A Study on the Functional Properties of Camellia(Camellia japonica L.) Seed Protein Isolate (분리 동백단백의 기능적 특성)

  • This study was carried out to investigate the functional properties such as nitrogen solubility, emulsifying property , foaming capapcity , water and oil absorption of Camellia (Camellia japonica .) seed protein isolate in condition of distilled water and 0.5M NaCl solution at pH 2.0∼10.0. Nitrogen solubility of Camellia protein isolate in distilled water showed the minimum value at pH 4.0 and increased at pH lower or higher than the isoelectric point(pH 4.0). It was 90.0 %at pH 10.0 Nitrogen solubility of 0.5M NaCl solution showed a similar pattern with that of distrille dwater but was higher than that of distilled water except pH 2.0 and pH 10.0. Emulsifying activity of Camellia seed protein islate showed the minimum value at pH 4.0, but was higher at ether value of pH. Emulsifying stability of protein isolate was stable by heat treatment for 30min, at 80℃ and increased in 0.5M NaCl solution more than that of distille dwater. Foaming capacity of Camellia seed protein isolate in distill3ed water showed the minimum value near the isoelectric point, While it changed little at other values of pH. Foaming stability slowly decreased as, but didn't make a significant difference as time was delayed . Oil absorption was 1.4ml per a sample of 1g and water absorption was 0.9ml per a sample of 1g. The former was higher than the latter . The content of total amino acid of Camellia protein isolate was 43.67% and the major total amino acid of Camellia protein isolate was 43.67% and the major total amino acid was in the order of glutamic acid , arginine, aspartic acid, and leucine.

Effects of Venlafaxine and Dexamethasone Treatment on HSP70 Expression in Rat C6 Glioma Cells (흰쥐 C6 신경교종 세포에서 Venlafaxine 그리고 Dexamethasone 처리가 열충격 단백질 70의 발현에 미치는 영향)

  • Object:The intracellular action of the antidepressant, venlafaxine, was studied in C6-gliomas using heat shock protein 70(HSP70) immunocytochemistry and HSP70 Western blots because HSP70 is associated with stress and depression. Methods:To examine how the glucocorticoid affects the expression of HSP70 in nerve cells, the rat C6 glioma cell was treated with dexamethasone for 6 hours. In addition, venlafaxine was administered to the experimental groups of C6 glioma cells for 1, 6, 24, and 72 hours each, after which the expression of HSP70 was investigated. Finally, venlafaxine and dexamethasone were simultaneously administered to the experimental groups for 1, 6, 24, and 72 hours, followed by an investigation of the expression of HSP70. Results:The short term(1 hour) venlafaxine treatment significantly increased the level of HSP70 expression. The short term treatment of venlafaxine with dexamethasone also increased the level of HSP70 expression but this reduction was not statistically significant. The long term(72 hours) venlafaxine with dexamethasone treatment significantly reduced the level of HSP70 expression. The long term treatment of venlafaxine also reduced the level of HSP70 expression but this reduction was not statistically significant. Dexamethasone(10uM, 6hours) did not affect the level of HSP70 expression compared with controls. Conclusion:Venlafaxine increases the expression of HSP70 at short term treatment, but prolonged treatment with dexamethasone suppresses the expression of HSP70.

Anatomical Proportions and Chemical and Amino Acid Composition of Common Shrimp Species in Central Vietnam

  • This investigation was conducted to evaluate the shrimp flesh (SF) and shrimp by-product (SB) of the most abundant shrimp species (Metapenaeus affinis, Penaeus semisulcatus and Penaeus monodon) caught in Central Vietnam, with the emphasis on yield, gross and amino acid (AA) composition and effect of heat treatment. The results showed that the mean edible SF and SB (head and shells with tail) yields of the three shrimp species averaged 56.7 and 43.3%, respectively, of the total wet body weight, with the M. affinis generating the highest by-product yield (45.7%) and P. semisulcatus (40.6%) the lowest. Significant differences in dry matter (DM), crude protein (CP) and ash content were found between SF and SB. The DM content of SF (21.5%) was lower than of SB (24.9%) and the ash content (on a DM basis) of the SB in all shrimp species was more than three times that of the SF (p<0.05), whereas the CP content was almost twice as high in the SF as compared with the SB (p<0.05). The SB of the three species contained (on a DM basis) between 44.0 and 49.8% CP (p<0.05) and between 13.5 and 18.1% chitin (p<0.05). The Ca content of SB differed also between species (p<0.05). On average, the sum of AA in SB corresponded to 89.3% of the CP and essential AA accounted for about 50% of the total AA. The most abundant AA were arginine, aspartic and glutamic acids, which accounted for 33% of the total AA. Minor, but significant differences in some AA concentrations of SB between species were observed (p<0.05). With the exception of the DM and ether extract content, all other chemical constituents of entire shrimp, SF and SB were not significantly affected by heat treatment (p>0.05).

