation of the other MCSs and prevent the resolution of the issues related to the other inappropriate MCSs. In addition, we propose a resolution process for each of the issues caused by these inappropriate MCSs and suggest an overall procedure for resolving them. The results of this study will contribute to the understanding and resolution of the inappropriate MCSs that may appear in the quantification of fire events PSA models.

Vital Area Identification of Nuclear Facilities by using PSA (PSA기법을 이용한 원자력시설의 핵심구역 파악)

  • The urgent VAI method development is required since "The Act of Physical Protection and Radiological Emergency that is established in 2003" requires an evaluation of physical threats in nuclear facilities and an establishment of physical protection in Korea. The VAI methodology is developed to (1) make a sabotage model by reusing existing fire/flooding/pipe break PSA models, (2) calculate MCSs and TEPSs, (3) select the most cost-effective TEPS among many TEPSs, (4) determine the compartments in a selected TEPS as vital areas, and (5) provide protection measures to the vital areas. The developed VAI methodology contains four steps, (1) collecting the internal level 1 PSA model and information, (2) developing the fire/flood/pipe rupture model based on level 1 PSA model, (3) integrating the fire/flood/pipe rupture model into the sabotage model by JSTAR, and (4) calculating MCSs and TEPS. The VAT process is performed through the VIPEX that was developed in KAERI. This methodology serves as a guide to develop a sabotage model by using existing internal and external PSA models. When this methodology is used to identify the vital areas, it provides the most cost-effective method to save the VAI and physical protection costs.

Rigorous dynamic simulation and determination of initial operating conditions for two-bed PSA processes (두 탑 PSA공정의 상세 동적모사 및 초기운전조건 결정)

  • A rigorous dynamic simulation was performed in binary gas mixture H$_{2}$/CO (70:30 vol.%) to determinate start-up operating conditions of PSA(Pressure Swing Adsorption) processes. The rigorous dynamic model for the PSA process contains an Ergun equation for expressing the pressure drop in a bed, and valve equations to compute the boundary pressure change of the bed. As the result of the continuous dynamic simulation of 100 operating cyles in various initial conditions, the unsteady-state appeared in the early period and the cyclic steady-state came out about 20th cycle in feed condition and vaccum condition, and 30th cycle in pure H$_{2}$ condition. As time goes by valve equations made change the pressure at each end of the bed in ressurization, countercurrunt-depressurization and pressure equalization steps. The H$_{2}$ purity and the recovery is 99.99% and 86.73% respectively, which is slightly higher than the experimental data. Main contributiion of this study includes supplying fundamental technologies of handling combined variables PSA processes by developing rigorous models.

A study on the improvement of indefiniteness in PRPDA by use of PSA (PSA를 이용한 PRPDA의 불명확성 개선에 대한 연구)

  • This paper deals with the improvement of indefiniteness in PRPDA(Phase Resolved Partial Discharge Analysis) through analyzing the relation between PRPDA and PSA(Pulse Sequence Analysis). For the analysis we adopted PD data from artificial defects in XLPE HV cable systems and performed PRPDA and PSA. As a results, we confirmed that PSA was more useful than PRPDA in classifying PD patterns with the degradation stages. Therefore, it is possible to improve the indefiniteness of PD pattern recognition using the relation between PRPDA and PSA.
