Computational Analysis of Neighboring Genes on Arabidopsis thaliana Chromosomes 4 and 5: Their Genomic Association as Functional Subunits

  • The genes related to specific events or pathways in bacteria are frequently localized proximate to the genome of their neighbors, as with the structures known as operon, but eukaryotic genes seem to be independent of their neighbors, and are dispersed randomly throughout genomes. Although cases are rare, the findings from structures similar to prokaryotic operons in the nematode genome, and the clustering of housekeeping genes on human genome, lead us to assess the genomic association of genes as functional subunits. We evaluated the genomic association of neighboring genes on chromosomes 4 and 5 of Arabidopsis thaliana with and without respectively consideration of the scaffold/matrix­attached regions (S/MAR) loci. The observed number of functionally identical bigrams and trig rams were significantly higher than expected, and these results were verified statistically by calculating p-values for weighted random distributions. The observed frequency of functionally identical big rams and trig rams were much higher in chromosome 4 than in chromosome 5, but the frequencies with, and without, consideration of the S/MAR in each chromosome were similar. In this study, a genomic association among functionally related neighboring genes in Arabidopsis thaliana was suggested.

국내산 프로폴리스의 화학적 특성에 관한 연구 (Studies on the Chemical Characteristics of Korean Propolis)

  • 본 연구는 국내산 프로폴리스의 화학적 특성을 조사하기 위하여 실시하였다. 위한 기초 자료를 얻고자 실시하였다. 예천과 영월 프로폴리스의 수분, 조단백질, 조지방, 화분, 그리고 탄수화물의 함량은 각각 3.25와 3.97, 9.82와 7.04, 48.25와 50.7, 3.91과 5.89, 그리고 34.77과 32.4%였다. 미네랄에서 Se은 검출이 되지 않았으며, P, Ca 및 Cu은 영월 프로폴리스가 184.15, 128.32, 1.77ppm으로 가장 높았으며, 예천산은 1130.6ppm, 그리고 영월산은 1049.1ppm이었다. 지방산 중 linoleic acid와 linolenic acid는 예천 프로폴리스가 47.6 및 70.0mg/100g이었으며, 영월 프로폴리스는 37.3과 47.0mg/100g이었다.

Improving Nutritional Quality of Cocoa Pod (Theobroma cacao) through Chemical and Biological Treatments for Ruminant Feeding: In vitro and In vivo Evaluation

  • Cocoa pod is among the by-products of cocoa (Theobroma cacao) plantations. The aim of this study was to apply a number of treatments in order to improve nutritional quality of cocoa pod for feeding of ruminants. Cocoa pod was subjected to different treatments, i.e. C (cocoa pod without any treatment or control), CAm (cocoa pod+1.5% urea), CMo (cocoa pod+3% molasses), CRu (cocoa pod+3% rumen content) and CPh (cocoa pod+3% molasses+Phanerochaete chrysosporium inoculum). Analysis of proximate and Van Soest's fiber fraction were performed on the respective treatments. The pods were then subjected to an in vitro digestibility evaluation by incubation in rumen fluid-buffer medium, employing a randomized complete block design (n = 3 replicates). Further, an in vivo evaluation of the pods (35% inclusion level in total mixed ration) was conducted by feeding to young Holstein steers (average body weight of $145{\pm}3.6kg$) with a $5{\times}5$ latin square design arrangement (n = 5 replicates). Each experimental period lasted for 30 d; the first 20 d was for feed adaptation, the next 3 d was for sampling of rumen liquid, and the last 7 d was for measurements of digestibility and N balance. Results revealed that lignin content was reduced significantly when cocoa pod was treated with urea, molasses, rumen content or P. chrysosporium (p<0.01) with the following order of effectiveness: CPh>CAm>CRu>CMo. Among all treatments, CAm and CPh treatments significantly improved the in vitro dry matter and organic matter digestibility (p<0.05) of cocoa pod. Average daily gain of steers receiving CAm or CPh treatment was significantly higher than that of control (p<0.01) with an increase of 105% and 92%, respectively. Such higher daily gain was concomitant with higher N retention and proportion of N retention to N intake in CAm and CPh treatments than those of control (p<0.05). It can be concluded from this study that treatment with either urea or P. chrysosporium is effective in improving the nutritive value of cocoa pod.