An ESR Study of Amino Acid and Protein Free Radicals in Solution Part Ⅴ. an ESR Study of Gamma-Irradiated Lysozyme in Frozen Aqueous Solutions

  • An electron spin resonance study has been made on lysozyme in. frozen aqueous solutions irradiated with $_{60}Co$ r-rays in air at $77^{circ}K.$ Water resonances are dominant when the concentration and the temperature are both below 20% and $130^{circ}K$ respectively. More solute radicals are produced in the solution of higher concentration. Majority of the solute radicals results from direct hit of the radiation. The same types of radicals are induced at $77^{circ}K$ whether the substances are irradiated in the dry state or in frozen aqueous solution. Based on these results, it is assumed that the number of ESR centers produced by the secondary intermolecular radical reacions and stabilized in aqueous solutions may depend on the concentration of the solution, and the presence of water may facilitate the secondary radical reactions occuring in the solute molecules after heat treatment. Majority of the solute radicals above around $193^{circ}K$ are believed to react with oxygen to form peroxytype radicals. However, when the solution is subiected to heat-treatment at $265^{circ}K$ after irradiation at $195^{circ}K$ the peroxy-type resonance was not observed, suggestin that an appreciable amount of oxygen is condensed into the ice, at $77^{circ}K.$ in addition to the oxygen that has already been dissolved in solution and react with solute free radicals during the process of heat-treatment. When the solution contains $H_2O_2$, no water resonance but $HO_2$, type resonance was observed probably indicating that the radiation-induced OH radicals are trapped in $H_2O_2$ aggregates and react readily with $H_2O_2$ molecules to poroduce $HO_2$ type radicals even at $77^{circ}K.$.

Changes of Phytic acid and Minerals by Heat Treatment in Korean Soybeans (한국산 대두의 열처리에 의한 피트산과 무기성분의 함량변화)

  • In this study, effect of heat treatment on phytic acid, phosphorus compounds, and minerals in Korean soybean varieties was investigated. Results were summarized as follwo: 1. In the soybeans tested, protein content ranged from 34.6 to 44.6%, lipid content from 15.4 to 20.2%, fiber content from 4.8 to 6.1% and ash content from 4.5 to 5.9%. 2. Content range for phytic acid in soybean varieties was 1,300 to 1,542mg/100g and its mean was measured 1,392mg/100g. With increasing of the temperature, the phytic acid tends to be destroyed, especially at 6$0^{\circ}C$ the loss was averaged about 20%. 3. Total phosphorus content in soybean ranged from 607 to 681mg/100g and the decending order of phosphors content in soybean varieties was Millyang > Hwangkeum > Kwangkyo > Danyoup > Hill > Jangyoup. It was also destroyed with increasing temperature. 4. Phytate phosphorus content range in soybean was from 315.6 to 318.0mg/100g and decreased with increasing temperature. 5. Inorganic phosphorus content ranged from 95.5 to 110.0mg/100g and it was increased by temperature rising among soybean varieties. 6. Phytate phosphorus to total phosphorus ratio ranged from 5.2 to 5.7 and decreased by temperature rising. 7. The content of nonphytate phosphorus varied widely between soybean varieties and decreased with temperature increase. 8. The content of calcium, iron and magnesium ranged from 15.7 to 25.7mg/100g from 8.8 to 16.8mg/100g and from 121.9 to 143.6mg/100g respectively. The content of Mg showed small difference among soybean varieties. The change of mineral content with heat treatment in soybean did not give any meaningful change mineral content.

Comparison of Physicochemical Properties and Antioxidant Activity between Raw and Heat-Treated Vegetables (열처리한 채소류의 이화학적 특성 및 항산화 활성 비교)

  • This study examines the changes in the physicochemical property and antioxidant activity of six types of vegetables (carrots, crown daisy, mugwort, cabbages, onions, and garlic) based on heat-treated cooking. According to the results, proximate contents (crude protein, fat, and ash) were lower in blanched samples than in untreated samples. Untreated cabbages showed the highest level of total dietary fiber content, which decreased by blanching and increased by high-temperature/high-pressure (HTHP) treatment. Noteworthy is that, in the case of soluble dietary fiber, blanched crown daisy and mugwort showed significantly high levels of 12.0 g and 7.3 g per 100 g(dry basis). There was no significant change in tocopherol content in heat-treated samples. The highest levels of total polyphenol and flavonoid content were 6.73 g and 5.51 g per 100 g, respectively, in the mugwort sample with HTHP treatment at $130^{\circ}C$ for 2 h. The water extract of mugwort with HTHP treatment had the strongest antioxidant effect based on three bioassays (SOD, DPPH, and ABTS). These results indicate the relative correlation between the level of physiologically active content and antioxidant activity and suggest new insights into ingredients for developing functional foods.

  • This study was conducted to purify and characterize a polysaccharide fraction from whole parts of Duchesnea indica (Andr.) Focke, which was previously reported to show an antitumor activity by us. The plant was extracted with 5% ethanol for 3 days in the room temperature. The extract was dialyzed by 5% ethanol, and then concentrated by extraction with $excess\;_N-butanol$, followed by gel filtration named Fr. a and b. Each was composed of gallic acid, hexose, pentose, uronic acid and protein, indicating that both fractions were tannic polysaccharide containing protein. Heat treatment of them yielded gallic acid less polysaccharides. They showed a colony stimulating factor activity.

