Application of Response Surface Analysis for Predicting Moisture Content of Binary Mixture (다중 회귀분석에 의한 이상혼합물(二相混合物)의 수분함량 예측)

  • The water sorption isotherms of binary mixtures, prepared by corn starch and isolated soybean protein (ISP) or casein, were measured and analyzed. Simple equations to predict moisture content from knowledge of composition and water activity of the mixture were derived by applying Response Surface Analysis. Comparison between predicted and experimental moisture content for 13 combinations of corn starch-lSP mixture at the range of $a_w$ 0.25-0.87 resulted in a maximum error of only 6.06% and an absolute mean error of 2.60%, and for the mixture of corn starch-casein the error was -4.39% and 2.12%, respectively. The agreement between experimental and predicted water sorption isotherms was shown to be 'highly acceptable' for the binary mixtures of 50% corn starch-50% ISP and 50% corn starch -50% casein.

THE USE OF MULTINUTRIENT BLOCK SUPPLEMENTED TO UREA TREATED RICE STRAW BASE DIET FOR DAIRY HEIFERS

  • Eighteen Holstein Friesian ${\times}$ Native heifers with an average live weight of $175.4{\pm}27.8kg$ were allotted to 3 dietary groups. All animals were kept individually in a stanchion barn with free access to water and urea-treated rice straw (UTS). The supplemented feeds were as follows: Group (Gr) 1 - concentrate mixture (15% CP) at 1% body weight (BW), Gr 2 - concentrate mixture at 0.7% BW + free licking of multinutrient block (MNB), Gr 3 - as Gr 2 + 0.3% BW ground com. Multinutrient block composed of 20% mineral mixture, 10% urea, 18% molasses, 20% soybean meal, 22% sesame meal, 10% cement, 0.15 million IU and 50 IU/kg MNB of vitamin A and E. The experimental period lasted 12 weeks. The result revealed that MNB enhanced intake of UTS and total dry matter intake. Animals in Gr 3 consumed a higher amount of feed than the other 2 groups, thus resulting in the highest live weight gain and tended to possess the best feed conversion ratio. Heifers in Gr 2 also performed better than Gr 1 although the difference was not significant. Average MNB intake found to be 0.65 kg/day. Feed cost per kg gain was not significantly different amon groups. However if the cost of MNB could be lower, it would benefit the production cost of animals. The advantage of MNB is the safety of using a high level of urea. Although it was supplemented to UTS which also contained non-protein nitrogen (NPN), no toxic sign was shown. The results indicated that MNB could be partially substituted to concentrate mixture and it was even better when used incombination with soluble carbohydrate feed.

The Effects of Dietary Enzyme Mixture Reinforced with ${\beta}$-Glucanase Activity on Mini Production and the Change of Somatic Cell Count in Lactating Dairy Cows (사료내 ${\beta}$-glucanase 활성 강화 고역가 복합효소제 첨가급여가 착유우의 유생산 및 체세포수 변화에 미치는 영향)

  • In recent years, many researches are actively undertaken for environmental-friendly animal production according to the increased understanding about food safety because of the outbreak of various diseases such as mad cow disease, Foot and mouth disease and Poultry Influenza virus. However, high quality(higher safety)- animal production may not be successful without increasing of disease resistance of animal and the improvement of feeding environment. To increase the disease resistance is able to be accomplished by stimulating the immune function. The present study was undertaken to investigate the effects of enzyme mixture reinforced with ${\beta}$-glucanase activity which degrade polysaccharide to release ${\beta}$-glucan known as stimulator of immune function on the change of milk production and somatic cell count. After 12weeks of experimental feeding, milk production tended to be increased and somatic cell count was decreased from average $227{\times}10^4$ to $37.1{\times}10^4$. Milk protein and solid-fat content were tended to increase but milk fat showed decreasing tendency by the feeding of enzyme mixture. All together, it has been suggest6d that the improvement of high quality milk production may be possible through the dietary addition of immune modulating enzyme mixture in lactating dairy cows.

Stability of Casein-Pectin Mixtures in Apple Juice (사과주스에서 카제인-펙틴 혼합물의 안정성)

  • The effect of addition of pectin on the stability of casein solution was studied to apply casein-pectin mixture to apple juice. The solubility of 0.1% casein solution was below 20% at pH 3-5. However, the solubility of 0.1% casein-0.1% pectin mixtures was over 70% at pH 2-10. The increase in the concentration of casein-pectin mixture showed adverse effect on the solubility. The apple juice (pH 3.4), containing 0.1-0.5% casein-pectin mixtures, remained stable without the precipitation of casein. The stability of apple juice including casein-pectin mixture was maintained upon heating at $100^{\circ}C$ for 10 minutes or refrigerating for a week. This study suggested the possibility of protein fortification to acidic beverages since casein-pectin mixture maintained stability in apple juice.