온열욕 겸용 숯가마에서 생산된 백탄의 특성 (Characteristics of White Charcoal Produced from the Charcoal Kiln for Thermotherapy)

  • 본 연구에서는 온열욕 겸용을 위해 제작한 숯가마와 전통식 숯가마에서 제조한 백탄의 특성에 대해서 비교, 검토하였다. 온열욕 겸용 숯가마는 밀폐된 숯가마에서 발생하는 미세먼지 및 유해가스와 같은 환경문제를 최소화하고 편안함과 안전성을 고려하여 제작하였다. 온열욕 겸용 숯가마에서 제조한 백탄은 전통식 숯가마의 백탄보다 회분과 휘발분이 다소 높았고 고정탄소가 다소 낮았다. 온열욕 겸용 숯가마에서 생산된 백탄의 밀도는 기존의 전통식 숯가마 백탄보다 약간 높았지만, 평형함수율과 pH는 거의 차이가 없었다. 발열량, 정련도, 경도와 해부학적 구조는 숯가마에 따른 차이가 없었다. 결론적으로 온열욕 겸용을 위해 개량된 숯가마에서 제조한 백탄은 국립산림과학원의 품질인증기준을 충족하였고, 온열욕 겸용 숯가마는 목탄제조에도 적합한 것으로 판단되었다.

Nutritional Evaluation of Distillery Sludge and Its Effect as a Substitute of Canola Meal on Performance of Broiler Chickens

  • The study was conducted to investigate the chemical composition of distillery yeast sludge and its inclusion in broiler diets to replace canola meal. Raw distillery yeast sludge was washed with water using water and sludge in the ratio 6:1, respectively. Proximate analysis of raw distillery yeast sludge and washed distillery sludge was carried out for crude protein (CP), true protein (TP), ether extract (EE), ash, acid insoluble ash and nitrogen free extract (NFE) determination. Mineral contents and amino acid profile of raw distillery yeast sludge and washed distillery sludge were also determined. After chemical evaluation, four iso-caloric and iso-nitrogenous broiler starter and finisher diets were prepared in mash form using 0 (control), 4, 8 and 12% levels of washed distillery sludge replacing canola meal. One hundred and twenty day-old broiler chicks were randomly distributed into 12 experimental units in such a way that each diet was offered to three experimental units, each comprising of 10 chicks. It was observed that washing affected the nutrients either by decreasing or increasing their concentration. It decreased the total mineral contents whereas CP, TP, EE and NFE contents increased. Washing also increased amino acid profile. Average feed intake and weight gain were higher in birds fed diet containing 8% washed distillery sludge and lower in birds fed diet containing 0% washed distillery sludge. Feed cost per kg live weight gain decreased significantly as the level of washed distillery sludge was increased in the diet. Average heart, liver and pancreas weights decreased with increased level of washed distillery sludge in the diet. The study revealed that after washing, distillery yeast sludge can be used successfully in broiler diets up to the level of 8% without any adverse effect on broiler's performance.