Studies on the effect of heating conditions on the quality of soybean flours (대두분 품질에 미치는 가열처리조건에 관한 연구)

  • Experiments were carried out for the development of a processing method of soybean into full-fat or defatted flour, using two varieties of soybean (kwangkyo produced in Korea and Bragg produced in India) Samples were subjected to dry dehulling, size reduction and wet heat treatment processes to make soybean flours. The quality of soybean flours were evaluated, and the results obtained were summarized as follows. 1. Crude fiber content of dehulled soybean was under 3.0% which indicated satisfactory dehulling, and there was no significant difference in crude fibre content between two varieties. 2. When dehulled soybean was cracked into soy grits by a hammer mill, 98.71∼98.86% of the soy grit was in the range of 10∼18 mesh which was the optimum size of particle for quick and uniform penetration of heat into the intra-particle air spaces. 3. Moisture content of soy flour after steam treatment at 15 psig for 5 to 30 min was only 0.29∼1.68% which did not hinder the next milling operation. 4. From the color analysis of soy flours, it was observed that the dominant wavelength for all the samples are in a very narrow range from 575 to 581 nm and the color variation was from yellow to yellowish orange. Twenty to twenty five % higher reflectance was observed in the defatted flours than full fat flours. The % chroma of the defatted flour slightly increased as the duration of steaming of soy grits increased, whereas that of the full-fat flour did not. 5. The protein extractability in the defatted flour at pH 7.6 showed progressive decrease in solubility from 48.40% (Bragg), 75.20% (kwangkyo) for untreated flours to 9.75% (Bragg), 26.27% (kwangkyo) for 30 min steaming. But Kwangkyo variety showed twice higher protein extractability than Bragg variety.

Film-Forming Properties of Proteinaceous Fibrous Material Produced from Soybean Fermented by Bacillus natto

  • The effectiveness of a proteinaceous fibrous material formed during commercial fermentation of soy protein (PFSP) and cysteine addition were evaluated in order to improve on the properties of soy protein-based films. Nine types of films were prepared at pH 7, 9, and 11, with heat treatments at $70^{\circ}C\;and\;90^{\circ}C$ for 30 min, by casting 5% (w/w) PFSP aqueous solution, containing 2.25% (w/w) glycerol, on to polystyrene plates. The tensile strength (TS) of films ranged from 3.88 to 6.87 MPa. The highest puncture strength (PS) was observed with pH 7.0 films prepared from PFSP solution heated at $70^{\circ}C$ (P<0.05). Alkaline pH and temperature caused a decrease in both the TS and PS of the films. The thickness of films ranged from $58\;to\;74{\mu}m$. Water vapor permeabilities of the films decreased with increasing pH and temperature. To produce films from PFSP, pH value of 7.0 to 9.0 and heat treatment of $70^{\circ}C\;to\;90^{\circ}C$ were needed. A soluble nature of PFSP films in water might be useful for preparation of hot water-soluble pouches. Cysteine addition could be necessary to produce films with increased TS and enhanced barrier properties. The combination treatment that provided the best combination of barrier and mechanical properties was the PFSP film prepared at pH 7.0 with addition of 1% cysteine. The films were good oxygen barriers.

Changes of Vascular Contractility of isolated Rat Aorta treated with Salt Stress (Salt 스트레스에 의한 흰쥐 적출대동맥의 수축력 변화양상)

  • To examine whether salt stress would alter or not contractility of isolated rat aorta, under anesthesia with sodium pentobarbital(50 mg kg-1 i.p.), male Sprague Dawley rats(300-330 g) were subjected to 0, 50, and 150 mM of sodium chloride at 37$^{\circ}C$ for 60 min. where as the sham group was left at modified Krebs-bicarbonate solution. To measure contractile response of vascular ring preparation isolated from rat was determined in organ bath and was recorded on physiograph connected to isometric transducer. And the strip was checked for expression of heat shock protein(Hsp) by Western blotting. One, three and eight hours later, we measured vascular contractility of isolated rat aorta treated with KCI, phenylephrine from organ bath study. The dose-vascular responses of potassium chloride and phenylephrine showed a little augmentation by NaCl concentration in the strips exposed to NaCl for 8 hours. And the response of relaxation induced by nitroprusside and acetylcholine was not influenced by NaCl stress in isolated aorta ring for 8 hours, respectively. Expression pattern of Hsp 70 of vascular muscle in isolated rat aorta showed a little increase in 150 mM NaCl group at 8 hours after NaCl treatment but not at 3 hours, and Hsp 60 expression of rat aorta was markedly increased in 50 mM NaCl group at 8 hours after NaCl treatment. Taken together, NaCl induced dose-and time dependent accumulation of the Hsp but not affected contraction of rat aorta. These data suggest that short term high salt stress was not sufficient to induce hypertension of rat aorta.