Dietary encapsulated essential oil mixture influence on apparent nutrient digestibility, serum metabolic profile, lymphocyte histochemistry and intestinal morphology of laying hens

  • Objective: The study aimed to evaluate the effects of a mixture of encapsulated essential oils (EOs) addition on nutrient digestion, serum biochemical parameters, peripheral blood alpha-naphthyl acetate esterase (ANAE), and acid phosphatase (ACP-ase) positive lymphocyte ratios and intestinal morphology in laying hens. Methods: A total of 320 laying hens of 48-wk-old were randomly allotted into 4 treatment groups with 10 replicates of 8 birds in each replicate. The birds were fed a basal diet (control) or the diet added with mixture of EOs (which consist of eugenol, nerolidol, piperine, thymol, linalool, and geraniol) at 50, 100, and 200 mg/kg for period of 84 days. Results: The addition of EOs at 100 or 200 mg/kg increased the dry matter, organic matter, and crude protein digestion as compared to control. The addition of all doses of EOs did not affect serum gamma glutamyl transferase, alanine aminotransferase, and P but increased serum asparate aminotransferase (AST) concentration. The addition of 200 mg/kg EOs increased serum creatinine, while 100 mg/kg decreased Ca concentration. The addition of 100 and 200 mg/kg EOs generally improved ANAE and ACP-ase positive peripheral blood lymphocyte ratios and intestinal morphology. Conclusion: It can be concluded that, the addition of 100 or 200 mg/kg encapsulated EOs generally increased apparent nutrient digestion and serum AST concentration, improved ANAE and ACP-ase positive peripheral blood lymphocytes and intestinal morphology in laying hens.

Stability of Protein Colloids in the Mixture of Cheese Whey and Soy Milk (유청(乳淸) 및 두유(豆乳) 혼합액에서의 단백질 콜로이드 안정성)

  • To investigate effectors on the colloidal stability of whey and soybean proteins, characteristics of tofu-gel formation, effects of heat treatment and salt composition on the colloidal stability, and effects of heat treatment on storage stability were analyzed. When experimental tofus were made from the mixture of whey and soybean, the calcium in the whey precipitated the soy proteins, and disrupted the gel formation, which resulted in the curd of poor texture. In the heat treatment at $60{\sim}100^{\circ}C$, whey and the whey proteins dialyzed against distilled water were readily preciptated at over $70^{\circ}C$, but the mixture of whey and soy extract as well as soy extract were stable at the range of temperature. The proteins of soy extract, whey dialyzed against sodium phosphate buffer, and the mixture were stable at the same heat treatment, and this suggested that phosphates in the soy extract stabilize specialty the whey proteins. Soy proteins were easily destabilized by adding $CaCl_2(0.05{\sim}0.07M)$ at $80{\circ}C$ and $70{\sim}85%$ of the proteins in soy extract and the mixture were preciptated, while only $30{\sim}55%$ of the proteins in whey dialyzed against distilled water were destabilized at the same conditions. Storage stability at $4^{\circ}C$ of the mixture was increased when the mixture was treated at $63^{\circ}C$ and lower temperature.

A Study of the Additional Toxicity of Mixed Food Additives to Rat (혼합(混合)된 식품첨가물(食品添加物)이 흰쥐의 생리(生理)에 미치는 상승적(相乘的) 독성(毒性) 효과(效果)에 관(關)한 연구(硏究))

  • To improve the food qualities in Korea, two hundred and fourtynine kinds of food additives have been allowed in food processing, of which one hundred and nineteen kinds could be used under the limitted conditions. Hence, in practical uses in food processing, many kinds of them are mixed at random within the permitted amounts for their special purposes. For last several years, many kinds of the food additives were prohibited because they have been proved to be toxic even with the single dose. Until recently a few studies on the toxicity in the mixture of food additives were reported, however, they were shown to be no severe additional effect on the animal. This study was performed to see if any elevation of chronic or subacute toxicity of food additives occur especially when they are mixed with each other, using three kinds of food additives (DHA, AF-2, BHT) most widely used as food preservatives, antiseptics and antioxidants. One hundred and fifty young male rats were taken and divided into ten feeding groups, one first control group (food additives blank), three second control groups (DHA 0.1%, AF-2 0.1%, BHT 0.5%), three mixture groups of low level (mixture of each 60% of two second control level) and three mixture groups of high level (mixture of each 90% of two second control level). As the methods of biological and clinical tests, the change of body weight (growth rate), daily intake of diets, organ to body weight ratio, histopathological findings of organs, hematological observation, liver and kidney function tests were checked three times during the periods of 24 weeks. The following results were obtained. 1. The low level group of DHA, AF-2 mixture and DHA, BHT mixture revealed a little retardation in growth rate than the first control group, however, they were similar to the second controls, while all the mixture groups of high level showed a more remarkable retardation than the first and second controls. 2. Average daily intake of the diets was the same in each group, showing a similar decreasing tendency (70-100g/kg of body weight) in accordance with the growth rate. It was observed that there are no differences in the taste and appetite in each group of rats. 3. Abnormal enlargements of kidney and lung were not seen in all the mixture groups compared with the controls, while a slight hepatomegaly was observed in all mixture groups of low level as in the second controls. Significant differences (almost 1% level) were observed between the high level groups and the first control group. 4. Histopathological effects of the food additives on lung, kidney and liver tissues were found in all mixture group of high level. The less frequencies of the same effects were also seen in the low level groups. 5. The esterified cholesterol to total cholesterol ratio in the mixture groups of high level showed a little lower values, and the activities of serum glutamate oxaloacetate transaminase and alkaline phosphatase decreased almost with significance of 5% level compared with the first control group. The serum A/G ratio in the mixture groups also decreased. The results demonstrated that the liver function was decreased in the mixture groups compared with the controls. 6. In all groups throughout the test period, kidney functions (concentration of protein and creatinine excreated per hour in urine and renal filtration rate) were shown almost as normal as the first control. 7. Average values of hematocrit, erythrocytes and leucocytes in the mixture groups were in the normal ranges as in the controls, which denotes that the production of blood cells in bone marrow were also normal in all groups. With the above results, it is concluded that when the food additives (DHA, AF-2, BHT) were given together to the rats in several combinations even in less amount, they showed more toxic signs than the single doses.