Predicting the Digestible Energy of Rapeseed Meal from Its Chemical Composition in Growing-finishing Pigs

  • Two experiments were conducted to establish a digestible energy (DE) content prediction model of rapeseed meal for growing-finishing pig based on rapeseed meal's chemical composition. In experiment 1, observed linear relationships between the determined DE content of 22 rapeseed meal calibration samples and proximate nutrients, gross energy (GE) and neutral detergent fiber (NDF) were used to develop the DE prediction model. In experiment 2, 4 samples of rapeseed meal selected at random from the primary rapeseed growing regions of China were used for testing the accuracy of DE prediction models. The results indicated that the DE was negatively correlated with NDF (r = -0.86) and acid detergent fiber (ADF) (r = -0.73) contents, and moderately correlated with gross energy (GE; r = 0.56) content in rapeseed meal calibration samples. In contrast, no significant correlations were found for crude protein, ether extract, crude fiber and ash contents. According to the regression analysis, NDF or both NDF and GE were found to be useful for the DE prediction models. Two prediction models: DE = 16.775-0.147${\times}$NDF ($R^2$ = 0.73) and DE = 11.848-0.131${\times}$NDF+0.231${\times}$GE ($R^2$ = 0.76) were obtained. The maximum absolute difference between the in vivo DE determinations and the predicted DE values was 0.62 MJ/kg and the relative difference was 5.21%. Therefore, it was concluded that, for growing-finishing pigs, these two prediction models could be used to predict the DE content of rapeseed meal with acceptable accuracy.

Chemical Composition of Thermal Treatment Yam (Dioscorea batatas DECNE.)

  • In this study, the thermal treatment yam (Dioscorea batatas DECNE.) was analyzed for its proximate and nutritional compositions including mineral, vitamin, amino acids and fatty acids. Thermal treatment yam is mainly consisted of carbohydrate (70.3%) with small amounts of crude protein (15.8%), crude ash (5.1%) and crude fat (1.6%). It was found to be a good source of essential minerals such as K (1100.2 mg/100 g), Mg (99.4 mg/100 g), Ca (54.8 mg/100 g) and Na (44.9 mg/100 g) but Zn (4.8 mg/100 g) and Fe (0.2 mg/100g) content was low. Little amounts of vitamin E (8.3 mg/100g), vitamin C (3.5 mg/100g) and vitamin B1 (2.1 mg/100g) could be found. The amino acid analysis revealed that the yam was superior with respect to serine (1454.2 mg%), lysine (684.6 mg%) and histidine (684.6 mg%). Essential amino acids were calculated to be 2849.3 mg%. The amino acid profiles showed that thermal treatment yam is limiting in isoleucine and phenylaline. Linoleic acid was the most predominant fatty acids with the value of 47.3% followed by palmitic acid (24.7%) and oleic acid (12.7%). And the unsaturated fatty acids including oleic acid and linoleic acid were present in big quantities in thermal treatment yam.

Protein quality, hematological properties and nutritional status of albino rats fed complementary foods with fermented popcorn, African locust bean, and bambara groundnut flour blends

  • The objective of this study was to determine protein quality and hematological properties of infant diets formulated from local food materials. The food materials were obtained locally, fermented, and milled into flour. The flours were mixed as 70% popcorn and 30% African locust bean (FPA), 70% popcorn and 30% bambara groundnut (FPB), and 70% popcorn, 20% bambara groundnut, and 10% African locust bean (FPAB). Proximate analysis, protein quality, hematological properties, and anthropometric measurements of the animals fed with the formulations were investigated. The protein contents of the formulated diets were significantly higher than that of Cerelac (a commercial preparation) ($15.75{\pm}0.01g$/100 g) and ogi (traditional complementary food) ($6.52{\pm}0.31g$/100 g). The energy value of FPAB ($464.94{\pm}1.22\;kcal$) was higher than those of FPA ($441.41{\pm}3.05\;kcal$) and FPB ($441.48{\pm}3.05\;kcal$). The biological value (BV) of FPAB (60.20%) was the highest followed by FPB (44.24%) and FPA (41.15%); however, BV of the diets was higher than that of ogi (10.03%) but lower than that of Cerelac (70.43%). Net protein utilization (NPU) of the formulations was 41.16-60.20%, whereas true protein digestibility was 41.05-60.05%. Metabolizable energy (232.98 kcal) and digestible energy (83.69 kcal) of FPAB were the highest, whereas that of FPA had the lowest values. The protein digestibility values corrected for amino acid score of the diets (0.22-0.44) were lower than that of Cerelac (0.52), but higher than that of ogi (0.21). The growth patterns and hematological properties (packed cell volume, red blood cells, hemoglobin, mean corpuscular hemoglobin concentration, mean corpuscular hemoglobin, and mean corpuscular volume) of the formulated diets were higher than those of ogi, but lower than those of Cerelac. In conclusion, we established that the FPAB food sample was rated best in terms of protein quality over the other formulated diets. Therefore, a FPAB blend may be used as a substitute for ogi.

Growth Performance, Carcass Composition and Meat Quality of Jiulong-yak (Bos grunniens)

  • Estimated liveweights, carcass characteristics, the proximate composition and amino-acid composition of the whole rib cut of the 10th/12th thoracic vertebrae have been determined for Jiulong-yak. The animal grows rapidly up to puberty at 2-3 years of age. At those ages, females reached 143.0$\pm$34.7 kg and 184.0$\pm$30.8 kg (SD) respectively, which was 60 to 80% of their mature weight at 6 to 7 yr old, then the rate of growth significantly slowed down. Males continued to rapidly increase body weight until an older age possibly due to the selection procedures. Animals included 20 males aged from 2.5 to 6.5 yr, seven females aged of 4.5 yr and 11 steers aged of 4.5-5.5 yr were slaughtered for carcass determination and chemical analysis of meat samples. Dressing-out percentage and ribeye area ranged from 48.53-55.04% and 48.02-68.56 $cm^2$ respectively, both of which differed by age and sex. Yak meat is scarlet in color and intramuscular marbling is poor. The 10th/12th rib-cut contained 24.99-31.93% dry matter; 19.98-22.58% protein; 2.52-10.86% fat and 0.93-1.00% ash. Meat from females and steers contained a higher percentage of fat than those from bulls. The pH of M. Longissimus dorsi measured at 24 h post-mortem ranged from 5.84-6.11. Amino acid composition was similar to that for other red meat, except for a lower content of methionine (1.26 g/100 g meat). The results indicated that yak meat can make a valuable contribution to the diets for the highlanders.

Effects of Gamma Irradiation on Nutrient Composition, Anti-nutritional Factors, In vitro Digestibility and Ruminal Degradation of Whole Cotton Seed

  • Whole cotton seed (WCS) has become one of the major feed ingredients in TMR for dairy cattle in Korea, and WCS for feed use is mostly imported from abroad. Since this genetically modified oil seed is usually fed to the animal in raw state, its germination ability, if last long, often causes concerns about ecological disturbances. In the process of looking for effective conditions to remove germination ability of WCS this study had the objectives to evaluate the nutritional effects of gamma irradiation at doses of 8, 10 and 12 kGy on changes in nutrient contents, anti-nutritional factors, in vitro digestibility and ruminal degradability. No significant differences were found in proximate analysis of nutrients between raw WCS and gamma irradiated one. Glycine and threonine contents significantly increased when the WCS was exposed to gamma ray as compared to untreated WCS (p<0.05). As for fatty acid composition, no significant differences were observed with the irradiation treatment. Free gossypol in WCS was decreased (p<0.05) by gamma irradiation treatment. Of the 3 different levels of gamma irradiation, a dose of 12 kGy was found to be the most effective in reducing free gossypol concentration. Results obtained from in situ experiment indicated that gamma irradiation at a dose of 10 kGy significantly (p<0.05) lowered rumen degradability of both dry matter and crude protein as compared with raw WCS. However, there were no significant differences in rapidly degradable and potentially degradable fractions of crude protein due to 10 kGy gamma irradiation. Overall, this study show that gamma irradiation at a dose of 10 kGy is the optimum condition for removing germination ability of WCS, and could improve nutritive value for the ruminant with respect to the decrease in both ruminal protein degradability and gossypol content of WCS.