Green Composites. II. Environment-friendly, Biodegradable Composites Using Ramie Fibers and Soy Protein Concentrate (SPC) Resin

  • Fully biodegradable and environment-friendly green composite specimens were made using ramie fibers and soy protein concentrate (SPC) resin. SPC was used as continuous phase resin in green composites. The SPC resin was plasticized with glycerin. Precuring and curing processes for the resin were optimized to obtain required mechanical properties. Unidirectional green composites were prepared by combining 65% (on weight basis) ramie fibers and SPC resin. The tensile strength and Young's modulus of these composites were significantly higher compared to those of pure SPC resin. Tensile and flexural properties of the composite in the longitudinal direction were moderate and found to be significantly higher than those of three common wood varieties. In the transverse direction, however, their properties were comparable with those of wood specimens. Scanning electron microscope (SEM) micrographs of the tensile fracture surfaces of the green composite indicated good interfacial bonding between ramie fibers and SPC resin. Theoretical values for tensile strength and Young's modulus, calculated using simple rule of mixture were higher than the experimentally obtained values. The main reasons for this discrepancy are loss of fiber alignment, voids and fiber compression due to resin shrinking during curing.

Textural and Organoleptic Properties of Tofu Manufactured with Micronized Full-fat Soyflour Fortified with Food Ingredients

  • Textural properties of tofu manufactured with micronized full-fat soyflour (MFS) were enhanced by the addition of soy protein isolate, whey protein concentrate, chitosan oligosaccharide and mushroom powder. The MFS solution (14.2% solid content) was converted to semi-solid tofu by a two-stage heat treatment with the addition of 4% coagulant mix. The MFS tofu was evaluated by a compression test as well as sensory evaluation. To produce the semi-solid gel (MFS tofu) with reasonably high strength and toughness, the MFS solution with 14.2% solid content and 7.0% protein had to be heat treated at 121$^{\circ}C$ for 3min. The relative toughness of MFS tofu was increased by the addition of SPI, showing a 144% increase. The toughness of MFS tofu prepared with the MFS/SPI mixture was greatly increased by the addition of WPC at the level of 0.7% and the water separation from MFS tofu was greatly reduced. Furthermore, the toughness and strength of MFS/SPI tofu was enhanced by the addition of 0.1% chitosan oligosaccharide and 0.2% mushroom powder. The sensory evaluation of the tofu fortified with SPI, chitosan oligosaccharide and mushroom powder was superior to that of MFS tofu, with a higher score for overall preference.

$^{125}I$ Labelling of Protein Using Immobilized Enzyme (고정화효소에 의한 단백질의 $^{125}I$ 표지반응)

  • For an effective solid-phase labelling of protein with $^{125}I$, studies on the immobilization of lactoperoxidase(LPO) on the inner wall of polystyrene tubes were carried out. Labelling of bovine serum albumin(BSA) and insulin was also practiced using the LPO immobilized tubes. The immobilized enzyme of about $2.5{\mu}g/tube$ was sufficient for small scale labelling since the results of radio-paper chromatography of the labelling mixture of insulin indicated that the yields were sufficiently high(80%) even in the reactions conducted at room temperature for 60 sec. The results of the Sephadex column chromatography indicated that the labelled products were not contaminated with $LPO-^{125}I$, and the radiochemical purity of the products was more than 90%. In considering the general trend that the $^{125}I$ labelled protein obtained by using LPO maintains its intactness better than those obtained by using chloramine-T, together with the tendency of yield enhancing with increase of reactants-concentration, the LPO immobilized tube method is estimated to be one of the simple methods of labelling. The product might be applicable without further purification.
